Inhibitory properties of bambara groundnut protein hydrolysate and peptide fractions against angiotensin-converting enzymes, renin and free radicals.

BACKGROUND: An increased rate of high blood pressure has led to critical human hypertensive conditions in most nations. In the present study, bambara protein hydrolysates (BPHs) obtained using three different proteases (alcalase, trypsin and pepsin) and their peptide fractions (molecular weight: 10, 5, 3 and 1 kDa) were investigated for antihypertensive and antioxidant activities. RESULTS: Alcalase hydrolysate contained the highest amount of low molecular weight (LMW) peptides compared to pepsin and trypsin hydrolysates. LMW peptides fractions (<1 kDa) exhibited the highest inhibitory activit

An animal-derived protein hydrolysate alleviates osmotic stress-induced oxidative damage in tomato by activating ascorbate and glutathione cycle.

BACKGROUND: Protein hydrolysates, as a major group of biostimulants, have gained increasing attention because of their remarkable efficacy in enhancing plant stress resistance. However, the mechanism underlying protein hydrolysate-mediated tolerance to osmotic stress remains poorly understood. In this study, the alterations in oxidative damage and the ascorbate and glutathione (AsA-GSH) cycle were analyzed to investigate the potential effects of protein hydrolysates derived from pig blood (PP) against PEG-induced osmotic stress in tomato. RESULTS: Osmotic stress triggers excessive accumulatio

Chemical acylation of pea protein isolate hydrolysate with fatty acid N-hydroxysuccinimide esters: Effect on structure and functional properties.

Applications of pea protein in the food industry have been greatly restricted by its poor functional properties. In order to solve this problem, a novel technique combining enzymatic hydrolysis and fatty acid acylation has been applied in this work to construct a pea protein-fatty acid covalent complex that aims to improve its functional properties. The processed pea protein with increased water solubility tends to decrease the chance of self-aggregation. Additionally, emulsifying and antioxidant properties have also been found after this process. On top of that, the modified pea protein has b

Dietary tryptophan-rich protein hydrolysate can acutely impact physiological and psychological measures of mood and stress in healthy adults.

BACKGROUND: Tryptophan is the precursor to the mood regulating neurotransmitter serotonin. Its brain bioavailability from food can be dependent on the dietary source. Egg protein hydrolysate (EPH), a dietary supplement rich in tryptophan, has previously shown to acutely impact cognition, mood and stress benefits at 2 g dose. No data exist on the acute effects of lower doses in a food matrix. METHODS: This exploratory study tested the acute effects of low-doses EPH (0.5, 1 g) in a food matrix on cognition, mood and stress. The study employed a double-blinded randomized controlled parallel desi

Separation and identification of ACE inhibitory peptides from lizard fish proteins hydrolysates by metal affinity-immobilized magnetic liposome.

Purification of peptides responsible for angiotensin I-converting enzyme (ACE) inhibitory activity from highly complex protein hydrolysates is difficult. Affinity chromatography is a powerful method for purification of peptides. In this study, a metal affinity-immobilized magnetic liposome (MA-IML) was prepared using lipid, N-hexadecyl iminodiacetic acid (HIDA) and magnetic nanoparticles made of FeCl3·6H2O and FeCl2·4H2O as main materials. MA-IML was used to adsorb ACE inhibitory peptides from lizard fish proteins hydrolysates. The optimal pH of adsorption solution was 8.5. The peptide sample

Toxicity study and blood pressure-lowering efficacy of whey protein concentrate hydrolysate in rat models, plus peptide characterization.

We evaluated the acute (single-dose) and subacute (repeated-dose) oral toxicity of alcalase-hydrolyzed whey protein concentrate. Our acute study revealed no death or treatment-related complications, and the median lethal dose of whey protein concentrate hydrolysate was >2,500 mg/kg. In the subacute study, when the hydrolysate was fed at 3 different concentrations (200, 400, and 800 mg/kg), no groups showed toxicity changes compared with controls. Then, whey protein concentrate hydrolysate was orally administered to spontaneously hypertensive rats. Results revealed significant reductions in blo

Enhanced antibacterial performance of chili pepper seed protein hydrolysates by high hydrostatic pressure assisted enzymatic hydrolysis on Staphylococcus aureus: structural basis and mechanistic insights.

Antibiotic resistance of Staphylococcus aureus threatens public health. Plant by-product protein hydrolysates contain antibacterial peptides as antibiotic alternatives, but compact native protein structures restrict hydrolysis efficiency and antibacterial fragment release. This study applied high hydrostatic pressure (HHP) to assist papain hydrolysis of chili pepper seed protein (PSP) to improve hydrolysis efficiency and antibacterial activity. Pressurisation (100-500 MPa) enhanced PSP hydrolysis degree by 4.48-28.84%. Hydrolysis at 100 MPa gave rise to hydrolysate optimal antibacterial activi

Comparative evaluation of liposome and ergosterol-niosome systems for encapsulation of goat milk protein hydrolysates: Structural properties, in vitro digestive stability, and storage stability.

This study developed and compared liposome and niosome delivery systems to improve the stability and bioaccessibility of goat milk protein hydrolysates. The results showed that the niosome delivery system incorporating ergosterol exhibited superior encapsulation efficiency (96.5 ± 0.04%) and smaller average particle size (216 ± 4.0 nm) compared with the liposome system (p < 0.05). Atomic force microscope results showed that both delivery systems had blunt cone morphology. FTIR spectral analysis also confirmed the successful encapsulating of the enzyme hydrolysates within the delivery systems.

Systematic Analysis of Peptide Mapping and Structural and Functional Identification of Protein Hydrolysates Derived from Six Kinds of Livestock/Poultry Meats Based on Peptidomics.

The purpose of this study was to investigate the effects of cooking treatment on the structural properties and potential bioactive peptide release of six meat protein hydrolysates using a cooking-assisted enzymatic hydrolysis model. FTIR showed that cooking reduced the contents of α-helix and β-sheet structures and promoted the formation of β-turn and random helix structures. The enzymatic hydrolysates demonstrated evident antioxidant activity in vitro, both before and after cooking, which was affected by species differences. LC-MS/MS combined with bioinformatics analysis indicated that cookin

Multi-immobilization of viscozyme, alcalase and flavourzyme by nanomagnetic combi-CLEAs method for oil and protein hydrolysates extraction from rice bran in aqueous phase.

Edible oil isolation from oil seeds by hexane faces pivotal challenges for human health and the environment. The aqueous enzymatic extraction procedure has been remarked as a preferable technique for concurrent oil and protein extraction. A one-pot combined cross-linked enzyme aggregates of Viscozyme, Alcalase and Flavourzyme in nanoscale for coinciding isolation of oil and protein hydrolysates is presented. The ideal parameters for immobilization were 1:1:1 ratio of enzymes in ethanol as aggregator, 0.75:1 ratio of lysine/enzyme mixture, 15 mM glutaraldehyde at temp = 2-3 °C, ultrasonic waves

Discovery of a novel dual-functional peptide from oyster protein hydrolysates with antioxidant and osteogenic properties: in vitro, in silico, and zebrafish model evaluation.

Bioactive peptides are emerging as promising agents for promoting osteogenesis and alleviating oxidative stress - both critical factors in osteoporosis development. In this study, we aimed to discover peptides from oyster protein hydrolysates with dual antioxidant and osteogenic activities. Hydrolysates were prepared from oyster mantle, non-mantle, and whole-body tissues, among which the <1 kDa fraction of the mantle hydrolysate showed the highest bioactivity. Peptides in this fraction were identified by LC-MS/MS and screened via in silico bioactivity prediction and molecular docking with BMPR

Production of Fish Protein Hydrolysates from Scyliorhinus canicula Discards with Antihypertensive and Antioxidant Activities by Enzymatic Hydrolysis and Mathematical Optimization Using Response Surface Methodology.

Fish discards are of major concern in new EU policies. Alternatives for the management of the new biomass that has to be landed is compulsory. The production of bioactive compounds from fish protein hydrolysates (FPH) has been explored in recent years. However, the viability of Scyliorhinus canicula discards, which might account for up to 90-100% of captures in mixed trawler, gillnet, and longline industrial fisheries, to produce FPH from the muscle with bioactivities has still not been studied in terms of the optimization of the experimental conditions to enhance its production. The effect of
