Effect of an n-3 fatty acid-deficient diet on the adenosine-dependent melatonin release in cultured rat pineal.

We studied the effect of a diet deficient in n-3 fatty acids on the adenosine-dependent melatonin release from cultured rat pineal gland after stimulation by 5'-N-ethylcarboxamidoadenosine (NECA), an A2 adenosine agonist. Experiments were conducted with 2-month-old rats raised on semipurified diets containing either peanut oil (n-3 deficients) or peanut plus rapeseed oil (controls). The proportion of docosahexaenoic acid (22:6 n-3) in the pineal total lipid fraction and in phosphatidylcholine and phosphatidylethanolamine was significantly decreased in n-3-deficient rats. This was compensated f

Diet-induced RKIP downregulation disrupts PC/PE-ER homeostasis to drive MASLD.

High-fat diet (HFD) is a risk factor for metabolic dysfunction-associated steatotic liver disease (MASLD), yet the molecular pathways that connect dietary fats to liver dysfunction remain unclear. Here, we discover that hepatic downregulation of Raf kinase inhibitory protein (RKIP) in MASLD patients and male mice is linked to fatty acid uptake, which causes endoplasmic reticulum (ER)-associated degradation of RKIP by inhibiting its S-palmitoylation. Via facilitating the m6A-modified RNA binding of YTHDF1, RKIP is required for the efficient translation of PEMT, an essential enzyme in maintainin

Effect of N-acetylneuraminic acid on respiratory tract secretion and inflammation in the bronchitic rabbit.

Long-term exposure of rabbits to SO2 gas caused the following changes in the respiratory tract system: decreases in the respiratory rate and pO2 blood level; an increase in the viscosity of sputum associated with increases in protein, unsaturated phosphatidylethanolamine and sphingomyelin; subacute rhinitis, tracheitis and multiple bronchopneumonia. Pretreatment with repeated administrations (inhalations) of N-acetylneuraminic acid remarkably prevented these inflammatory changes. The significance of this finding is discussed.

Increased methylation of chloroform extractable products and CTP: cholinephosphate cytidylyltransferase in brain membrane preparations from triethyltin-intoxicated rats.

Rats were chronically intoxicated with triethyltin in the drinking water (0.002%) for a period of 15 days. Starting with day 5 of the intoxication period a decrease in the body weight was observed and, in parallel, the development of a cerebral oedema could be followed by measuring white matter density. At the same time, an increase of phosphatidylethanolamine-N-methyltransferase and cholinephosphate cytidylyltransferase activities was noted. This increase might be a compensatory mechanism for counteracting the membrane damages induced by triethyltin.

[Mechanism of icariin inhibiting the proliferation of human prostate cancer PC-3 cells: An exploration based on cell metabolomics].

OBJECTIVE: To study the mechanism of icariin inhibiting the proliferation of human PCa PC-3 cells based on cell metabolomics technology. METHODS: We determined the proliferation activity of human PC-3 cells by methyl thiazolyl tetrazolium(MTT) assay, and compared the proliferation of the PC-3 cells among the control, 5-fluorouracil and icariin intervention groups. Using the Bligh Dyer method, we extracted endogenous metabolites from the cells, analyzed the metabolic profile by ultra-high pressure liquid chromatography tandem quadrupole time-of-flight mass spectrometry, identified the differen

MALDI-MSI of lipids in a model of breast cancer brain metastasis provides a surrogate measure of ischemia/hypoxia.

Breast cancer brain metastasis (BCBM) has an incidence of 10-30%. It is incurable and the biological mechanisms that promote its progression remain largely undefined. Consequently, to gain insights into BCBM processes, we have developed a spontaneous mouse model of BCBM and in this study found a 20% penetrance of macro-metastatic brain lesion formation. Considering that lipid metabolism is indispensable to metastatic progression, our goal was the mapping of lipid distributions throughout the metastatic regions of the brain. Matrix-assisted laser desorption/ionization mass spectrometry imaging

The Vitamin D Receptor Regulates Glycerolipid and Phospholipid Metabolism in Human Hepatocytes.

The vitamin D receptor (VDR) must be relevant to liver lipid metabolism because VDR deficient mice are protected from hepatosteatosis. Therefore, our objective was to define the role of VDR on the overall lipid metabolism in human hepatocytes. We developed an adenoviral vector for human VDR and performed transcriptomic and metabolomic analyses of cultured human hepatocytes upon VDR activation by vitamin D (VitD). Twenty percent of the VDR responsive genes were related to lipid metabolism, including MOGAT1, LPGAT1, AGPAT2, and DGAT1 (glycerolipid metabolism); CDS1, PCTP, and MAT1A (phospholipid

From masochistic enzymology to mechanistic physiology and disease.

The pioneering work of Eugene Kennedy in the 1950s established the choline pathway for phosphatidylcholine (PC) biosynthesis. However, the regulation of PC biosynthesis was poorly understood at that time. When I started my lab at the University of British Columbia in the 1970s, this was the focus of my research. This article provides my reflections on these studies that began with enzymology and the use of cultured mammalian cells, and progressed to utilize the techniques of molecular biology and gene-targeted mice. The research in my lab and others demonstrated that the regulated and rate-lim

Protein-energy malnutrition during gestation and lactation in rats affects growth rate, brain development and essential fatty acid metabolism.

The influence of feeding a low protein diet to rat dams during gestation and lactation on lipid metabolism in pups was studied. Wistar rats were fed 5, 10, 15 and 25% dietary protein during gestation and lactation. Pup growth was monitored until weaning, and brain weight, protein concentration, proteolipid concentration and total lipid phosphorus concentration of brain were analyzed. The levels of fatty acids in dam milk as well as in pup liver phospholipids and brain prosphatidylcholine and phosphatidylethanolamine were determined. The progressive deprivation of maternal dietary protein produ

Brain damage and recovery in hyperphenylalaninemic rats.

Rats were made hyperphenylalaninemic by injecting a mixture of alpha-methylphenylalanine and phenylalanine. Brain development was measured by biochemical, histological and 31-P nuclear magnetic resonance (NMR) methods. In 17-day-old hyperphenylalaninemic rats, brain myelinogenesis was disturbed. Compared to controls, test animals had lower body weights, brain weights, cerebrosides, sulfatides, myelin basic protein (MBP) and reduced cerebroside sulfotransferase (CST) and 2'3'-cyclic nucleotide-3'-phosphohydrolase (CNP) activities. No changes were found in total proteins, total lipids, total pho

Dark Side of Escherichia coli Biogenic Inner Membrane: Overabundance of Three Main Phospholipids on Cytoplasmic Leaflet.

Transmembrane (TM) asymmetry of glycerophospholipids in the plasma membrane is well established, yet still a complex and evolving area of study regarding the exact mechanisms, magnitude, and composition. The emerging controversy challenges "Fluid Mosaic Sandwich" model simplicity by combining compositional (different types of lipids on each side) and numerical (different total amounts of phospholipids) asymmetries. While Gram-negative Escherichia coli phosphatidylethanolamine (PE) distribution is asymmetric (mostly cytoplasmic), the TM distribution of phosphatidylglycerol (PG) and cardiolipin

Bacterial cell membrane models: choosing the lipid composition.

The reasons for the wide diversity of lipids found in natural cell membranes are still not fully understood but could potentially be exploited in treating disease and infection. This study aims to establish whether charge alone or specific chemical structure of an anionic lipid headgroup determines the structure and properties of model bacterial cell membranes. We compare different compositions of a zwitterionic lipid di-myristoyl phosphatidylethanolamine (DMPE) and two anionic lipids, di-myristoyl phosphatidylglycerol (DMPG) and tetra-myristoyl cardiolipin (TMCL). TMCL has a distinct condensi

Integrating microfluidics and streamlined remote drug loading: one step closer to continuous manufacturing of liposomal injectables containing small drugs.

The industrial manufacturing of liposomal injectables faces significant technical challenges. Traditional batch manufacturing methods, like ethanol injection and extrusion, are time-consuming, prompting a shift towards continuous manufacturing. To improve process efficiency, this study tested microfluidics, a technique widely used in lipid nanoparticle (LNP) production for nucleic acid delivery, to optimize doxorubicin liposome manufacturing, focusing on vesicle formation and remote drug loading. The formulation consisted of neutral phospholipids with minimal DSPE-PEG content and no negatively

Delivery of Corn-Derived Nanoparticles with Anticancer Activity to Tumor Tissues by Modification with Polyethylene Glycol for Cancer Therapy.

PURPOSE: We recently reported that intratumoral injection of corn-derived nanoparticles (cNPs) affords anticancer activity in tumor-bearing mice. To increase their applicability in cancer therapy, we examined the tissue distribution of cNPs after intravenous injection in mice, modified their surface with polyethylene glycol (PEG) to improve tumor delivery, and examined tissue distribution and anticancer activity of PEG-cNPs in tumor-bearing mice. METHODS: N-(Carbonyl-methoxypolyethyleneglycol2000)-1,2-distearoyl-sn-glycero-3-phosphoethanolamine (DSPE-PEG) was added to cNPs by sonication to ob

The topology of red cell membrane lipids in hereditary spherocytosis.

The topology of membrane phospholipids in the red cells of patients with hereditary spherocytosis has been studied with the non-penetrating probe 2,3,5-trinitrobenzenesulphonate. There was no significant difference in the assymetric distribution of the aminophospholipids phosphatidylserine and phosphatidylethanolamine across the two halves of the membrane bilayer in red cells of five patients with hereditary spherocytosis as compared to seven normal controls. These studies indicated that the complex processes responsible for membrane lipid assymetry are intact in hereditary spherocytosis.

Seminal Plasma Metabolomics Reveals Potential Metabolic Pathways Linking Organophosphate Flame Retardant Exposure to Impaired Semen Quality.

Organophosphate flame retardants (OPFRs) may affect semen quality, but the mechanisms remain unclear. This study included 157 men from the SEARCH cohort and evaluated the associations between urinary OPFR levels and semen quality, and the role of the seminal plasma metabolome. Overall, bis(2-butoxyethyl) phosphate (BBOEP) was negatively associated with progressive motility (β [95% confidence interval (CI)]: -0.090 [-0.175, -0.005]), and some associations between OPFRs and semen quality were more pronounced among men with male-factor infertility. Furthermore, widespread associations were observ

Novel biallelic PISD missense variants cause spondyloepimetaphyseal dysplasia with disproportionate short stature and fragmented mitochondrial morphology.

Biallelic variants in PISD cause a phenotypic spectrum ranging from short stature with spondyloepimetaphyseal dysplasia (SEMD) to a multisystem disorder affecting eyes, ears, bones, and brain. PISD encodes the mitochondrial-localized enzyme phosphatidylserine decarboxylase. The PISD precursor is self-cleaved to generate a heteromeric mature enzyme that converts phosphatidylserine to the phospholipid phosphatidylethanolamine. We describe a 17-year-old male patient, born to unrelated healthy parents, with disproportionate short stature and SEMD, featuring platyspondyly, prominent epiphyses, and

The tegumental surface membranes of Schistosoma mansoni are enriched in parasite-specific phospholipid species.

The complex surface structure of adult Schistosoma mansoni, the tegument, is essential for survival of the parasite. This tegument is syncytial and is covered by two closely-apposed lipid bilayers that form the interactive surface with the host. In order to identify parasite-specific phospholipids present in the tegument, the species compositions of the major glycerophospholipid classes, phosphatidylcholine, phosphatidylserine, phosphatidylethanolamine and phosphatidylinositol, including lysophospholipid species, were analysed in adult S. mansoni worms, isolated tegumental membranes and hamste

Alboserpin, a factor Xa inhibitor from the mosquito vector of yellow fever, binds heparin and membrane phospholipids and exhibits antithrombotic activity.

The molecular mechanism of factor Xa (FXa) inhibition by Alboserpin, the major salivary gland anticoagulant from the mosquito and yellow fever vector Aedes albopictus, has been characterized. cDNA of Alboserpin predicts a 45-kDa protein that belongs to the serpin family of protease inhibitors. Recombinant Alboserpin displays stoichiometric, competitive, reversible and tight binding to FXa (picomolar range). Binding is highly specific and is not detectable for FX, catalytic site-blocked FXa, thrombin, and 12 other enzymes. Alboserpin displays high affinity binding to heparin (K(D) ~ 20 nM), but
