A tree peony INDETERMINATE DOMAIN transcription factor PrIDD7 inhibits seed oil accumulation.

PrIDD7 acts as a key negative regulator of tree peony seed oil accumulation by downregulating the FA biosynthesis genes PrHAD and PrKASII. Seed oils serve not only as a critical dietary source of nutrients but also as a key raw material for industrial manufacturing. However, the transcriptional regulatory mechanisms underlying seed oil accumulation remain largely unclear. The tree peony (Paeonia rockii) is characterized by high seed oil content and a high proportion of polyunsaturated fatty acids, which are essential for human health. Here, we identified a tree peony INDETERMINATE DOMAIN trans

The pathogenic effector PHYL1JWB interacts with ZjCURT1A to mediate jujube witches' broom-induced photosynthesis impairment of jujube trees.

Photosynthesis impairment and the accompanying leaf yellowing represent a prominent and physiologically critical symptom of phytoplasma infection. However, the underlying molecular mechanisms remain poorly understood. In this study, we identified the Jujube Witches' Broom pathogenic effector PHYL1JWB, which was previously reported to induce phyllody in jujube, as a key inducer of photosynthesis impairment. Overexpression of PHYL1JWB in sour jujube and Arabidopsis thaliana disrupted photosynthesis by causing severe disorganization of chloroplast ultrastructure. PHYL1JWB directly interacts with

MtMAP3Kδ6 Modulates the Growth and Development through Sugar Metabolism Regulation in Medicago truncatula.

Plant growth and development are intricately regulated by molecular mechanisms, with the mitogen-activated protein kinase (MAPK) signaling cascade and its associated modules being pivotal. In this study, we identified and characterized a member of the MAPKKK family, MtMAP3Kδ6, from Medicago truncatula. This gene, classified within the B3 subgroup of the MAPKKK family, was expressed across various tissues during plant growth. The knockout mutant of MtMAP3Kδ6 displayed dwarfism, characterized by reduced branching and smaller leaf size, whereas overexpression of MtMAP3Kδ6 in Medicago truncatula l

Ethylene response factor 113 regulates the growth and development of poplar through the tryptophan-dependent auxin metabolism.

ERF transcription factors play a pivotal role in orchestrating plant growth and development processes. In this study, PagERF113 was isolated from the 84 K poplar (Populus alba × P. glandulosa) and its function was detected using molecular and genetic methods. Subcellular localization and trans-activation assays indicated that PagERF113 lacks transcriptional activation capability, residing in both the cell membrane and nucleus. Phenotypic analyses revealed that transgenic poplar lines overexpressing PagERF113 (OE) exhibited weedy growth compared to wild-type (WT), characterized by small leaves,

Genetic manipulation of the genes for clonal seeds results in sterility in cotton.

BACKGROUND: Heterosis is a common phenomenon in plants and has been extensively applied in crop breeding. However, the superior traits in the hybrids can only be maintained in the first generation but segregate in the following generations. Maintaining heterosis in generations has been challenging but highly desirable in crop breeding. Recent study showed that maternally produced diploid seeds could be achieved in rice by knocking out three meiosis related genes, namely REC8, PAIR1, OSD1 to create MiMe in combination with egg cell specific expression of BBM transcription factor, a technology c

Suppression of microRNA159 impacts multiple agronomic traits in rice (Oryza sativa L.).

BACKGROUND: microRNAs (miRNAs) are important regulators in plant growth and development. miR159 is a conserved miRNA among different plant species and has various functions in plants. Studies on miR159 are mostly done on model plant, Arabidopsis thaliana. In rice, studies on miR159 were either based upon genome-wide expression analyses focused upon responses to different nitrogen forms and abiotic stress or upon phenotypic studies of transgenic plants overexpressing its precursor. STTM (Short Tandem Target Mimic) is an effective tool to block the activity of endogenous mature miRNA activity in

Arabidopsis thaliana gonidialess A/Zuotin related factors (GlsA/ZRF) are essential for maintenance of meristem integrity.

Observation of a differential expression pattern, including strong expression in meristematic tissue of an Agave tequilana GlsA/ZRF ortholog suggested an important role for this gene during bulbil formation and developmental changes in this species. In order to better understand this role, the two GlsA/ZFR orthologs present in the genome of Arabidopsis thaliana were functionally characterized by analyzing expression patterns, double mutant phenotypes, promoter-GUS fusions and expression of hormone related or meristem marker genes. Patterns of expression for A. thaliana show that GlsA/ZFR genes

Overexpression of the grapevine PGIP1 in tobacco results in compositional changes in the leaf arabinoxyloglucan network in the absence of fungal infection.

BACKGROUND: Constitutive expression of Vitis vinifera polygalacturonase-inhibiting protein 1 (Vvpgip1) has been shown to protect tobacco plants against Botrytis cinerea. Evidence points to additional roles for VvPGIP1, beyond the classical endopolygalacturonase (ePG) inhibition mechanism, in providing protection against fungal infection. Gene expression and biochemical datasets previously obtained, in the absence of infection, point to the cell wall, and particularly the xyloglucan component of transgenic VvPGIP1 lines as playing a role in fungal resistance. RESULTS: To elucidate the role of

Pest resistance function of BtACO8 gene from Balsas teosinte.

The peach aphid (Myzus persicae) is a major agricultural pest that causes significant yield losses and transmits numerous plant viruses. We profiled BtACO8 expression in Balsas teosinte and found that aphid infestation induced its transcripts 12.6-fold at 12 h post-infestation relative to non-infested controls. We therefore cloned BtACO8, encoding 1-aminocyclopropane-1-carboxylate oxidase, and investigated its role in aphid resistance by heterologous overexpression in Arabidopsis thaliana. Three homozygous BtACO8-OE lines were obtained and used for functional characterization. Compared with wi

A rapid and efficient in planta assay for assessment of RNA editing factor performance.

Land plant mitochondria and plastids are endosymbiotic organelles that possess their own, reduced genomes. Gene expression is largely regulated at the transcript level, predominantly by nuclear-encoded proteins. Notably, up to thousands of cytidines in organellar RNAs are specifically edited to uridines, mostly restoring conserved genetic information. Key players in this process are nuclear-encoded pentatricopeptide repeat (PPR) proteins, which bind target RNAs in a one-PPR:one-nucleotide manner, following a two-amino-acid-based code. Cytidine deamination is catalyzed by intrinsic or trans-act

Identification, functional and interaction proteins analysis of BjuBBX25b involved in development regulation and Cd responsiveness in Brassica juncea.

B. juncea (Brassica juncea var. latipa Li) is an important leafy vegetable in China, which is suitable for both fresh consumption and processing. In this study, 71 BjuBBXs were identified and categorized into five subfamilies with segmental duplication driving family expansion. Expression profiling by RNA-seq and qRT-PCR revealed that BjuBBX25b exhibits distinct organ-specific expression patterns and was significantly up-regulated in roots under cadmium (Cd) stress, prompting further functional investigation. To elucidate its role in development and heavy metal response, three BjuBBX25b overex

The transcription factor LbYABBY1 of Limonium bicolor raises salt sensitivity by repressing the LbSAD2 pathway.

The recretohalophyte Limonium bicolor produces salt glands on its leaf surface and can thrive in saline soils. YABBY (YAB) family transcription factors participate in plant development and stress responses; our previous single-cell transcriptome data revealed high expression of LbYAB1 (Lb2G09016) in salt gland subclusters. Here, we confirmed the LbYAB1 expression in salt glands and leaf primordia using RNA in situ hybridization and promoter-driven GUS histological staining; moreover, LbYAB1 expression was induced under 6-benzylaminopurine treatment. We explored LbYAB1 function by generating ov

The effector SJP3 interferes with pistil development by sustaining SHORT VEGETATIVE PHASE 3 expression in jujube.

Jujube witches' broom (JWB) is a phytoplasma disease that causes severe damage to jujube (Ziziphus jujuba) crops worldwide. Diseased jujube plants show enhanced vegetative growth after floral reversion, including leafy flower structures (phyllody) and the fourth whorl converting into a vegetative shoot. In previous research, secreted JWB protein 3 (SJP3) was identified as an inducer of phyllody. However, the molecular mechanisms of SJP3-mediated pistil reversion remain unknown. Here, the effector SJP3 was found to interact with the MADS-box protein SHORT VEGETATIVE PHASE 3 (ZjSVP3). ZjSVP3 was

Expression of a Chlorophyll b Reductase Gene from Zoysia japonica Causes Changes in Leaf Color and Chlorophyll Morphology in Agrostis stolonifera.

The NYC-like (NOL) enzyme is considered as an essential enzyme for chlorophyll b degradation, which catalyzes the formation of 7-hydroxymethyl chlorophyll a from chlorophyll b. The ZjNOL gene was cloned from Zoysia japonica with a completed coding sequence of 981-bp in length, encoding 326 amino acids. ZjNOL was localized on the stroma side of the thylakoid membrane, and co-localized with ZjNYC in the chloroplasts. Multiple photoregulatory elements and hormone regulatory elements were identified in the promoter region of the ZjNOL gene, and the expression level of the ZjNOL gene was dramatical

MKK4-MPK3-WRKY17-mediated salicylic acid degradation increases susceptibility to Glomerella leaf spot in apple.

Glomerella leaf spot (GLS), a fungal disease caused by Colletotrichum fructicola, severely affects apple quality and yield, yet few resistance genes have been identified in apple (Malus domestica Borkh.). Here we found a transcription factor MdWRKY17 significantly induced by C. fructicola infection in the susceptible apple cultivar "Gala." MdWRKY17 overexpressing transgenic "Gala" plants exhibited increased susceptibility to C. fructicola, whereas MdWRKY17 RNA-interference plants showed opposite phenotypes, indicating MdWRKY17 acts as a plant susceptibility factor during C. fructicola infectio

Benefits and concerns associated with biotechnology-derived foods: can additional research reduce children health risks?

The development of techniques devised for the genetic manipulation of foods poses new risks for children with food allergy (FA). The introduction of foreign allergenic proteins from different foods into previously tolerated foods may trigger allergic reactions, often complicating with anaphylactic shock in a subset of allergic babies. Children with FA, even if subjected to preventative diets, always challenge the risk of developing allergic manifestations after unintentional intake of a non tolerated food in restaurant settings, with relatives or schoolmates, etc, where product labelling is ne

Genetic engineering of eggplant accumulating β-carotene in fruit.

Genetic engineering of eggplant using fruit-specific EEF48 promoter-driven bacterial PSY gene, crtB, confers β-carotene accumulation in fruit. Eggplant (Solanum melongena L.) is globally cultivated especially in Asia and is an important source of nutrients in the diets of low-income consumers in developing countries. Since fruits of eggplant have low provitamin A carotenoid content, it is expected to develop eggplant with high carotenoid content for combatting vitamin A deficiency. To achieve this, the present study implemented a metabolic engineering strategy to modify the carotenoid biosynth

Genetic manipulation of gibberellin metabolism in transgenic rice.

The 'green revolution' was fueled by the introduction of the semi-dwarf trait into cereal crop cultivars. The semi-dwarf cultivars--which respond abnormally to the plant growth hormone gibberellin (GA)--are more resistant to wind and rain damage and thus yield more grain when fertilized. To generate dwarf rice plants using a biotechnological approach, we modified the level of GA by overproduction of a GA catabolic enzyme, GA 2-oxidase. When the gene encoding GA 2-oxidase, OsGA2ox1, was constitutively expressed by the actin promoter, transgenic rice showed severe dwarfism but failed to set grai

The bHLH Transcription Factor DkLHW Is a Positive Regulator of Plant Height in Persimmon.

Dwarfism is an important agronomic trait in perennial fruit trees, although its molecular regulatory mechanism in persimmon remains poorly understood. In this study, we identified a bHLH transcription factor, DkLHW, which functions as a positive regulator of plant height in persimmon. Yeast two-hybrid (Y2H), bimolecular fluorescence complementation (BiFC), and pull-down assays demonstrated that DkLHW interacts with DkMYB117. Electrophoretic mobility shift (EMSA) assays showed that DkLHW enhances the DNA-binding ability of DkMYB117 to the DkGA2ox2 promoter. Dual-luciferase reporter assays furth

The combination of morphogenic regulators BABY BOOM and GRF-GIF improves maize transformation efficiency and promotes leaf regeneration.

Transformation is an indispensable tool for plant genetics and functional genomics. Although stable transformation in maize is no longer a major obstacle, there remains a need for accessible and efficient methods for academic laboratories. Here, we present the GGB system, a rapid and efficient approach optimized for immature embryo transformation in B104 and other maize lines. This system combines two distinct morphogenetic regulators, the wheat GRF4-GIF1 chimera and the maize BABY BOOM (BBM) transcription factor (hence the name "GGB") with a modified QuickCorn protocol, enabling regeneration
