Self-amplifying hypoxia cascade in covalent organic framework/metal organic framework nanoreactors for synergistic cancer therapy.

Tumor hypoxia, a hallmark of the tumor microenvironment (TME), drives malignancy and therapeutic resistance but also offers a target for selective therapy. Hypoxia-activated prodrugs (HAPs) like TH-302 exploit this niche, yet their efficacy is often limited by insufficient hypoxia levels. To overcome this limitation, a hypoxia-responsive nanoplatform COF@MOF/TH-302@HA was designed to intensify tumor hypoxia and trigger a self-amplifying therapeutic cascade. The platform was constructed by growing a hypoxia-sensitive copper-based metal-organic framework (Cu-MOFs) on a nitrogen-rich porphyrinic

Developmental toxicity of acephate by gavage in mice.

Acephate (O,S - dimethyl acetyl phosphoramidothioate), an organophosphate insecticide, was evaluated for its potential to produce developmental toxicity in mice after oral administration. Pregnant ICR (CD-1) mice were given sublethal doses of 0 (distilled water), 7, 14, and 28 mg/kg/day acephate by gavage on Gestation Days 6 through 15. Maternal effects in the 28 mg/kg/day dose group included cholinergic signs, decreased body weight at 15 and 18 days of gestation, and decreased absolute and relative brain weight. Placental weight was also decreased and liver weight was increased in the high do

Investigation of the immunomodulatory effects and molecular mechanisms of cichoric acid on cyclophosphamide-induced immunosuppression in mice.

Immunosuppressive diseases (ISD) are relatively common in veterinary clinical practice. The lack of therapeutic drugs for ISD severely impacts the healthy breeding of livestock and poultry. Cichoric Acid (CA), a natural active ingredient derived from traditional medicinal plants, has demonstrated a notable immunoenhancing effect. Nevertheless, the underlying mechanisms by which CA attenuates cyclophosphamide (CTX)-mediated immunosuppressive effects in mice have not yet been fully elucidated. In vivo, CA demonstrated a significant increase in body weight of CTX-induced immunosuppressed mice, el

Protective effect of Huangqi-Guizhi-Wuwutang against cyclophosphamide-induced spermatogenesis dysfunction in mice by promoting steroid hormone biosynthesis.

ETHNOPHARMACOLOGICAL RELEVANCE: The primary adverse effect of cyclophosphamide (CTX) chemotherapy to cancer in male patients of reproductive age is a significant impairment of reproductive function. Huangqi-Guizhi-Wuwutang (HGW), a classical traditional Chinese medicine formula, is designed to exert a salutary effect on qi and promote blood circulation, thereby eliminating blood stasis and promoting spermatogenesis, and it has been recorded as a treatment for oligospermia. However, its potential in mitigating the adverse impact of CTX on male spermatogenesis remains unexplored. AIM OF THE STU

Solid state and solution study of some phosphoramidate derivatives containing the P(O)NHC(O) bifunctional group: crystal structures of CCl(2)HC(O)NHP(O)(NCH(3)(CH(2)C(6)H(5)))(2), p-ClC(6)H(4)C(O)NHP(O)(NCH(3)(CH(2)C(6)H(5)))(2), CCl(2)HC(O)NHP(O)(N(CH(2)C(6)H(5))(2))(2) and p-BrC(6)H(4)C(O)NHP(O)(N(CH(2)C(6)H(5))(2))(2).

Synthetic methods for several novel phosphoramidate compounds containing the P(O)NHC(O) bifunctional group were developed. These compounds with the general formula R(1)C(O)NHP(O)(N(R(2))(CH(2)C(6)H(5)))(2), where R(1)=CCl(2)H, p-ClC(6)H(4), p-BrC(6)H(4), o-FC(6)H(4) and R(2)=hydrogen, methyl, benzyl, were characterized by several spectroscopic methods and analytical techniques. The effects of phosphorus substituents on the rotation rate around the P-N(amine) bond were also investigated. (1)H NMR study of the synthesized compounds demonstrated that the presence of bulky groups attached to the p

AT-527, a Double Prodrug of a Guanosine Nucleotide Analog, Is a Potent Inhibitor of SARS-CoV-2 In Vitro and a Promising Oral Antiviral for Treatment of COVID-19.

The impact of severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2), the causative agent of COVID-19, is global and unprecedented. Although remdesivir has recently been approved by the FDA to treat SARS-CoV-2 infection, no oral antiviral is available for outpatient treatment. AT-527, an orally administered double prodrug of a guanosine nucleotide analog, was previously shown to be highly efficacious and well tolerated in hepatitis C virus (HCV)-infected subjects. Here, we report the potent in vitro activity of AT-511, the free base of AT-527, against several coronaviruses, including SAR

Immunotoxicity of acute acephate exposure in control or IL-1-challenged rats: correlation between the immune cell composition and corticosteroid concentration in blood.

Corticosterone concentration and the immune cell composition were measured in rats exposed by intraperitoneal (i.p.) injection to different doses (10-500 mg kg(-1)) of acephate (Ace) and 250 micro g kg(-1) of interleukin 1 (IL-1), either alone or in combination. Two different combination protocols were used: IL-1 and Ace were administered simultaneously; and IL-1 was injected 60 min after Ace administration (sequential exposure). Ace, in a dose- and time-dependent manner, inhibited blood and brain acetylcholinesterase (AChE) activities, increased blood corticosterone concentrations, suppressed

Genome-scale dissection of phase-variable gene function in Campylobacter jejuni using a stabilized phasotype library.

UNLABELLED: Phase variation (PV) allows bacterial pathogens to rapidly modify surface structures through reversible mutations-often driven by simple sequence repeats (SSRs)-thereby facilitating immune evasion and environmental adaptation. In Campylobacter jejuni, SSR-mediated PV regulates numerous surface-exposed molecules, including capsular polysaccharides and lipooligosaccharides, which play key roles in host interaction and immune resistance. However, the stochastic nature of PV has made it difficult to systematically analyze the functions of individual phase-variable genes (PVGs). Here, w

Irreversible activation of adenylate cyclase of toad erythrocyte plasma membrane by 5'-guanylylimidodiphosphate.

The irreversible activation of adenylate-cyclase by 5'guanylylimidodiphosphate, a phosphoramidate analog of 5'GTP, has been examined in toad (Bufus marinus) plasma membranes using the technique of preincubating the membranes with the nucleotide under various controlled conditions followed by washing and subsequent assay of enzyme activity. Activation of adenylate cyclase by Gpp(NH)p, but not GTP, is essentially permanent and persists following extensive washing, prolonged incubation at 30 degrees C in the absence of the nucleotide, and after dissolution of the membranes with Lubrol PX. Post-Transplant Cyclophosphamide for the Prevention of Graft-vs.-Host Disease in Allogeneic Hematopoietic Cell Transplantation: A Guide to Management for the Advanced Practitioner.

Cyclophosphamide remains a critical component to haploidentical transplant conditioning regimens. Post-transplant cyclophosphamide (PTCy) emerged as an effective component of graft-vs.-host disease (GVHD) prophylaxis in the nonmyeloablative haploidentical bone marrow transplant setting. The relative ease of administration compared with ex vivo manipulations and efficacy in reducing GVHD has led to increasing PTCy use in transplant centers around the world. The role of PTCy has expanded to haploidentical transplantation with myeloablative conditioning regimens and peripheral blood progenitor ce

Beyond DNA Damage: Exploring the Immunomodulatory Effects of Cyclophosphamide in Multiple Myeloma.

The alkylating agent cyclophosphamide has been used in the treatment of multiple myeloma for over 60 years. At low doses, cyclophosphamide also has significant immunomodulatory activity, which can be used to modify the immunosuppressive tumor microenvironment in order to augment responses to existing therapies. Immune-mediated therapies are becoming more widespread in modern approaches to myeloma treatment. In this review, we discuss the effects cyclophosphamide has on the immune system, and how it can be used synergistically with other treatment modalities including the immunomodulatory agent

The Protective Effects of Sesamin against Cyclophosphamide-Induced Nephrotoxicity through Modulation of Oxidative Stress, Inflammatory-Cytokines and Apoptosis in Rats.

Cyclophosphamide is an anticancer drug with a wide spectrum of clinical uses, but its typical side effects are multiple complications, including nephron toxicity. The possible molecular mechanism of the nephroprotective action of sesamin (SM) against cyclophosphamide (CP) induced renal toxicity was investigated in rats by understanding oxidative stress and inflammatory cytokines. In this study, rats were arbitrarily grouped into the following four groups: a normal control group (CNT); a CP-induced toxicity group; a treatment group with two doses of sesamin SM10 and SM20; a group with sesamin (

Sialyltransferase Inhibitors for the Treatment of Cancer Metastasis: Current Challenges and Future Perspectives.

Potent, cell-permeable, and subtype-selective sialyltransferase inhibitors represent an attractive family of substances that can potentially be used for the clinical treatment of cancer metastasis. These substances operate by specifically inhibiting sialyltransferase-mediated hypersialylation of cell surface glycoproteins or glycolipids, which then blocks the sialic acid recognition pathway and leads to deterioration of cell motility and invasion. A vast amount of evidence for the in vitro and in vivo effects of sialyltransferase inhibition or knockdown on tumor progression and tumor cell meta

Amelioration of the cyclophosphamide induced genotoxic damage in mice by the ethanolic extract of Equisetum arvense.

In the present study, we evaluated the potential of the plant E. arvense against the cytotoxic and mutagenic effects induced by cyclophosphamide (chemotherapeutic agent) in the bone marrow cells of mice using the Chromosome assay (CA) and Mitotic index (MI) in vivo as the biomarkers. The study was performed following 3 protocols: pre-treatment, simultaneous treatment and post-treatment with the ethanolic extract of the plant. The results demonstrated that the plant extract was not cytotoxic and mutagenic and has a protective effect against the mutagenicity induced by cyclophosphamide in pre, s

Targeting homologous recombination and telomerase in Barrett's adenocarcinoma: impact on telomere maintenance, genomic instability and tumor growth.

Homologous recombination (HR), a mechanism to accurately repair DNA in normal cells, is deregulated in cancer. Elevated/deregulated HR is implicated in genomic instability and telomere maintenance, which are critical lifelines of cancer cells. We have previously shown that HR activity is elevated and significantly contributes to genomic instability in Barrett's esophageal adenocarcinoma (BAC). The purpose of this study was to evaluate therapeutic potential of HR inhibition, alone and in combination with telomerase inhibition, in BAC. We demonstrate that telomerase inhibition in BAC cells incre

Advances in antisense oligonucleotide treatment for cancer.

RNA therapeutics, including antisense oligonucleotides (ASOs), have emerged as a promising class of drugs, with several already approved for clinical use. To date, most approved ASO-based RNA therapies target non-malignant disorders such as neurodegenerative diseases, and only a single therapy in this class has been approved for cancer. Notably, nearly half of existing RNA therapeutics act by modulating RNA splicing. Given the growing evidence implicating aberrant RNA splicing in cancer pathogenesis, the development of ASO-based therapeutics for oncologic indications is expected to accelerate.

Oral chemotherapy agents in cats: clinical uses, administration and side effects.

PRACTICAL RELEVANCE: Cancer is a very common disease in cats, and many feline patients will require chemotherapy as part of their treatment plan, either as a sole agent or as part of multimodality therapy. Although some general practitioners might be reluctant to undertake injectable chemotherapy, oral chemotherapy may feel like a more acceptable treatment option to manage in practice. CLINICAL CHALLENGES: The small body size of many cats means that tablet sizes made for the human market are less appropriate, which creates dosing challenges. The administration of oral medication can also be a
