Studies on the nutrition of marine flatfish. The thiamin requirement of turbot (Scophthalmus maximus).

1. Seven groups of young turbot (Scophthalmus maximus) were given diets containing graded levels of thiamin (0-19--50 mg/kg) for 16 weeks and their growth rate was measured during this period. 2. Good growth was obtained on all these treatments except in the group given the lowest dietary thiamin level (0-19 mg/kg). These fish grew normally until the 12th week but thereafter their weight did not increase. 3. Measurements of erythrocyte transketolase (sedoheptulose-7-phosphate: D-glyceraldehyde-3-phosphate glycolaldehydetransferase; EC 2.2.1.1) activity at the end of the experiment and of perce

Beriberi heart disease in Durban. A retrospective study.

A review of 41 possible cases of beriberi diagnosed in a 4-year period (1981-1985) revealed that the diagnosis was fairly certain in 23 patients, although measurements of red cell transketolase had not been performed. Fifteen patients had classic (high-output) beriberi and 8 acute beriberi (shoshin type). This article calls for an increased awareness of the condition so that appropriate and timely therapy may be instituted.

Biochemical and histological studies on thiamine-deficient and ethanol-fed rats.

Rats were separated into four groups and four different liquid diets were given to each group. Group 1: thiamine-sufficient diet with no ethanol, group 2: thiamine-sufficient diet with ethanol, group 3: thiamine-deficient diet with no ethanol, group 4: thiamine-deficient diet with ethanol. After four weeks, all rats were fasted for 24 hr and then ethanol was given orally to every rat. After one hour, every rat was sacrificed and biochemical and histological analyses were carried out. Transketolase activity in brain and liver decreased in groups 2, 3 and 4. There was significant decrease in tra

Thiamine deficiency encephalopathy in a specific-pathogen-free cat colony.

Thiamine deficiency encephalopathy occurred in a specific-pathogen-free cat colony where the diet consisted exclusively of commercially canned diet. This product was described by the manufacturer as a complete diet for cats. The clinical diagnosis was confirmed by the demonstration of bilaterally symmetrical hemorrhage and microvascular fibrinoid necrosis in the brain and low erythrocyte transketolase levels. Treatment of clinically ill cats with vitamin B complex was followed by recovery within 24 hours.

Thiamine and magnesium deficiencies: keys to disease.

Thiamine deficiency (TD) is accepted as the cause of beriberi because of its action in the metabolism of simple carbohydrates, mainly as the rate limiting cofactor for the dehydrogenases of pyruvate and alpha-ketoglutarate, both being critical to the action of the citric acid cycle. Transketolase, dependent on thiamine and magnesium, occurs twice in the oxidative pentose pathway, important in production of reducing equivalents. Thiamine is also a cofactor in the dehydrogenase complex in the degradation of the branched chain amino acids, leucine, isoleucine and valine. In spite of these well ac

Optimum nutrition: thiamin, biotin and pantothenate.

The metabolism of glucose is deranged in thiamin deficiency, but once any deficiency has been corrected there is no further effect of increased thiamin intake on the ability to metabolize glucose through either pyruvate dehydrogenase (EC 1.2.4.1) and the citric acid cycle, or the pentose phosphate pathway, in which transketolase (EC 2.2.1.1) is the thiamin-dependent step. It has been suggested that the Wernicke-Korsakoff syndrome is associated with a genetic variant of transketolase which requires a higher than normal concentration of thiamin diphosphate for activity. This finding would sugges

Effect of oral contraceptive agents on thiamin status.

A cross-sectional and follow up study of oral contraceptive agent (OCA) users revealed no adverse effects of OCA on erythrocyte transketolase activity (ETK) or thiamin pyrophosphate effect (TPP effect). In the cross-sectional study group, thiamin intake was below the recommended in only 19.2 per cent subjects and thiamin intake/100 kcal/day revealed a significant negative correlation with TPP effect. Routine prophylactic administration of thiamin was not indicated in OCA users.

The protective effect of hymochromone against ethanol-induced thiamin deficiency.

Intragastric administration of ethanol to male rats given at 6 gm/kg body weight, daily for two weeks, produced no significant change in the body or liver weights. Hepatic total lipid increased but this was no more than a trend. Treatment with ethanol, however, resulted in a marked decrease in transketolase activity (TK) of the haemolysed whole blood, associated with a pronounced rise in the stimulating effect of thiamin pyrophosphate on this enzyme (TPP effect). These changes mediated by ethanol were counteracted by hymochromone.

[Specific activity of thiamine and oxythiamine immobilized on modified cellulose].

The effect of thiamine and oxythiamine preparations on the activities of vitamin B1-dependent enzymes in the body of white mice was studied. It was found that the cellulose oxythiamine derivative was similar to the initial compound in the character and intensity of its antivitamin action. It was also shown that the cellulose thiamine derivative had a prolonged effect in the body, which was manifested in a slower increase in the activities of transketolase, pyruvate dehydrogenase and in the amount of thiamine diphosphate in tissues of vitamin B1-deficient animals, as compared to the effect of t

Clinical manifestations of chronic thiamine deficiency in rhesus monkey.

In the course of multiple episodes of thiamine deficiency in the rhesus monkey, the triad of anorexia, apathy, and hind limb weakness is the earliest clinical manifestation. In later episodes, nystagmus, abducens paresis, midline ataxia, dysmetria, and congestive heart failure are also seen. With the exception of dysmetria, the neurologic signs promptly respond to thiamine administration. Pair-fed controls showed no clinical signs. Neither peripheral neuropathy nor edema was observed. Thiamine-deficiency in the experimental animals was confirmed by blood transketolase assays.

Imatinib resistance associated with BCR-ABL upregulation is dependent on HIF-1alpha-induced metabolic reprograming.

As chronic myeloid leukemia (CML) progresses from the chronic phase to blast crisis, the levels of BCR-ABL increase. In addition, blast-transformed leukemic cells display enhanced resistance to imatinib in the absence of BCR-ABL-resistance mutations. In this study, we show that when BCR-ABL-transformed cell lines were selected for imatinib resistance in vitro, the cells that grew out displayed a higher BCR-ABL expression comparable to the increase seen in accelerated forms of the disease. This enhanced expression of BCR-ABL was associated with an increased rate of glycolysis but with a decreas

The vitamin B1 status among young Egyptian males in relation to infection with parasites.

Erythrocytic transketolase activity and thiamin pyrophosphate effect (TPP%) were used to define the nutritional thiamin status of an age stratified sample of 66 young males with parasitic infection. The results revealed that 6% of the control group were biochemically deficient in vitamin B1 compared with a percentage incidence of 42% among the parasitized subjects. Intestinal infection with Entamoeba histolytica, Hymenolepsis nana or with more than one parasite had the most depleting effect on the tissue thiamin status.

Regulating TKT activity inhibits proliferation of human acute lymphoblastic leukemia cells.

Among pediatric blood cancers, acute lymphoblastic leukemia (ALL) is the most common hematologic malignancy. Within ALL, T-cell acute lymphoblastic leukemia (T-ALL) accounts for 10 to 15% of all pediatric cases, and ~25% of adult cases. For T-ALL, its recurrence and relapse after treatment remain problematic. Therefore, it is necessary to develop new therapies for T-ALL. Recent studies suggested regulating energy metabolism is a novel approach to inhibit tumor growth, likely a promising treatment. Transketolase (TKT) is an important enzyme for modulating glucose metabolize in the pentose phosp

Cardiac and renal pentose phosphate pathway activity in thiamine deficiency.

Thiamine deficiency was produced in young rats by feeding a thiamine deficient diet. At a time when neurological symptoms were severe, and cardiac and renal transketolase activities were decreased, the animals were sacrificed. Glucose-6-phosphate dehydrogenase and 6-phosphogluconate dehydrogenase activities, and flux through the pentose phosphate pathway were similar in pair-fed control and thiamine deficient rats. These data suggest that altered pentose phosphate pathway activity is not a vital feature of murine thiamine deficiency.

The authors present one case of Wernicke's encephalopathy in a patient with obstructive pyloric syndrome caused by gastric adenocarcinoma, without previous history of alcoholism or malnutrition. They comment on the importance of physiological aspects of thiamine as a co-enzyme of transketolase and pyruvate-decarboxilase in the metabolic pathway for ATP production and the possibility of existing an interaction between genetical and environmental factors in the onset the symptoms. They draw attention for the clinical diagnosis, which should be precocious for the therapy with thiamine to be effec

Pyrithiamine-induced thiamine deficiency: effects on rat myelination.

Pyrithiamine was administered to newborn rats throughout the vulnerable period for myelinogenesis. A major metabolic defect was produced in the cerebral activities of the thiamine-dependent enzymes, transketolase and pyruvate decarboxylase. In spite of a defect in carbohydrate metabolism which is lethal in adult rats, overall development, and myelination as indicated by biochemical and morphological criteria, proceeded at an essentially normal rate. These findings indicate alternative metabolic pathways may be operational in newborn rat brain enabling it to circumvent major blockage in thiamin

Catechol biosynthesis from glucose in Escherichia coli anthranilate-overproducer strains by heterologous expression of anthranilate 1,2-dioxygenase from Pseudomonas aeruginosa PAO1.

BACKGROUND: The aromatic compound catechol is used as a precursor of chemical products having multiple applications. This compound is currently manufactured by chemical synthesis from petroleum-derived raw materials. The capacity to produce catechol is naturally present in several microbial species. This knowledge has been applied to the generation of recombinant Escherichia coli strains that can produce catechol from simple carbon sources. RESULTS: Several strains derived from E. coli W3110 trpD9923, a mutant that overproduces anthranilate, were modified by transforming them with an expressi
