The brain levels of the endogenous excitotoxin quinolinic acid (QUIN) and its bioprecursor, the free radical generator 3-hydroxykynurenine (3-HK), are elevated in early stage Huntington disease (HD). We now examined the status of these metabolites in three mouse models of HD. In R6/2 mice, 3-HK levels were significantly and selectively elevated in the striatum, cortex and cerebellum starting at 4 weeks of age. In contrast, both 3-HK and QUIN levels were increased in the striatum and cortex of the full-length HD models, beginning at 8 months (YAC128) and 15 months (Hdh(Q92) and Hdh(Q111)), resp

Although the involvement of oxidative mechanisms in the cytotoxicity of excitatory amino acids has been well documented, it is not known whether the intrastriatal injection of quinolinic acid (QA) induces changes in glutathione (GSH) metabolism. In this work, the activities of the enzymes GSH reductase (GRD), GSH peroxidase (GPX), and GSH S-transferase (GST), as well as the GSH content, were studied in the striatum, hippocampus, and frontal cortex of rats 1 and 6 weeks following the intrastriatal injection of QA (225 nmol). One group of animals remained untreated. This lesion resulted in a 20%

Quinolinic acid (QA) induced clonic and tonic convulsions in mice when it was injected into the cerebral ventricle. Pretreatment with L-arginine (L-Arg), a substrate of nitric oxide (NO) synthase (NOS), and/or 5,6,7,8-tetrahydrobiopterin (THB), cofactor of NOS, tended to potentiate QA-induced convulsion. NG-monomethyl-L-arginine (NMMA), a competitive NOS inhibitor, diminished QA-induced convulsion. This effect of NMMA was attenuated by coadministration of L-Arg or THB. Sodium nitroprusside (SNP), which spontaneously releases NO, did not potentiate, but diminished QA-induced convulsion. These f

Quinolinic acid (QUIN), an excitotoxic compound present in the mammalian CNS and periphery, has been hypothetically linked to human neurodegenerative disorders such as Huntington's disease and epilepsy. Quinolinic acid phosphoribosyltransferase (QPRT), the catabolic enzyme of QUIN, is found in the CNS and peripheral organs where it may be a major influence on the tissue levels of QUIN. We have measured QPRT activity in human blood as a means of assessing one aspect of QUIN metabolism in humans. The enzyme was present in blood cells, platelets having a sixfold greater activity than erythrocytes

D & C Yellow No. 11 and Quinoline Yellow belong to a group of quinophtalone dyes with a common basic structure. D & C Yellow No. 11 is used mostly in plastics, spirit lacquers, coloured smokes and cosmetics, but it is also increasingly used as a dye in soaps and shampoos. Quinoline Yellow is used for dyeing wool, silk and nylon in cases where good light fastness is not required, but is most commonly used as a drug and food colouring agent. A patient, extremely sensitive to D & C Yellow No. 11 with a positive patch test reaction down to 0.00001%, also reacted to Quinoline Yellow at a test conce

A comparison of the biological activity of isomers with varying alkyl substitutions on the heterocyclic nitrogen of benzhydro[f]quinoline derivatives was made. The secondary amines did not inhibit adrenergic transmission. The trans-isomer of the secondary amine was 0.5 as active as norepinephrine when evaluated for positive chronotropic action in anesthetized cats. The trans-isomers of the N-alkyl derivatives produce inhibition of responses produced by stimulation of cardioaccelerator nerves with doses of 1--5 mumol/kg. Likewise potent emetic activity (dog) and rotational behavior (rat) was ob

Here, a facile and efficient protocol for the synthesis of 3-hydroxyquinolin-8-yl propanoates via Rh(III)-catalyzed C(8)-H activation of 2-substituted quinolines has been developed. The reaction proceeds via C(8)-H activation, functionalization with acrylates, followed by intramolecular migration of the oxygen atom from quinoline N-oxides to the acrylate moiety. In this approach, N-oxide plays a dual role of a traceless directing group as well as a source of an oxygen atom for hydroxylation. This catalytic method involves simultaneous formation of new C-C and C-O bonds and is applicable only f

We present a convenient and efficient protocol to synthesize quinolines and quinazolines in one pot under mild conditions. A variety of substituted quinolines were synthesized in good to excellent yields (up to 97% yield) from the dehydrogenative cyclizations of 2-aminoaryl alcohols and ketones catalyzed by readily available Co(OAc)2·4H2O. This cobalt catalytic system also showed high activity in the reactions of 2-aminobenzyl alcohols with nitriles, affording various quinazoline derivatives (up to 95% yield). The present protocol offers an environmentally benign approach for the synthesis of

Antibiotic-resistant bacteria and surface-attached biofilms continue to play a significant role in human health and disease. Innovative strategies are needed to identify new therapeutic leads to tackle infections of drug-resistant and tolerant bacteria. We synthesized a focused library of 14 new halogenated quinolines to investigate the impact of ClogP values on antibacterial and biofilm-eradication activities. During these investigations, we found select polar appendages at the 2-position of the HQ scaffold were more well-tolerated than others. We were delighted to see multiple compounds disp

RIOK2 is a promising therapeutic target in cancer due to its pivotal role in tumor progression. Based on our previously identified RIOK2 inhibitor CQ211, we conducted a comprehensive pharmacokinetic optimization campaign, leading to the discovery of CQ3196 as a potent and orally bioavailable RIOK2 inhibitor. CQ3196 demonstrates remarkable RIOK2 binding affinity, with a Kd value of 14 nM, and exhibits potent proliferative inhibitory activities in gastric cancer cell lines. Furthermore, CQ3196 displays favorable PK properties, it achieves an AUC(0-t) value of 3.5 × 103 μg/L·h following oral admi

The effects of the novel acylquinoline derivative, 1-(2-methylphenyl)-4-[(3-hydroxypropyl)amino]-6-trifluoromethoxy-2,3- dihydropyrrolo[3,2-c]quinoline (AU-006) on experimental ulcer models and on gastric secretion were examined. AU-006 prevented dose dependently gastric lesions induced by 95% ethanol when given orally (30-300 mg/kg). Similarly, gastric lesions caused by 0.3 N NaOH were inhibited by oral pretreatment with AU-006. To investigate the anti-ulcer mechanism of AU-006, the effect of AU-006 on gastric acid secretion was tested. Intraduodenal administration of AU-006 reduced in vivo g

The multi-step access to versatile N-heterocyclic scaffolds is highly dependent on non-renewable fossil-based feedstock over different catalysts for each step. Here, a mesoporous NaY zeolite-encapsulated atomically dispersed cobalt catalyst (Co@NaY-M) is deliberately designed to be robust for single-step, three-component upgrading of biomass-derived lactic acid, aldehydes, and anilines into 3,4-unsubstituted quinolines via cascade dehydrogenation, annulation-aromatization, and decarboxylation. Na+ co-dopant in zeolitic hexagonal prism adjacent to single-atom Co in the supercage acts as an elec

In this work the synthesis and characterization of new gold(III) complexes with quinoline ligands are described. These complexes contain different steric and electronic properties of the donor atom at 8-position of the quinoline in order to modulate their stability and their biological activity. Their redox potential, stability in organic and aqueous solvents, and their biological activity in a panel of six different human tumor cell lines are also presented. In addition, interaction studies of the complexes with model biological molecules (pBR322 and L-acetyl-N-cysteine) were carried out, sug

A novel method for the preparation of 2-carboxyl-3-aryl quinoline derivatives from anilines, ethyl glyoxalate and enol ethers as phenylacetaldehyde surrogates is reported. The three-component coupling reaction occurs rapidly under mild conditions in dichloromethane catalysed by TFA. The method allows a more direct access to 3-aryl quinolines, sidestepping issues encountered with phenylacetaldehyde derivatives. This chemistry was used to prepare quinolines with 3-diarylether functionality that showed low micromolar efficacy (IC50 range: 5-26 μM) against in vitro Toxoplasma gondii coupled with l

In the title compound, [CdCl(NCS)(C(9)H(8)N(2))](n), the Cd(II) atom is in a distorted octa-hedral coordination environment defined by two chloride anions, two N atoms from an 8-amino-quinoline ligand, one N atom from one thio-cyanate anion and one S atom from a symmetry-related thio-cyanate anion. Two Cd(II) atoms are bridged by two chloride anions, forming an inversion-related Cd(2)Cl(2) unit; these units are further linked through thio-cyanate anions, leading to a chain structure extending parallel to [010]. With the aim to design dual-acting antiprotozoals, a set of hybrid-molecules was prepared. Amino-substituted 2-aza- and 3-azabicyclo[3.2.2]nonanes with antiplasmodial and antitrypanosomal activity were linked via aminoalkyl linkers to the 2,8-bis(trifluoromethyl)quinoline scaffold of mefloquine. All newly synthesized compounds were characterized by IR spectroscopy, NMR spectroscopy, and HRMS. Their antiplasmodial activities were examined in vitro against the NF54 strain of Plasmodium falciparum and their antitrypanosomal activity against the STIB900 strain of Trypanosoma brucei rhodesiense. The ageing population and the increasing prevalence of age-related diseases underscore the urgent need for targeted therapeutic strategies. Accumulating evidence indicates that quinolinic acid (QA), a neuroinflammatory neurotoxin, contributes to the pathogenesis of neurodegenerative disorders. In this study, using Caenorhabditis elegans as a model organism, we demonstrate that chronic QA exposure acts as a robust driver of accelerated aging, significantly reducing overall healthspan. This pro-aging effect is accompanied by the premature onset of decreased locomotor function, enhanced lipofusci
