The lipoprotein electrophoregrams and the cholesterol and triglyceride levels of the pleural fluid were evaluated for patients with chylous pleural effusions, as defined by the presence of a distinctive band of chylomicrons on the lipoprotein electrophoregram, and in patients with nonchylous effusions of various causes. Endothelial cells regulate intercellular and inter-organ communication through extracellular vesicles (EVs), which have diagnostic and therapeutic potential. We describe single-particle interferometric reflectance imaging directly from culture supernatants, followed by ultracentrifugation isolation of EVs. We determine EV size and concentration by nanoparticle tracking analysis and characterize EVs by western blotting and transmission electron microscopy. Bacterial extracellular vesicles (bEVs) participate in regulating both intra-bacterial community interactions and bacteria-host cell interactions. Here, we propose a universal and highly reproducible protocol for the extraction of bEVs from bacterial culture. The workflow combines differential centrifugation, ultracentrifugation, and enrichment, allowing efficient impurity removal and bEV enrichment. By sucrose density gradient ultracentrifugation and standard hemolytic complement assays, complement fixing activity of about 19 S and greater was found in blister fluids of one patient with pemphigus and three of five patients with bullous pemphigoid. Control blister fluids, including experimentally induced blisters, lacked such activity. Complement fixing activity was apparent, however, in 6 of 10 pemphigus sera and in two of five bullous pemphigoid sera tested. The material present in both pemphigus and bullous pemphigoid appears to activate the classical complement pathway.
