Prevalence of Toxocara eggs in sandpits in Tokushima city and its outskirts.

To demonstrate the prevalence of Toxocara eggs in sandpits in Tokushima Prefecture, Japan, sand samples were examined. Of 46 sandpits surveyed, 29 sandpits (63.3%) had Toxocara eggs. Sandpits in public parks and playgrounds in residential areas were more contaminated (87.5%) with the eggs than those in kindergartens, schools, and children's centers (36.4%). To examine the seasonal change in the contamination, sand samples obtained from five sandpits were examined monthly from May 1990 to April 1991. Toxocara eggs were found in the majority of sandpits (3/5-5/5) in the spring, early summer, and

The life cycle of Toxocara vitulorum in Asian buffalo (Bubalus bubalis).

A procedure for labelling hatched Toxocara vitulorum larvae with 75 selenium is described. Labelled larvae are infective when administered into the small intestine and portal vein of buffalo of all ages. Only very young calves are infected after oral administration. The labelled larvae are used with an enhanced fluorographic autoradiographic procedure to study the dynamics of the infection in non-pregnant and pregnant buffalo. Larvae penetrate the wall of the small intestine between 2 and 8 h after administration. Most larvae go straight to the liver via the portal vein but a few enter the mes

Toxocara canis: proteolytic enzymes secreted by the infective larvae in vitro.

Second-stage larvae of the dog nematode Toxocara canis are infective to man and cause the syndromes of visceral larva migrans and ocular toxocariasis. Larvae cultured in vitro secrete proteases which degrade components of a model of extracellular matrix and basement membranes. These enzymes have been characterized using a variety of techniques. Multiple enzyme activities were demonstrated by substrate gel electrophoresis, associated with proteins of molecular weights of 120 and 32 kDa. The enzyme activity was inhibited both in substrate gels and in a radiogelatin microplate assay by phenylmeth

The impact of single versus repeated exposure of mice to Toxocara canis.

Infection with T. canis can alter dramatically the brain and behavior of the host. Previous results suggest that if the mammalian host is exposed either simultaneously to lead, or has a history of prior exposure to that toxic substance, the magnitude of the behavioral reaction to T. canis may be modified or even reduced. The present data suggest that the magnitude of both the behavioral and tissue/immune reactions may be less if the organism has multiple, instead of a single exposure, to T. canis. Lead, and perhaps other environmental toxicants may alter neurotropic products of the parasite, t

Radiolabeling and autoradiographic tracing of Toxocara canis larvae in male mice.

Artificially hatched infective larvae of Toxocara canis were labeled with 75Se in Medium 199 (Gibco) containing 75Se-methionine. Male CD-1 mice were infected with radiolabeled larvae by intragastric intubation or by intraperitoneal injection. At intervals of 3-56 days mice were killed and the organs prepared for compressed organ autoradiography. Radioactivity of parasitic larvae showed an exponential decrease with time, reflecting catabolism of label with a biological half life of 26 days (effective half life of 21 days) making possible experiments lasting several months. Total body larva coun

Effects of irradiation on the biology of the infective larvae of Toxocara canis in the mouse.

Mice were infected with either 2,000 normal or irradiated embryonated eggs of Toxocara canis and the number of larvae in their livers, lungs, brains, and carcasses investigated at 5, 20, and 33 days of infection. Mortality of mice infected with normal eggs was 33% between day 4 and 8 postinfection but there was no mortality among mice infected with irradiated eggs. Irradiation with 60, 90, or 150 kr of X-rays inhibited the migration of larvae from the livers and lungs and their accumulation in brain and carcass in proportion to the irradiation dose. By day 33 of infection, the ratio of larvae

[The larva precipitation test with Ascaris suum and Toxocara canis larvae].

Investigations were made by means of a larvae precipitation test (LPT) on different laboratory animals (pigs and white mice, and pigs, rabbits, guinea pigs and white mice respectively) experimentally infected with Ascaris suum and Toxocara canis respectively for the serological evidence of specific A. suum and T. canis antibodies. Living A. suum larvae isolated from the lungs of experimentally infected white mice forming oral, excretion porus and cuticula precipitates proved to be a suitable antigen for the intravital evidence of prepatent A. suum infections in pigs and mice. By using living T

Effect of fenbendazole on Toxocara canis larvae in tissues of infected dogs.

The effect of fenbendazole therapy was studied in six dogs fed 10,000 embryonated Toxocara canis eggs. At 47 days after they were fed T canis, four dogs were given fenbendazole in two divided doses totaling 50 mg/kg of body weight each day for 14 days. Two infected dogs were not given fenbendazole. All dogs were necropsied at the end of treatment and the foci were counted in the lungs; their skeletal muscles were digested in 1% trypsin for the recovery of larvae. The T canis larvae were not recovered from the skeletal muscle of the four infected dogs treated with fenbendazole; 15 and 42 larvae

[The significance of fluorescein angiogarphy in ocular toxocara granuloma (author's transl)].

Four serologically-proved cases of intraocular toxocara infections are demonstrated. Three different manifestations in the following way were found: 1. Solitary granuloma on the posterior pole (2 patients). 2. Anterior and posterior uveitis of both eyes with granuloma near the ora (1 patient). 3. Iridocyclitis of both eyes (1 patient). The fluorescein angiography examination in two cases with solitary granuloma have been repeated over a period of time and the relation of the satisfactory results to the further development of the disease is shown.

Effect of Irradiated Toxocara Canis Embryonated Eggs: A Promising Vaccination Strategy.

Toxocariasis is a global zoonotic parasitic disease caused by Toxocara canis that poses significant public health challenges. The development of a vaccine would provide a promising method to control toxocariasis. This study aimed to evaluate the protective efficacy of gamma- and UV-irradiated embryonated T. canis eggs in mice. Seventy Swiss albino mice were classified into four groups: Group (I) the normal control; Group (II) was subdivided into a positive control once (IIA) and positive control twice (IIB); Group (III) was subdivided into a gamma-vaccinated control (IIIA) and gamma-vaccinated

Kinetics of Foxp3-expressing regulatory cells in experimental Toxocara canis infection.

Foxp3-expressing cells have recently been recognized as a cornerstone for the homeostasis of the immune system, and as key cells in many infectious diseases. Moreover, they have been found to contribute to the regulation of parasite-induced immunopathology in many parasitic infections. However, their role in Toxocara-induced immunopathology has not yet been investigated. The aim of this study is to assess the kinetics of Foxp3-expressing regulatory cells during the course of experimental infection by Toxocara canis (T. canis). Foxp3+ cells were identified in the liver by immunohistochemistry,

Toxocara canis in experimentally infected pigs: migratory pattern and tissue lesions.

Fifteen Yorkshire female pigs were inoculated with 100,000 infective T. canis eggs. Three animals were used as uninfected controls. Groups of three infected pigs were euthanized by accepted methods on days 7, 14, 21, 28 and 126 p.i., respectively. Larvae were recovered from all animals included in each group slaughtered on days 7 and 14 p.i.; on day 21 p.i. from two pigs, on day 28 p.i. from one, and no larvae were found on day 126 p.i. Differences in the mean number of larvae per gram in lymph nodes, liver and lungs between slaughter days, were significant for livers on day 7 p.i. and for lun
