Distinct mechanism of syntaxin3-dependent tight junction assembly in keratinocyte model cells.

In the epidermis, differentiated keratinocytes form tight junctions (TJs) at a specific layer of the stratified epithelium, contributing to the skin barrier function. Previously, we demonstrated that the t-SNARE protein Syntaxin3 (Stx3) is necessary for TJ assembly in differentiated HaCaT keratinocytes. In simple epithelial cells, the apical targeting signal (ATS), located within helix a of Stx3, appears to be essential for TJ formation. Here, using Stx3-knockout HaCaT cells with inducible expression of various Stx3 mutants, we present a distinct mechanism of Stx3-dependent TJ assembly in epid

Nanoscopic analysis of tight junction organization in in vitro blood-brain barrier models.

BACKGROUND: The composition of tight junctions (TJs) in epithelia is well understood, whereas that one in endothelial cells at the blood-brain barrier (BBB) is controversial. Although freeze fracture EM remains the gold standard for assessing TJ nano-structures, advances in fluorescence microscopy, such as stimulated emission depletion (STED) together with image analysis tools, allow immunostaining-based quantitative analysis of the protein composition and the nano-organization of TJ strands. METHODS: Here, we present two approaches for quantitative analysis of the BBB TJ meshwork utilizing c

Claudin-23 reshapes epithelial tight junction architecture to regulate barrier function.

Claudin family tight junction proteins form charge- and size-selective paracellular channels that regulate epithelial barrier function. In the gastrointestinal tract, barrier heterogeneity is attributed to differential claudin expression. Here, we show that claudin-23 (CLDN23) is enriched in luminal intestinal epithelial cells where it strengthens the epithelial barrier. Complementary approaches reveal that CLDN23 regulates paracellular ion and macromolecule permeability by associating with CLDN3 and CLDN4 and regulating their distribution in tight junctions. Computational modeling suggests th

Baicalin alleviates endometrial inflammatory injury through regulation of tight junction proteins.

Endometritis is the foremost reason for reduced reproductive performance, which impedes the establishment of pregnancy in ruminants. Baicalin is extensively acknowledged as a tocolytic drug. However, the preventive effect of baicalin on endometrial inflammatory injury remains unclear. The present study aimed to determine the potential benefits of baicalin on endometrial inflammatory injury in animal and cellular models. The results showed that baicalin alleviated the impairment of tight junctions (TJs) and inflammation in the endometrium induced by LPS treatment. Baicalin increased claudin 3 (

Irsogladine maleate alters expression of a tight junction protein in portal hypertensive gastropathy.

BACKGROUND AND AIM: Portal hypertensive gastropathy (PHG) is characterized by noninflammatory edema and vasodilatation of the lamina propria of the mucosal epithelium. In addition, the alterations of intercellular junction proteins and dilatation of the endothelial gaps have been reported. In this study, we examined whether irsogladine maleate (IM), a gastric mucosal protective agent, has the potential to improve PHG by restoration of tight junctions (TJs). METHODS: Twenty-four patients with PHG were registered and randomly assigned into two groups: 12 patients in the IM-administration group

Enterotoxigenic Escherichia coli infection induces tight junction proteins expression in mice.

Enterotoxigenic Escherichia coli (ETEC) causes diarrhea in travelers, young children and piglets, but the precise pathogenesis of ETEC induced diarrhea is not fully known. Recent investigations have shown that tight junction (TJ) proteins and aquaporin 3 (AQP 3) are contributing factors in bacterial diarrhea. In this study, using immunoblotting and immunohistochemistry analyses, we found that ETEC increases the protein abundance of TJ proteins (occludin, claudin-1, zonula occludens-1) in mice. Enterotoxigenic Escherichia coli induced the expression of TJ proteins in mice through pathways by in

[Functional Modulation of Tight Junction to Enhance the Permeability of Biological Products (Biologics) in Biomembranes].

In recent years, biological products (biologics), including blood components, recombinant therapeutic proteins, antibodies, gene therapeutic materials, and so on, have been produced by biotechnology methods and other novel technologies. These products are essential therapeutic materials in progressive medicine. However, we often encounter the lower permeability of these biologics through biomembranes, due to their high molecular mass. In the last three decades, we have investigated drug delivery systems, including several enhancement methods for the permeability of biologics such as recombinan

Glucocorticoids Regulate Tight Junction Permeability of Lung Epithelia by Modulating Claudin 8.

The lung epithelium constitutes a selective barrier that separates the airways from the aqueous interstitial compartment. Regulated barrier function controls water and ion transport across the epithelium and is essential for maintaining lung function. Tight junctions (TJs) seal the epithelial barrier and determine the paracellular transport. The properties of TJs depend especially on their claudin composition. Steroids are potent drugs used to treat a variety of airway diseases. Therefore, we addressed whether steroid hormones directly act on TJ properties in lung epithelia. Primary human trac

Giardia disrupts the arrangement of tight, adherens and desmosomal junction proteins of intestinal cells.

Giardia duodenalis is a parasitic protozoan that causes diarrhea and other symptoms which together constitute a disease known as giardiasis. Although the disease has been well defined, the mechanisms involving the establishment of the infection have not yet been fully elucidated. In this study, we show that after 24h of interaction between parasites and intestinal Caco-2 cells, there was an alteration of the paracellular permeability, as observed by an approximate 42% of reduction in the transepithelial electrical resistance and permeation to ruthenium red, which was concomitant with ultrastru

Paracellin-1, a renal tight junction protein required for paracellular Mg2+ resorption.

Epithelia permit selective and regulated flux from apical to basolateral surfaces by transcellular passage through cells or paracellular flux between cells. Tight junctions constitute the barrier to paracellular conductance; however, little is known about the specific molecules that mediate paracellular permeabilities. Renal magnesium ion (Mg2+) resorption occurs predominantly through a paracellular conductance in the thick ascending limb of Henle (TAL). Here, positional cloning has identified a human gene, paracellin-1 (PCLN-1), mutations in which cause renal Mg2+ wasting. PCLN-1 is located i

Role of tight-junctional pathways in bile salt-induced increases in colonic permeability.

The effects of a dihydroxy bile salt, taurochenodeoxycholate (TCDC), on the permeability and conductance of isolated, short-circuited segments of the rabbit descending colon were examined using conventional Ussing chamber techniques. Increasing concentrations of TCDC (1-4 mM) produced dose-dependent increases in sodium backflux (JNas leads to m) and tissue conductance (Gt) when applied to either the mucosal or serosal salines. However, mucosal addition was twice as potent in increasing JNas leads to m and Gt at 4 mM. Tracer experiments indicated that the transepithelial serosal-to-mucosal flux

Targeting Tight Junction Proteins to Restore Airway Epithelial Barrier Function in Asthma.

PURPOSE OF REVIEW: Asthma is increasingly recognized not only as an inflammatory disorder but also as a disease rooted in airway epithelial barrier dysfunction. This review examines tight junction proteins (TJPs) including occludin, claudins, and ZO family members, as central regulators of epithelial integrity and potential therapeutic targets. RECENT FINDINGS: Disruption of TJPs is an early and pivotal event in asthma, enabling allergen penetration, amplifying immune responses, and promoting airway remodeling. Mechanistic studies reveal TJP regulation involves intricate crosstalk with inflam

Syntaxin-3 Regulates Tight Junction Assembly in Human Retinal Pigment Epithelium.

The retinal pigment epithelium (RPE) consists of polarized epithelial cells, serving as a support system for photoreceptor maintenance, where the polarity is contributed by the distribution of syntaxins (STX) within the cell. STX3, a known regulator of apical trafficking in epithelial cells, was previously understood to be absent in human RPE cells, with its functions thought to be compensated by STX1A. However, our results on SNARE mRNA expression profile in RPE detected the presence of 2 splice variants of STX3. Further investigation in donor retina, primary hRPE, and ARPE-19 cells revealed

AURKA modulates tight junction functionality to influence the proliferation and metastasis of lung adenocarcinoma.

Aberrant overexpression of AURKA (aurora kinase A) is strongly associated with various malignancies. However, limited research exists regarding its clinical significance and pathogenic mechanisms in lung adenocarcinoma. This study elucidates clinical correlations and provides experimental evidence supporting AURKA's potential as a therapeutic target for lung adenocarcinoma (LUAD). We analyzed AURKA expression levels and prognostic value using data from TCGA and GEO databases. qRT-PCR compared AURKA expression in LUAD tissues and adjacent normal specimens. siRNA-mediated knockdown efficiency wa

Microbial communities and tight junction protein expression in the gastrointestinal tract of feedlot cattle.

The gastrointestinal tract (GIT) of cattle plays a vital role in nutrient absorption, immune function, and microbial homeostasis. While the importance of the GIT microbiome and epithelial barrier integrity has been increasingly recognized, the typical composition of microbial communities and the expression of tight junction proteins (TJPs) in feedlot cattle remains poorly characterized. We investigated microbial community structure and TJP expression at three GIT sites: the rumen (RU), small intestine (SI), and large intestine (LI) in 21 finish-fed feedlot steers sourced from 21 commercial fee

Duodenal low-grade inflammation and expression of tight junction proteins in functional dyspepsia.

BACKGROUND: Duodenal changes in functional dyspepsia (FD) might be related to the development of symptoms. However, relationships among low-grade inflammation, Helicobacter pylori infection, and protein expression by tight junctions (TJs) in the duodenum are unclear. We therefore aimed to determine whether duodenal inflammation and genes associated with TJ proteins are associated with FD. METHODS: We evaluated inflammatory cell infiltration of the duodenum, H pylori infection, and genes associated with TJ proteins in duodenal biopsy specimens from 35 patients with FD according to the Rome III
