Understanding the dynamic behavior of wild-type and mutant LARS1 proteins: insight from comparative molecular dynamics simulations.

Leucyl-tRNA synthetase 1 (LARS1) is a multi-domain enzyme essential for translational fidelity and amino acid sensing, with its anticodon-binding domain (ABD) playing a critical role in tRNA recognition. Missense variants in LARS1 have been associated with infantile liver failure syndrome type 1 (ILFS1), yet the structural mechanisms underlying their functional impact remain poorly understood. In this study, we investigated the structural and dynamical effects of the patient derived D794G mutation located in the anticodon-binding domain of LARS1. To evaluate its potential pathogenicity, compar

Rational design of k-casein peptides to modulate GSK-3B dynamics for Alzheimer's therapy.

GSK-3β is an important therapeutic target in Alzheimer's disease due to its central role in tau hyperphosphorylation, and synaptic dysfunction. In this study, a κ-casein-derived peptide (LALTLPFLGA) was identified via HADDOCK and introduced to MD simulations for MM/PBSA per-residue analysis, revealing four unfavorable mutation sites (L12, L14, T15, F18). Using MCSM and ΔΔG-based substitution, a 48-peptide library was generated; 22 candidates satisfied toxicity/allergenicity filters. Docking and MD simulations prioritized four leads-PEP8, PEP36, PEP40, and PEP44-with PEP8 (- 89.1 kcal·mol⁻¹) an

In silico evaluation of bioactive compounds as dual inhibitors targeting ALK and EGFR for non-small cell lung cancer therapy.

Non-Small Cell Lung Cancer (NSCLC) is the most common type of lung cancer, accounting for around 85% of all lung cancer cases. Limitations of current therapeutics, including inefficient pharmacokinetics, poor selectivity, resistance development, and a lack of dual inhibitors, highlight the need for new drugs with improved efficacy and biocompatibility. This study used a comprehensive in silico approach against two major NSCLC targets, anaplastic lymphoma kinase (ALK) and epidermal growth factor receptor (EGFR), to identify potential dual inhibitors. After virtual screening of more than 12,200

Structural and dynamic characterization of MirA-like xenosiderophore transporters in Fusarium: in silico modeling and in vitro validation.

Fusarium, an omnipresent fungus is known for its pathogenic potential. Pathogenic fungi like all other organisms need iron and are known to produce siderophores for iron acquisition. Siderophore producers possess transporters for their own siderophore as well as for siderophores secreted by other organisms known as xenosiderophore. Although iron is an essential nutrient it can also cause oxidative stress and cell-death at high concentrations. While siderophore utilization is employed by the fungi for efficient iron acquisition, it can be exploited against them by supplying xenosiderophore exog

Antiplatelet therapy through inhibition of P2Y12 and phosphodiesterase receptors by novel synthesis 1,3-dicyclohexylpyrimidine-2,4(1H,3H)-dione derivatives with computational evaluation.

In this study, we report the synthesis of a new class of 1,3-dicyclohexylpyrimidine-2,4-dione derivatives with potential antiplatelet activity. The novelty of this work lies in the development of unprecedented pyrimidine-based scaffolds, prepared through simple synthetic routes, and their evaluation as dual inhibitors of P2Y12 and phosphodiesterase 5 receptors. Biological assays demonstrated that compound 10 was the most active P2Y12 inhibitor (IC50 = 0.271 ± 0.009 μg/ml), surpassing reference drug clopidogrel (IC50 = 0.327 ± 0.011 μg/ml). Compound 10 also exhibited potent PDE5 inhibition, usi

In-silico screening to identify phytochemical inhibitor for hP2X7: A crucial inflammatory cell death mediator in Parkinson's disease.

The second most prevalent neurological disease among the elderly is Parkinson's disease, where neuroinflammation plays a significant role in its pathology. Purinergic signaling mediated by P2X7 plays a significant role in neuroinflammation and pyroptotic cell death pathways through mediators like NLRP3, Caspase-1, and Caspase-3, instigating pyroptotic cell death. No synthetic agent advanced in late-stage clinical trials due to their inefficacy and toxicity. Hence, in this study, we aimed to identify a phytoconstituent inhibitor against the hP2X7 receptor to ameliorate the inflammatory processe

Computational approach towards repurposing of FDA approved drug molecules: strategy to combat antibiotic resistance conferred by Pseudomonas aeruginosa.

Antimicrobial resistance is recognized as a major worldwide public health dilemma in the current century. Pseudomonas aeruginosa, a Gram-negative opportunistic pathogen, causes nosocomial infections like respiratory tract infections, urinary tract infections, dermatitis, and cystic fibrosis. It manifests antibiotic resistance via intrinsic, acquired, and adaptive pathways, where efflux pumps function in the extrusion of antibiotics from the cell. MexB protein, part of the tripartite efflux pumps MexAB-OprM present in P.aeruginosa, expels the penems and β-lactam antibiotics, thereby enhancing P

Organotin Antifouling Compounds and Sex-Steroid Nuclear Receptor Perturbation: Some Structural Insights.

Organotin compounds (OTCs) are a commercially important group of organometallic compounds of tin used globally as polyvinyl chloride stabilizers and marine antifouling biocides. Worldwide use of OTCs has resulted in their ubiquitous presence in ecosystems across all the continents. OTCs have metabolic and endocrine disrupting effects in marine and terrestrial organisms. Thus, harmful OTCs (tributyltin) have been banned by the International Convention on the Control of Harmful Antifouling Systems since 2008. However, continued manufacturing by non-member countries poses a substantial risk for a
