Contaminant-induced immunotoxicity in harbour seals: wildlife at risk?

Persistent, lipophilic polyhalogenated aromatic hydrocarbons (PHAHs) accumulate readily in the aquatic food chain and are found in high concentrations in seals and other marine mammals. Recent mass mortalities among several marine mammal populations have been attributed to infection by morbilliviruses, but a contributing role for immunotoxic PHAHs, including the polychlorinated biphenyls (PCBs), polychlorinated dibenzo-p-dioxins (PCDDs), and polychlorinated dibenzofurans (PCDFs) was not ruled out. We addressed this issue by carrying out a semi-field study in which captive harbour seals were fe

Disease conditions affecting common seals (Phoca vitulina) around the Scottish mainland, September-November 1988.

Common seal populations around the Scottish coast were less severly devastated by phocine distemper than those on the Continent. Nevertheless, over a 4-month period, 59 common seals were examined postmortem. The macroscopic and histopathological lesions are described. Forty-two per cent of these seals were considered to be suffering from phocine distemper. Serology on 29 of the 59 seals was undertaken and the results are discussed in relation to the histopathological findings. Bordetella bronchiseptica proved to be an important secondary invader in phocine distemper virus infected seals. Vermi

The pineal gland in newborn southern elephant seals, Mirounga leonina.

In the newborn southern elephant seal the pineal gland is very large, and both pineal and plasma melatonin concentration is elevated. The pineal gland was investigated during the first 24 h, and up to 20 days of age, in elephant seal pups. A primary aim of this investigation was to determine whether there are obvious ultrastructural characteristics of pinealocytes that are exhibiting extraordinarily high levels of activity. Blood and pineal glands were collected from thirty seven pups of known age which were sampled at random from early September to early November (1985) at Macquarie Island. T

Lethal Encephalitis in Seals with Japanese Encephalitis Virus Infection, China, 2017.

We isolated Japanese encephalitis virus (JEV) from brain samples of 2 seals with lethal encephalitis at Weihai Aquarium, Weihai, China, in 2017. We confirmed our findings by immunohistochemical staining and electron microscopy. Phylogenetic analysis showed this virus was genotype I. Our findings suggest that JEV might disseminate though infected zoo animals.

Entrance and survival of Brucella pinnipedialis hooded seal strain in human macrophages and epithelial cells.

Marine mammal Brucella spp. have been isolated from pinnipeds (B. pinnipedialis) and cetaceans (B. ceti) from around the world. Although the zoonotic potential of marine mammal brucellae is largely unknown, reports of human disease exist. There are few studies of the mechanisms of bacterial intracellular invasion and multiplication involving the marine mammal Brucella spp. We examined the infective capacity of two genetically different B. pinnipedialis strains (reference strain; NTCT 12890 and a hooded seal isolate; B17) by measuring the ability of the bacteria to enter and replicate in cultur

Auditory masking in three pinnipeds: aerial critical ratios and direct critical bandwidth measurements.

This study expands the limited understanding of pinniped aerial auditory masking and includes measurements at some of the relatively low frequencies predominant in many pinniped vocalizations. Behavioral techniques were used to obtain aerial critical ratios (CRs) within a hemianechoic chamber for a northern elephant seal (Mirounga angustirostris), a harbor seal (Phoca vitulina), and a California sea lion (Zalophus californianus). Simultaneous, octave-band noise maskers centered at seven test frequencies (0.2-8.0 kHz) were used to determine aerial CRs. Narrower and variable bandwidth masking no

Lipopolysaccharide heterogeneity in Brucella strains isolated from marine mammals.

Smooth lipopolysaccharides (S-LPSs) from Brucella strains isolated from seals, dolphins, porpoises, an otter and a minke whale were characterized by ELISA using monoclonal antibodies (mAbs) directed against seven previously defined O-polysaccharide (O-PS) epitopes and by Western blot after SDS-PAGE. All strains studied were A-dominant as shown by specific polyclonal sera and this was also confirmed by the mAbs. However, binding patterns in ELISA of mAbs to the specific common (C) epitopes were rather heterogeneous, and for some strains, such as those isolated from striped dolphins, binding of

Do descending influences alternate to produce episodic breathing?

This study examines the episodic breathing patterns of three disparate groups of vertebrates. In an in vitro bullfrog brainstem-spinal cord preparation, episodic breathing was replaced by uniformly spaced breaths following transection caudal to the optic chiasma. The same effect was produced in hibernating squirrels by inhalation of mild anesthesia. Preliminary data suggest that a similar conversion is also produced in hibernating squirrels by vagotomy, in conjunction with blockade of central NMDA-type glutamate receptors. In all cases, even though overall breathing frequency increased, due to

Adaptations to prolonged fasting in free-living weaned gray seal pups.

The water and energy metabolism of 12 free-living gray seal pups during their postweaning fast was investigated using tritiated water for determination of rates of energy expenditure and water turnover and changes in body composition. Changes in serum composition and urinary output were also measured. The duration of the postweaning fast ranged from 10 to greater than 28 days. The mean daily energy expenditure of pups was 12.0 +/- 1.0 (SE) MJ, equivalent to 2.7 times the basal metabolic rate predicted from M. Kleiber (The Fire of Life. Huntington, NY: Krieger, 1975). On average, lipid cataboli

Far away from home: gastrointestinal parasites trace the journey of a grey seal (Halichoerus grypus) from the Wadden Sea to the Mediterranean Sea.

Grey seals, Halichoerus grypus (GSs), inhabit cold temperate and subarctic waters along the North Atlantic Ocean. Individuals of GS can regularly disperse towards southern areas (ca. 38°N-39°N) but occurrence at lower latitudes is exceptional. On 18 February 2022, a 217-cm-long male of GS was detected in waters off the SW Atlantic coast of Spain (37°N), then entered the western Mediterranean Sea and wandered for 15 days until he died. Here, we use gastrointestinal parasites to investigate the geographical origin of the GS and the length of the journey towards Mediterranean waters. Seven helmin

Use of stable isotope ratio analysis to investigate the biology and clinical significance of seal parasites.

Stranded seals are often infected with a broad range of parasites, although whether they are the cause of significant morbidity or an incidental finding is usually unclear. In this study we used stable isotope ratio analysis, a method frequently used to investigate food webs, to explore the extent to which common seal parasites feed directly on host tissue and fluids or absorb host-derived metabolites, which in turn may give an indication of their potential impact on the host's health. The trophic discrimination factor Δ15N for the nasal mite, Halarachne halichoeri, was mostly positive, rangin

Dynamics of mercury stable isotope compounds in Arctic seals: New insights from a controlled feeding trial on hooded seals Cystophora cristata.

Accurate interpretation of mercury (Hg) isotopic data requires the consideration of several biotic factors such as age, diet, geographical range, and tissue metabolic turnover. A priori knowledge of prey-predator isotopic incorporation rates and Hg biomagnification is essential. This study aims to assess Hg stable isotopes incorporation in an Arctic species of Phocidae, the hooded seal Cystophora cristata, kept in human care for 24 months (2012-2014) and fed on a constant diet of Norwegian Spring Spawning herring Clupea harengus. We measured THg, MMHg and iHg levels, as well as Hg stable isoto

Male grey seal commits fatal sexual interaction with adult female harbour seals in the German Wadden Sea.

Males of several seal species are known to show aggressive copulating behaviour, which can lead to injuries to or suffocation of females. In the North Sea, grey seal predation on harbour seals including sexual harassment is documented and represents violent interspecific interaction. In this case series, we report pathological and molecular/genetic findings of 11 adult female harbour seals which were found dead in Schleswig-Holstein, Germany, within 41 days. Several organs of all animals showed haemorrhages and high loads of bacteria, indicating their septic spread. All females were pregnant o

Patterns of age-related change in reproductive effort differ in the pre-natal and post-natal periods in a long-lived mammal.

Age-related changes in maternal reproductive allocation for long-lived species are a key prediction from life-history theory. Theoretical and empirical work suggests that allocation may increase with age due to constraint (increases with experience) or restraint (increases with age in the face of declining residual reproductive value), and may decrease among the oldest aged animals due to senescence in reproductive function. Here, we use a hierarchical modelling approach to investigate the age-related patterns of change in maternal reproductive effort in the Weddell seal, a long-lived marine m

Morbillivirus in common seals stranded on the coasts of Belgium and northern France during summer 1998.

Sixteen common seals (Phoca vitulina) were stranded on the Belgian and northern French coasts during the summer of 1998. Eleven (10 pups and one adult) were sampled for histopathological, immunohistochemical, serological, bacteriological, parasitological and virological investigations. The main gross findings were severe emaciation, acute haemorrhagic enteritis, acute pneumonia, interstitial pulmonary emphysema and oedema, and chronic ulcerative stomatitis. Microscopical lung findings were acute to subacute pneumonia with interstitial oedema and emphysema. Severe lymphocytic depletion was obse

Seroconversion and abortion in cattle experimentally infected with Brucella sp. isolated from a Pacific harbor seal (Phoca vitulina richardsi).

Previously unrecognized Brucella species have been isolated from a number of marine mammals, including harbor seals (Phoca vitulina richardsi) in the Puget Sound area of the state of Washington. Because of the presence of dairy herds in proximity to the harbor seal populations, a study was conducted to determine the effects of the harbor seal Brucella isolate in experimentally inoculated cattle. Six pregnant cattle were exposed by intravenous injection (n = 3) or intraconjunctival inoculation (n = 3). Two pregnant cows were intravenously injected with saline and served as controls. [Use of recording methods and sound analysis of cough in the study of sound phenomena associated with respiration].

The objective registration of the human body functions is one of the main tasks of the modern and prospective medicine. The registration of the heart, brain, muscles etc. activity have a long tradition. The registration of sound processes, for instance coughing was not solved completely despite their diagnostic importance. The authors worked out a new non invasive and precise method for cough sound registration and analysis which allows evaluation of the sound pattern, intensity and time duration. Application of this method for registration and analysis of the cry of new-borns gave rise to dou

Experimental Challenge of Atlantic Cod (Gadus morhua) with a Brucella pinnipedialis Strain from Hooded Seal (Cystophora cristata).

Pathology has not been observed in true seals infected with Brucella pinnipedialis. A lack of intracellular survival and multiplication of B. pinnipedialis in hooded seal (Cystophora cristata) macrophages in vitro indicates a lack of chronic infection in hooded seals. Both epidemiology and bacteriological patterns in the hooded seal point to a transient infection of environmental origin, possibly through the food chain. To analyse the potential role of fish in the transmission of B. pinnipedialis, Atlantic cod (Gadus morhua) were injected intraperitoneally with 7.5 x 107 bacteria of a hooded s

A new multi-host species indirect ELISA using protein A/G conjugate for detection of anti-Toxoplasma gondii IgG antibodies with comparison to ELISA-IgG, agglutination assay and Western blot.

Toxoplasma gondii is a zoonotic protozoan parasite which can cause significant disease and losses in livestock and wild animals. It is increasingly recognized as an important foodborne pathogen in a broad range of food animals and products. Effective control strategies require rapid, reliable and cost-effective detection methods for large scale surveys and diagnostic applications in a broad range of warm-blooded animals. To overcome one or more of these shortcomings in the currently available detection methods for T. gondii infection a non-species-specific protein A/G conjugate was used in the
