Comparative transcriptome and proteome analysis of resistant and susceptible Talaromyces marneffei clinical isolates reveals a systemic multitiered defense network against azoles.

Talaromyces marneffei is a thermally dimorphic fungus causing life-threatening systemic mycosis in immunocompromised patients. The emergence of azole-resistant clinical isolates, especially those with reduced susceptibility to voriconazole (VOC) and fluconazole (FLC), presents a critical therapeutic challenge. Although target-gene alterations and efflux-related mechanisms have been reported in pathogenic fungi, the broader molecular basis distinguishing high-minimum-inhibitory concentration (MIC) and low-MIC T. marneffei clinical isolates remains incompletely defined. Here, we performed integr

Sequential Talaromyces marneffei and Legionella pneumophila infections leading to the diagnosis of anti-interferon-γ autoantibody-associated immunodeficiency: a case report.

BACKGROUND: Opportunistic infections caused by uncommon intracellular pathogens can serve as important clues to underlying immune dysfunction. CASE PRESENTATION: We report a 57-year-old male who presented with chronic cough, dyspnea, fever, and weight loss. Chest imaging revealed diffuse pulmonary infiltrates and mediastinal lymphadenopathy, mimicking tuberculosis or lymphoma. Despite empirical anti-tuberculosis and broad-spectrum antibiotic therapy, his condition deteriorated. Metagenomic next-generation sequencing (mNGS) and culture confirmed Talaromyces marneffei infection. During antifung

Wortmannilactones I-L, new NADH-fumarate reductase inhibitors, induced by adding suberoylanilide hydroxamic acid to the culture medium of Talaromyces wortmannii.

With the aim of finding more potential inhibitors against NADH-fumarate reductase (specific target for treating helminthiasis and cancer) from natural resources, Talaromyces wortmannii was treated with the epigenome regulatory agent suberoylanilide hydroxamic acid, which resulted in the isolation of four new wortmannilactones derivatives (wortmannilactones I-L, 1-4). The structures of these new compounds were elucidated based on IR, HRESIMS and NMR spectroscopic data analyses. These four new compounds showed potent inhibitory activity against NADH-fumarate reductase with the IC50 values rangin

Development of a Droplet Digital PCR Assay Targeting the Internal Transcribed Spacer Gene for Rapid Detection of Talaromyces marneffei in AIDS Patients.

Talaromycosis caused by Talaromyces marneffei is a life-threatening mycosis in patients with acquired immunodeficiency syndrome (AIDS). The gold-standard diagnostic method relies on time-consuming cultures, which delay treatment and increase mortality. In this study, we developed a rapid and sensitive droplet digital PCR (ddPCR) assay targeting the internal transcribed spacer (ITS) gene for detecting T. marneffei and compared its performance with blood culture and quantitative PCR (qPCR) assays. The ddPCR assay had a detection limit of one copy/reaction, making it 10-fold more sensitive than q

Integration of Transcriptomics and Proteomics to Elucidate Inhibitory Effect and Mechanism of Antifungalmycin B from Marine Streptomyces hiroshimensis in Treating Talaromyces marneffei.

Talaromyces marneffei (TM) is an opportunistic pathogenic fungus that mainly infects immunocompromised patients. Currently, the global prevalence of talaromycosis caused by TM is increasing, leading to an increased demand for anti-TM drugs. In our previous study, a novel 28-membered macrolide compound, antifungalmycin B (ANB), was isolated from Streptomyces hiroshimensis GXIMD 06359, exhibiting significant antifungal properties. However, its in vivo mechanisms and direct antifungal effects warrant further investigation. In this study, we employed a mouse model in conjunction with transcriptomi

Coinfection by Talaromyces marneffei and Mycobacterium abscessus in a human immunodeficiency virus-negative patient with anti-interferon-γ autoantibody: a case report.

Patients with anti-interferon (IFN)-γ autoantibodies have weakened immune defenses against intracellular pathogens. Because of its low incidence and non-specific symptoms, diagnosis of anti-IFN-γ autoantibody syndrome is difficult to establish during the early stages of infection. Here, we report a patient with high titers of serum anti-IFN-γ autoantibodies suffering from opportunistic infections. The patient presented with intermittent fever for 2 weeks. During his first hospitalization, he was diagnosed with Talaromyces marneffei pulmonary infection and successfully treated with antifungal t

Penicillium and Talaromyces diversity in cystic fibrosis patient sample and the description of a new species, Penicillium subluteum sp. nov. (Eurotiales, Aspergillaceae).

Penicillium and Talaromyces species are frequently isolated from the respiratory tracts of cystic fibrosis (CF) patients, yet their diversity, ecological roles, and clinical significance remain poorly understood. In this study, we analyzed 521 fungal isolates (482 Penicillium and 39 Talaromyces) obtained from Dutch CF patients to investigate species diversity and prevalence. Using a combination of AFLP fingerprinting and DNA sequences analysis, we identified 57 Penicillium and 18 Talaromyces species, including a putatively new species named Penicillium subluteum sp. nov. The most commonly isol

Sporopollenin-chitosan microspheres loaded with an endophytic fungus Talaromyces neorugulosus R-209 for promoting development and controlling root rot in pigeon pea.

Plant-beneficial microbes can be effective as biological agents for promoting development and controlling diseases in plants. However, direct inoculation of non-encapsulated plant-beneficial microbes into the soils can affect their vitality and efficacy. A novel bio-based encapsulant, sporopollenin-chitosan microspheres (SCMs), was developed to load an endophytic fungus Talaromyces neorugulosus R-209 with antagonistic activities against the root rot pathogen (Rhizoctonia solani AG4) and plant growth-promoting functions. The results showed that T. neorugulosus R-209 encapsulated in SCMs (TnR-20

Challenges in Diagnosis and Treatment of HIV-Negative Host Pulmonary Talaromyces marneffei in Non-Endemic Areas: A Case Report with a History of Pulmonary Tuberculosis.

Talaromyces marneffei is an important opportunistic fungal pathogen closely related to acquired immunodeficiency syndrome (AIDS), and its infection is relatively rare in human immunodeficiency virus (HIV)-negative populations. Although HIV-related immunosuppression remains the main risk factor, the history of treated tuberculosis and subsequent structural lung damage may constitute an underrecognized predisposing condition in non-epidemic areas, especially in elderly patients. This report described a 69-year-old HIV-negative male pulmonary infection case from a non-endemic area of T. marneffei
