Sphingolipids as a Novel Therapeutic Target in Radiation-Induced Lung Injury.

Radiation-induced lung injury (RILI) is a potential complication of thoracic radiotherapy that can result in pneumonitis or pulmonary fibrosis and is associated with significant morbidity and mortality. The pathobiology of RILI is complex and includes the generation of free radicals and DNA damage that precipitate oxidative stress, endothelial cell (EC), and epithelial cell injury and inflammation. While the cellular events involved continue to be elucidated and characterized, targeted and effective therapies for RILI remain elusive. Sphingolipids are known to mediate EC function including man

Relationships between the changes of skin temperature and radiation skin injury.

Background: Radiation skin injury (RSI) causes changes in skin temperature, but detailed information on the thermographic responses is currently lacking. We investigated thermographic patterns after radiotherapy. We hypothesized that skin temperature may be used as a diagnostic and early predictor of RSI severity.Method: All breast cancer patients received radiotherapy after unilateral postmastectomy. The contralateral supraclavicular area served as control, and the frontal thermal image of torso was taken by a thermal infrared imager weekly. We defined areas of interest on bilateral symmetric

Role of betaine in liver injury induced by the exposure to ionizing radiation.

Oxidative stress, apoptosis, and fibrosis may play a major role in the development of radiation-induced liver damage. Betaine, a native compound widely present in beetroot, was reported to possess hepato-protective properties. The objective of this study was to investigate the influence of betaine on radiation-induced liver damage. Animals were exposed to 9 Gy applied in 3 doses of 3 Gy/wk. Betaine (400 mg/kg/d), was orally supplemented to rats after the first radiation dose, and daily during the irradiation period. Animals were sacrificed 1 day after the last dose of radiation. The results sh

Prophylactic Bevacizumab May Mitigate Radiation Injury: An Experimental Study.

BACKGROUND: Stereotactic radiosurgery (SRS) is widely used to treat brain pathologies alone or in concert with other treatment modalities. However, there are some side effects, such as radiation injury characterized by edema and necrosis in peripheral tissues, that must be managed. A new treatment agent against this side effect is bevacizumab, which targets increased vascular endothelial growth factor (VEGF) as a prominent etiologic factor in radiation injury. In this study, we created a rat experimental model to describe the effects of both radiation and the anti-VEGF monoclonal antibody beva

Radiation injury to cardiac arteries and myocardium is reduced by soy isoflavones.

OBJECTIVE: The negative effects of incidental radiation on the heart and its vessels, particularly in the treatment of locally advanced non-small cell lung cancer, esophageal cancer, left-sided breast cancer, and lymphoma, are known. Late cardiac events induced by radiotherapy including coronary artery disease, ischemia, congestive heart failure, and myocardial infarction can manifest months to years after radiotherapy. We have previously demonstrated that soy isoflavones mitigate inflammatory responses induced in lungs by thoracic irradiation resulting in decreased vascular damage, inflammati

Oxidative stress mediates radiation lung injury by inducing apoptosis.

PURPOSE: Apoptosis in irradiated normal lung tissue has been observed several weeks after radiation. However, the signaling pathway propagating cell death after radiation remains unknown. METHODS AND MATERIALS: C57BL/6J mice were irradiated with 15 Gy to the whole thorax. Pro-apoptotic signaling was evaluated 6 weeks after radiation with or without administration of AEOL10150, a potent catalytic scavenger of reactive oxygen and nitrogen species. RESULTS: Apoptosis was observed primarily in type I and type II pneumocytes and endothelium. Apoptosis correlated with increased PTEN expression, in

Hepatic radiation injury in distal esophageal carcinoma: a case report.

The liver is an organ at particular risk for toxicity after radiotherapy for distal esophageal cancer. We report the case of a 58-year-old man with distal esophageal carcinoma, who underwent follow-up FDG-PET/CT 6 weeks after completion of preoperative neoadjuvant chemoradiotherapy. The left hepatic lobe and caudate showed new, round areas of hypoenhancement on CECT and increased FDG-uptake, which resembled metastases. However an, as yet unreported, linear configuration of FDG uptake was noted and found to correlate with the anterior posterior-posterior anterior opposing field radiotherapy por

[Experimental study on brachial plexus injury induced by radiation in rats].

OBJECTIVE: To explore the changes of morphology and ventricornual motor neurons in SD rats' ventral horn of spinal cord after radiated as the therapy protocol for breast cancer, to discover the rule of radiation-induced injury of brachial plexus, and also if there exits the reversible conversion in neurons. METHODS: Twenty SD rats were selected. The left side of the rats was used as the radiation side, and the right side as the control side. The RIBPI animal models were established by divided-dose of radiation. Using 2 Gy/time and 5 times/week, a total administered dose reached 30 Gy after 3

Glycine betaine, a beer component, protects radiation-induced injury.

Human whole-blood was exposed to 137Cs gamma-rays or 50 keV/microm carbon ions in the presence or absence of glycine betaine, a beer component in vitro. The dicentrics of chromosome aberrations in human lymphocytes were significantly (p < 0.05) reduced by glycine betaine after irradiation with 4 Gy of either gamma-rays or carbon ions. The maximum protection by glycine betaine for gamma-rays or carbon ions was 37% and 20%, respectively. C3H/He female mice, aged 14 weeks, received an i.p. injection of glycine betaine 15 min before whole-body irradiation with gamma-rays or 50 keV/microm carbon io

Intrarectal application of amifostine for the prevention of radiation-induced rectal injury.

Clinically symptomatic late injury to the rectal wall occurs in about one third of patients with prostate cancer treated with external beam irradiation. Reducing the physical dose to the anterior rectal wall without a similar reduction in the posterior peripheral zone is difficult because of the proximity of these structures. Based on our previous observations that intrarectal application of amifostine resulted in very high concentrations of amifostine and its active metabolite WR-1065 in the rectal wall of Copenhagen rats, the authors initiated a phase I clinical trial in 1998. Radiation therapy for heterotopic ossification in a patient with traumatic brain injury.

Radiation therapy has been known to have a prophylactic effect for heterotopic ossification (HO), but until now it has not been known to have a therapeutic effect for established HO. We report a case of established HO compounded with a sudden increase in activity, that was improved with radiation therapy. A patient with traumatic brain injury had HO in both hips and thighs two months after the initial trauma. The existing level of HO activity suddenly increased seven months after the initial trauma, and was accompanied by severe pain that was refractory to indomethacin. We assumed that the pai

Hyperbaric oxygen therapy for radiation-induced brain injury in children.

BACKGROUND: Radiation-induced necrosis (RIN) of the brain is a complication associated with the use of aggressive focal treatments such as radioactive implants and stereotactic radiosurgery. In an attempt to treat patients with central nervous system (CNS) RIN, ten patients received hyperbaric oxygen treatment (HBOT). METHODS: Patients presented with new or increasing neurologic deficits associated with imaging changes after radiotherapy. Necrosis was proven by biopsy in eight cases. HBOT was comprised of 20-30 sessions at 2.0 to 2.4 atmospheres, for 90 minutes-2 hours. Sites of RIN included

Pathogenesis of acute radiation injury to the rectum. A prospective study in patients.

In a prospective investigation of the clinical and pathological effects of pelvic radiotherapy on the rectum, nine patients (age range 58-77 years) had symptoms assessed weekly during radical treatment for bladder and prostatic tumours, and at 2, 4 and 12 weeks after treatment. Stool frequency increased in all patients from a mean of 1.7 per day before treatment to 5.0 per day at 4 weeks (P < 0.05). Seven patients developed liquid stools during treatment; 6 had pain on defaecation, 8 had urgency of defaecation, all experienced tenesmus and 3 had episodes of faecal incontinence. One patient had

Severe progressive periodontal destruction due to radiation tissue injury.

Cancer radiotherapy to the head and neck region results in short- and long-term radiation tissue injuries. Radiation bone injury is a long-term manifestation which could progress to osteoradionecrosis. A case of radiation tissue injury to the periodontium is presented. The possible pathogenesis of these events is described as they relate to the sequential radiographic changes observed over a period of 6 years until the involved teeth were exfoliated. The post-irradiation management of the teeth with advancing periodontal disease in the path of irradiation was by conservative means, including g

Isolated congenital hemianopia caused by prenatal injury to the optic radiation.

Isolated congenital hemianopias are typically caused by developmental abnormalities of the occipital lobe cortex. We describe two patients with an incidental partial hemianopia associated with unilateral periventricular leukomalacia that was acquired prenatally. Magnetic resonance imaging scans suggest a late second or early third trimester unilateral cerebral ischemic event.

Radiation injury in the neonatal canine kidney. II. Quantitative morphology.

Quantitative morphology was used to investigate injury induced in the developing canine kidney by ionizing radiation. Irradiation at 2 days of age destroyed nephrogenic tissue in the outer renal cortex and significantly reduced kidney weight and volume through 70 days of age. However, as early as 14 days of age and continuing through 200 days of age, hypertrophy was demonstrated in the surviving middle and inner cortical nephrons of these kidneys. Both corpuscular and tubular components of the nephrons grew at an accelerated rate as compared to controls. The corpuscular volume per nephron incr

Respiratory-gated micro-computed tomography imaging to measure radiation-induced lung injuries in mice following ultra-high dose-rate and conventional dose-rate radiation therapy.

PURPOSE: Ultra-high dose-rate radiotherapy (FLASH-RT) shows the potential to eliminate tumors while sparing healthy tissues. To investigate radiation-induced lung damage, we used in vivo respiratory-gated micro-computed tomography (micro-CT) to monitor mice that received photon FLASH-RT or conventional RT on the FLASH irradiation research station at TRIUMF. APPROACH: Thirty healthy male C57BL/6 mice received baseline micro-CT scans followed by radiation therapy targeting the thorax. Treatments administered included no irradiation, 10-MV photon FLASH-RT, and 10-MV conventional RT with either 1

1,2-propanediol ameliorated radiation-induced intestinal injury in mice.

Radiotherapy is constricted by collateral normal tissue injury during treatment, particularly the gastrointestinal tracts, which is usually referred as radiation-induced gastrointestinal syndrome (RIGS). Currently, there is no FDA-approved agent for the prevention or treatment of RIGS. By using a mice model of RIGS, we demonstrated that 1,2-propanediol (1,2-PD) prevents radiation-induced fatal intestinal injury and significantly increases mice survival following lethal doses of radiation. 1,2-PD pretreatment also enhanced the survival of Lgr5+ISCs and improved crypts regeneration after radiati
