Anti-estrogen prevents xenoestrogen-induced testicular pathology of eelpout (Zoarces viviparus).

Estrogenic alkylphenols have been shown to affect the reproductive system of male fish causing induction of vitellogenin synthesis and altered testis structure. However, it is still unknown whether the histopathological effects on the testes is mediated by the estrogen receptor or if it represent general toxicopathological effects. In the present study, the effects of different concentrations of the estrogenic chemical 4-tert-octylphenol on vitellogenin (Vtg) synthesis and testicular structure were investigated in the eelpout Zoarces viviparus during spermatogenesis. Adult male eelpout were ex

De novo assembly and characterization of the Trichuris trichiura adult worm transcriptome using Ion Torrent sequencing.

Infection with helminthic parasites, including the soil-transmitted helminth Trichuris trichiura (human whipworm), has been shown to modulate host immune responses and, consequently, to have an impact on the development and manifestation of chronic human inflammatory diseases. De novo derivation of helminth proteomes from sequencing of transcriptomes will provide valuable data to aid identification of parasite proteins that could be evaluated as potential immunotherapeutic molecules in near future. Herein, we characterized the transcriptome of the adult stage of the human whipworm T. trichiura

Gonadal Lipid Storage in Mytilus coruscus: A Comprehensive Gene Network and Key Gene Discovery.

The hard-shelled mussel (Mytilus coruscus), a commercially vital bivalve in China, accumulates lipids predominantly in its gonads, the species' primary edible tissue. Understanding the molecular mechanisms underlying gonad-specific lipid storage is critical for improving reproductive efficiency and aquaculture yield. This study employs comparative transcriptomic analysis of multiple tissues (gonad, gill, mantle, foot, hemolymph) to pinpoint key regulatory genes involved in lipid deposition. Through weighted gene co-expression network analysis (WGCNA), vitellogenin (VG), perilipin (PLIN), and t

Knockdown of LmSPARC influences lipid metabolic homeostasis involved ovarian development in Locusta migratoria.

Reproduction, a fundamental feature of all known life, is closely correlated with energy homeostasis. Lipids, which can accumulate, provide essential energy support for reproduction. Secreted Protein, Acidic and Rich in Cysteine (SPARC) is a cellular matrix glycoprotein involved in the regulation of extracellular matrix synthesis. However, the precise coordination of lipid homeostasis by SPARC during insect reproductive development remains poorly understood. Our study revealed that when the expression of LmSPARC was downregulated by RNA interference (RNAi) in 4th and 5th instar nymphs, molting

Estrogens as immunotoxicants: 17α-ethinylestradiol exposure retards thymus development in zebrafish (Danio rerio).

Estrogenic endocrine disrupting compounds (EEDCs) can cause alterations in sexual development and reproductive function of fish. Growing evidence suggests that EEDCs can also interfere with development and function of innate immunity of fish. The present study examined a potential disruptive effect of EEDCs at field-relevant concentrations on the development of adaptive immunity, more specifically the thymus. Zebrafish (Danio rerio) were exposed from fertilization until 64 days post-fertilization (dpf) to environmentally relevant (3 and 10 ng/L) concentrations of the synthetic estrogen 17α-eth

Thyroid hormone-induced demineralisation of the vertebral skeleton of the eel, Anguilla anguilla.

The role of thyroid hormones (TH) in bone remodelling is controversial. Indeed, in humans, while they are necessary for normal growth and development, their overproduction can induce important mineral bone loss and osteoporosis. Intense bone resorption is a natural phenomenon also observed in some teleosts, during reproductive migration and fasting. Our work aimed at investigating the effects of chronic treatments with TH (thyroxin, T4 or triiodothyronine, T3) on bone resorption in a migratory fish, the European eel (Anguilla anguilla), a representative species of an ancient group of teleosts

Acute and chronic effects of polymetallic nodule leachate in the marine copepod Tigriopus koreanus.

Polymetallic nodules containing manganese, iron, and other metals are found in the seafloor. Leachates of polymetallic nodules can be discharged into seawater during ocean mining, disrupting marine ecosystems and causing adverse effects on marine organisms. Here, we investigate the acute and chronic effects of two polymetallic nodule leachates on the life-history parameters (mortality, development, and fecundity) and transcriptional differences of detoxification, antioxidant, and reproduction-related genes for cytochrome P450, glutathione S-transferase, and vitellogenin in the marine copepod T

Phototoxicity and oxidative stress responses in Daphnia magna under exposure to sulfathiazole and environmental level ultraviolet B irradiation.

Sulfonamide antibiotics frequently occur in aquatic environments. In this study, phototoxicity of sulfathiazole (STZ) and its mechanism of action were investigated using Daphnia magna. We evaluated the changes of molecular level stress responses by assessing gene expression, enzyme induction and lipid peroxidation, and the related organism-level effects in D. magna. In the presence of ultraviolet B (UV-B) light (continuous irradiation with 13.8+/-1.0microWcm(-2)d(-1)), STZ (at the nominal concentration of 94.9mg/L) caused a significant increase in reactive oxygen species (ROS) generation and l

Stage-specific transcriptomic analysis reveals insights into the development, reproduction and biological function of allergens in the European house dust mite Dermatophagoides pteronyssinus.

BACKGROUND: House dust mites (HDMs) such as Dermatophagoides pteronyssinus are major allergy elicitors worldwide, yet their gene expression across developmental stages remains underexplored. Herein, we report a comprehensive RNAseq analysis of larvae, nymphs, and adult males and females, mapped to a recently published high-quality genome with extended functional annotations. RESULTS: Analysis of differentially expressed genes (DEG) revealed that female-biased expression was the most prevalent profile (16% of genes), while males exhibited the highest fold-change differences. DEG data, combined

A Glycoprotein Hormone Orchestrates Ovarian Development in the Fall Armyworm Spodoptera Frugiperda.

Glycoprotein hormones serve as critical regulators of diverse physiological processes in both vertebrates and invertebrates, yet their functional roles in insect reproduction remain poorly understood. Here, we identified two glycoprotein hormone subunits, SfLFT1 and SfLFT2, and their cognate receptor SfLFTR (a leucine-rich-repeat-containing GPCR) in Spodoptera frugiperda. Liposome-encapsulated siRNA-mediated knockdown of these genes severely disrupted ovarian development as evidenced by suppressed vitellogenin biosynthesis and reduced mature follicle formation. Identification and functional characterization of a novel ovarian-specific Kelch domain-containing gene involved in the ecdysteroid signaling pathway of Penaeus monodon.

In crustaceans, vitellogenin (Vg) synthesis is regulated by complex hormonal and molecular networks. This study investigates the role of the ecdysone receptor (PmEcR) in regulating Vg expression and identifies downstream effectors in Penaeus monodon. Silencing of PmEcR using RNA interference resulted in a two-fold increase in Vg expression and a significant rise in the gonadosomatic index (GSI), suggesting PmEcR acts as a suppressor of vitellogenesis. Through a suppression subtractive hybridization (SSH) technique, an uncharacterized LOC 113805388 gene, was identified as being downregulated up

Biological Functions of Growth Differentiation Factor 9 in Early Ovarian Development of Japanese Eel (Anguilla japonica).

Growth differentiation factor 9 (GDF9) is a member of the transforming growth factor-β (TGF-β) superfamily and is expressed in an oocyte-specific manner. It plays a crucial role in the early stage of ovarian development. Japanese eel (Anguilla Japonica), a spawning migration teleost, has artificial reproduction still under investigation. Aimed at developing a novel method for the successful artificial reproduction of Japanese eel, in the present study, the role of GDF9 in the regulation of early ovarian development was investigated. The Gdf9 gene in Japanese eel was 1293 bp in length, coding f

17α-Ethynylestradiol alters the peritoneal immune response of gilthead seabream.

17α-Ethynylestradiol (EE2), a synthetic estrogen used in most oral contraceptives pills and hormone replacement therapies, is found in many water bodies, where it can modulate the fish immune response. EE2 acts as an endocrine disruptor in gilthead seabream, Sparus aurata L., a marine teleost fish of great economic value in Mediterranean aquaculture, as it induces hepatic vitellogenin gene (vtg) expression. Moreover, EE2 also alters the capacity of gilthead seabream to appropriately respond to infection although it does not behave as an immunosuppressor. Nevertheless, these previous studies ha

Endocrine Disruption throughout the Hypothalamus-Pituitary-Gonadal-Liver (HPGL) Axis in Marine Medaka (Oryzias melastigma) Chronically Exposed to the Antifouling and Chemopreventive Agent, 3,3'-Diindolylmethane (DIM).

Despite being proposed as a promising antifouling and chemopreventive agent, the environmental risks of 3,3'-diindolylmethane (DIM) are scarcely investigated. Therefore, this study used adult marine medaka (Oryzias melastigma) as a model organism to examine the toxicological effects and underlying mechanism of DIM throughout the hypothalamus-pituitary-gonadal-liver (HPGL) axis following 28 days of exposure to low DIM concentrations (0 and 8.46 μg/L). The results showed that altered gene transcription in the hypothalamus, pituitary, and gonads contributed to the great imbalance in hormone homeo

Tissue responsiveness to estradiol and genistein in the sea bass liver and scale.

As in mammals, estrogens in fish are essential for reproduction but also important regulators of mineral homeostasis. Fish scales are a non-conventional target tissue responsive to estradiol and constitute a good model to study mineralized tissues effects and mechanisms of action of estrogenic compounds, including phytoestrogens. The responsiveness to estradiol and the phytoestrogen genistein, was compared between the scales and the liver, a classical estrogenic target, in sea bass (Dicentrarchus labrax). Injection with estradiol and genistein significantly increased circulating vitellogenin (

Alternations in neuroendocrine and endocrine regulation of reproduction in male goldfish (Carassius auratus) following an acute and chronic exposure to vinclozolin, in vivo.

The fungicide vinclozolin (VZ) is in use globally and known to disrupt reproductive function in male. The present study tested the hypothesis that VZ disrupts testicular function in goldfish (Carassius auratus) by affecting brain-pituitary-testis axis. Goldfish were exposed to 100, 400 and 800 μg/L VZ and 5 μg/L 17β-estradiol (E2) for comparison. In VZ treated goldfish, 11-ketotesteosterone (11-KT) secretion was changed depending on dose and duration period of treatment. Following 7 days of exposure, 11-KT was decreased in goldfish exposed to 800 μg/L VZ, while it was increased in goldfish exp

Derivation of an aquatic predicted no-effect concentration for the synthetic hormone, 17 alpha-ethinyl estradiol.

17alpha-Ethinyl estradiol (EE2) is a synthetic estrogen widely used in combination with other steroid hormones in oral contraceptives and in the contraceptive patch. EE2 has been detected in sewage treatment plant effluents in the low nanogram -per-liter range and occasionally in surface waters in the U.S., U.K., Canada, Brazil, Germany, and elsewhere. The mode of action is receptor-mediated, and estrogen receptors exist in mammals and other vertebrates. A large number of studies on the effects of EE2 on aquatic organisms exist. One hundred English language studies published between 1994 and 2

Transgenic tomato strategies targeting whitefly eggs from apoplastic or ovary-directed proteins.

BACKGROUND: Transgenic plants expressing proteins that target the eggs of the ubiquitous plant pest Bemisia tabaci (whitefly) could be an effective insecticide strategy. Two approaches for protein delivery are assessed using the mCherry reporter gene in transgenic tomato plants, while accommodating autofluorescence in both the plant, phloem-feeding whitefly and pedicle-attached eggs. RESULTS: Both transgenic strategies were segregated to homozygous genotype using digital PCR. The first strategy uses a glycotransferase secretion signal peptide. Despite bright apoplastic accumulation, mCherry i

Exposure to difenoconazole induces reproductive toxicity in zebrafish by interfering with gamete maturation and reproductive behavior.

Difenoconazole (DCZ) is a triazole fungicide that negatively affects aquatic organisms and humans. However, data regarding the reproductive toxicity of DCZ are insufficient. In this study, we used zebrafish (from 2 h post-fertilization [hpf] to adulthood) as a model to evaluate whether DCZ at environmentally relevant concentrations (0.1, 1.0, and 10.0 μg/L) induces reproductive toxicity. After exposure to DCZ, egg production and fertilization rates were reduced by 1.0 and 10.0 μg/L. A significant decrease in gamete frequency (late vitellogenic oocytes and spermatozoa) was observed at 10.0 μg/L

Effects of Imidacloprid and Varroa destructor on survival and health of European honey bees, Apis mellifera.

There has been growing concern over declines in populations of honey bees and other pollinators which are a vital part to our food security. It is imperative to identify factors responsible for accelerated declines in bee populations and develop solutions for reversing bee losses. While exact causes of colony losses remain elusive, risk factors thought to play key roles are ectoparasitic mites Varroa destructor and neonicotinoid pesticides. The present study aims to investigate effects of a neonicotinoid pesticide Imidacloprid and Varroa mites individually on survivorship, growth, physiology,
