Opioid and non-opioid central cardiovascular effects of ketamine.

The cardiovascular responses to ketamine injected intracisternally were examined in chloralose anaesthetized cats. Blood pressure and heart rate were recorded at different time intervals after intracisternal injection of drug or saline vehicle. The low doses of ketamine (0.5 or 1.0 mg) elicited dose dependent increase in blood pressure and heart rate. In contrast the high dose of ketamine (4 mg), produced a fall in blood pressure and heart rate. The cardiovascular response elicited by the low dose was naloxone insensitive and completely blocked by haloperidol, but not by dopamine antagonist pi

Within-individual variation in steady state plasma levels of different neuroleptics and prolactin.

Eleven male chronic schizophrenics were given, serially, oral pimozide, fluphenazine, and flupenthixol; the two latter were also given intramuscularly as decanoates in oil. Oral haloperidol was given before and after each drug. Analysis of variance of steady state plasma levels of the different neuroleptics showed considerable within-individual variation in such levels, probably due to differences in absorption and metabolism and between routes of administration. The findings suggest that if a patient fails to respond to one neuroleptic, there may be good pharmacokinetic reasons for switching

Compulsive gnawing in rats after implantation of drugs into the ventral thalamus. A contribution to the mechanism of morphine action.

1 Implantation of morphine into various parts of the corpus striatum of rats evokes only weak gnawing responses.2 Deposition of apomorphine, morphine or methadone in the region of the nucleus ventralis thalami produces a biphasic response, i.e. general excitation, followed by a period of intense gnawing.3 The effect of both apomorphine and morphine is blocked by chlorpromazine, haloperidol and pimozide. However, pretreatment with alpha-methyltyrosine methyl ester or alpha-methyldopa prevents only the gnawing response to morphine, but not to apomorphine.4 Systemic nalorphine, morphine or pethid

Intravenous self-administration of cocaine and norcocaine by dogs.

The potency of cocaine, relative to d-amphetamine, to initiate and maintain intravenous self-administration behavior by dogs (n = 5) was determined. Response-contingent infusions of cocaine (at unit doses of 0.15, 0.30 and 0.60 mg/kg/infusion) and d-amphetamine (at unit doses of 0.05 and 0.10 mg/kg/infusion) were available during daily 4-h sessions on a FR1 reinforcement schedule. By comparing the dose-response curves of the two drugs, it was found that 1 mg of amphetamine is equivalent to 5.3 mg of cocaine (95% confidence limits = 3.8--9.1 mg). In a second experiment, pretreatment with the al

Delusions of parasitosis.

The fixed belief that one is infested with living organisms, in the absence of any objective evidence that such infestation exists, is termed "delusions of parasitosis." For most patients, this is a "monosymptomatic" disorder because there is no evidence of psychotic ideation in other areas of their lives. This overall normality of thought processes, together with exceptional strength of conviction regarding infestation, makes confirmation of a suspected diagnosis difficult. We want, and need, to be certain that a true infestation is not missed. This requires careful history taking, thorough e

Activation of 5-hydroxytryptamine4 receptors causes calcium influx in adrenocortical cells: involvement of calcium in 5-hydroxytryptamine-induced steroid secretion.

5-Hydroxytryptamine (5-HT) stimulates corticosteroid secretion from adrenal cells through activation of 5-HT4 receptors positively coupled to adenylyl-cyclase. In the present study, we investigated in frog adrenocortical cells the effect of 5-HT4 receptor agonists on cytosolic calcium concentration ([Ca2+]i) and determined the sequence of events associated with 5-HT4 receptor agonist zacopride (10[-8] to 10[-5]M each in the vicinity of cultured adrenocortical cells caused a dose-dependent increase in [Ca2+]i. Pharmacotherapy for Tourette Syndrome.

Tourette syndrome (TS) is associated with dysregulated cortico-striatal-thalamo-cortical neural circuitry, of which the primary implicated neurotransmitters include dopamine, glutamate, and gamma-aminobutyric acid. Pharmacologic intervention for tics should be considered when tics are causing psychological, functional, or physical impairment, and behavioral treatment is either inaccessible or ineffective. Only 3 medications have Food and Drug Administration approval for TS, including 2 typical antipsychotics (pimozide and haloperidol) and 1 atypical antipsychotic (aripiprazole). As such, off-l

The role of dopamine neurotransmission in ethanol dependence development and ethanol withdrawal in rats.

This study was undertaken to evaluate the influence of acute doses of ethanol, chronic ethanol administration and the withdrawal syndrome on central dopaminergic neurotransmission in 700 rats. The ethanol effects were differentiated by their influence on D1 and D2 receptors using following dopaminergic ligands. SKF 38393 (2.5 mg/kg), SCH 23,390 (0.5 mg/kg), pimozide (1 mg/kg), PPHT (2 mg/kg). It was found that ethanol in a low single dose (1.0 g/kg p.o.) increased DA level in rats' brain. In chronic alcoholized rats SKF 38,393 increased the D1 receptor answer in biochemical and behavioural exp

Antiemetic specificity of dopamine antagonists.

Twelve antagonists of apomorphine-induced emesis in dogs were studied in different tests to evaluate their antiemetic specificity. Ten of these antagonists were neuroleptics: benzquinamide, clebopride, bromopride, prochlorperazine, haloperidol, chlorpromazine, thiethylperazine, metoclopramide, droperidol, and pimozide blocked conditioned responding in dogs and apomorphine-induced stereotyped behavior in rats. The use of these compounds as anti-emetics entails a risk of neurological side effects. Metopimazine and domperidone were devoid of neuroleptic activity. Metopimazine, however, showed pot

[Drug treatment of obsessive-compulsive disorder. With proper drugs and some patience many patients can be helped].

Pharmacotherapy which, together with behavior therapy, is a major pillar in the treatment of obsessive-compulsive disorder (OCD), can bring about a distinct symptomatic improvement in 50-70% of the patients. In addition to the classical tricyclic antidepressant, clomipramine, a number of new selective serotonin reuptake inhibitors have now been approved for the treatment of OCD. In comparison with depression, pharmacotherapy of OCD is characterized by a long latency of effect of eight to twelve weeks, and by the high doses required by some patients. A combination of pharmacotherapy and behavio

Separation anxiety in children and adolescents treated with risperidone.

Risperidone is an atypical antipsychotic that has been investigated as a treatment for several severe psychiatric disorders in children and adults. A low dose of risperidone was added to other medications to treat two adolescent boys with obsessive compulsive disorder (OCD) and a childhood history of separation anxiety disorder, and one prepubertal boy with a history of attention deficit hyperactivity disorder and escalating behavior suggestive of mania. Each patient developed severe separation anxiety that resolved when risperidone was discontinued. The response was similar to that described

Effects of dopamine on the gastrointestinal tract of chicks.

The effects of dopamine on the gastrointestinal tract of chicks have been investigated. Dopamine caused concentration-related contractions of the upper oesophagus, crop or lower oesophagus but relaxed the duodenum, ileum, large intestine or caecum in a concentration-related fashion. By means of specific antagonists, the presence of muscarinic, histamine (H1), alpha-adrenergic, 5-HT-ergic and dopaminergic receptors were established in the lower oesophagus and their stimulation caused contraction while beta-adrenergic-receptor stimulation caused relaxation. Contraction of the lower oesophagus in

[Dysmorphophobia (body dysmorphic disorder)].

This article reviews the historical and terminological origins of dysmorphophobia from Herodotus to today. It explains the differences pointed out by many authors, including the DSM-III-R, between body dismorphic disorder and delusional disorder somatic type, which are referred to as monosymptomatic hypochondriacal psychoses in Europe. Epidemiological data, clinical characteristics and outcome are discussed. Explicative theories and neurobiological, developmental and analytical aspects of body image are presented. The association between body dismorphic disorder and other disorders is analyzed

Tourette's syndrome: update.

Tourette's syndrome is a widely misunderstood chronic disorder that develops in childhood and is usually lifelong. It is characterized by waxing and waning of involuntary motor and phonic tics. The features and differential diagnosis are discussed in this paper. The estimated prevalence rate of Tourette's syndrome, 0.05%, implies that this disorder is not rare. The reasons for diagnostic confusion are outlined, and the genetic and neurotransmitter features discussed. The management of Tourette's syndrome has become more effective with the availability of at least two psychoactive drugs, halope

Non-antiepileptic drugs for trigeminal neuralgia.

BACKGROUND: Trigeminal neuralgia was defined by the International Association for the Study of Pain as a sudden, usually unilateral, severe, brief, stabbing recurrent pain in the distribution of one or more branches of the fifth cranial nerve. Standard treatment is with anti-epileptic drugs. Non-antiepileptic drugs have been used in the management of trigeminal neuralgia since the 1970s. This is an update of a review first published in 2006 and previously updated in 2011. OBJECTIVES: To systematically review the efficacy and tolerability of non-antiepileptic drugs for trigeminal neuralgia. S

Separation of drug effects on timing and behavioral inhibition by increased stimulus control.

Impulsive behavior may represent, in part, a failure of behavioral inhibition (the ability to delay or inhibit a response). In this study, use of a multiple signaled-unsignaled differential-reinforcement-of-low-rates (DRL) 15-s schedule allowed examination of drug effects in conditions in which level of stimulus control differed. Results showed that whereas diazepam increased premature responding during signaled and unsignaled DRL components, amphetamine and delta9-tetrahydrocannabinol increased premature responding primarily during unsignaled components when timing was necessary for efficient

Weekly Supervised Administration of Oral Antipsychotics: An Alternative to Long-Acting Injections?

Maintenance of response in schizophrenia is largely dependent on compliance with antipsychotic treatment. When people with schizophrenia are responsible for their own treatment, partial or non-adherence is common and usually results in relapse. Assured compliance with antipsychotic treatment is possible when long-acting injectable antipsychotics are given by healthcare staff, but some patients may not consent to treatment for a variety of reasons. An alternative to long-acting injections is the use of supervised oral administration of long-acting antipsychotics. This method assures compliance
