Differences in leaf characteristics between ozone-sensitive and ozone-tolerant hybrid aspen (Populus tremula x Populus tremuloides) clones.

The authors analyzed a suite of leaf characteristics that might help to explain the difference between ozone-sensitive and ozone-tolerant hybrid aspen (Populus tremula L. x Populus tremuloides Michx.) clones. An open-field experiment comprising ambient ozone and 1.5x ambient ozone concentration (about 35 ppb) and two soil nitrogen regimes (60 and 140 kg N ha(-1) year(-1)) was conducted over two growing seasons on potted plants of eight hybrid aspen clones. Four of the clones had previously been determined to be ozone sensitive based on impaired growth in response to elevated ozone concentratio

Fate and effects of heavy metals in salt marsh sediments.

The fate and effects of selected heavy metals were examined in sediment from a restored salt marsh. Sediment cores densely covered with Spartina patens were collected and kept either un-amended or artificially amended with nickel (Ni) under standardized greenhouse conditions. Ni-amendment had no significant effect on the fate of other metals in sediments, however, it increased root uptake of the metals. Metal translocation into the shoots was small for all metals. Higher Ni concentrations in plants from amended cores were accompanied by seasonal reductions in plant biomass, photosynthetic capa

Mass spectrometry-based cross-linking study shows that the Psb28 protein binds to cytochrome b559 in Photosystem II.

Photosystem II (PSII), a large pigment protein complex, undergoes rapid turnover under natural conditions. During assembly of PSII, oxidative damage to vulnerable assembly intermediate complexes must be prevented. Psb28, the only cytoplasmic extrinsic protein in PSII, protects the RC47 assembly intermediate of PSII and assists its efficient conversion into functional PSII. Its role is particularly important under stress conditions when PSII damage occurs frequently. Psb28 is not found, however, in any PSII crystal structure, and its structural location has remained unknown. In this study, we u

S-state dependence of the calcium requirement and binding characteristics in the oxygen-evolving complex of photosystem II.

The functional role of the Ca (2+) ion in the oxygen-evolving complex of photosystem II is not yet clear. Current models explain why the redox cycle of the complex would be interrupted after the S 3 state without Ca (2+), but the literature shows that it is interrupted after the S 2 state. Reinterpretation of the literature on methods of Ca (2+) depletion [Miqyass, M., van Gorkom, H. J., and Yocum, C. F. (2007) Photosynth. Res. 92, 275-287] led us to propose that all S-state transitions require Ca (2+). Here we confirm that interpretation by measurements of flash-induced S-state transitions in

Evidence for an association of the early light-inducible protein (ELIP) of pea with photosystem II.

The precursor to the nuclear-coded 17 kDa early light-inducible protein (ELIP) of pea has been transported into isolated intact chloroplasts. The location of the mature protein in the thylakoid membranes was investigated after using cleavable crosslinkers such as DSP and SAND in conjunction with immunofractionation methods and by application of mild detergent fractionation. We show that ELIP is integrated into the membranes via the unstacked stroma thylakoids. After isolation of protein complexes by solubilization of membranes with Triton X-100 and sucrose density-gradient centrifugation the c

Involvement of the Light Signalling Components HY5 and BIC1,2 and SPA1 in Plant Responses to Elevated Daytime UV-B Doses.

Plants respond to ultraviolet B radiation (280-320 nm) with an integrated reaction that includes the reception of the acting stress factor, followed by the generation of reactive oxygen species and damage to macromolecules and membrane structures, as well as changes in cellular metabolism and the formation of protective systems. However, the involvement of key UV-B-related signalling components such as HY5, SPA1 and BIC1 or BIC2 proteins in physiological, biochemical and molecular responses remains insufficiently understood. The effects of 8, 16 and 24 h of UV-B exposure (within an 8 h photope

A novel insight into the mode of action of glufosinate: how reactive oxygen species are formed.

Glufosinate targets glutamine synthetase (GS), but its fast herbicidal action is triggered by reactive oxygen species (ROS). The relationship between GS inhibition and ROS accumulation was investigated in Amaranthus palmeri. Glufosinate's fast action is light-dependent with no visual symptoms or ROS formation in the dark. Inhibition of GS leads to accumulation of ammonia and metabolites of the photorespiration pathway, such as glycolate and glyoxylate, as well as depletion of other intermediates such as glycine, serine, hydroxypyruvate, and glycerate. Exogenous supply of glycolate to glufosina

Effect of CO2 content in air on functioning of Arabidopsis thaliana photosynthetic electron transport chain.

The functioning of the photosynthetic electron transport chain and the proceeding of accompanying processes were studied in Arabidopsis thaliana plants acclimated during 2weeks to reduced (150ppm) or elevated (1000ppm) CO2 concentrations in air. Measured at ambient CO2 , the quantum yields of both photosystems were lower in plants acclimated to these CO2 concentrations as compared with control plants grown at ambient CO2 . The difference was more pronounced at the beginning of the illumination. It is discussed that this difference resulted from the difference in Rubisco content, which at both

Cyanide binding at the non-heme Fe2+ of the iron-quinone complex of photosystem II: at high concentrations, cyanide converts the Fe2+ from high (S = 2) to low (S = 0) spin.

The primary electron acceptor complex of photosystem II, QAFe2+, can bind a number of small molecules at the iron site, including cyanide [Koulougliotis, D., Kostopoulos, T., Petrouleas, V., & Diner, B. A. (1993) Biochim. Biophys. Acta 1141, 275-282)]. In the presence of NaCN (30-300 mM) at pH 6.5, the reduced state, QA-Fe2+, produced either by illumination at < or = 200 K or by reduction in the dark with sodium dithionite, is characterized by a g = 1.98 EPR signal. The light- or dithionite-induced g = 1.98 signal decays with increasing pH above 6.5 and is almost totally absent at pH 8.1 and N

Harnessing salinity induced adaptive potential of extracellular polymeric substances producing rice-field cyanobacterium Neowestiellopsis sp. VKB03: a physiochemical, morphological, and biomass characterization.

The toxicity of salinity in irrigated land has significantly affected global rice production. Cyanobacteria are a core atmospheric nitrogen-fixer in the rice-field community. However, extracellular polymeric substances (EPS) play a distinct role in the adaptive fitness of cyanobacteria under salinity stress. This study analyzed physiochemical, morphological, biomass, and polymer characteristics of rice-field cyanobacterium Neowestiellopsis sp. VKB03. Our findings showed that high salt content elevated the levels of carbohydrates, EPS, lipids, proline, stress biomarkers (H2O2 and MDA), and anti

Light stress-regulated two-helix proteins in Arabidopsis thaliana related to the chlorophyll a/b-binding gene family.

The chlorophyll a/b, chlorophyll a/c, and chlorophyll a/a light-harvesting proteins are part of an extended gene family that also includes the transiently expressed stress proteins, the Elips (early light-induced proteins). Four Elip homologue proteins, encoded by single-copy nuclear genes, have been identified in the Arabidopsis thaliana database. These proteins were divided into two groups according to the expression pattern under light-stress conditions and the predicted secondary structure. Group one included two members of the Elip family with three predicted transmembrane helices and a g

Photosynthetic carbon reduction and carbon oxidation cycles are the main electron sinks for photosystem II activity during a mild drought.

Stomatal closure can explain the inhibition of net CO2 uptake by a leaf subjected to a mild drought: the photosynthetic apparatus appears resistant to lack of water. Changes in both the water content of leaves maintained in a constant environment and the ambient CO2 molar fraction during measurements on well-hydrated leaves lead to similar effects on net CO2 uptake and whole chain electron transport as estimated by leaf chlorophyll fluorescence measurements. In particular, it is shown that photosystem II (PSII) functioning and its regulation are not qualitatively changed during desiccation and

Magnesium and manganese affect photosynthesis, essential oil composition and phenolic compounds of Tanacetum parthenium.

The accumulation of plant defense metabolites is closely associated with the concentration of nutrient elements, yet data related to the interactive effects of two nutrients on the deployment of phenolics and terpenoids are scare. In the present study, the interaction between magnesium (Mg) and manganese (Mn) on nutrient uptake, photosynthesis, oxidative status and the accumulation of phenolics and terpenoids in the leaves of feverfew plants grown at different concentrations of Mg and Mn was investigated. Nutrient uptake and photosynthesis were associated with the amount of applied Mg but coul

The interplay between thiol-compounds against chromium (VI) in the freshwater green alga Monoraphidium convolutum: toxicology, photosynthesis, and oxidative stress at a glance.

In this paper, the multifaceted Cr(VI) toxicity over the freshwater green alga Monoraphidium convolutum was assessed by concomitantly monitoring thiol-dependent redox balances, photosynthesis activity and growth-survival scores. Control group showed exponential growth rate at (5.78±0.29) division/day until 8th day with linear increasing chlorophyll a/protein ratios (CHLa/PROT) throughout the period. Cultures of M. convolutum were exposed for 5 days to Cr(VI) concentrations from 0 up to 100mg/L showing that CHLa/PROT ratios were sensibly affected, in agreement to the calculated LC(50,48 h) (5.3

Production of reactive oxygen species in chloride- and calcium-depleted photosystem II and their involvement in photoinhibition.

Mixed photosystem II (PSII) samples consisting of Cl(-)-depleted and active, or Ca(2+)-depleted and active PSII enriched membrane fragments, respectively, were investigated with respect to their susceptibility to light. In the presence of Cl(-)-depleted PSII, active centers were damaged more severely, most likely caused by a higher amount of reactive oxygen species formed in the nonfunctional centers. Cl(-) depletion led to an increased H(2)O(2) production, which seemed to be responsible for the stimulation of PSII activity loss. To distinguish between direct H(2)O(2) formation by partial wate

HY5 enhances Arabidopsis tolerance to combined high light and heat stress by coordinating photoprotection and hormone signaling.

High light (HL) and heat stress (HS) are two major abiotic factors that commonly co-occur in nature and severely impair photosynthetic performance when combined. The bZIP transcription factor HY5 is a well-known integrator of light and temperature cues, but its role under combined HL + HS stress remains largely unexplored. Here, we investigated the role of HY5 in Arabidopsis tolerance to HL + HS using wild-type (Col-0), HY5-deficient (hy5-215), and HY5-overexpressing (HY5OX) lines. Physiological and biochemical analyses revealed that HY5OX plants maintained higher photosynthetic efficiency, lo

Decreased and increased expression of the subunit CHL I diminishes Mg chelatase activity and reduces chlorophyll synthesis in transgenic tobacco plants.

The chelation of Fe2+ and Mg2+ ions forms protoheme IX and Mg-protoporphyrin IX, respectively, and the latter is an intermediate in chlorophyll synthesis. Active magnesium protoporphyrin IX chelatase (Mg-chelatase) is an enzyme complex consisting of three different subunits. To investigate the function of the CHL I subunit of Mg-chelatase and the effects of modified Mg-chelatase activity on the tetrapyrrole biosynthetic pathway, we characterized N. tabacum transformants carrying gene constructs with the Chl I cDNA sequence in antisense and sense orientation under the control of the CaMV 35S pr

Dimeric chlorite dismutase from the nitrogen-fixing cyanobacterium Cyanothece sp. PCC7425.

It is demonstrated that cyanobacteria (both azotrophic and non-azotrophic) contain heme b oxidoreductases that can convert chlorite to chloride and molecular oxygen (incorrectly denominated chlorite 'dismutase', Cld). Beside the water-splitting manganese complex of photosystem II, this metalloenzyme is the second known enzyme that catalyses the formation of a covalent oxygen-oxygen bond. All cyanobacterial Clds have a truncated N-terminus and are dimeric (i.e. clade 2) proteins. As model protein, Cld from Cyanothece sp. PCC7425 (CCld) was recombinantly produced in Escherichia coli and shown to

Transcriptome for photobiological hydrogen production induced by sulfur deprivation in the green alga Chlamydomonas reinhardtii.

Photobiological hydrogen production using microalgae is being developed into a promising clean fuel stream for the future. In this study, microarray analyses were used to obtain global expression profiles of mRNA abundance in the green alga Chlamydomonas reinhardtii at different time points before the onset and during the course of sulfur-depleted hydrogen production. These studies were followed by real-time quantitative reverse transcription-PCR and protein analyses. The present work provides new insights into photosynthesis, sulfur acquisition strategies, and carbon metabolism-related gene e
