Borrelia burgdorferi, the causative agent of Lyme disease, is the most common vector-borne disease in the United States. Compared with other bacterial pathogens, B. burgdorferi has many unique features. For instance, its highly segmented genome was predicted to encode very few proteins directly dedicated to gene expression regulation. Yet, the spirochete continuously reprograms its transcriptome and proteome to promote survival and pathogenesis as spirochetes traverse the enzootic lifecycle between ticks and mammals. Signal sensing systems, a unique alternative sigma factor cascade, and multi-

Bacillus thuringiensis and other members of the Bacillus cereus family are resistant to many β-lactams. Resistance is dependent upon the extracytoplasmic function sigma factor σP. We used label-free quantitative proteomics to identify proteins whose expression was dependent upon σP. We compared the protein profiles of strains which either lacked σP or overexpressed σP. We identified 8 members of the σP regulon which included four β-lactamases as well as three penicillin-binding proteins (PBPs). Using transcriptional reporters, we confirmed that these genes are induced by β-lactams in a σP-depe

Oxidative stress causes damage to nucleic acids, membrane lipids and proteins. One striking effect is the metal-catalyzed, site-specific carbonylation of proteins. In the gram-positive soil bacterium Bacillus subtilis, the PerR-dependent specific stress response and the sigmaB-dependent general stress response act together to make cells more resistant to oxidative stress. In this study, we analyzed the carbonylation of cytoplasmic proteins in response to hydrogen peroxide stress in B. subtilis. Furthermore, we asked whether the sigmaB-dependent response to oxidative stress also confers protect

Hallmarks of the developmental cycle of the obligate intracellular pathogenic bacterium Chlamydia are the primary differentiation of the infectious elementary body (EB) into the proliferative reticulate body (RB) and the secondary differentiation of RBs back into EBs. The mechanisms regulating these transitions remain unclear. In this report, we developed an effective novel strategy termed dependence on plasmid-mediated expression (DOPE) that allows for the knockdown of essential genes in Chlamydia. We demonstrate that GrgA, a Chlamydia-specific transcription factor, is essential for the secon

BACKGROUND: Since the RpoN-RpoS regulatory network was revealed in the Lyme disease spirochete Borrelia burgdorferi a decade ago, both upstream and downstream of the pathway have been intensively investigated. While significant progress has been made into understanding of how the network is regulated, most notably, discovering a relationship of the network with Rrp2 and BosR, only three crucial virulence factors, including outer surface protein C (OspC) and decorin-binding proteins (Dbps) A and B, are associated with the pathway. Moreover, for more than 10 years no single RpoS-controlled gene

UNLABELLED: Elongation factor P (EF-P) is conserved across the three kingdoms and alleviates ribosome stalling at the XPPX motif of a protein. A genome-wide ribosome-stalling atlas was previously created for the ancestral strain NCIB3610. However, reportedly, ribosome-stalling does not always decrease protein levels. To determine changes in protein levels in the efp mutant, we performed a proteomic analysis of the laboratory strain-based efp mutant. To evaluate its effects on translation, the amount of protein per mRNA molecule must be determined. Therefore, we performed the RNA-seq experiment

The opportunistic pathogen Legionella pneumophila alternates between two states: replication within phagocytes and transmission between host amoebae or macrophages. In broth cultures that model this life cycle, during the replication period, CsrA inhibits expression of transmission traits. When nutrients become limiting, the alarmone (p)ppGpp accumulates and the sigma factors RpoS and FliA and the positive activators LetA/S and LetE promote differentiation to the transmissible form. Here we show that when cells enter the postexponential growth phase, RpoS increases expression of the transmissi

A newly described coronavirus named severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), which is the causative agent of coronavirus disease 2019 (COVID-19), has infected over 2.3 million people, led to the death of more than 160,000 individuals and caused worldwide social and economic disruption1,2. There are no antiviral drugs with proven clinical efficacy for the treatment of COVID-19, nor are there any vaccines that prevent infection with SARS-CoV-2, and efforts to develop drugs and vaccines are hampered by the limited knowledge of the molecular details of how SARS-CoV-2 infects c

Bacillus anthracis, a rod-shaped, spore-forming bacterium, is the causative agent of anthrax. The life cycle of B. anthracis involves sporulation and germination processes that are precisely regulated by distinct sigma factors and associated proteins. Its pathogenicity is primarily attributed to a tripartite toxin consisting of lethal factor (LF), edema factor (EF) and protective antigen (PA), as well as an antiphagocytic capsule of Poly-γ-D-Glutamate. The virulence of B. anthracis is further regulated by various post-translational modifications (PTMs), including protein phosphorylation, acety

Color instability in anterior composite restorations remains a primary cause for clinical replacement. While pre-polymerization thermal modification is increasingly utilized in daily practice, its precise impact on phase-specific and long-term optical resistance against continuous thermal aging and aggressive dietary staining remains unclarified. This in vitro study evaluated the impact of pre-polymerization thermal modification (4 °C, 23 °C, and 55 °C) on the phase-specific and cumulative color stability (ΔE00) of four anterior composite resins subjected to sequential thermal aging and prolon

Leptospira are emerging zoonotic pathogens transmitted from animals to humans typically through contaminated environmental sources of water and soil. Regulatory pathways of pathogenic Leptospira spp. underlying the adaptive response to different hosts and environmental conditions remains elusive. In this study, we provide the first global Transcriptional Start Site (TSS) map of a Leptospira species. RNA was obtained from the pathogen Leptospira interrogans grown at 30°C (optimal in vitro temperature) and 37°C (host temperature) and selectively enriched for 5' ends of native transcripts. A tota

BACKGROUND: Respiratory motion remains a major challenge in radiotherapy for non-small cell lung cancer (NSCLC), often requiring a balance between target coverage and normal tissue sparing.
INTRODUCTION: This study aimed to develop and validate a clinically implementable, individualized PTV margin strategy by integrating principal component analysis (PCA) with key clinical predictors.
METHODS: A cohort of 61 NSCLC patients (Stage III) underwent 4DCT simulation. 4DCT datasets were used both for motion modeling and as a reference planning approach (ITV-based). The proposed method was compared

Methanotrophs require copper for their activity as it plays a critical role in the oxidation of methane to methanol. To sequester copper, some methanotrophs secrete a copper-binding compound termed methanobactin (MB). MB, after binding copper, is reinternalized via a specific outer membrane TonB-dependent transporter (TBDT). Methylosinus trichosporium OB3b has two such TBDTs (MbnT1 and MbnT2) that enable M. trichosporium OB3b to take up not only its own MB (MB-OB3b) but also heterologous MB produced from other methanotrophs, e.g., MB of Methylocystis sp. strain SB2 (MB-SB2). Here, we show that

Tetanus neurotoxin (TeNT) invades the central nervous system (CNS) via retrograde transport through motoneurons. Whether a host factor is needed to mediate TeNT trafficking has remained unknown. Here, using pull-down and mass spectrometry analysis, and by developing a luciferase-based in vivo transport assay, we identify receptor protein tyrosine phosphatase sigma (PTPRS; PTPσ) as the retrograde transport receptor for TeNT. Global or motoneuron-specific PTPRS knockout (KO) abolishes TeNT entry into the CNS in mouse models. PTPRS-TeNT interactions occur in endosomes, prevent toxin translocation

Chlamydia, an obligate intracellular bacterial pathogen, has a unique developmental cycle involving the differentiation of invading elementary bodies (EBs) to noninfectious reticulate bodies (RBs), replication of RBs, and redifferentiation of RBs into progeny EBs. Progression of this cycle is regulated by three sigma factors, which direct the RNA polymerase to their respective target gene promoters. We hypothesized that the Chlamydia-specific transcriptional regulator GrgA, previously shown to activate σ66 and σ28, plays an essential role in chlamydial development and growth. To test this hypo

Since the beginning of the coronavirus disease (COVID)-19 pandemic, the need for effective treatments for COVID-19 led to the idea of "repurposing" drugs for antiviral treatment. Several antipsychotics and antidepressants have been tested for in vitro activity against the severe acute respiratory syndrome coronavirus 2. Chlorpromazine, other phenothiazine antipsychotics, and the antidepressant fluoxetine were found to be rather potent in these studies. However, whether effective plasma concentrations can be obtained with clinically accepted doses of these drugs is not clear. Data of COVID-19 p

The crystal structure of PR3, a serine proteinase from the azurophilic granules of human polymorphonuclear neutrophils, has been solved by molecular replacement using the human leukocyte elastase structure. The PR3 structure has been refined to an R-factor (= sigma parallel Fo magnitude of-Fc parallel/sigma magnitude of Fo) of 0.201 for all data in the range of 10.0 to 2.2 A resolution. The enzyme was crystallized in space group P21 with four molecules in the asymmetric unit (Vm approximately equal to 2.6 A/Da). The overall fold consists of two domains of beta-barrel structures typical of the

Photorhabdus (Enterobacteriaceae) bacteria are pathogenic to insects and mutualistic with entomopathogenic Heterorhabditis nematodes. Photorhabdus luminescens subsp. akhurstii LN2, associated with Heterorhabditis indica LN2, shows nematicidal activity against H. bacteriophora H06 infective juveniles (IJs). In the present study, an rpoS mutant of P. luminescens LN2 was generated through allelic exchange to examine the effects of rpoS deletion on the nematicidal activity and nematode development. The results showed that P. luminescens LN2 required rpoS for nematicidal activity against H06 nemato

Burkholderia pseudomallei, the causative agent of melioidosis, is a facultative intracellular Gram-negative bacillus which can survive and multiply in both phagocytic and nonphagocytic cells. This bacterium could also induce apoptosis in various cell types. In the present study, we extend our finding to demonstrate the role of RpoS of B. pseudomallei in apoptosis induction. Unlike the wild-type strain, the B. pseudomallei rpoS mutant strain failed to induce cytotoxicity in mouse macrophages (RAW264.7). Furthermore, the mutant produced less extensive mitochondrial membrane potential changes and

Mycobacterium tuberculosis is exposed to a variety of stresses during a chronic infection, as the immune system simultaneously produces bactericidal compounds and starves the pathogen of essential nutrients. The intramembrane protease, Rip1, plays an important role in the adaptation to these stresses, at least partially by the cleavage of membrane-bound transcriptional regulators. Although Rip1 is known to be critical for surviving copper intoxication and nitric oxide exposure, these stresses do not fully account for the regulatory protein's essentiality during infection. In this work, we demo
