Spatio-temporal dynamics and diversity of hematophagous flies (Diptera: Muscidae, Tabanidae, and Glossinidae) in livestock areas of Southern Benin: dominance of Stomoxys and occurrence of Glossina tachinoides with implications for vector surveillance.

African animal trypanosomiases (AAT) are vector-borne diseases transmitted by hematophagous flies, causing substantial economic losses in sub-Saharan African livestock systems. No multisite, inter-seasonal entomological study has previously characterized vector communities in southern Benin. This study characterizes their spatio-temporal distribution and diversity to inform AAT surveillance. A multisite survey was conducted across 6 communes during 2 contrasting seasons in 2025, using Vavoua, biconical, and Nzi traps deployed for 72 consecutive hours per session. Flies were identified morpholo

An atlas of tsetse and bovine trypanosomosis in Sudan.

BACKGROUND: After a long period of neglect, initiatives are being implemented in Sudan to control tsetse and trypanosomosis. Their planning, execution and monitoring require reliable information on the geographic distribution of the disease and its vectors. However, geo-referenced and harmonized data at the national level are lacking, despite the fact that a number of epidemiological studies were conducted over the years. The Atlas of tsetse and bovine trypanosomosis in Sudan tries to fill this gap. METHODS: The present study includes both a review of historical datasets on tsetse flies and b

The antiviral drug valacyclovir successfully suppresses salivary gland hypertrophy virus (SGHV) in laboratory colonies of Glossina pallidipes.

Many species of tsetse flies are infected with a virus that causes salivary gland hypertrophy (SGH) symptoms associated with a reduced fecundity and fertility. A high prevalence of SGH has been correlated with the collapse of two laboratory colonies of Glossina pallidipes and colony maintenance problems in a mass rearing facility in Ethiopia. Mass-production of G. pallidipes is crucial for programs of tsetse control including the sterile insect technique (SIT), and therefore requires a management strategy for this virus. Based on the homology of DNA polymerase between salivary gland hypertroph

Studies of trypanosomiasis in the Luangwa valley, north-eastern Zambia.

BACKGROUND: The present study, conducted in Zambia's Luangwa valley where both animal African trypanosomiasis (AAT) and human African trypanosomiasis (HAT) are endemic, combined the use of microscopy and molecular techniques to determine the presence of trypanosome species in cattle, goats and tsetse flies. METHODS: This study was conducted between 2008 and 2010 in Petauke, Chama and Isoka districts, north-eastern Zambia. A total of 243 cattle, 36 goats and 546 tsetse flies, were examined for presence of trypanosome species using microscopy, PCR and loop-mediated isothermal amplification (LAM

The population structure of Glossina fuscipes fuscipes in the Lake Victoria basin in Uganda: implications for vector control.

BACKGROUND: Glossina fuscipes fuscipes is the primary vector of trypanosomiasis in humans and livestock in Uganda. The Lake Victoria basin has been targeted for tsetse eradication using a rolling carpet initiative, from west to east, with four operational blocks (3 in Uganda and 1 in Kenya), under a Pan-African Tsetse and Trypanosomiasis Eradication Campaign (PATTEC). We screened tsetse flies from the three Ugandan PATTEC blocks for genetic diversity at 15 microsatellite loci from continental and offshore populations to provide empirical data to support this initiative. METHODS: We collected

Determinants of Human African Trypanosomiasis Elimination via Paratransgenesis.

Human African trypanosomiasis (HAT), transmitted by tsetse flies, has historically infected hundreds of thousands of individuals annually in sub-Saharan Africa. Over the last decade, concerted control efforts have reduced reported cases to below 10,000 annually, bringing complete elimination within reach. A potential technology to eliminate HAT involves rendering the flies resistant to trypanosome infection. This approach can be achieved through the introduction of transgenic Sodalis symbiotic bacteria that have been modified to produce a trypanocide, and propagated via Wolbachia symbionts, wh

Study on the sequential tsetse-transmitted Trypanosoma congolense, T. brucei brucei and T. vivax infections to African buffalo, eland, waterbuck, N'Dama and Boran cattle.

Susceptibility of African buffalo, eland, waterbuck, N'Dama and Boran cattle to sequential Glossina morsitans centralis-transmitted infections of Trypanosoma congolense, T. brucei brucei and T. vivax was compared, and their possible role as reservoirs of these parasites for G. moristans centralis, G. pallidipes, G. austeni, G. brevipalpis and G. longipennis determined. The buffalo, eland, waterbuck and N'Dama controlled T. congolense parasitaemias and were able to prevent anaemia. By contrast, one Boran became severely anaemic whilst the other controlled parasitaemia and anaemia. When the abov

An agent-based model of sleeping sickness: simulation trials of a forest focus in southern Cameroon.

An agent-based model (AMB) used to simulate the spread of Human African Trypanosomiasis is presented together with the results of simulations of a focus of the disease. This model is a completely spatialized approach taking into account a series of often overlooked parameters such as human behaviour (activity-related movements), the density and mobility of the disease vectors--tsetse flies (Glossina spp.)--and the influence of other tsetse feeding hosts (livestock and wild animal populations). The agents that represent humans and tsetse flies move in a spatially structured environment managed

Oxidative Phosphorylation Is Required for Powering Motility and Development of the Sleeping Sickness Parasite Trypanosoma brucei in the Tsetse Fly Vector.

The single-celled parasite Trypanosoma brucei is transmitted by hematophagous tsetse flies. Life cycle progression from mammalian bloodstream form to tsetse midgut form and, subsequently, infective salivary gland form depends on complex developmental steps and migration within different fly tissues. As the parasite colonizes the glucose-poor insect midgut, ATP production is thought to depend on activation of mitochondrial amino acid catabolism via oxidative phosphorylation (OXPHOS). This process involves respiratory chain complexes and F1Fo-ATP synthase and requires protein subunits of these c

Optimum land cover products for use in a Glossina-morsitans habitat model of Kenya.

BACKGROUND: Tsetse flies are the primary vector for African trypanosomiasis, a disease that affects both humans and livestock across the continent of Africa. In 1973 tsetse flies were estimated to inhabit 22% of Kenya; by 1996 that number had risen to roughly 34%. Efforts to control the disease were hampered by a lack of information and costs associated with the identification of infested areas. Given changing spatial and demographic factors, a model that can predict suitable tsetse fly habitat based on land cover and climate change is critical to efforts aimed at controlling the disease. In vivo experimental drug resistance study in Trypanosoma vivax isolates from tsetse infested and non-tsetse infested areas of Northwest Ethiopia.

Ethiopia, particularly in the Northwest region, is affected by both tsetse fly and non-tsetse fly transmitted trypanosomosis with a significant impact on livestock productivity. The control of trypanosomosis in Ethiopia relies on either curative or prophylactic treatment of animals with diminazene aceturate (DA) or isometamidium chloride (ISM), respectively. However, since these two trypanocides have been on the market for more than 40 years, this may have resulted in drug-resistance. Therefore, in vivo drug resistance tests on two Ethiopian isolates of Trypanosoma vivax were completed, one fr

[Control of vectors by trapping and its impact on sleeping sickness in a zone of high density of Glossina fuscipes quanzensis (Lefini River, People's Republic of the Congo)].

Tsetse control by trapping and its impact on the transmission of sleeping sickness in the high density area of Glossina fuscipes quanzenis (Léfini river, People's republic of Congo). The focus of human sleeping sickness of the Léfini river is of strategic importance for the People's Republic of Congo as it is located at an important junction which allows the trypanosomiasis to spread over the whole country. A campaign of tsetse control by trapping was undertaken in this focus, where an adjacent game reserve provides a continuous source of flies. The only practicable strategy was the use of a n

Tsetse salivary gland hypertrophy virus: hope or hindrance for tsetse control?

MANY SPECIES OF TSETSE FLIES (DIPTERA: Glossinidae) are infected with a virus that causes salivary gland hypertrophy (SGH), and flies with SGH symptoms have a reduced fecundity and fertility. The prevalence of SGH in wild tsetse populations is usually very low (0.2%-5%), but higher prevalence rates (15.2%) have been observed occasionally. The successful eradication of a Glossina austeni population from Unguja Island (Zanzibar) using an area-wide integrated pest management approach with a sterile insect technique (SIT) component (1994-1997) encouraged several African countries, including Ethiop

Limited impact of vector control on the population genetic structure of Glossina fuscipes fuscipes from the sleeping sickness focus of Maro, Chad.

Tsetse flies (genus Glossina) transmit deadly trypanosomes to human populations and domestic animals in sub-Saharan Africa. Some foci of Human African Trypanosomiasis due to Trypanosoma brucei gambiense (g-HAT) persist in southern Chad, where a program of tsetse control was implemented against the local vector Glossina fuscipes fuscipes in 2018 in Maro. We analyzed the population genetics of G. f. fuscipes from the Maro focus before control (T0), one year (T1), and 18 months (T2) after the beginning of control efforts. Most flies captured displayed a local genetic profile (local survivors), bu

Mapping African Animal Trypanosomosis risk from the sky.

In Burkina Faso, African Animal Trypanosomosis (AAT) is still a major hindrance to cattle breeding, especially in the Mouhoun river basin, which was identified as a priority area for tsetse control. The attempt of the present work was to assess the abundance of tsetse flies and AAT risk using remote sensing coupled to field environmental data, along a Mouhoun river section of 234 km long, harbouring an open riverine forest where G. tachinoides Westwood is the predominant tsetse species. The water course was classified into three epidemiological landscapes, corresponding to a "disturbed", "natu

Host-seeking efficiency can explain population dynamics of the tsetse fly Glossina morsitans morsitans in response to host density decline.

Females of all blood-feeding arthropod vectors must find and feed on a host in order to produce offspring. For tsetse-vectors of the trypanosomes that cause human and animal African trypanosomiasis-the problem is more extreme, since both sexes feed solely on blood. Host location is thus essential both for survival and reproduction. Host population density should therefore be an important driver of population dynamics for haematophagous insects, and particularly for tsetse, but the role of host density is poorly understood. We investigate the issue using data on changes in numbers of tsetse (Gl

RNA-seq de novo Assembly Reveals Differential Gene Expression in Glossina palpalis gambiensis Infected with Trypanosoma brucei gambiense vs. Non-Infected and Self-Cured Flies.

Trypanosoma brucei gambiense (Tbg), causing the sleeping sickness chronic form, completes its developmental cycle within the tsetse fly vector Glossina palpalis gambiensis (Gpg) before its transmission to humans. Within the framework of an anti-vector disease control strategy, a global gene expression profiling of trypanosome infected (susceptible), non-infected, and self-cured (refractory) tsetse flies was performed, on their midguts, to determine differential genes expression resulting from in vivo trypanosomes, tsetse flies (and their microbiome) interactions. An RNAseq de novo assembly was

Innate immunity in the tsetse fly (Glossina), vector of African trypanosomes.

Tsetse flies (Glossina sp.) are medically and veterinary important vectors of African trypanosomes, protozoan parasites that cause devastating diseases in humans and livestock in sub-Saharan Africa. These flies feed exclusively on vertebrate blood and harbor a limited diversity of obligate and facultative bacterial commensals. They have a well-developed innate immune system that plays a key role in protecting the fly against invading pathogens and in modulating the fly's ability to transmit African trypanosomes. In this review, we briefly summarize our current knowledge on the tsetse fly innat

Spatial and temporal variations relevant to tsetse control in the Bipindi focus of southern Cameroon.

BACKGROUND: Human African Trypanosomiasis (HAT) remains a public health problem in many poor countries. Due to lack of financial resources in these countries, cost-effective strategies are needed for efficient control of this scourge, especially the tsetse vector. It was shown that perennial water sources maintain a favourable biotope for tsetse flies and thus the transmission dynamics of sleeping sickness. The present paper aimed at assessing the transmission dynamics of HAT in a forest environment where the hydrographic network is important. METHODS: Two entomological surveys were carried o
