Acinetobacter baumannii, a notorious nosocomial pathogen, exhibits enhanced antibiotic resistance through biofilm formation. However, a comprehensive understanding of the heterogeneity and regulatory dynamics underlying biofilm development at single-cell resolution is lacking. This study outlines the transcriptional landscape of A. baumannii biofilm formation at single-cell resolution and explores potential therapeutic targets. We monitored the dynamic formation process of biofilms using single-cell RNA sequencing (scRNA-seq) technology. Subsequently, we conducted characteristic genes, gene on

Agricultural freshwater wetlands and their drainage networks represent significant yet underrepresented components of the global carbon cycle. Across Europe, particularly in France, extensive marshlands are managed through seasonal water-level regulation combined with agricultural production, yet their carbon dynamics remain poorly constrained. This study investigates temporal water pCO₂ dynamics and planktonic food-web (PFW) structures in a drainage ditch within an organically managed coastal wetland in Charente-Maritime, representative of widespread agro-wetland management practices. Infections caused by methicillin-resistant Staphylococcus aureus (MRSA), particularly those associated with biofilms on invasive medical devices such as venous catheters, represent a significant public health challenge due to increased antimicrobial resistance. This study evaluated the anti-biofilm potential of geraniol, a natural monoterpene, against MRSA strains. In a catheter-based in vitro model, geraniol substantially inhibited initial biofilm formation and effectively disrupted pre-formed mature MRSA biofilms on catheter surfaces. Scanning electron microscopy (SEM) corroborated structura

Anammox bacteria play a pivotal role in natural and engineered ecosystems, yet their physiological characterization remains severely limited by the lack of pure cultures. Highly enriched planktonic anammox cultures are essential for physiological studies, but conventional cultivation is extremely time-consuming (200-500 d) and hampered by uncontrolled cell aggregation and persistent non-anammox contaminants. Here, we developed a novel two-stage strategy for the rapid cultivation of planktonic anammox bacteria. Anammox granules were first subjected to magnetic-mechanical hybrid dispersion to sl

Multi-drug resistant Gram-negative pathogens are increasingly difficult-to-treat perpetrators of infections. New, innovative, and more multifaceted therapies for the treatment of multi-drug resistant strains are thus urgent to hinder further drug resistance and mitigate deadly, untreatable infections. Our study aimed to investigate the efficacy of cefiderocol against Gram-negative aerobic bacteria alone and in combination with phages. The minimum inhibitory concentration (MIC) of cefiderocol was determined using the microdilution broth method, while the minimum biofilm bactericidal concentrati

The National Institutes of Health of the United States stated that over 80% of microbial infections in the body are due to biofilms, many of which are resilient to traditional antimicrobial treatments and need surgery to resolve the chronic infection. Pseudomonas aeruginosa is a Gram-negative pathogen responsible for biofilm-related infection in immunocompromised patients, including those suffering from cystic fibrosis. Here, we attempted to understand the potential relationship between the structure of bacterial surface glycans and the resistance to antibiotics. We focused the attention on li

Derazantinib (DZB), a pan-fibroblast growth factor receptor (FGFR) inhibitor, exhibits potent activity against FGFR1-3 kinases and has been clinically approved for antitumor therapy. However, its antibacterial properties remain unknown. Here, we demonstrated that DZB displays broad-spectrum activity against Staphylococcus aureus (S. aureus), with minimum inhibitory concentrations (MICs) ranging from 6.25 to 25 μM. DZB exhibited more rapid and stronger bactericidal activity against planktonic cells of both MSSA and MRSA compared to vancomycin. DZB at 6.25 μM robustly inhibited biofilm formation

The global antimicrobial resistance crisis has stimulated the development of innovative therapeutics. Methicillin-resistant Staphylococcus aureus (MRSA), a critical pathogen responsible for skin and soft tissue infections. MRSA biofilms exhibit greater resistance to antibiotics compared to planktonic cells. Antimicrobial peptides (AMPs) are potential alternatives but face challenges like high costs, salt sensitivity, toxicity, and protease degradability. This study developed highly potent, salt-tolerant, nontoxic, and proteolytically stable membranolytic AMPs: d-WRL (composed of all d-amino ac

Managing bacterial infections and the spread of microbial resistance is one of the most critical and complex tasks of modern healthcare infrastructures. Antiseptics and disinfectants such as biocides play a significant role in controlling microbial resistance by reducing the microbial load on surfaces, skin, and environments, thereby limiting the opportunity for pathogens to proliferate and develop resistance. Herein, we tested the different interactions of quaternary ammonium compound (QAC)-based biocide compositions in pursuit of a better antimicrobial performance. An extensive microbiologic

Porphyrins play an extremely important role in both photodynamic (PDT) and sonodynamic therapy (SDT). These techniques, which have a lot in common, are largely based on the interaction between the sensitizer and light or ultrasounds (US), respectively, resulting in the formation of reactive oxygen species (ROS) that have the ability to destroy target cells. SDT requires the use of an appropriate frequency of US waves that are able to excite the chemical compound used. In this study, five porphyrin complexes were used: free-base meso-tetra(N-methyl-pyridinium-4-yl)porphyrin (TMPyP) and its tran

The microbial stability of wine can be compromised by the presence of lactic acid bacteria (LAB) and acetic acid bacteria (AAB), which can cause spoilage via off flavour production, increased acetic acid production, or biofilm formation. To manage the growth of LAB in winemaking, fumaric acid (FA) has been proposed as an alternative to traditional antimicrobial agents, such as sulfur dioxide (SO2). This study aimed to evaluate the inhibitory effects of FA on the growth of LAB and AAB based on the influence of pH and ethanol in a synthetic wine-like medium. The research involved the determinati

Epigallocatechin-3-gallate (EGCG), a polyphenol derived from Green Tea, is one of the sources of natural bioactive compounds which are currently being developed as medicinal ingredients. Besides other biological activities, this natural compound exhibits anti-cariogenic effects. However, EGCG has low physical-chemical stability and poor bioavailability. Thus, the purpose of this study was to develop and characterize lipid-chitosan hybrid nanoparticle with EGCG and to evaluate its in vitro activity against cariogenic planktonic microorganisms. Lipid-chitosan hybrid nanoparticle (LCHNP-EGCG) wer

Antimicrobial photodynamic inactivation (aPDI) is a treatment for the eradication of drug-resistant micro-organisms. One of the advantages of this technique, is that there is minimal possibility of microbial resistance. Hence, herein, the preparation and characterization of novel neutral and cationic morpholine containing Schiff base phthalocyanines are reported. The cationic complexes gave moderate singlet oxygen quantum yields (ΦΔ) of ∼0.2 in aqueous media. Conversely, the neutral complexes generated very low ΦΔ values making them very poor candidates for antimicrobial studies. The cationic

Surfaces in highly anthropized environments are frequently contaminated by both harmless and pathogenic bacteria. Accidental contact between these contaminated surfaces and people could contribute to uncontrolled or even dangerous microbial diffusion. Among all possible solutions useful to achieve effective disinfection, ultraviolet irradiations (UV) emerge as one of the most "Green" technologies since they can inactivate microorganisms via the formation of DNA/RNA dimers, avoiding the environmental pollution associated with the use of chemical sanitizers. To date, mainly UV-C irradiation has

In recent years, the frequency of nosocomial infections has increased. Hospital water systems support the growth of microbes, especially opportunistic premise plumbing pathogens. In this study, planktonic prokaryotic communities present in water samples taken from hospital showers and hand basins, collected over three different sampling phases, were characterized by 16S rRNA gene amplicon sequencing. Significant differences in the abundance of various prokaryotic taxa were found through univariate and multivariate analysis. Overall, the prokaryotic communities of hospital water were taxonomica

Monitoring of planktonic salmon louse (Lepeophtheirus salmonis salmonis) abundance and parameterization of key life-history traits has been hindered by labour-intensive and error-prone quantification using traditional light microscopy. Fluorescence illumination has been proposed as a means of improving visualization, but prior to this study adequate investigation of the relevant fluorescence profiles and measurement conditions has not been undertaken. We investigated the fluorescence profiles of L. salmonis and non-target copepod spp. with excitation and emission matrices (200-600 nm) and iden

Surface disinfection in health-care facilities is critical to prevent dissemination of Clostridioides difficile (C. difficile). Tetracyclines (TCs) are broad-spectrum antibiotics that are associated with a low risk of development of C. difficile infection (CDI) and are used as photosensitizers (PS) in photodynamic therapy (PDT). We evaluated whether TCs may be useful environmental cleansing agents. We compared the in vitro ability to kill C. difficile of four TCs (TC, doxycycline, minocycline, and tigecycline) combined with PDT using ultraviolet A (UVA). We included chitosan, a cationic materi

Cryptococcus neoformans/Cryptococcus gattii are fungal pathogens that affect the central nervous system, mainly in immunocompromised individuals. Due to the limited pharmacological arsenal available for the treatment of cryptococcosis associated with cases of antifungal resistance of Cryptococcus spp. reported in some studies, the search for new compounds with antifungal potential becomes relevant. Thus, the objective of this study was to evaluate the inhibitory effect of phenothiazines (promethazine and chlorpromazine) on C. neoformans/C. gattii planktonic cells and biofilms. In vitro plankto
