Visualization of incidentally imaged pituitary gland on three-dimensional arterial spin labeling of the brain.

OBJECTIVE: To evaluate the visualization of incidentally imaged normal pituitary gland on three-dimensional (3D) pseudo continuous arterial spin labeling (PCASL) perfusion imaging of the brain. METHODS: Ninety-three patients with a normal pituitary gland who underwent 3D PCASL for suspected brain diseases were retrospectively included. Visualization of the pituitary gland on PCASL cerebral blood flow (CBF) maps was assessed independently by two observers using a three-point grading system: Grade 1, pituitary CBF ≤ CBF of the cerebral white matter (WM); Grade 2, CBF of WM < pituitary CBF ≤ CBF

The potent radioprotective agents: Novel nitronyl nitroxide radical spin-labeled resveratrol derivatives.

It is commonly known that radiotherapy is still a key modality for treatment of cancer. Though this effect is desirable during radiotherapy, it leads to radiotoxicity on normal healthy cells. In the present research, we designed, synthesized and analyzed a series of nitronyl nitroxide radical (NITR) spin-labeled resveratrol (RES) derivatives. The cytotoxicity of the newly synthesized substances was tested on Jurkat T cells. The derivatives were studied as reactive oxygen species (ROS) scavenger to protect ionizing radiation of Jurkat T cells upon 6 Gy X-irradiation. The experimental results re

Pseudo-continuous arterial spin labeling MRI study of patients with obsessive-compulsive disorder.

Arterial spin labeling (ASL) magnetic resonance imaging is a novel technique that can measure regional cerebral blood flow (rCBF). Here we used pseudo-continuous ASL (pCASL) to examine the structural and functional imaging data in patients with obsessive-compulsive disorder (OCD). We estimated the gray matter volume imaging and pCASL imaging data by means of a voxel-by-voxel statistical analysis. We evaluated the differences of rCBF and gray matter volume between the OCD patients and healthy subjects. We detected a significant rCBF reduction in OCD patients in the right posterior cingulate ext

The utility of arterial spin labeling in the presurgical evaluation of poorly defined focal epilepsy in children.

OBJECTIVE: The authors sought to assess the utility of arterial spin labeling (ASL) perfusion 3T-MRI for the presurgical evaluation of poorly defined focal epilepsy in pediatric patients. METHODS: Pseudocontinuous ASL perfusion 3T-MRI was performed in 25 consecutive children with poorly defined focal epilepsy. ASL perfusion abnormalities were detected qualitatively by visual inspection and quantitatively by calculating asymmetry index (AI) maps and significant z-score cluster maps based on successfully operated cases. ASL results were prospectively compared to scalp EEG, structural 3T-MRI, FD

Arterial spin labeling perfusion changes of the frontal lobes in children with posterior fossa syndrome.

OBJECTIVE: Posterior fossa syndrome (PFS) is a common complication following the resection of posterior fossa tumors in children. The pathophysiology of PFS remains incompletely elucidated; however, the wide-ranging symptoms of PFS suggest the possibility of widespread cortical dysfunction. In this study, the authors utilized arterial spin labeling (ASL), an MR perfusion modality that provides quantitative measurements of cerebral blood flow without the use of intravenous contrast, to assess cortical blood flow in patients with PFS. METHODS: A database of medulloblastoma treated at the author

[Ictal arterial spin labeling MRI findings in two cases of acute confusional migraine].

Arterial spin labeling (ASL) is a magnetic resonance imaging (MRI) technique that enables visualizing of cerebral blood flow without need of a contrast medium. In recent years, there have been reports from outside Japan related to ASL use in migraine attacks. We report two cases of acute confusional migraine (ACM) in children. At time of confusion, ASL imaging showed reduced blood flow: for the first patient, in both cerebral hemispheres from the occipital lobe through the parietal lobe to the temporal lobe; for the second patient, throughout the left cerebral hemisphere. T1-, T2-, diffusion-w

An Integrated Spin-Labeling/Computational-Modeling Approach for Mapping Global Structures of Nucleic Acids.

The technique of site-directed spin labeling (SDSL) provides unique information on biomolecules by monitoring the behavior of a stable radical tag (i.e., spin label) using electron paramagnetic resonance (EPR) spectroscopy. In this chapter, we describe an approach in which SDSL is integrated with computational modeling to map conformations of nucleic acids. This approach builds upon a SDSL tool kit previously developed and validated, which includes three components: (i) a nucleotide-independent nitroxide probe, designated as R5, which can be efficiently attached at defined sites within arbitra

Diagnostic classification of arterial spin labeling and structural MRI in presenile early stage dementia.

Because hypoperfusion of brain tissue precedes atrophy in dementia, the detection of dementia may be advanced by the use of perfusion information. Such information can be obtained noninvasively with arterial spin labeling (ASL), a relatively new MR technique quantifying cerebral blood flow (CBF). Using ASL and structural MRI, we evaluated diagnostic classification in 32 prospectively included presenile early stage dementia patients and 32 healthy controls. Patients were suspected of Alzheimer's disease (AD) or frontotemporal dementia. Classification was based on CBF as perfusion marker, gray m

Orthogonal spin labeling and Gd(III)-nitroxide distance measurements on bacteriophage T4-lysozyme.

We present the first example of chemoselective site-specific spin labeling of a monomeric protein with two spectroscopically orthogonal spin labels: a gadolinium(III) chelate complex and a nitroxide radical. A detailed analysis of the performance of two commercially available Gd(III) ligands in the Gd(III)-nitroxide pulse double electron-electron resonance (DEER or PELDOR) experiment is reported. A modification of the flip angle of the pump pulse in the Gd(III)-nitroxide DEER experiment is proposed to optimize sensitivity.

Arterial spin labeling MRI: an emerging biomarker for Alzheimer's disease and other neurodegenerative conditions.

PURPOSE OF REVIEW: The search for biomarkers that detect and track disease progression in early Alzheimer's disease has been a major pursuit for two decades. Functional measures reflecting alterations of synaptic activity associated with neuronal dysfunction have displayed promise for following disease course in early stages. Although fluorodeoxyglucose positron emission tomography (FDG PET) and blood oxygen level-dependent functional MRI (BOLD fMRI) have been studied extensively for this purpose, this review will discuss the emergence and potential role of arterial spin labeling (ASL) MRI, wh

Monitoring the aggregation processes of amyloid-β using a spin-labeled, fluorescent nitroxyl radical.

Amyloid nitroxyl radical (nitroxide) ligands were used to detect amyloid-β fibrils, the main constituents of senile plaques in Alzheimer's disease, using anisotropic ESR spectra, and were found to affect the aggregation process due to the radical functionality. These compounds have great potential as novel and multifunctional probes, combining spin labels, spin probes, and fluorescence probes.

Interactions of anthrax lethal factor with protective antigen defined by site-directed spin labeling.

The protective antigen (PA) moiety of anthrax toxin forms oligomeric pores that translocate the enzymatic moieties of the toxin--lethal factor (LF) and edema factor (EF)--across the endosomal membrane of mammalian cells. Here we describe site-directed spin-labeling studies that identify interactions of LF with the prepore and pore conformations of PA. Our results reveal a direct interaction between the extreme N terminus of LF (residues 2-5) and the Φ-clamp, a structure within the lumen of the pore that catalyzes translocation. Also, consistent with a recent crystallographic model, we find tha

Hypertension and cerebral vasoreactivity: a continuous arterial spin labeling magnetic resonance imaging study.

Hypertension is associated with microvascular and macrovascular brain injury but its direct influence on the cerebral circulation is not fully clear. Our objective was to investigate the association of hypertension with global and regional cerebral vasoreactivity to CO(2) using continuous arterial spin labeling MRI, independent of stroke and white matter hyperintensities. Participants (n=62; mean age: 66.7±1.0 years, 55% women, 84% white, 65% hypertension, 47% stroke) underwent arterial spin labeling perfusion MRI during normal breathing, 5% CO(2) rebreathing, and hyperventilation, as well as

Territorial arterial spin labeling in the assessment of collateral circulation: comparison with digital subtraction angiography.

BACKGROUND AND PURPOSE: Collateral circulation plays a vital role in patients with steno-occlusive disease, in particular for predicting stroke outcome. Digital subtraction angiography (DSA) is the gold standard for the assessment of collateral circulation, despite its invasive nature. Recently, the development of a new class of arterial spin labeling (ASL) methods allowed independent measurement of territorial flow information without the need for contrast media injection. Here, we compared combined territorial ASL (TASL) and MR angiography (MRA) against DSA in the assessment of collateral ci

Synthesis and properties of umbelliferone-nitroxide radical hybrid compounds as fluorescence and spin-label probes.

Two hybrid compounds 6 and 7, linked via an ester-bond between the 7-hydroxyl residue of an umbelliferone 1 and a carboxylic acid residue of two nitroxide radicals 3 and 4, and one hybrid compound 8, linked via an ester-bond between a 3-carboxylic acid residue of umbelliferone 2 and a hydroxyl residue of nitroxide radical 5, were synthesized in good yields, and their fluorescence and ESR spectra before and after the addition of L-ascorbic acid sodium salt in PBS (pH 7.0) were measured. The ESR intensities of 6 and 7 were proportionally reduced after the addition of ascorbic acid sodium salt, a

Validation study of a pulsed arterial spin labeling technique by comparison to perfusion computed tomography.

Abnormalities in cerebral blood flow (CBF) are believed to play a significant role in the development of major neonatal neuropathologies. One approach that would appear ideal for measuring CBF in this fragile age group is arterial spin labeling (ASL) since ASL techniques are noninvasive and quantitative. The purpose of this study was to assess the accuracy of a pulsed ASL method implemented on a 3-T scanner dedicated to neonatal imaging. Cerebral blood flow was measured in nine neonatal piglets, the ASL-CBF measurements were acquired at two inversion times (TI) (1,200 and 1,700 ms), and CBF wa

Efficient visualization of vascular territories in the human brain by cycled arterial spin labeling MRI.

Intracranial vascular territories are usually visualized with the use of angiographic techniques. However, because it is difficult to visualize the distal vascular bed with any angiographic technique, it is also difficult to infer the parenchymal borders of vascular territories. Arterial spin labeling (ASL) MRI provides information on cerebral perfusion, and a regional ASL (regASL) approach offers the potential to visualize perfusion of the anterior and posterior circulation separately. Current techniques perform the labeling in each feeding artery as a separate experiment, which is very time-
