Evaluating the cross-reactivity of a stool methylated syndecan-2 (meSDC2) test in colorectal cancer detection.

BACKGROUND: A stool methylated syndecan-2 (meSDC2) test has demonstrated high sensitivity and specificity in diagnosing colorectal cancer (CRC). We aimed to evaluate the cross-reactivity of the meSDC2 test through multi-cancer analysis. METHODS: This study is a prospective, single-centre study. We recruited patients with 13 different types of cancers, including CRC, as positive control. Excluding those with CRC, participants did not undergo colonoscopy. Stool samples were collected from each patient before starting definitive therapy for each cancer and sent to the central lab for methylation

Regulation of organogenesis. Common molecular mechanisms regulating the development of teeth and other organs.

Vertebrate organs develop from epithelial and mesenchymal tissues, and during their early development they share common morphological features. These include condensation of the mesenchymal cells and thickening, folding or branching of epithelial sheets. Sequential and reciprocal interactions between the epithelial and mesenchymal tissues play central roles in regulation of the morphogenesis of all organs. During recent years increasing amounts of molecular data have accumulated from studies describing developmental changes in expression patterns of molecules, as well as from functional in vit

Proteoglycan-based diversification of disease outcome in head and neck cancer patients identifies NG2/CSPG4 and syndecan-2 as unique relapse and overall survival predicting factors.

BACKGROUND: Tumour relapse is recognized to be the prime fatal burden in patients affected by head and neck squamous cell carcinoma (HNSCC), but no discrete molecular trait has yet been identified to make reliable early predictions of tumour recurrence. Expression of cell surface proteoglycans (PGs) is frequently altered in carcinomas and several of them are gradually emerging as key prognostic factors. METHODS: A PG expression analysis at both mRNA and protein level, was pursued on primary lesions derived from 173 HNSCC patients from whom full clinical history and 2 years post-surgical follo

Changes in heparan sulfate are associated with delayed wound repair, altered cell migration, adhesion and contractility in the galactosyltransferase I (beta4GalT-7) deficient form of Ehlers-Danlos syndrome.

Reduced activity of beta4-galactosyltransferase 7 (beta4GalT-7), an enzyme involved in synthesizing the glycosaminoglycan linkage region of proteoglycans, is associated with the progeroid form of Ehlers-Danlos syndrome (EDS). In the invertebrates Drosophila melanogaster and Caenorhabditis elegans, mutations in beta4GalT-7 affect biosynthesis of heparan sulfate (HS), a modulator of several biological processes relevant to wound repair. We have analyzed structural alterations of HS and their functional consequences in human beta4GalT-7 Arg270Cys mutant EDS and control fibroblasts. HS disaccharid

Integrative Single-Cell RNA Sequencing and Spatial Transcriptomics Uncover Distinct Fibroblast-Macrophage Crosstalk in Human Hip Synovium Between Patients With Femoroacetabular Impingement and Osteoarthritis.

OBJECTIVE: Femoroacetabular impingement (FAI) and synovitis have been recognized as essential factors for developing osteoarthritis (OA) in the hip joints. However, little is known about altered synovial cellular compositions, their associated transcriptomic profiles, and cell-cell interactions in FAI and hip OA. METHODS: Synovial samples from a sex-matched cohort of patients with FAI and hip OA (n = 6 per condition) were analyzed using integrative single-cell RNA sequencing and spatial transcriptomics. RESULTS: Compared to FAI, epiregulin (EREG)-enriched lining synovial fibroblast-like syno

* Glioblastoma Exosomes for Therapeutic Angiogenesis in Peripheral Ischemia.

Peripheral ischemia as a result of occlusive vascular disease is a widespread problem in patients older than the age of 65. Angiogenic therapies that can induce microvascular growth have great potential for providing a long-lasting solution for patients with ischemia and would provide an appealing alternative to surgical and percutaneous interventions. However, many angiogenic therapies have seen poor efficacy in clinical trials, suggesting that patients with long-term peripheral ischemia have considerable therapeutic resistance to angiogenic stimuli. Glioblastoma is one of the most angiogenic

TLR4 dependent heparan sulphate-induced pancreatic inflammatory response is IRF3-mediated.

BACKGROUND: Degraded extracellular matrix can stimulate the innate immune system via the Toll-Like Receptor-4 (TLR4). In the pancreas, syndecan-anchored heparan sulphate (HS) on the ductal epithelium can be cleaved off its protein cores by the proteases (trypsin and elastase) and potentially activate TLR4 signalling. METHODS: To investigate this signalling event, a low sulphated HS (500 μg/ml) was infused into the biliary-pancreatic duct of C57BL/6J wild-type mice. Phosphate buffered saline (PBS) and lipopolysaccharide (LPS) were used as negative and positive controls, respectively. Mice were

Xin, an actin binding protein, is expressed within muscle satellite cells and newly regenerated skeletal muscle fibers.

Xin is a muscle-specific actin binding protein of which its role and regulation within skeletal muscle is not well understood. Here we demonstrate that Xin mRNA is robustly upregulated (>16-fold) within 12 h of skeletal muscle injury and is localized to the muscle satellite cell population. RT-PCR confirmed the expression pattern of Xin during regeneration, as well as within primary muscle myoblast cultures, but not other known stem cell populations. Immunohistochemical staining of single myofibers demonstrate Xin expression colocalized with the satellite cell marker Syndecan-4 further support

Anti-inflammatory and neuroprotective effect of bryodulcosigenin against cerebral ischemia/reperfusion injury via modulation of inflammatory signaling pathways.

BACKGROUND: During acute ischemic stroke, a cascade of pathophysiological reactions leads to brain cell injury, primarily via disruptions in energy metabolism and increased oxidative stress. In this study, we scrutinized the neuroprotective effects of bryodulcosigenin (BRY) against acute cerebral ischemia/reperfusion (CIR) injury in rats. MATERIAL AND METHODS: To induce middle cerebral artery occlusion (MCAO) in rats, a nylon monofilament suture with a silicon-coated tip was inserted into the internal carotid artery. The cerebral infarct volume, brain water content, neurological deficits, bra

SUMO2-mediated SP1 SUMOylation promotes M2 macrophage polarization to facilitate diabetic foot ulcer healing.

Diabetic foot ulcers (DFU) present a severe clinical challenge, marked by poor healing, chronic inflammation, and elevated amputation risk. The M2 macrophage is known to support the wound healing of DFU, and syndecan-4 (SDC4) contributes to this process. However, the upstream regulatory mechanisms involving post-translational modifications and transcriptional networks remain unclear. The related molecule levels were measured by RT-qPCR and Western blot. The M2 macrophage biomarkers (IL-10, Arg-1) were detected by ELISA kits, while the number of F4/80+CD206+ cells was quantified via flow cytome

Methyl eugenol increases neural microcirculation to promote nerve repair by regulating endothelial cell and Schwann cell communication via Ptn/Sdc4.

Neuromicrocirculatory disorders often accompany nerve injury. Improving neural microcirculation can promote the repair of injured nerves. Methyl eugenol is a major bioactive component of Asarum heterotropoides and has been reported to exert analgesic effects in chronic pain. This study aimed to demonstrate that methyl eugenol can enhance neural microcirculation and thereby promote nerve repair. A combination of single-cell sequencing and mRNA sequencing revealed that pleiotrophin (Ptn)/syndecan-4 (Sdc4) is vital for coupling between endothelial cells and Schwann cells. A total of 32 rats were

Glycoprotein Nonmetastatic B⁺ Myeloid-Derived Suppressor Cells Mediate Radiation-Induced Immune Suppression in Prostate Cancer and Are Targetable to Improve Systemic Control.

PURPOSE: Stereotactic body radiation therapy (SBRT) provides excellent local control for localized prostate cancer (PC); however, systemic relapse remains the primary cause of death in high-risk patients, underscoring the need to understand and therapeutically address radiation-induced immune suppression. Here, we identify a previously unrecognized myeloid checkpoint pathway driven by glycoprotein nonmetastatic B (GPNMB⁺) myeloid-derived suppressor cells (MDSCs) as a dominant systemic response to clinical SBRT and demonstrate a tractable strategy to counter it. METHODS AND MATERIALS: We used

Dynamics of Circulating Endothelial Injury Markers Following Kidney Transplantation.

INTRODUCTION: Endotheliopathy is well studied in chronic kidney disease, brought on by the chronic stressors of proinflammatory cytokines and reactive oxygen species. However, the endothelial response to renal transplantation has not been well investigated. This study aimed to evaluate circulating biomarkers of endothelial injury acutely after renal transplantation. METHODS: Serum samples were collected from 51 renal transplant patients preoperatively, immediately postoperatively, and at 24 and 72 h postoperatively. Serum was then analyzed for biomarkers of endotheliopathy, including E-select
