Chlorpromazine and pimozide alter reinforcement efficacy and motor performance.

This study evaluated the effects of chlorpromazine and pimozide on reinforced responding. In each session, rats were exposed to a series of five variable-interval reinforcement schedules. The response requirement was a lever press, the reward was a small portion of water, and the reinforcement rate varied from about 20 to 660 reinforcers per hour. Response rate was a negatively accelerated function of reinforcement rate, and the relationship between the two variables was described by the equation for a rectangular hyperbola (the matching law). One parameter of the hyperbola is equivalent to th

Morphine withdrawal decreases responding reinforced by sucrose self-administration in progressive ratio.

Previous studies have shown that withdrawal from psychostimulant drugs such as d-amphetamine or methamphetamine decreases motivation to work for a natural reinforcement, which is thought to be associated with the withdrawal-induced depressive state and hypofunction of the mesolimbic dopamine system. However, to our knowledge, studies exploring the effect of morphine withdrawal on motivation for a natural reinforcement are lacking. The purpose of the present study was to examine whether motivation to work for a natural reinforcement changes during morphine withdrawal. Three groups of male Sprag

Demand for heroin in rats: effects of non-drug alternative substitutes and complements.

Given the central role of economic demand for drugs in substance use disorder, identifying factors that increase or decrease drug value is of high importance. The present study investigated how the concurrent availability of non-drug reinforcers previously shown to be heroin substitutes or complements affected demand for heroin in rats. In Exp. 1, three groups of rats pressed one lever for intravenous heroin infusions on a series of prices that increased over sessions. The groups differed with respect to what they received for pressing a second lever: timeout-from-avoidance (TOA) reinforcers,

Can a Chatbot Help Heal a Wound? Context-Aware Prompts for Boosting Adherence in Diabetic Foot Ulcers.

BACKGROUND: Smart offloading technologies enable the real-time, objective monitoring of adherence in patients with diabetic foot ulcers (DFUs). Although remote tracking may reinforce adherence and improve wound healing, effectiveness depends on sustained device use, particularly as devices are often removed during rest periods. Real-time, behavior-contingent feedback informed by sensor data, including AI-supported messaging capable of detecting nonadherence, may enhance reinforcement. However, the feasibility and behavioral impact of such strategies remain unclear. METHODS: We conducted a pro

Long- vs short-access cocaine alters behavioral inhibition for cocaine in male rats.

Impulsivity and behavioral inhibition are measures commonly associated with substance misuse, particularly cocaine use disorder. However, patterns of impulsive behaviors have been shown to differ based on cocaine use history and level of cocaine dependence. Extended cocaine access, which more closely models neural and behavioral changes that take place during the development of problematic cocaine use, has been shown to decrease behavioral inhibition in comparison to limited cocaine access. However, previous preclinical studies investigating these relationships have been mostly correlational a

Acute inhalation exposure to cyclohexane and schedule-controlled operant performance in rats: comparison to d-amphetamine and chlorpromazine.

Adult male rats pressed a lever on a multiple fixed ratio-fixed interval (FR20-FI120 sec) schedule of food presentation, and after attaining a stable baseline subjects received an acute inhalation exposure to cyclohexane vapor (0 ppm, 500 ppm, 2000 ppm, or 7000 ppm) for 6 hr. During the operant session that began 30 min after termination of exposure, FR running rate for the 7000 ppm group decreased 11% relative to performance on the previous day. FR post-reinforcement pause duration and the rate and pattern of FT performance were unaffected. Cyclohexane exposures of 500 or 2000 ppm had no dete

Interaction of sigma and PCP-like drugs on operant behaviors in the rat.

Many PCP-like drugs interact with at least two types of binding sites in the CNS, one of which is linked to excitatory amino acid transmission and the other with an unknown function. The present experiments were designed to further clarify the mechanism of action of drugs in this class. Assessment was made of the effects of PCP, MK-801, (+)-pentazocine, (+)- and (-)-N-allyl-normetazocine (NANM), (+)-amphetamine and BMY-14802 in rats responding under a multiple timeout 600 s (TO), differential reinforcement of low rates 10 s (DRL), fixed ratio (FR) 10 schedule of reinforcement. Comparison of the effects of clozapine, haloperidol, chlorpromazine and d-amphetamine on performance on a time-constrained progressive ratio schedule and on locomotor behaviour in the rat.

Performance on progressive ratio schedules has been proposed as a means of assessing the effects of drugs on "reinforcer efficacy". It has been proposed that the effects of neuroleptic drugs on operant behaviour are mediated by a reduction of "reinforcer efficacy". We examined the effects of two "conventional" neuroleptics (haloperidol and chlorpromazine) and an "atypical" neuroleptic (clozapine) on progressive ratio schedule performance; d-amphetamine was used as a comparison compound. In experiment 1, rats responded for a sucrose reinforcer on a time-constrained progressive ratio schedule (7

Adjunctive polydipsia as a model of alcoholism.

Schedule-induced polydipsia or adjunctive drinking as been proposed as an animal model of alcoholism. Procedures employed to modify adjunctive drinking are reviewed here in order to evaluate their usefulness in the control of schedule-induced alcohol consumption. The major procedures employed to alter adjunctive drinking are: (1) physiological, such as pre-loading the animal with water, desalivating the animal, or adding a sweet solution to the water; (2) pharmacological, such as evaluating the effects of amphetamines or other drugs on adjunctive drinking; and (3) behavioral, such as changing

Amineptine improves the performance of dogs in a complex temporal regulation schedule.

Amineptine is a tricyclic antidepressant with activating properties, that stimulates spontaneous locomotor activity in rodents and elevates mood in humans. It mainly inhibits dopamine uptake and weakly increased dopamine release. Formulating the hypothesis that this drug would decrease waiting capacity, we decided to test amineptine in a Differential Reinforcement of Response Duration schedule (DRRD 9 s Limited Hold 1.5 s) in the dog. The drug was administered orally at 2.5, 5.0, 7.5, 10 and 20 mg/kg, 1 h before the experimental session. Between 2.5 and 10 mg/kg, amineptine improved the perfor

Effects of gabapentin on heroin and cocaine self-administration in rats.

Reports of gabapentin misuse among people with opioid use disorder have increased, as has the presence of gabapentin in opioid overdose deaths. Despite these trends and known interactions between these drugs in other settings (e.g. gabapentin as an 'opioid-sparing' agent), few studies have evaluated the impact of gabapentin on reinforcing effects of opioids. Intravenous self-administration in rats was used to determine whether gabapentin modifies the reinforcing effects of heroin or cocaine. Rats self-administered drugs under a progressive ratio schedule of reinforcement after saline or gabape

Use of the timeout ribbon procedure during community-based instruction.

Involvement in community-based instruction can be adversely affected when students engage in behavior that interferes with participation in instruction. A multiple probe across settings design with an embedded reversal was used to investigate the effectiveness of the nonseclusionary timeout ribbon procedure for two middle-school students with moderate mental retardation in community and school settings. An athletic wristband served as the timeout ribbon, which functioned as the stimulus for the availability of reinforcement. When the student was wearing the wristband, he could earn reinforcers

Activity changes in rats following acute trimethyltin exposure.

The aliphatic organo-metal derivative, trimethyltin, causes marked morphological damage to the central nervous system (CNS), when the compound is administered by the intragastric route. This report describes certain behavioral consequences of [CH3]3Sn treatment. Either 7.0 mg/kg [CH3]3SnCl or 0.9% saline was injected intragastrically in male rats of Long-Evans strain divided into two equal groups of nine each. Forty days later open-field activity during a 2-min interval was measured for each rat. Following activity testing, the animals were trained to press a lever for food reinforcement on an

Chronic levetiracetam (Keppra®) treatment increases the reinforcing strength of cocaine in rhesus monkeys.

BACKGROUND: Drugs that increase inhibitory neuronal activity in the brain have been proposed as potential medications for stimulant use disorders. OBJECTIVE: The present study assessed the ability of chronically administered levetiracetam (Keppra®), a clinically available anticonvulsant drug that increases GABA by binding to synaptic vesicle glycoprotein 2A, to modulate the reinforcing strength of cocaine in monkeys. METHODS: Three adult male rhesus monkeys (Macaca mulatta) self-administered cocaine intravenously each day under a progressive-ratio (PR) schedule of reinforcement. Two monkeys

Captopril alters schedule induced polydipsia, urination, and defecation in rats.

Schedule induced polydipsia, urination and defecation were examined in rats that received training on a fixed interval 2 min schedule of food reinforcement. In Phase I of the experiment, animals received peripheral injections of captopril (an angiotensin conversion enzyme blocker, 0.5 or 50 mg/kg), or equivalent volumes of 0.9% saline. The results showed that low doses of captopril (0.5 mg/kg) significantly increased both operant responding and the adjunctive behaviors. High peripheral doses of captopril significantly reduced responding and schedule induced behavior. In Phase II of the experim

The effects of chlorpromazine and imipramine on rate and stimulus control of matching to sample.

Pigeons were trained to perform simultaneous, two-color matching to sample under a multiple fixed-ratio fixed-interval schedule of food presentation. The sequence terminating with a peck on the matching key (a "match") was treated as a unit, analogous to a single key peck in conventional schedules. Except for intermittent reinforcement of matches, no consequent stimulus distinguished matches from mismatches (sequences terminating with pecks on the nonmatching key). The pattern of matches during nondrug sessions resembled that of simpler operants maintained by similar schedules. Matches increas

Prolonged behavioral effects of in utero exposure to lead or methyl mercury: reduced sensitivity to changes in reinforcement contingencies during behavioral transitions and in steady state.

Postnatal exposure to lead or methyl mercury results in mental retardation, learning deficits, and other neurobehavioral effects in humans, and adverse consequences of prenatal exposure have been clearly documented with methyl mercury. To examine the developmental neurotoxicity of these metals, especially lead, concurrent schedules of food reinforcement were used to identify learning deficits in squirrel monkeys exposed during gestation to either methyl mercury or lead. Pregnant squirrel monkeys were administered methyl mercury (0.7 to 0.9 ppm in maternal blood) or lead (21 to 79 micrograms/dl

Acute and chronic nicotine effects on multiple-schedule behavior: oral and SC routes.

For rats responding on a 3 h FI 4 min FR 20 schedule of food reinforcement, presession SC nicotine doses (0.1-0.8 mg/kg) produced depression in all responding followed by stimulation of FI responding that was dependent upon both time and dose. With daily presession 0.8 mg/kg SC nicotine injections for 9 days, no tolerance to the depressive or stimulatory effects of nicotine occurred. Prediction and control of food rewards modulate endogenous pain inhibitory systems.

Tail-flick latencies were measured in food-deprived rats submitted to various schedules of food reinforcement with experimental and yoked animals differing by the possibility of either predicting the occurrence of food by means of a pre-food signal, controlling its delivery by lever-pressing or developing adjunctive activities (schedule-induced drinking). In the first two cases, yoked animals that could not predict or control food deliveries displayed higher tail-flick latencies at the end of sessions than did experimental animals. In the polydipsia experiment, rats that did not develop schedu

Effect of dopamine agents on schedule- and deprivation-induced drinking in rats.

The dopamine agonist, apomorphine, or its antagonist, haloperidol, was administered to rats whose drinking was induced by fixed-interval schedules of pellet delivery or by water deprivation. The first study revealed that both drugs produced dose-dependent decreases in bar-pressing and schedule-induced polydipsia (SIP). At higher doses, haloperidol also depressed the rate of pellet delivery. The second study demonstrated that the suppression in SIP obtained in the first study was primarily due to the direct effect of the drugs and not to changes they produced on the underlying food reinforcemen
