Penicillin anaphylaxis: a review of sensitization, treatment, and prevention.

Penicillin and its derivatives are the commonest cause of drug-induced anaphylaxis, accounting for some 500 deaths per year in the United States. Penicillins are, however, sometimes withheld unnecessarily in patients who give spurious histories of penicillin allergy. A properly performed skin test will detect patients at risk of serious anaphylaxis. Many physicians do not fully appreciate the value of penicillin skin testing, however, and this has contributed to the withholding of penicillin and the use of other, often more expensive medications. Skin testing with a mixture of benzylpenicilloy

Characterization of an antigen in acute anaphylactic dialysis reactions: ethylene oxide-altered human serum albumin.

Anaphylactic reactions to dialysis have been associated with antibody to human serum albumin (HSA) that has been reacted with ethylene oxide (ETO). ETO, an agent used to sterilize dialyzers, is a potent alkylator of proteins. We have studied the antigenic characteristics of ETO-HSA. By use of immunoelectrophoresis we have demonstrated differences in electrophoretic mobility between HSA and ETO-HSA. By gel chromatography we have observed an apparent increase in the molecular weight of ETO-HSA as compared to HSA. Antigenic activity of ETO-HSA as determined by the technique of Lidd and Farr is fo

Tunable adhesive hydrogel enhances diabetic wound closure through bacterial inactivation and immune microenvironment modulation.

Diabetic wound healing remains a complex scientific challenge due to the extremely complex microenvironment, including sustained hypoxia, high levels of reactive oxygen species (ROS), repeated bacterial infection, and abnormal immune regulation. Herein, a tunable adhesive hydrogel (PTA@Hydrogel) is constructed that regulates the immune microenvironment to accelerate diabetic wound healing. ε-Polylysine (EPL) was cross-linked with N-isopropylacrylamide (NIPAM), polydopamine (PDA) coated microalgae system (PTA), and α-lipoic acid sodium (LA-Na) under 365 nm irradiation to fabricate PTA@Hydrogel.

Multifunctional Antiswelling Hydrogel for Underwater Sensing Achieved through Ultrafast Protocatechualdehyde-Fe3+ Autocatalytic Gelation.

Hydrogel-based sensors typically exhibit pronounced swelling underwater, leading to the accelerated deterioration of mechanical properties and signal distortion, thereby limiting their applicability. In this study, we developed an antiswelling hydrogel comprising N, N'-methylenebis (acrylamide)-cross-linked poly(acrylic acid) (PAAc) and polylysine (PLL) in a water/glycerol (Gl) binary solvent system via a protocatechualdehyde (PA)-Fe3+ autocatalytic system, inspired by natural catechol/metal-ion coordination and solvent-exchange strategies. The PA-Fe3+ system rapidly initiates AAc polymerizati

A polymeric triple-enzyme nanoparticle by light activatable crosslinking for cascade catalytic therapy of osteosarcoma.

Multi-enzymatic cascade reactions play fundamental roles in biosynthesis, energetic metabolism, cell signaling, proliferation and apoptosis in living cells. Such reactions have been proposed for cancer therapy via overproducing reactive oxygen species, depleting nutrients for tumor starvation, or remodeling immunosuppressive microenvironment, however, co-delivery of multiple enzymes with optimal cascade catalytic activity remains challenging. Here, we propose a polymer nanoparticle loaded with glucose oxidase (GOx), lactate oxidase (LOx), and chloroperoxidase (CPO) for synergistic cancer catal

Block Copolymer Micellization of DNA Origami Promotes Solubility in Organic Solvents.

The DNA origami technique allows the precise synthesis of complex, biocompatible nanomaterials containing small molecules, biomolecules, and inorganic nanoparticles. The negatively charged phosphates in the backbone make DNA highly water-soluble and require salts to shield its electrostatic repulsion. DNA origamis are therefore not soluble in most organic solvents. While this is not problematic for applications in biochemistry, biophysics, or nanomedicine, other potential applications, processes, and substrates are incompatible with saline solutions, which include the synthesis of many nanomat

Effect of insoluble extracellular matrix molecules on Fas expression in epithelial cells.

Fas, which functions to initiate a signal causing apoptosis, is expressed in epithelia, thus, suggesting a role in controlling cell number during states of cell and matrix turnover. In view of this, we hypothesized that cell-matrix interactions may be an important determinant of Fas expression in epithelial cells. To investigate this, we examined the effect of insoluble extracellular matrix molecules on Fas expression in murine lung epithelial (MLE) cells, a transformed mouse lung epithelial cell line. We report that 1) insoluble extracellular matrices increased Fas mRNA in a time and concentr

Biodistribution of anti-CEA F(ab')2 fragments conjugated with chelating polymers: influence of conjugate electron charge on tumor uptake and blood clearance.

F(ab')2 fragments of anti-carcinoembryonic antigen (CEA) monoclonal antibody (mAb) were modified with three chain-terminal polylysine-based chelating polymers so as to carry different electron charges. Immunoreactive conjugates labeled with 111In up to a specific radioactivity of 120-140 microCi/micrograms were injected into nude mice bearing human colorectal carcinoma, and the biodistribution patterns were compared with each other and with that of an anti-CEA F(ab')2-DTPA control. Immunoconjugate modified with positively-charged polymer produced the highest tumor uptake [up to 20% injected do

ε-poly-l-lysine-grafted nanocellulose prepared by enzymatic oxidation for high-internal-phase Pickering emulsions and nutraceutical delivery.

This study presents a strategy for preparing ε-poly-l-lysine-grafted nanocellulose via TEMPO-laccase/O2 oxidation followed by aldehyde-directed Schiff-base coupling. The resulting nanofibrils, with lengths of 0.8-1.2 μm, exhibited zwitterionic behavior, with ζ-potentials ranging from +49 to -28 mV, while Schiff-base grafting at pH 9 afforded the highest surface amino-group density of 0.915 ± 0.08 mmol g-1. Owing to the combination of high-aspect-ratio fibrillar morphology and amphoteric surface functionality, ε-PL-TLOCN enabled the formation of ultra-stable oil-in-water high-internal-phase emu

Effects of Oligolysine-Polyethylene Glycol Coating on the Biodistribution of Wireframe DNA Origami Nanosheets in Zebrafish Embryos.

DNA origami-based nanotechnology is a versatile tool for exploring fundamental biological questions and holds significant promise for future biomedical applications. Here, we leverage the optical transparency of the embryonic zebrafish to analyze live embryos injected intravenously with fluorescently labeled wireframe DNA origami nanosheets. Our approach integrated long-term, high-resolution imaging of transgenic live zebrafish embryos with single-cell RNA sequencing to elucidate the effects of oligolysine-polyethylene glycol copolymer (K-PEG) coating on the biodistribution of fluorescence sig

Effect of redox-responsive DTSSP crosslinking on poly(l-lysine)-grafted-poly(ethylene glycol) nanoparticles for delivery of proteins.

Therapeutic proteins are utilized in a variety of clinical applications, but side effects and rapid in vivo clearance still present hurdles. An approach that addresses both drawbacks is protein encapsulation within in a polymeric nanoparticle, which is effective but introduces the additional challenge of destabilizing the nanoparticle shell in clinically relevant locations. This study examined the effects of crosslinking self-assembled poly(l-lysine)-grafted-poly(ethylene glycol) nanoparticles with redox-responsive 3,3'-dithiobis(sulfosuccinimidyl propionate) (DTSSP) to achieve nanoparticle de

Microencapsulation of pancreatic islet cells: a bioartificial endocrine pancreas.

It was about two decades ago that Chang proposed the use of microencapsulated islets as artificial beta cells. By using alginate-poly(L-lysine)-alginate membranes, biocompatible, durable capsules containing viable islet cells can be produced which are impermeable to cells and effector molecules of the immune system, thus providing a total protection to transplanted islets against rejection. The capsule wall contains 93% (w/w) water and can be classified as a hydrogel. Many hydrogels have gained general acceptance as being biocompatible materials. Complexation of Polypeptides and Polypeptoids with Nucleic Acids: Does Chirality Matter for Salt Stability and Morphology?

Liquid-liquid phase separation (LLPS), especially via coacervation, offers a novel drug delivery strategy by encapsulating therapeutic agents within phase-separated droplets, thereby improving stability, solubility, and controlled release. Polypeptides and polypeptoids are ideal biomaterials for these systems due to their versatility and tunable properties. Polypeptoids are particularly advantageous, offering enhanced enzymatic resistance and greater control over molecular interactions, making them suitable for complexation studies. This research explores the binary complexation of L-, D,L-, a

Ultrasound-Guided Injectable Adhesive Hydrogel Enabling Targeted Hemostasis in Deep and Incompressible Wounds.

Conventional hemostatic materials struggle to rapidly control bleeding in deep, incompressible visceral wounds, especially when surgical access is unavailable. In response, we designed an injectable, bio-based hydrogel for ultrasound-guided hemostasis. By chemically modifying natural polymers: hyaluronic acid, chitosan, and polylysine, and inducing a Schiff base reaction between aldehyde and amino groups at room temperature, the hydrogel forms instantly using a dual-syringe system without requiring additional stimuli such as heat or light. After optimizing the formulation, the hydrogel (OPH) a

Promicrobial mucoadhesive micro-composites enable delivery of beneficial oral bacteria to restore and modulate oral biofilm communities.

Imbalances within the oral microbiome, composed of over 700 phylotypes, drive both local diseases, including periodontitis, and systemic conditions, such as rheumatoid arthritis and cardiovascular disease. Given the overuse of conventional antimicrobial agents to manage oral diseases and the relapsing nature associated with current intervention strategies, innovative promicrobial approaches to oral biofilm community restoration are needed. Importantly, there is a critical unmet clinical need for active restoration and sustained delivery of beneficial oral commensals rather than continued disru

A Facile, Scalable, and Universal Strategy to Construct Densely Packed l-Lysine-Derived Zwitterionic Brushes for Blood-Contacting Catheters.

Thrombus formation associated with blood-contacting catheters remains a prevalent clinical issue. Antithrombotic coatings capable of inhibiting coagulation activation without interfering with hemostasis are highly desired. Herein, we constructed thick and densely packed poly(ε-acryloyl l-lysine) (p(LysAA)) zwitterionic brushes on the inner wall of chemical inert polyethylene (PE) catheters via a recently developed universal, substrate-independent coating strategy that enables facile scaling-up for catheters with complex geometries. Enhanced antifouling performance of ultra-thin antimicrobial peptide membrane through chemical-crosslinking layer-by-layer assembly.

Biofouling and microbial penetration are still the important issues for surface water purification during gravity-driven microfiltration process, where biofouling has the tendency to deteriorate microbial rejection efficiency. Herein, ultra-thin antimicrobial layer composed of ε-poly-l-Lysine (EPL) and tannic acid (TA) biomacromolecules was successfully chemically crosslinked onto polyvinylidene fluoride (PVDF) membrane via covalent bond-involved layer-by-layer (LbL) method. As-prepared PVDF@EPL membrane could kill bacteria through the contact sterilization effect of AMP. Besides, PVDF@EPL mem

Pseudomonas aeruginosa Adhesion and Biofilm Formation on Poly(l-lysine)-Tethered Hydrogels: Synergistic Effect of Substrate Stiffness and Positive Charge Density.

Infections associated with antibacterial-resistant Pseudomonas aeruginosa (P. aeruginosa) are the major cause of morbidity and mortality of patients, presenting one of the greatest therapeutic challenges for treatment of community-acquired and nosocomial infections. To develop antimicrobial hydrogel coatings to control the adhesion and subsequent biofilm formation of P. aeruginosa, we have used photo-cross-linked poly(ethylene glycol) diacrylate (PEGDA) hydrogels with varied cross-linking densities and covalently grafted poly(l-lysine) (PLL) at different weight compositions (ϕPLL). Grafting of cationic molecules to hyaluronic acid improves adsorption and cartilage lubrication.

Synovial fluid lubricates articular joints by forming a hydrated layer between the cartilage surfaces. In degenerative joint diseases like osteoarthritis (OA), the synovial fluid is compromised, which leads to less effective innate lubrication and exacerbated cartilage degeneration. Studies over the years have led to the development of partially or fully synthetic biolubricants to reduce the coefficient of friction with cartilage in knee joints. Cartilage-adhering, hydrated lubricants are particularly important to provide cartilage lubrication and chondroprotection under high normal load and s
