Determining the Advantages, Costs, and Trade-Offs of a Novel Sodium Channel Mutation in the Copepod Acartia hudsonica to Paralytic Shellfish Toxins (PST).

The marine copepod Acartia hudsonica was shown to be adapted to dinoflagellate prey, Alexandrium fundyense, which produce paralytic shellfish toxins (PST). Adaptation to PSTs in other organisms is caused by a mutation in the sodium channel. Recently, a mutation in the sodium channel in A. hudsonica was found. In this study, we rigorously tested for advantages, costs, and trade-offs associated with the mutant isoform of A. hudsonica under toxic and non-toxic conditions. We combined fitness with wild-type: mutant isoform ratio measurements on the same individual copepod to test our hypotheses. A

Current decontamination challenges and potentially complementary solutions to safeguard the vulnerable seafood industry from recalcitrant human norovirus in live shellfish: Quo Vadis?

Safeguarding the seafood industry is important given its contribution to supporting our growing global population. However, shellfish are filter feeders that bioaccumulate microbial contaminants in their tissue from wastewater discharged into the same coastal growing environments leading to significant human disease outbreaks unless appropriately mitigated. Removal or inactivation of enteric viruses is very challenging particularly as human norovirus (hNoV) binds to specific histo-blood ligands in live oyster tissue that are consumed raw or lightly cooked. The regulatory framework that sets ou

Tracking marine phycotoxins: spatial distribution and source elucidation of domoic acid and lipophilic shellfish toxins in the Beibu Gulf, South China Sea.

Harmful algal blooms (HABs) pose escalating threats to marine ecosystems and seafood safety worldwide, particularly in nearshore waters. The Beibu Gulf, located in the northwestern South China Sea, supports rich biodiversity and serves as a major fishing area. To comprehensively understand the current status of phycotoxin pollution, we conducted a basin-wide cruise survey in August 2021, collecting 60 phytoplankton samples to characterize the spatial distribution, compositional profile, and molecular origins of 12 target marine phycotoxins, along with their key environmental drivers. Domoic ac

Occurrence of potentially toxic microalgae and diarrhetic shellfish toxins in the digestive tracts of green sea turtles (Chelonia mydas) from southern Brazil.

Algal toxins are involved in the mortality and/or illness of marine organisms via consumption of contaminated prey, or upon direct exposure to toxic cells. In this study, the presence of potentially toxic microalgal cells was investigated within the digestive tract contents of a threatened species of green turtle (Chelonia mydas). Additionally, lipophilic toxins were determined by LC-MS/MS in tissue samples (liver, stomach and/or intestine) of selected animals (n = 39 individuals) found dead-stranded in southern Brazil, from winter/2015 to autumn/2016. Thirteen potentially toxic species of mic

Rapid and sensitive detection of domoic acid in shellfish using a magnetic bead-based competitive ELISA with a high-affinity peptide as a molecular binder.

Addressing the critical health concerns posed by domoic acid (DA), a neurotoxic compound produced by toxic marine algae and bioaccumulated in shellfish, necessitates the development of a rapid, precise, and robust detection system. Traditional DA detection methods have stability and sensitivity issues, which hinder effective toxin detection. To overcome these limitations, we developed a novel direct competitive enzyme-linked immunosorbent assay (dc-ELISA) platform that utilizes peptide-immobilized magnetic beads (MGBs/peptide). The affinity peptides identified through phage display and chemica

A Sub-Acute Dosing Study of Saxitoxin and Tetrodotoxin Mixtures in Mice Suggests That the Current Paralytic Shellfish Toxin Regulatory Limit Is Fit for Purpose.

Paralytic shellfish poisoning is a worldwide problem induced by shellfish contaminated with paralytic shellfish toxins. To protect human health, a regulatory limit for these toxins in shellfish flesh has been adopted by many countries. In a recent study, mice were dosed with saxitoxin and tetrodotoxin mixtures daily for 28 days showing toxicity at low concentrations, which appeared to be at odds with other work. To further investigate this reported toxicity, we dosed groups of mice with saxitoxin and tetrodotoxin mixtures daily for 21 days. In contrast to the previous study, no effects on mous

Simultaneous determination of 12 lipophilic shellfish toxins in human plasma, serum, and urine using UPLC-MS/MS combined with PRiME HLB μElution platform.

Lipophilic shellfish toxins (LSTs) pose significant health risks through contaminated seafood consumption, necessitating robust monitoring methods, which is critical for early and definitive diagnosis. However, trace analysis of LSTs in human biological fluids remains challenging due to complex matrices and ultra-low concentrations. For this purpose, a rapid, sensitive, and high-throughput UPLC-MS/MS method was developed and validated for the simultaneous determination of 12 LSTs in human plasma, serum, and urine using a μElution 96-well plate for sample preparation. Use of two-dimensional gel electrophoresis to differentiate morphospecies of Alexandrium minutum, a paralytic shellfish poisoning toxin-producing dinoflagellate of harmful algal blooms.

Contamination of shellfish with paralytic shellfish poisoning toxins (PST) produced by toxic harmful algal blooms (HABs) have been negatively affecting the shellfish and aquaculture industries worldwide. Therefore, accurate and early identification of toxic phytoplankton species is crucial in HABs surveillance programs that allow fish-farmers to take appropriate preventive measures in shellfish harvesting and other aquaculture activities to overcome the negative impacts of HABs on human health. The identification of toxic dinoflagellates present in the water is currently a time-consuming opera

Contrasted occurrence and temporal trends of legacy and alternative flame retardants in marine shellfish from the French coasts over 10 years of monitoring.

Following regulations on flame retardant usage during the last few decades, alternative flame retardants (a-FRs) have been introduced to the market, increasing the diversity and quantities of FR use, and raising concerns about their occurrence in the marine environment. In this study, both legacy and alternative brominated and chlorinated FRs were determined in filter-feeding shellfish over a ten-year period (2012-2022) at 20 sites located on the French coastline. The studied contaminants included some legacy FRs, such as polybrominated diphenylethers (PBDEs), hexabromobenzene (HBB), hexabromo

Detection, occurrence and monthly variations of typical lipophilic marine toxins associated with diarrhetic shellfish poisoning in the coastal seawater of Qingdao City, China.

In recent years, related research has mainly examined lipophilic marine toxins (LMTs) in contaminated bivalves or toxic algae, whereas the levels of LMTs in seawater remain largely unexplored. Okadaic acid (OA), yessotoxin (YTX), and pectenotoxin-2 (PTX2) are three typical LMTs produced by certain marine algae that are closely linked to diarrhetic shellfish poisoning. In this study, a new method of solid phase extraction combined with liquid chromatography - electrospray ionization ion trap tandem mass spectrometry was developed to determine the presence of OA, YTX, and PTX2 in seawater simult

Arsenic, cadmium, and manganese levels in shellfish from Map Ta Phut, an industrial area in Thailand, and the potential toxic effects on human cells.

Map Ta Phut Industrial Estate is a major industrial area in Thailand for both petrochemical and heavy industries. The release of hazardous wastes and other pollutants from these industries increases the potential for contamination in foods in the surrounding area, especially farmed shellfish. This study determined the arsenic (As), cadmium (Cd), and manganese (Mn) concentrations in the edible flesh of farmed shellfish, including Perna viridis, Meretrix meretrix, and Scapharca inaequivalvis, around the Map Ta Phut area using inductively coupled plasma mass spectrometry. The results showed that

Developing a detection strategy for ten paralytic shellfish poisonings in urine, combining high-throughput DESI-MS screening and accurate UPLC-QqQ/MS quantification.

Paralytic shellfish poisoning (PSP) is the most widespread and harmful form of shellfish poisoning with high mortality rate. In this study, a combined desorption electrospray ionization mass spectrometry (DESI-MS) and ultra-performance liquid chromatography triple quadrupole mass spectrometry (UPLC-QqQ/MS) method was established for the detection of PSPs in urine. The method was optimized using a spray solution of methanol and water (1:1, v/v) containing 0.1 % FA, at a flow rate of 2.5 µL·min-1 and an applied voltage of 3 kV. The limit of detection (LOD) for PSPs detection by DESI-MS was in th

Rapid and sensitive detection of noroviruses by using TaqMan-based one-step reverse transcription-PCR assays and application to naturally contaminated shellfish samples.

Noroviruses (NoV), which are members of the family Caliciviridae, are the most important cause of outbreaks of acute gastroenteritis worldwide and are commonly found in shellfish grown in polluted waters. In the present study, we developed broadly reactive one-step TaqMan reverse transcription (RT)-PCR assays for the detection of genogroup I (GI) and GII NoV in fecal samples, as well as shellfish samples. The specificity and sensitivity of all steps of the assays were systematically evaluated, and in the final format, the monoplex assays were validated by using RNA extracted from a panel of 84

Development of fluorescence sensor and test paper based on molecularly imprinted carbon quantum dots for spiked detection of domoic acid in shellfish and lake water.

Excessive levels of domoic acid (DA) in edible shellfish and water can seriously threaten human health and even cause death. Herein, we have developed a fluorescence sensor and test paper based on molecularly imprinted carbon quantum dots (B-CDs@MIPs) for DA analysis. In this research, the prepared carbon quantum dots (B-CDs) with good optical properties were used as the sensitive fluorescent signal probes, and the molecularly imprinted polymers (MIPs) instead of biological antibodies were employed as the specific recognition components because of their high stability and low cost. Due to the

A systematic review and meta-analysis indicates a substantial burden of human noroviruses in shellfish worldwide, with GII.4 and GII.2 being the predominant genotypes.

Human noroviruses (HuNoVs) have been found as the leading cause of acute gastroenteritis outbreaks in all age groups and are significantly correlated with the consumption of shellfish. In this study, the contamination of HuNoVs in shellfish was estimated through a systematic review and meta-analysis. Studies on the contamination of HuNoVs in shellfish were searched from PubMed, Web of Science, Embase, and Cochrane Library from January 2000 to August 2021. A total of 75 studies were included, and the pooled HuNoVs prevalence in shellfish was 29% (95% CI: 23-35) worldwide. As revealed by the res

The value of toxin profiles in the chemotaxonomic analysis of paralytic shellfish toxins in determining the relationship between British Alexandrium spp. and experimentally contaminated Mytilus sp.

Although phytoplankton is ubiquitous in the world's oceans some species can produce compounds that cause damaging effects in other organisms. These include the toxins responsible for paralytic shellfish poisoning, which, in UK waters, are produced by dinoflagellates from the Alexandrium genus. Within Great Britain (GB) a monitoring programme exists to detect this harmful genus as well as the Paralytic Shellfish Poisoning (PSP) toxins in the flesh of shellfish from classified production areas. The techniques used for toxin analysis allow for detailed analysis of the toxin profiles present in co

Implication of brevetoxin B1 and PbTx-3 in neurotoxic shellfish poisoning in New Zealand by isolation and quantitative determination with liquid chromatography-tandem mass spectrometry.

Brevetoxin B1 (BTX-B1) was isolated from Austrovenus stutchburyi following the 1992-1993 outbreak of neurotoxic shellfish poisoning (NSP) in New Zealand. We report here the first isolation of PbTx-3 from the same shellfish and the development of a procedure for quantitative determination of PbTx-3 and BTX-B1. PbTx-3 was isolated by chromatography on columns of SiO2, ODS, and LH-20, followed by reverse-phase HPLCs. In mass spectrometry (MS) with an electrospray ionization (ESI) interface operating in the positive or negative ion mode, the abundant protonated ion [M+H]+ of PbTx-3 (m/z 897) and t

Pulsed ultraviolet (PUV) disinfection of artificially contaminated seawater seeded with high levels of pathogen disease indicators as an alternative for the shellfish industry depuration systems.

The increase in pathogen levels in seawater threatens the safety of entire aquatic ecosystems. Foodborne pathogens can potentially accumulate in shellfish, especially in filter feeders such as bivalves, requiring an efficient depuration process before consumption. Alternative approaches to promote a cost-efficient purge at depuration plants are urgently needed. A small prototype pulsed ultraviolet (PUV) light recirculation system was designed, and its depuration potential was tested in a seawater matrix artificially contaminated with high levels of microbial pathogens Escherichia coli, Staphyl

[Non-industrial labor, internal conditions and repetitive strain injury: a case study in a shellfish-rearing community on Maré Island, State of Bahia, Brazil].

Non-industrial labor is still widespread in Brazil. An appropriate approach ought to consider different social specificities from those of salaried-work relations. The scope was to examine non-industrial work practices, specifically those of extractive shellfishing, and their relation to health conditions in a small-scale fishing community. It focused on the cadences and time contingencies, as well as other working conditions, related to the development of ailments such as repetitive strain injury -RSI.
