PARP1: A Potential Molecular Marker to Identify Cancer During Colposcopy Procedures.

Despite efforts in prevention, cervical cancer still presents with a high worldwide incidence and remains a great problem in public health, especially in low-income countries. Screening programs, such as colposcopy with Papanicolaou testing, have greatly improved mortality rates. However, the agents currently used to delineate those lesions (topical application of acetic acid or Lugol iodine) lack specificity and sometimes can lead to unnecessary biopsies or even cervical excisions. A tool to enable in vivo histology to quickly and quantitatively distinguish between tumor, dysplastic tissue, a

Association study of hsa_circ_0001946, hsa-miR-7-5p and PARP1 in coronary atherosclerotic heart disease.

BACKGROUND: Our previous work identified an aberrant expression of hsa_circ_0001946 in coronary atherosclerotic heart disease (CHD). Here we aimed to verify the role of hsa_circ_0001946 as a biomarker for CHD, and explore the clues of its downstream regulation. METHODS: The hsa_circ_0001946 expression in CHD patients (n = 120) and controls (n = 120) were confirmed with qRT-PCR. CircBank and miRDB were used for target analysis in silico. Spearman correlation test was performed to infer potential interrelationships among the nucleic acid molecular biomarkers, and their predictive abilities were

Inhibition of PARP1 Dampens Pseudorabies Virus Infection through DNA Damage-Induced Antiviral Innate Immunity.

Pseudorabies virus (PRV) is the causative pathogen of Aujeszky's disease in pigs. Although vaccination is currently applied to prevent the morbidity of PRV infection, new applications are urgently needed to control this infectious disease. Poly(ADP-ribose) polymerase 1 (PARP1) functions in DNA damage repair. We report here that pharmacological and genetic inhibition of PARP1 significantly influenced PRV replication. Moreover, we demonstrate that inhibition of PARP1 induced DNA damage response and antiviral innate immunity. Mechanistically, PARP1 inhibition-induced DNA damage response resulted

Modulation of PARP activity by Monomethylarsonous (MMA+3) acid and uranium in mouse thymus.

Arsenic exposure is well established to impair the function of zinc finger proteins, including PARP-1. Previous studies from our lab show that early developing T cells in the thymus are very sensitive to arsenite (As+3)-induced genotoxicity mediated through PARP-1 inhibition. Additionally, it has been shown that uranium (in the form of uranyl acetate, UA) also suppresses PARP-1 activity in HEK cells. However, very little is known about whether the As+3 metabolite, monomethylarsonous acid (MMA+3), also inhibits PARP-1 activity and if this is modified by combined exposures with other metals, suc

Nicotinamide and the skin.

Nicotinamide, an amide form of vitamin B3, boosts cellular energy and regulates poly-ADP-ribose-polymerase 1, an enzyme with important roles in DNA repair and the expression of inflammatory cytokines. Nicotinamide shows promise for the treatment of a wide range of dermatological conditions, including autoimmune blistering disorders, acne, rosacea, ageing skin and atopic dermatitis. In particular, recent studies have also shown it to be a potential agent for reducing actinic keratoses and preventing skin cancers.

Tumor cell intrinsic dsRNA innate immune response triggered by PARP inhibitor is compromised in BRCA1-deficient breast cancer by repressing IRF3.

Poly(ADP-ribose) polymerase 1 (PARP1) inhibition represents a promising targeted therapy for BRCA-deficient cancer patients based on the synthetic lethality theory. Recent evidence shows that the efficacy of DNA damage drugs depends on two aspects: DNA repair signaling and immune response. Applying a functional proteomics approach, we find that the function of the spliceosome is perturbed by PARP inhibitors via enhancing interaction between PARP1 and SF3B1, a key factor of the spliceosome. We demonstrate that differential alternative spliced mRNA and accumulation of double-stranded RNA (dsRNA)

PARP1 interacts with WDR5 to enhance target gene recognition and facilitate tumorigenesis.

Poly (ADP-ribose) polymerase-1 (PARP1) is a nuclear protein that attaches negatively charged poly (ADP-ribose) (PAR) to itself and other target proteins. While its function in DNA damage repair is well established, its role in target chromatin recognition and regulation of gene expression remains to be better understood. This study showed that PARP1 interacts with SET1/MLL complexes by binding directly to WDR5. Notably, although PARP1 does not modulate WDR5 PARylation or the global level of H3K4 methylation, it exerts locus-specific effects on WDR5 binding and H3K4 methylation. Interestingly,

PARP1 inhibition in naïve mouse embryonic stem cells induces viral mimicry.

PARP1/2 inhibitors (PARPi) are effective in cancer therapy due to their synthetic lethality in cells with defects in DNA double-strand break repair (DSBR). Here, we show that DSBR-proficient, naïve pluripotent mouse embryonic stem cells (mESC) exhibit high sensitivity towards PARP1/2 inhibition by talazoparib and olaparib. This sensitivity results from a two-tiered response of mESC to PARPi, starting with the activation of DNA stress signalling via ATM and followed by a p53-controlled, TET-TDG-dependent transcriptional response, including the de-repression of endogenous retroviral elements (ER

Evidence for the efficacy of Iniparib, a PARP-1 inhibitor, in BRCA2-associated pancreatic cancer.

Pancreatic cancer is an aggressive, frequently fatal malignancy that strikes 37,000 patients annually in the U.S.A. It is poorly responsive to standard chemotherapies such as gemcitabine. Approximately 5-10% of pancreatic cancer occurs in the setting of a BRCA2 mutation. Breast and ovarian carcinomas that harbor BRCA2 mutations are susceptible to the effects of an emerging class of targeted agents, namely, poly(ADP-ribose) polymerase (PARP) inhibitors. This report describes the case of a patient with a germline BRCA2 mutation and an associated pancreatic cancer treated with iniparib (BSI-201),

PARP-1 deficiency increases the severity of disease in a mouse model of multiple sclerosis.

Poly(ADP-ribose) polymerase-1 (PARP-1) has been implicated in the pathogenesis of several central nervous system (CNS) disorders. However, the role of PARP-1 in autoimmune CNS injury remains poorly understood. Therefore, we studied experimental autoimmune encephalomyelitis (EAE), a model for multiple sclerosis in mice with a targeted deletion of PARP-1. We identified inherent physiological abnormalities in the circulating and splenic immune composition between PARP-1(-/-) and wild type (WT) mice. Upon EAE induction, PARP-1(-/-) mice had an earlier onset and developed a more severe EAE compared

Towards resolving the enigma of the dichotomy of resveratrol: cis- and trans-resveratrol have opposite effects on TyrRS-regulated PARP1 activation.

Unlike widely perceived, resveratrol (RSV) decreased the average lifespan and extended only the replicative lifespan in yeast. Similarly, although not widely discussed, RSV is also known to evoke neurite degeneration, kidney toxicity, atherosclerosis, premature senescence, and genotoxicity through yet unknown mechanisms. Nevertheless, in vivo animal models of diseases and human clinical trials demonstrate inconsistent protective and beneficial effects. Therefore, the mechanism of action of RSV that elicits beneficial effects remains an enigma. In a previously published work, we demonstrated st

PARP1 inhibition affects pleural mesothelioma cell viability and uncouples AKT/mTOR axis via SIRT1.

Malignant Pleural Mesothelioma (MMe) is a rare but increasingly prevalent, highly aggressive cancer with poor prognosis. The aetiology of MMe is essentially a function of previous exposure to asbestos fibres, which are considered to be an early-stage carcinogen. Asbestos is toxic to human mesothelial cells (HMCs), that activate the nuclear enzyme poly(ADP-ribose) polymerase-1 (PARP1) to repair DNA. The targeting of PARP1 is showing considerable potential for delivering selective tumour cell kill while sparing normal cells, and offers a scientifically rational clinical application. We investiga

Metformin Induces PARP1-mediated Cell Death in NPC/HK1 Human Nasopharyngeal Carcinoma Cells.

BACKGROUND/AIM: Nasopharyngeal carcinoma (NPC) in endemic regions remains prone to treatment failure and poor prognosis due to distant metastasis, underscoring the need for novel therapeutic strategies. Metformin exhibits anticancer activity in NPC; however, the mechanism underlying its single-agent inhibition of tumor cell viability remains incompletely defined. This study investigated the molecular basis of metformin-induced viability inhibition in NPC cells. MATERIALS AND METHODS: NPC/HK1 cells were used in this study. Cell viability was quantified using the MTT assay. Apoptosis- and autop

Computational design, synthesis, and in vitro evaluation of thiadiazole-derived PARP-1 inhibitors.

BACKGROUND: Poly (ADP-ribose) polymerase-1 (PARP-1) is a crucial enzyme in DNA repair pathways and a significant target for cancer therapy. Due to their favorable physicochemical and biological characteristics, heterocyclic scaffolds like 1,3,4-thiadiazoles are good candidates for PARP-1 inhibitor design. OBJECTIVES: The purpose of this work was to use an integrated computational and experimental strategy to design, manufacture, and assess new 1,3,4-thiadiazole derivatives as possible PARP-1 inhibitors. METHODS: MM-GBSA binding-energy calculations and molecular docking were used to create an

PARP inhibitors induce a senescence phenotype in non-small cell lung carcinoma cell lines.

Several anticancer treatments have been shown to activate the DNA damage repair pathway but also, in some cases, to lead to therapy-induced senescence. Senescent cells can either exert protumoral or antitumoral effects. However, it remains poorly characterized which treatments lead to a senescent state. Our findings identify Talazoparib, a PARP1 inhibitor, as the most potent inducer of senescence in nonsmall cell lung carcinoma cell lines among a variety of PARP1 inhibitors. In the absence of PARP1, no senescence phenotype was observed, thus demonstrating that PARP1 is necessary for the induct

Role of AIF in cardiac apoptosis in hypertrophic cardiomyocytes from Dahl salt-sensitive rats.

AIMS: The caspases are thought to be central mediators of the apoptotic program, but recent data indicate that apoptosis may also be mediated by caspase-independent mechanisms such as apoptosis-inducing factor (AIF). The role of AIF-induced apoptosis in heart, however, is currently not well understood. The aim of this study was to investigate the presence of and conditions for AIF-induced cardiac apoptosis in vitro. METHODS AND RESULTS: Hypertrophic cardiomyocyte (H-CM) cultures were prepared from the hearts of Dahl salt-sensitive rats fed a high salt diet. Apoptotic stimulation induced by hy

PARP1 during embryo implantation and its upregulation by oestradiol in mice.

Pregnancy requires successful implantation of an embryo, which occurs during a restricted period defined as 'receptivity of the endometrium' and is influenced by the ovarian steroids progesterone and oestradiol. The role of poly(ADP-ribose)polymerase-1 (PARP1) in apoptosis is well established. However, it is also involved in cell differentiation, proliferation and tissue remodelling. Previous studies have described the presence of PARP in the uterus, but its exact role in embryo implantation is not yet elucidated. Hence, in this study, we studied the expression of PARP1 in the uterus during em

Poly(ADP-ribose)-mediated interplay of XPA and PARP1 leads to reciprocal regulation of protein function.

Poly(ADP-ribose) (PAR) is a complex and reversible post-translational modification that controls protein function and localization through covalent modification of, or noncovalent binding to target proteins. Previously, we and others characterized the noncovalent, high-affinity binding of the key nucleotide excision repair (NER) protein XPA to PAR. In the present study, we address the functional relevance of this interaction. First, we confirm that pharmacological inhibition of cellular poly(ADP-ribosyl)ation (PARylation) impairs NER efficacy. Second, we demonstrate that the XPA-PAR interactio

PJ34, a PARP1 inhibitor, promotes endothelial repair in a rabbit model of high fat diet-induced atherosclerosis.

Objective: Atherosclerosis involves endothelial injury caused by oxidative stress. Endothelial progenitor cells (EPCs) play important roles in preventing the early stages of atherosclerosis. Meanwhile, poly (ADP-ribose) polymerase 1 (PARP1) utilizes nicotinamide adenine dinucleotide (NAD+) to repair DNA damage. PARP1 overactivation results in excessive NAD+ consumption in the presence of pathological DNA damage. PJ34 is a PARP1 inhibitor that attenuates cellular NAD+ depletion and can prevent endothelial dysfunction. However, few studies have been conducted on its effects on EPCs. In this stud
