The Dynamic Interface of Viruses with STATs.

Viruses commonly antagonize the antiviral type I interferon response by targeting signal transducer and activator of transcription 1 (STAT1) and STAT2, key mediators of interferon signaling. Other STAT family members mediate signaling by diverse cytokines important to infection, but their relationship with viruses is more complex. Importantly, virus-STAT interaction can be antagonistic or stimulatory depending on diverse viral and cellular factors. While STAT antagonism can suppress immune pathways, many viruses promote activation of specific STATs to support viral gene expression and/or produ

Insights into the pathogenesis of varicella viruses.

PURPOSE OF REVIEW: Varicella zoster virus (VZV) is a highly contagious, neurotropic alpha herpes virus that causes varicella (chickenpox). VZV establishes lifelong latency in the sensory ganglia from which it can reactivate to induce herpes zoster (HZ), a painful disease that primarily affects older individuals and those who are immune-suppressed. Given that VZV infection is highly specific to humans, developing a reliable in vivo model that recapitulates the hallmarks of VZV infection has been challenging. Simian Varicella Virus (SVV) infection in nonhuman primates reproduces the cardinal fea

[The mosquito-borne viruses in Europe].

Epidemiologic changes of vector-borne diseases in recent years have multiple causes, including climate change. There are about 3500 species of mosquitoes worldwide, three-quarters of which live in tropical and subtropical wetlands. Main viruses transmitted by mosquitoes in Europe belong to the genus Flavivirus; some of them have been recently reported in Italy (Usutu and Japanese encephalitis virus), while others have been circulating for years and autochthonous transmission has been documented (West Nile virus). Mosquito-borne viruses can be classified according to the vector (Aedes or Culex)

In vivo reassortment of influenza viruses.

The genetic material of influenza A virus consists of eight negative-sense RNA segments. Under suitable conditions, the segmented structure of the viral genome allows an exchange of the individual gene segments between different strains, causing formation of new reassorted viruses. For reassortment to occur, co-infection with two or more influenza virus strains is necessary. The reassortment is an important evolutionary mechanism which can result in antigenic shifts that modify host range, pathology, and transmission of the influenza A viruses. In this process, the influenza virus strain with

[Persistence of bacteria and viruses in Ixodes].

Behaviour of viruses and salmonellas in ticks after their single or combined contamination was thoroughly studied on laboratory animals with bacteriemia or virusemia. When Ixodes ricinus was contaminated simultaneously with forest-spring encephalitis virus and salmonellas there were observed a decrease in the virus titer by the 30th-40th days and its death in 60 days. In case of the I. ricinus nymphs contamination, the virus titer after the combined contamination was by a factor of 10(2) lower in 60 days vs. the contamination with the virus alone and did not reach the contamination dose. [Influenza viruses--a challenge for vaccinations].

The clinical picture of influenza may vary from mild respiratory infection to pneumonia requiring intensive care. Annual epidemics are most commonly caused by H3N2 or H1N1 type influenza A or influenza B viruses. The population's immune protection against a new virus type is low, whereupon morbidity and mortality may be high. Vaccinations are the most important means to decrease influenza morbidity. Annual variation and quick intercontinental migration of influenza viruses, combined with the possibility of the creation of reassortant viruses, are significant challenges for the development of i

Viruses associated with pneumonia in adults.

Viral pneumonia, which is typically associated with disease in childhood, is increasingly recognized as causing problems in adults. Certain viruses, such as influenza virus, can attack fully immunocompetent adults, but many viruses take advantage of more-vulnerable patients. The latter include patients receiving immunosuppressive therapy and elderly subjects, particularly those residing in long-term care facilities. The range of viruses producing pneumonia in adults includes common agents, such as varicella-zoster virus and influenza virus, as well as respiratory syncytial virus, human metapne

New respiratory viruses and the elderly.

The diagnostics of respiratory viral infections has improved markedly during the last 15 years with the development of PCR techniques. Since 1997, several new respiratory viruses and their subgroups have been discovered: influenza A viruses H5N1 and H1N1, human metapneumovirus, coronaviruses SARS, NL63 and HKU1, human bocavirus, human rhinoviruses C and D and potential respiratory pathogens, the KI and WU polyomaviruses and the torque teno virus. The detection of previously known viruses has also improved. Currently, a viral cause of respiratory illness is almost exclusively identifiable in ch

Keeping the memory of influenza viruses.

Protection against pathogens is mediated by both humoral responses (neutralizing antibodies) and cellular immunity, both CD4+ and CD8+ cells. In the case of influenza viruses, circulating strains contain both variable and conserved T and B cell epitopes that are challenged after vaccination and/or infection. During infection, the role of T cells is to prevent viral dissemination in the organism by killing the infected cells and helping B cell antibody production to neutralize the virus. The threat of influenza virus increases the preparedness of protective immunity to pandemic and seasonal inf

The viruses of Australia and the risk to tourists.

Australia is a climatically diverse country varying from a tropical climate in the north to arid central desert and grassland regions, and to temperate climates in the south. There are many viral infections found in Australia that are common to developed countries worldwide, but this article will focus on those that pose a special risk for travellers to Australia, especially the mosquito-borne viruses. The commonest are the members of the alphavirus genus, particularly Ross River virus and Barmah Forest virus, which cause predominantly arthralgia with or without fever or rash. Less frequent bu

[Viruses and bats: rabies and Lyssavirus].

Recent emerging zoonoses (hemorrhagic fevers due to Ebola or Marburg virus, encephalitis due to Nipah virus, severe acute respiratory syndrome due to SRAS virus...) outline the potential of bats as vectors for transmission of infectious disease to humans. Such a potential is already known for rabies encephalitis since seven out of the eight genotypes of Lyssavirus are transmitted by bats. In addition, phylogenetic reconstructions indicate that Lyssavirus have evolved in chiropters before their emergence in carnivores. Nevertheless, carnivores remain the most critical vectors for public health,

Brazilian vaccinia viruses and their origins.

Although the World Health Organization (WHO) declared global smallpox eradicated in 1980, concerns over emergent poxvirus infections have increased. Most poxvirus infections are zoonotic; exploring their genetic diversity will illuminate the genetic and evolutionary aspects of poxvirus infections, ecology, and epidemiology. In recent decades, several strains of the orthopoxvirus vaccinia virus (VACV) have been isolated throughout Brazil, including genetically distinct isolates within the same outbreak. To further investigate the diversity and origins of these viruses, we analyzed molecular dat

Viruses in periodontal disease - a review.

The purpose of this review was to evaluate the evidence supporting the hypothesis that viral infection plays a role in the development of periodontitis. An involvement in periodontal diseases has been suspected specifically for human immunodeficiency virus (HIV) and herpes viruses. An association has been demonstrated between HIV infection and some distinct forms of periodontal infection, i.e. necrotizing lesions. Furthermore, reports of increased prevalence and severity of chronic periodontitis in HIV-positive subjects suggests that HIV infection predispose to chronic periodontitis. Several s

Infection of patients by bloodborne viruses.

BACKGROUND: Despite taking precautions, healthcare workers performing invasive procedures run a small risk of infection by bloodborne viruses. When injury occurs, the viral status of the patient is often unknown and testing requires informed consent, which may be refused. On the other hand, although the chance of transmission of infection from a healthcare worker to a patient is extremely small, such personnel have an obligation of disclosure and, if seropositive, are barred from performing invasive procedures. METHODS: The medical literature on bloodborne virus transmission between carers an

The role of viruses in human diabetes.

Viruses have long been considered a major environmental factor in the aetiology of Type I (insulin-dependent) diabetes mellitus and recent work has greatly confirmed and extended this role. In addition to the enteroviruses, there are several other viruses which, from time to time, have been considered potential causal agents for human diabetes. With the exception of rubella, their role is not clear. The relation of enteroviruses with Type I diabetes has only been properly clarified by the use of new technologies, especially those based on polymerase chain reaction methods to identify them in b

Genes and viruses in hepatobiliary neoplasia.

Hepatobiliary neoplasms comprise a significant portion of the worldwide cancer burden. Advances in basic science research have led to rapid progress in our understanding of the molecular events responsible for these dreaded diseases. The genetic changes associated with hepatocellular carcinoma (HCC) have received the most attention. Aflatoxin B1 exposure leads to mutations in the p53 tumor suppressor gene, most commonly a transversion in codon 249 that leads to a substitution of serine for arginine in the p53 protein. Numerous other tumor suppressor genes, oncogenes, and tumor gene pathways ar

An update on transfusion-transmitted viruses.

Advances in blood donor screening have advanced the concept that a zero-risk blood supply may be possible in the future. Although the application of a large number of serologic tests and more stringent donor questioning has certainly aided our ability to screen out donors, risks still remain. In this article, the literature on some transfusion transmitted diseases is reviewed.

Viruses transmitted by sandflies in Italy.

Five different phlebotomus-transmitted viruses have been isolated in Italy. Four of them belong to the Bunyaviridae family (genus Phlebovirus). Sandfly fever Sicilian and Sandfly fever Naples viruses were isolated from sick persons during World War II, Toscana and Arbia viruses have been isolated more recently from Phlebotomus perniciosus and P. perfiliewi. Radi virus is the only phlebotomus-transmitted virus isolated in Italy from P. perfiliewi, which belongs to the Rhabdoviridae family (genus Vesiculovirus). The circulation and the natural cycle of these viruses on the basis of seroepidemiol

[Persistence of viruses in rheumatoid arthritis].

Humoral and cell-mediated immune response to antigens of hepatitis B (HBsAg and HBeAg), delta-virus, rubella, and measles viruses was studied. Detection in a number of patients with rheumatoid arthritis of HBsAg and HBeAg in blood sera permits a conclusion that productive virus infection is formed in such patients. At the same time in patients with rheumatoid arthritis areactivity of immunocytes to HBsAg and HBeAg was found while marked sensitization of lymphocytes to rubella virus antigens was demonstrated. The patients with rheumatoid arthritis, however, do not develop mere long-term carrier
