Dietary intake of table olives exerts antihypertensive effects in association with changes in gut microbiota in spontaneously hypertensive rats.

Arbequina table olive (AO) consumption lowers blood pressure (BP) in spontaneously hypertensive rats (SHR). This study evaluates whether dietary supplementation with AO induced changes in the gut microbiota that are consistent with the purported antihypertensive effects. Wistar-Kyoto rats (WKY-c) and SHR-c received water, while SHR-o were supplemented by gavage with AO (3.85 g kg-1) for 7 weeks. Faecal microbiota was analysed by 16S rRNA gene sequencing. SHR-c showed increased Firmicutes and decreased Bacteroidetes compared to WKY-c. AO supplementation in SHR-o decreased BP by approximately 19

Gut microbiota dysbiosis exaggerates ammonia-induced tracheal injury Via TLR4 signaling pathway.

Ammonia is a toxic air pollutant that causes severe respiratory tract injury in animals and humans. Gut microbiota dysbiosis has been found to be involved in the development of respiratory tract injury induced by air pollutants, however, the specific mechanism requires investigation. Here, we found that, inhaled ammonia induced tracheal injury by reducing expression of claudin-1, increasing expression of muc5ac, TLR4, MyD88, NF-κB and cytokines (TNF-α, IL-1β, IL-6 and IL-10), and also altering tracheal microbiota composition. Spearman correlation analysis indicated that gut microbiota dysbiosi

The association between early-life gut microbiota and childhood respiratory diseases: a systematic review.

Data from animal models suggest a role of early-life gut microbiota in lung immune development, and in establishing susceptibility to respiratory infections and asthma in humans. This systematic review summarises the association between infant (ages 0-12 months) gut microbiota composition measured by genomic sequencing, and childhood (ages 0-18 years) respiratory diseases (ie, respiratory infections, wheezing, or asthma). Overall, there was evidence that low α-diversity and relative abundance of particular gut-commensal bacteria genera (Bifidobacterium, Faecalibacterium, Ruminococcus, and Rose

Lacticaseibacillus rhamnosus DACN152 effectively ameliorated letrozole-induced polycystic ovary syndrome: sex hormone homeostasis, microbiota and metabolite profiles.

This study investigated the potential role of Lacticaseibacillus rhamnosus DACN152 (DACN152) in ameliorating polycystic ovary syndrome (PCOS). Using a letrozole-induced PCOS murine model, we demonstrated that DACN152 attenuated ovarian histopathological damage, restored estrous cyclicity, and normalized sex hormone levels. Furthermore, DACN152 enhanced ovarian steroidogenesis by regulating key steroidogenic genes (StAR, CYP17A1, and CYP19A1) and sex hormone receptors (Lhr and Pgr), while concurrently suppressing ovarian apoptosis through modulation of apoptotic regulators (Bax and Bcl-2). Microbial metabolite hyodeoxycholic acid induces macrophage immunometabolic reprogramming and alleviates ulcerative colitis.

Host-gut microbiota metabolic interactions are implicated in the pathogenesis of ulcerative colitis (UC), but the underlying mechanisms of certain metabolites remain ambiguous. Here, we revealed an impaired bile acid homeostasis in UC with a significant deficiency of hyodeoxycholic acid (HDCA), which was inversely correlated with the severity of UC. Ruminococcus callidus was linked to altered HDCA generation, and colonization of R. callidus increased HDCA concentrations via bile salt hydrolase. Single-cell RNA sequencing (ScRNA-seq) indicated that HDCA reshaped the intestinal macrophage landsc

Polypeptides synthesized by common bacteria in the human gut improve rodent metabolism.

The human gut microbiota has the potential to synthesize proteins that may influence host metabolism. Here we report two polypeptides, RUMTOR-derived peptide (RORDEP) 1 and RORDEP2, circulating in human blood and synthesized by specific strains of gut commensal Ruminococcus torques that correlate inversely with adiposity in humans. Oral gavage with RORDEP-expressing strains improved glucose tolerance, increased bone density and reduced fat mass with an enhanced expression of genes and proteins involved in thermogenesis and lipolysis in lean mice on a high-fat diet and diet-induced obese mice.

Fructus Mori polysaccharides modulate the axial distribution of gut microbiota and fecal metabolites to improve symptoms of hyperglycemia in type 2 diabetic mice.

To understand the gut microbiota composition in different intestinal segments is essential to clarify the structure-hypoglycemic relationship of Fructus Mori Polysaccharides (MFP). In this study, the spatial distribution of gut microbiota and fecal metabolites affected by MFP in type 2 diabetes mice was investigated using the 16S rRNA high-throughput sequencing technology and gas chromatography. The results showed that MFP could control body weight and reduce the blood glucose level. After MFP intervention, the abundance of Lactobacillus and Bifidobacterium increased by 3.43 and 33.47 times in

Detection of cell-free microbial DNA using a contaminant-controlled analysis framework.

BACKGROUND: The human microbiome plays an important role in cancer. Accumulating evidence indicates that commensal microbiome-derived DNA may be represented in minute quantities in the cell-free DNA of human blood and could possibly be harnessed as a new cancer biomarker. However, there has been limited use of rigorous experimental controls to account for contamination, which invariably affects low-biomass microbiome studies. RESULTS: We apply a combination of 16S-rRNA-gene sequencing and droplet digital PCR to determine if the specific detection of cell-free microbial DNA (cfmDNA) is possibl

Modulation of the fecal bile acid profile by gut microbiota in cirrhosis.

BACKGROUND & AIMS: The 7α-dehydroxylation of primary bile acids (BAs), chenodeoxycholic (CDCA) and cholic acid (CA) into the secondary BAs, lithocholic (LCA) and deoxycholic acid (DCA), is a key function of the gut microbiota. We aimed at studying the linkage between fecal BAs and gut microbiota in cirrhosis since this could help understand cirrhosis progression. METHODS: Fecal microbiota were analyzed by culture-independent multitagged-pyrosequencing, fecal BAs using HPLC and serum BAs using LC-MS in controls, early (Child A) and advanced cirrhotics (Child B/C). A subgroup of early cirrhotic

Multi-omics reveals that rumen fluid transfer affects the immune function of Xizang sheep by regulating enterohepatic metabolic homeostasis.

The gut microbiota plays a crucial role in immune regulation and metabolic homeostasis in ruminants; however, the systemic effects of rumen fluid transfer (RFT) on the gut-liver axis remain unclear. In this study, Xizang sheep were used as the animal model to investigate the effects of RFT using multi-omics approaches, including transcriptomics, metabolomics, and 16S rRNA sequencing. The results showed that RFT was associated with changes in immune- and metabolism-related genes (e.g., TLR4, NOD2, PPARA, and ABCG2) and alterations in several key metabolites, including tryptophan-related compoun
