Advancing human metapneumovirus vaccine development through fusion protein stabilization.

Human metapneumovirus (hMPV) is a major respiratory pathogen that causes a substantial global disease burden, particularly in infants, older adults, and immunocompromised individuals. Despite more than two decades of research, no licensed vaccines or antiviral therapies are available, underscoring a persistent unmet clinical need. The viral fusion (F) glycoprotein is the leading target for vaccine and immunotherapeutic development due to its essential role in viral entry and its ability to elicit neutralizing antibodies. However, hMPV-F is highly metastable and undergoes conformational transit

The RNA-binding protein RALY promotes ATM activation by stabilizing Tip60.

A rapid DNA damage response (DDR) and efficient DNA repair are essential for maintaining genome integrity. As a central apical kinase, ATM phosphorylates multiple downstream substrates to initiate DDR signaling and coordinate DNA repair following double-strand breaks (DSBs). However, the precise molecular mechanisms underlying ATM activation remain incompletely understood. Here, we identify the RNA-binding protein RALY as a critical regulator of ATM activation. We show that RALY directly interacts with and stabilizes the acetyltransferase Tip60, a key activator of ATM. Mechanistically, RALY co

LncRNA THOR regulates spermatogenesis by interacting with and stabilizing Sycp3 protein in zebrafish.

THOR (testis-associated highly conserved oncogenic long non-coding RNA) is a highly conserved and testis-enriched lncRNA across vertebrates that plays diverse roles in various cancers. However, its physiological function and regulatory mechanism in testes remain largely unknown. Here, we investigated the genomic location and expression pattern of THOR in the model organism zebrafish, and generated a homozygous THOR knockout model using CRISPR-Cas9 technology. Loss of THOR in zebrafish impaired spermatogenesis, leading to oligospermia (38.7% reduction in sperm count), reduced sperm motility, sp

Impact of lactose and soluble milk salts on the heat stability of milk proteins.

Our objective was to determine the separate effects of lactose, soluble mineral concentration, and added dipotassium phosphate (DKP) on the heat stability of milk protein beverages made from rehydrated milk protein concentrate (MPC) with 85% crude protein on a DM basis (MPC85). Seven batches of rehydrated (7.5% protein) liquid MPC were made from each of 2 lots of MPC85 powder. Treatment 1 was 7.5% MPC85 in deionized (DI) water. Treatment 2 was 7.5% MPC85 that was rehydrated with 4°C Jenness-Koops (JK) buffer, a mix of salts that simulate milk salts. Treatment 3 was 7.5% MPC85 with JK buffer wi

Binding affinities for 2D protein dimerization benefit from enthalpic stabilization.

Dimerization underpins all macromolecular assembly processes both on and off the membrane. While the strength of dimerization, K D , is commonly quantified in solution (3D), many proteins like the soluble BAR domain-containing proteins also reversibly dimerize while bound to a membrane surface (2D). The ratio of dissociation constants, h = K D 2 D K D 3 D , defines a lengthscale that is essential for determining whether dimerization is more favorable in solution or on the membrane surface, particularly for these proteins that reversibly transition between 3D and 2D. While purely ent

Covalently crosslinked coacervates: immobilization and stabilization of proteins with enhanced enzymatic activity.

Coacervates represent models for membrane-free protocells and thus provide a simple route to synthetic cellular-like systems that provide selective encapsulation of solutes. Here, we demonstrate a simple and versatile post-coacervation crosslink method using the thiol-ene click reaction in aqueous media to prepare covalently crosslinked coacervates. The crosslinking of the coacervate enables stability at extreme pH where the uncrosslinked coacervate fully disassembles. The crosslinking also enhances the hydrophobicity within the coacervate environment to increase the encapsulation efficiency o

USP3 deubiquitinates and stabilizes the adapter protein ASC to regulate inflammasome activation.

Inflammasomes are essential components of the innate immune system and its defense against infections, whereas the dysregulation of inflammasome activation has a detrimental effect on human health. The activation of inflammasomes is subjected to tight regulation to maintain immune homeostasis, yet the underlying mechanism remains elusive. Here, we identify USP3 as a direct deubiquitinating enzyme (DUB) for ASC, the central adapter mediating the assembly and activation of most inflammasomes. USP3 removes the K48-linked ubiquitination on ASC and strengthens its stability by blocking proteasomal

The adaptor protein chaperone AAGAB stabilizes AP-4 complex subunits.

Adaptor protein 4 (AP-4) is a heterotetrameric complex composed of ε, β4, μ4, and σ4 subunits that mediates export of a subset of transmembrane cargos, including autophagy protein 9A (ATG9A), from the trans-Golgi network (TGN). AP-4 has received particular attention in recent years because mutations in any of its subunits cause a complicated form of hereditary spastic paraplegia referred to as "AP-4-deficiency syndrome." The identification of proteins that interact with AP-4 has shed light on the mechanisms of AP-4-dependent cargo sorting and distribution within the cell. However, the mechanis

Dynamic aspects of pressure and temperature-stabilized intermediates of outer surface protein A.

Structural characterization of alternatively folded and partially disordered protein conformations remains challenging. Outer surface protein A (OspA) is a pivotal protein in Borrelia infection, which is the etiological agent of Lyme disease. OspA exists in equilibrium with intermediate conformations, in which the central and the C-terminal regions of the protein have lower stabilities than the N-terminal. Here, we characterize pressure- and temperature-stabilized intermediates of OspA by nuclear magnetic resonance spectroscopy combined with paramagnetic relaxation enhancement (PRE). We found

STAM-binding protein regulates melanoma metastasis through SLUG stabilization.

STAM-binding protein, STAMBP, is a JAMM-family deubiquitinating enzyme containing the microtubule-interacting/transport domain and STAM-binding domain. Although the biological importance of STAMBP in development has been recognized because the microcephaly-capillary malformation syndrome in human is caused by its somatic mutations, the role of STAMBP in cancer has not yet been determined. In this study, we demonstrate that STAMBP is a key molecule for regulating melanoma migration and invasion, but not survival, by knocking down STAMBP in vitro. STAMBP regulates SLUG expression through a post-

The Mechanistic Impact of N-Glycosylation on Stability, Pharmacokinetics, and Immunogenicity of Therapeutic Proteins.

N-glycosylation is one of major post-translational modifications in nature, and it is essential for protein structure and function. As hydrophilic moieties of glycoproteins, N-glycans play important roles in protein stability. They protect the proteins against proteolytic degradation, aggregation, and thermal denaturation through maintaining optimal conformations. There are extensive evidences showing the involvement of N-glycans in the pharmacodynamics and pharmacokinetics of recombinant therapeutic proteins and antibodies. Highly sialylated complex-type glycans enable the longer serum half-l

In vitro digestion of oil-in-water emulsions stabilized by whey protein nanofibrils.

The effect of pH (3 and 7) and varied energy density of a high-pressure homogenization process on the stability of oil-in-water (O/W) emulsions stabilized by whey protein fibrils was evaluated. A dynamic digestion model comprising the simulation of stomach, duodenum, jejunum and ileum, has been used to evaluate O/W emulsions' behavior under gastrointestinal (GI) conditions. The emulsions did not separate phases during the storage period (7 days). The emulsions stabilized by whey protein fibrils were stable under simulated gastric conditions but were destabilized in the simulated intestinal con

Stabilization of mutant BRCA1 protein confers PARP inhibitor and platinum resistance.

Breast Cancer Type 1 Susceptibility Protein (BRCA1)-deficient cells have compromised DNA repair and are sensitive to poly(ADP-ribose) polymerase (PARP) inhibitors. Despite initial responses, the development of resistance limits clinical efficacy. Mutations in the BRCA C-terminal (BRCT) domain of BRCA1 frequently create protein products unable to fold that are subject to protease-mediated degradation. Here, we show HSP90-mediated stabilization of a BRCT domain mutant BRCA1 protein under PARP inhibitor selection pressure. The stabilized mutant BRCA1 protein interacted with PALB2-BRCA2-RAD51, was

The methionine-aromatic motif plays a unique role in stabilizing protein structure.

Of the 20 amino acids, the precise function of methionine (Met) remains among the least well understood. To establish a determining characteristic of methionine that fundamentally differentiates it from purely hydrophobic residues, we have used in vitro cellular experiments, molecular simulations, quantum calculations, and a bioinformatics screen of the Protein Data Bank. We show that approximately one-third of all known protein structures contain an energetically stabilizing Met-aromatic motif and, remarkably, that greater than 10,000 structures contain this motif more than 10 times. α-Hemoglobin-stabilizing protein is a sensitive and specific marker of erythroid precursors.

α-Hemoglobin-stabilizing protein (AHSP) is an abundant erythroid-specific chaperone protein that facilitates incorporation of nascent α-globin into hemoglobin A. We characterized AHSP expression by immunohistochemistry in a panel of 100 neoplastic and reactive bone marrow biopsies and splenic tissue with extramedullary hematopoiesis and compared it with established erythroid markers CD71 and CD235a. In all cases, AHSP expression was limited to physiological nucleated erythroid precursors (EPs) and blasts in erythroid leukemias. Although CD71 also stained EPs, it additionally stained nonerythro

Protein stabilization in aqueous solutions of polyphosphazene polyelectrolyte and non-ionic surfactants.

Applications of polyelectrolytes as pharmaceutical excipients or biologically active agents generated an increased interest in formulations, in which ionic macromolecules share the same milieu with protein drugs or vaccine antigens. Macromolecular interactions, which often take place in such systems, can potentially impact formulation activity and stability. The present article reports that poly[di(carboxylatophenoxy)phosphazene], disodium salt (PCPP), which has been previously shown to be a potent vaccine adjuvant, also displays a strong protein stabilizing effect in aqueous solutions that ca

Chemometric evaluation of key residues affecting the stability of cold shock proteins.

Reduced properties, also known as amino acid descriptors, quantify changes in physicochemical properties of amino acids upon mutation. Combined with PLS (partial least squares) modeling, the impact of various mutations on the conformational stability of cold shock proteins (CSPs) was examined. Consistent with the initial evaluation of these systems, electrostatic properties at a select number of central residues were found to govern the conformational stability of CSP mutants. In addition, the current studies found that packing efficiency within the β-sheet core and flexibility of the peptide

SUMO1 modification stabilizes polymerase protein VP1 of Blotched Snakehead Virus.

SUMOylation, the covalent attachment of SUMO molecules to target proteins, is a post-translational modification (PTM) that plays essential roles in the replication of many viruses. In this study, we demonstrate that SUMO1 effectively conjugates to VP1, the RNA-dependent RNA polymerase (RdRp) of Blotched Snakehead Virus (BSNV). SUMO1 modification of BSNV VP1 requires the active sites of both SUMO1 and Ubc9. Site-directed mutagenesis revealed that the SIM motif (393IYIY396) of VP1 is essential for its SUMOylation. Notably, the SIM mutant VP1 exhibited significantly reduced protein stability comp

Quinoa protein-dextran conjugates as functional stabilizers for curcumin-loaded Nanoemulsions.

This study employed high-pressure microfluidization (HPM) to facilitate the Maillard reaction between quinoa protein (QP) and dextran (DX), systematically examining the effects of various pressures on the conjugate's physicochemical properties. Fourier transform infrared spectroscopy confirmed the formation of QP-DX conjugates, characterized by a new peak at 1149 cm-1 (covalent CN bond). Secondary and tertiary structure analyses revealed that HPM-assisted Maillard reaction partially unfolded QP molecules, enhancing conformational flexibility and interfacial properties. Pickering emulsions stabilized by protein/polysaccharide polyelectrolyte complexes for lipase catalysis.

The Pickering emulsion stabilized by ovalbumin/pectin polyelectrolyte complex with porcine pancreatic lipase (PPL) dissolved in the aqueous phase was investigated for its feasibility in biphasic catalysis. CLSM and QCM-D analysis revealed that PPL discretely adsorbed to emulsion interface, though they were both negatively charged. The hydrolysis of p-NPP was favored in low stabilizer concentrations and could be saturated by PPL, and the optimum conditions were pH 6.5, ovalbumin to pectin mass ratio 1:3, complex concentration 0.25 %, and PPL dosage 0.6 %. Under these conditions, the Pickering e
