Cassette EEG sleep recordings in Gilles de la Tourette syndrome.

Tourette syndrome (TS) patients often complain of sleep problems, and questionnaire studies indicate that sleep disturbance is frequent. Decreased slow wave sleep and increased awakenings have been reported in laboratory polysomnography in TS patients, and a serotoninergic disorder of arousal has been postulated. We recorded outpatient sleep in 20 patients newly diagnosed with TS utilizing a 4-channel cassette EEG system. The newly-diagnosed patients were predominantly male, and ranged in age from 10 to 36 years. Some had taken psychotropic medications in the past, but none had been treated sy

HLA class II-restricted binding of muramyl peptides to B lymphocytes of normal and narcoleptic subjects.

The propensity for narcolepsy, a clinical sleep disorder of unknown etiology, is virtually totally included within the HLA-DR2,DQw1 (DRw15,DQw6) phenotype. The disorder is characterized by decreased sleep latency, early onset of rapid eye movement sleep, and a paucity of nocturnal slow-wave sleep. Muramyl peptides, naturally occurring bacterial cell wall peptidoglycans, potently enhance the duration and amplitude of slow-wave sleep in animals, bind to murine mononuclear cells, and exhibit a major histocompatibility complex-restricted immunoadjuvant effect in mice. We examined the binding of mu

Thalamo-Cortical and Thalamo-Thalamic Coupling During Sleep and Wakefulness in Rats.

Introduction: The thalamus, a heterogeneous brain structure, is involved in the generation of sleep-related thalamo-cortical oscillations. Higher order nuclei might possess a distinct function compared with first-order nuclei in brain communication. Here it is investigated whether this distinction can also be found during the process of falling asleep and deepening of slow-wave sleep. Methods: A nonlinear version of Granger causality was used to describe changes in directed network activity between the somatosensory cortex and rostral reticular thalamic nucleus (rRTN) and caudal reticular thal

Multivariate deep phenotyping reveals behavioral correlates of non-restorative sleep in 22q11.2 deletion syndrome.

Converging evidence suggests that sleep disturbances can directly contribute to a transdiagnostic combination of behavior and neurocognitive difficulties characterizing most forms of psychopathology. However, it remains unclear how the growing comprehension of sleep neurophysiology should best inform sleep quality assessment in mental health patients. To address this fundamental question, we performed deep multimodal sleep and behavioral phenotyping in 37 individuals at high genetic risk for psychopathology due to 22q11.2 Deletion Syndrome (Mean age:19±8.17, M/F = 22/15) and 34 Healthy Control

REM sleep prefrontal high-frequency oscillation chains mediate distinct cortical - hippocampal reactivation patterns compared to NREM sleep.

REM (rapid eye movement) and non-REM (NREM) sleep stages contribute to systems memory consolidation in hippocampal-cortical circuits. However, the physiological mechanisms underlying REM memory processes remain relatively unclear compared to NREM memory reactivation. Here we report, in rodents, the existence of prefrontal cortical (PFC) high-frequency oscillation (HFO) chains in REM sleep during the consolidation of recently acquired spatial memory. High-density tetrode recordings in hippocampal area CA1 and PFC reveal that REM cortical HFOs occur in characteristic chains that are phase-modula

Synchrony of neuromodulatory systems during NREM sleep.

Neuromodulatory systems play an essential role in regulating brain states and functions. The canonical view is that the release of monoamines including norepinephrine (NE) and serotonin (5-HT) is high during wakefulness and attenuated during sleep, particularly during rapid eye movement (REM) sleep, whereas the cholinergic system is active during both wakefulness and REM sleep and quiescent during non-REM (NREM) sleep. Recent studies have revealed a slow and rhythmic release pattern of neuromodulators during NREM sleep that drives infraslow oscillation (ISO) (0.02 to 0.03 Hz) in the brain. A k

Neuronal firing rates diverge during REM and homogenize during non-REM.

Neurons fire at highly variable intrinsic rates and recent evidence suggests that low- and high-firing rate neurons display different plasticity and dynamics. Furthermore, recent publications imply possibly differing rate-dependent effects in hippocampus versus neocortex, but those analyses were carried out separately and with potentially important differences. To more effectively synthesize these questions, we analyzed the firing rate dynamics of populations of neurons in both hippocampal CA1 and frontal cortex under one framework that avoids the pitfalls of previous analyses and accounts for

Wake slow waves in focal human epilepsy impact network activity and cognition.

Slow waves of neuronal activity are a fundamental component of sleep that are proposed to have homeostatic and restorative functions. Despite this, their interaction with pathology is unclear and there is only indirect evidence of their presence during wakefulness. Using intracortical recordings from the temporal lobe of 25 patients with epilepsy, we demonstrate the existence of local wake slow waves (LoWS) with key features of sleep slow waves, including a down-state of neuronal firing. Consistent with a reduction in neuronal activity, LoWS were associated with slowed cognitive processing. Ho

Sleep in young adults with Asperger syndrome.

Asperger syndrome (AS) is a neurodevelopmental disorder belonging to autism spectrum disorders. Both children and adults with AS have subjective impairment in the initiation and continuity of sleep, and studies using objective assessment are sparse. Twenty young AS adults with frequent complaints of low sleep quality were compared to 10 age-, gender- and education-matched controls without sleep complaints using polysomnography and spectral power analysis of slow-wave sleep. AS subjects displayed a similar polysomnographic profile as compared with controls. In spectral power analysis, a statist

[Long-term follow-up of a case of Landau-Kleffner syndrome].

Long-term investigation of a male case of Landau-Kleffner syndrome with auditory, verbal and non-verbal agnosia was performed clinico-electroencephalographically and neuropsychologically. Atypical absences and secondarily generalized tonic-clonic seizures were observed. The spike-wave index of slow wave sleep from all-night EEG recordings was 96% in this case. It was impossible to differentiate this case from epilepsy with "continuous spikes and waves during slow wave sleep (CSWS)" except the presence of auditory agnosia. Auditory agnosia of this case disappeared after antiepileptic therapy. I

Clinical and EEG sleep changes in primary dysthymia and generalized anxiety: a comparison with normal controls.

All night EEG sleep recordings and clinical assessments of sleep quality were performed in normal controls, patients with generalized anxiety disorder and primary dysthymia. Patients were selected according to DSM-III R. Changes of sleep architecture, namely a reduction of slow wave sleep, are similar in generalized anxiety and dysthymia. Also the two groups do not exhibit the REM sleep disturbances usually observed in affective illness. Duration and continuity measures are unchanged in dysthymics, but anxious patients show some features of insomnia. The analysis of subjective aspects of sleep

Sleep disturbance in depression reconsidered.

The well documented observation of a characteristic sleep disturbance in depression has been accorded a good deal of significance. Several current hypotheses regarding the etiology of depression, are based on these findings. However, a review of the descriptive studies shows that care has not generally been taken either to restrict or objectively measure daytime sleep. Evidence is presented suggesting that daytime sleep may occur in depressed patients more frequently than commonly thought and that, in normal subjects, daytime napping can result in nocturnal sleep changes similar to those seen

Effects of nocturnal gamma-hydroxybutyrate on sleep/waking patterns in narcolepsy-cataplexy.

Continuous 48-hour polygraphic recordings of sleep/waking patterns were performed on 14 patients with narcolepsy-cataplexy before and after 7-10 days of treatment of their nocturnal sleep with gamma-hydroxybutyrate (GBH). GBH improved the quality of night sleep by increasing the amount of slow wave sleep, reducing stage I, increasing sleep efficiency (percentage of time in bed spent asleep), and reducing the number of periods of short sleep under 15 minutes. Also nighttime REM sleep was reduced in latency and became less fragmented. The daytime period contained less slow wave sleep and REM sle

Arousal responses to chemical stimuli during sleep.

Arousal from sleep is an important component of any co-ordinated response to an external stimulus. Respiratory stimuli imposed during sleep are capable of eliciting arousal responses as well as the classic ventilatory responses Considerable survival advantage may result as a consequence of intact arousal mechanisms. Developmental studies of arousal responses to respiratory stimuli are lacking. Arousal responses to chemoreceptor stimuli have been examined in adult tracheotomized dogs, both before and following bilateral surgical denervation of the carotid bodies. Prior to denervation of the car

Sleep deprivation and sleep intensity exert distinct effects on cerebral vasomotion and brain pulsations driven by the respiratory and cardiac cycles.

The flow of cerebrospinal fluid (CSF) through the brain is driven by cerebral vasomotion, along with respiratory and cardiac forces. Growing evidence suggests that sleep facilitates this flow, yet the role of homeostatic sleep mechanisms remains largely unknown. In a circadian-controlled sleep and sleep deprivation study in humans, we used accelerated neuroimaging to investigate how sleep pressure and slow-wave-rich sleep affect low-frequency brain pulsations (LFPs; 0.012-0.034 Hz) as well as brain pulsations originating from the respiratory and cardiac cycles. These pulsations cause movement

Depth-dependent changes of obstruction patterns under increasing sedation during drug-induced sedation endoscopy: results of a German monocentric clinical trial.

PURPOSE: Drug-induced sedation endoscopy (DISE) and simulated snoring (SimS) can locate the site of obstruction in patients with sleep-disordered breathing (SDB). There is clinical evidence for a change in collapsibility of the upper airway depending on the depth of sedation. So far, a dose-response relationship between sedation and collapsibility has not been demonstrated. METHODS: DISE and SimS were performed in 60 consecutive patients with SDB under monitoring of depth of sedation by BiSpectral Index® (BIS). Initially, SimS was conducted followed by DISE using bolus application of propofol

Modulation of morphine-induced EEG and behavioral effects by dynorphin A-(1-13) in non-tolerant and morphine-tolerant rats.

The purpose of the present study was to assess effects of dynorphin A-(1-13) on morphine-induced changes in electroencephalographic (EEG) spectral power and morphine-induced suppression of slow-wave sleep in non-tolerant and morphine-tolerant rats. Adult female Sprague-Dawley rats were implanted with chronic cortical EEG electrodes, electromyographic electrodes in the temporalis muscle and with intracerebroventricular (i.c.v.) cannulae and, in some cases, additional intravenous (i.v.) cannulae. Injections of morphine (i.c.v., 20 micrograms/rat) produced a biphasic EEG and behavioral response,

ECoG of both hemispheres, EOG, neck EMG and EKG were recorded in 2 white (age 10 days) and 2 gray pups (age 1 month) of harp seal. The active wakefulness occupied 23.4 +/- 3.8% of total recording time, the relaxed wakefulness--32.6 +/- 3.6%, drowsiness--4.8 +/- 1.1%, slow wave sleep--31.5 +/- 3.0%, paradoxical sleep--7.7 +/- 0.8%. The sleep cycle averaged 18.2 +/- 1.4 min. Interhemispheric asymmetry of the ECoG was not seen in all 4 pups. The respiration was fast and regular in the relaxed wakefulness, then long respiration pauses were alternated with episodes of hyperventilation during slow w

AQP4-mediated glymphatic clearance: Sleep, neurodegeneration, and the translational gap.

One-third of adults in industrialized societies are chronically sleep-deprived. If current evidence linking sleep disruption to glymphatic failure extends to human populations, this may represent not merely a productivity concern but a significant and underappreciated risk factor for neurodegeneration at the population scale. The glymphatic system, a brain-wide perivascular network that clears soluble amyloid-beta, tau, alpha-synuclein, and other neurotoxic metabolites through astrocytic aquaporin-4 water channels, operates predominantly during slow-wave sleep and is impaired when sleep is dis

Effects of opioid microinjections in the nucleus of the solitary tract on the sleep-wakefulness cycle states in cats.

BACKGROUND: Previous studies have shown that the region of the nucleus of the solitary tract (NST) is involved in the control of electrocortical activity and in sleep mechanisms. It also is well known that this region contains the highest concentration of opioid receptors within the medullary brainstem. The involvement of the NST opioid system in sleep-wakefulness states were evaluated. METHODS: Ten cats were implanted with electrodes for chronic polygraphic recordings of their sleep-wakefulness states and provided with an implanted guide cannula stereotaxically aimed at the NST region.
