Cell surface receptors for tick-borne encephalitis (TBE) virus are proposed to be identified by affinity chromatography with polyclonal antiidiotypic antibodies (AIA) as antireceptor antibodies. A single affinity chromatography procedure permitted us to obtain 33-150 times purified cell surface receptor from human embryonic renal cell membranes. Four polypeptides with molecular weight 43, 67, 110, and 210 kD were detected in highly purified fractions of TBE virus cell surface receptor. Antireceptor antibodies reacted in immunoblotting only with the 67 kD polypeptide. The molecular weight of pr

BACKGROUND: Single-chain antibody-based CAR-T cells targeting BAFF-R have demonstrated antitumor effects against human B-cell malignancies and can overcome CD19 antigen loss. However, CARs built on antigen-specific single-chain antibody variable fragments may have limitations, e.g., large CAR binding domain size. To improve the function of BAFF-R CAR-T cells, we designed CARs with only a fully human heavy-chain variable domain.
METHODS: A fully human antibody phage display library was used to select anti- BAFF- R clones using protein/cell alternate panning. The screened clones were grafted in

Although antibody sequences are highly diverse, they are constrained by requirements for expression and limited off-target reactivity. Describing which sequences violate such constraints has proven to be difficult. Here, we introduce a machine-learning framework to leverage a previously underutilized source of data for this problem. We use human single-cell sequencing data to find instances of allelic inclusion, a rare event where B cells express two different antibody light chains as mRNA. Previous studies suggest that one of these chains is either autoreactive or non-expressing as protein. W

Novel monoclonal antibodies that specifically recognize gamma-carboxyglutamyl (Gla) residues in proteins and peptides have been produced. As demonstrated by Western blot and time-resolved immunofluorescence assays the antibodies are pan-specific for most or all of the Gla-containing proteins tested (factors VII, IX, and X, prothrombin, protein C, protein S, growth arrest-specific protein 6, bone Gla protein, conantokin G from a cone snail, and factor Xa-like proteins from snake venom). Only the Gla-containing light chain of the two-chain proteins was bound. Decarboxylation destroyed the epitop

BACKGROUND: Severe fever with thrombocytopenia syndrome (SFTS) caused by severe fever with thrombocytopenia syndrome virus (SFTSV) has become a persistent threat to public health. Since neutralizing monoclonal antibody therapy can invoke an immediate and effective passive immunity, endeavours made to discover the therapeutic monoclonal antibodies should be worthwhile. This study aimed to screen and purify the human monoclonal antibodies (hmAbs) against SFTSV glycoprotein C and characterize the properties of the hmAbs.
METHODS: Human monoclonal antibodies against SFTSV glycoprotein C (Gc) were

Vaccine-induced immune thrombotic thrombocytopenia (VITT) is a rare but dangerous side effect of adenoviral-vectored COVID-19 vaccines. VITT had been linked to production of autoantibodies recognizing platelet factor 4 (PF4). Here, we characterize anti-PF4 antibodies obtained from a VITT patient's blood. Intact mass measurements indicate that a significant fraction of these antibodies represent a limited number of clones. MS analysis of large antibody fragments (the light chain and the Fc/2 and Fd fragments of the heavy chain) confirms the monoclonal nature of this component of the anti-PF4 an

The prevalence and variation of porcine reproductive and respiratory syndrome virus (PRRSV) in China are increasing. The rapid preparation of essential antibodies will effectively reveal the antigenicity, epitopes, and intracellular distribution of viral proteins. Single-B-cell antibody technology is a novel method for screening diverse functional monoclonal antibodies (mAbs). Herein, we successfully expressed PRRSV nonstructural protein 12 (Nsp12) in suspension-cultured Chinese hamster ovary (CHO) cells. Using single-B-cell antibody technology, we utilized fluorescence-activated cell sorting

1. DQ epitopes were defined by studying the distinct reaction profiles of 17 allosera and 35 DQ-specific mouse monoclonal antibodies to HLA class II single antigen beads coated with purified recombinant DQ molecules. 2. DQ molecules are heterodimers with a polymorphic alpha chain and beta chain. Sixteen of the defined epitopes correlated to various DQB specificities and one defined epitope correlated to DQA1*0201. 3. Among the 16 DQB epitopes, 7 sites were recognized by both alloantibodies and DQ monoclonal antibodies, 5 sites were identified by DQ monoclonal antibodies and 4 sites by alloanti

Yeast display platform has proven instrumental for directed evolution of diverse proteins towards altered binding specificity, affinity, or improved biophysical properties. Particularly in antibody display, this method has delivered variants that are featured today as integral parts of approved therapeutic agents. While broadly used for discovery of binding agents, yeast display has also been applied for optimization of properties of Fc fragment. Among the bispecific antibody formats, which come in a variety of different molecular architectures, the single-chain Fc is a less explored alternati

Graves' disease is characterized by production of agonist antibodies to the thyroid-stimulating hormone receptor (TSHR), but knowledge of the genetic and somatic events leading to their aberrant production is limited. We describe the genetic analysis of two monoclonal antibodies (mAbs) with thyroid-stimulating activity (TSAb) obtained from a single mouse with experimental Graves' disease. The mAbs were class switched, but used the same rearrangement of immunoglobulin heavy chain, variable region (IGHV) and immunoglobulin light chain, variable region (IGLV) germline genes, implying a clonal rel

BACKGROUND: Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) continues to evolve and mutate during transmission, making the development of effective broad-spectrum antibodies for the prevention and treatment of SARS-CoV-2 infections of great significance. In this study, peripheral blood mononuclear cells were isolated from 15 convalescent coronavirus disease 2019 (COVID-19) patients to construct a human-derived single-chain variable fragment (scFv) antibody library against SARS-CoV-2. Monoclonal antibodies targeting the conserved structural region of the spike glycoprotein subunit

Botulinum toxin is a protein toxin secreted by Clostridium botulinum that is strongly neurotoxic. Due to its characteristics of being super toxic, quick acting, and difficult to prevent, the currently reported antiviral studies focusing on monoclonal antibodies have limited effectiveness. Therefore, for the sake of effectively prevention and treatment of botulism and to maintain country biosecurity as well as the health of the population, in this study, we intend to establish a single chain antibody (scFv) targeting the carboxyl terminal binding functional domain of the botulinum neurotoxin he

CAR-T cell therapy is at the forefront of next-generation multiple myeloma (MM) management, with two B-cell maturation antigen (BCMA)-targeted products recently approved. However, these products are incapable of breaking the infamous pattern of patient relapse. Two contributing factors are the use of BCMA as a target molecule and the artificial scFv format that is responsible for antigen recognition. Tackling both points of improvement in the present study, we used previously characterized VHHs that specifically target the idiotype of murine 5T33 MM cells. This idiotype represents one of the m

Assisted reproductive technologies increasingly demand efficient approaches for sperm sexing in livestock, ideally compatible with scalable manufacturing and reduced resource use. This study serves as a proof-of-concept for a biocompatible electrospun poly (lactic acid) (PLA) biomaterial platform functionalized with a recombinant single-chain variable fragment antibody specific to bovine Y-sperm (BY-scFv), enabling selective separation of bovine Y-sperm from X-sperm populations. Electrospun mats were prepared by selection PLA concentration (6-10% w/w) and deposited solution volume (10-20 mL),

Cardiovascular diseases (CVDs) remain a significant global health challenge, leading to substantial morbidity and mortality. Despite recent advancements in CVD management, pharmaceutical treatments often suffer from poor pharmacokinetics and high toxicity. With the rapid progress of modern molecular biology and immunology, however, single-chain fragment variable (scFv) molecule engineering has emerged as a promising theranostic tool to offer specificity and versatility in targeting CVD-related antigens. To represent the latest development on the potential of scFv in the context of CVDs, this r

BACKGROUND: Celiac disease (CD) is one of the most common food-related chronic disorders. It is mediated by the dietary consumption of prolamins, which are storage proteins of different grains. So far, no therapy exists and patients are bound to maintain a lifelong diet to avoid symptoms and long-term complications. To support those patients we developed a tandem single chain Fragment variable (tscFv) acting as a neutralizing agent against prolamins. We recombinantly produced this molecule in E. coli, but mainly obtained misfolded product aggregates, so-called inclusion bodies, independent of

Rationale: CD38 is a target for the therapy of multiple myeloma (MM) with monoclonal antibodies such as daratumumab and isatuximab. Since MM patients exhibit a high rate of relapse, the development of new biologics targeting alternative CD38 epitopes is desirable. The discovery of single-domain antibodies (nanobodies) has opened the way for a new generation of antitumor therapeutics. We report the generation of nanobody-based humanized IgG1 heavy chain antibodies (hcAbs) with a high specificity and affinity that recognize three different and non-overlapping epitopes of CD38 and compare their c

OBJECTIVE: Multifocal motor neuropathy (MMN) and the Guillain-Barré syndrome (GBS) are immune-mediated motor neuropathies with antibodies against the ganglioside GM1. In GBS, these antibodies are induced by molecular mimicry, but in MMN their origin is elusive.
METHODS: We compared the light-chain use of anti-GM1 IgM antibodies in serum from 42 patients with MMN and 23 patients with GBS by ELISA.
RESULTS: Exclusive use of either κ or λ light chains was found in 38 (90%) patients with MMN and 9 (39%) with GBS (p<0.001).
CONCLUSIONS: Anti-GM1 IgM antibodies in most patients with MMN are produ

Two chimeric antibodies (ch) 13D6 and 10C2 against the glycoprotein E of tick-borne encephalitis virus (TBEV) were constructed by fusing variable regions of murine monoclonal antibodies (Mabs) 13D6 and 10C2 to human constant regions. Monovalent analogues of these antibodies in format of single-chain antibodies (scFv or sc) were developed, as well. The ch13D6, ch10C2, sc13D6 and sc10C2 exhibited binding characteristics similar to parental Mabs. Only the ch13D6 and sc13D6 were able to neutralize TBEV infectivity in vitro. The in vitro neutralization provided by ch13D6 suggests that this antibody

IgE antibodies in the sera of subjects allergic to beta-lactam antibiotics detect a spectrum of specificities ranging from side-chain groups to an entire penicillin or cephalosporin molecule. In addition to such structural heterogeneity of allergenic determinants, IgE antibodies in the sera of different allergic subjects show heterogeneous recognition responses. Detailed immunochemical studies were carried out on the sera of penicillin-allergic subjects that showed selective and unexpected reactions with the frequently prescribed penicillin, amoxicillin. Antibodies from one subject reacted onl
