The genus Diapus Chapuis was erected (Chapuis 1865) for four species of pinhole borer (Curculionidae: Platypodinae) from the Oriental region and New Guinea. It was distinguished from other platypodine genera primarily by the widely separated procoxae (Chapuis 1865). Hopkins (1914) designated Diapus quadrispinatus Chapuis, 1865 as the type species of the genus. The genus is currently placed in the platypodine tribe Tesserocerini, subtribe Diapodina (Alonso-Zarazaga & Lyal 2009). Only two genera are included in the Diapodina, Diapus and Genyocerus Motschulsky (Alonso-Zarazaga & Lyal 2009, Jordal

Coffee berry borer, Hypothenemus hampei (Ferrari) is the most serious insect pest of coffee around the world. Although it is already present in most of the world's major coffee growing regions, it is important to delay further spread and to prevent reintroductions that might include hyperparasites or improve the genetic base of existing populations. Green coffee is shipped around the world for custom blending and roasting and such shipments carry the risk of spreading H. hampei. We used heavily infested coffee berries as a surrogate for green coffee to test the freezing tolerance of H. hampei.

Cysteine proteinases from larvae of the common bean weevil, Acanthoscelides obtectus (Coleoptera: Bruchidae), were isolated by ion exchange affinity chromatography on a CM-Cellulose column and used to select mutant cystatins from a library made with the filamentous M13 phage display system. The library contained variant cystatins derived from the nematode Onchocerca volvulus cystatin through mutagenesis of loop 1, which contains the QVVAG motif that is involved in binding to proteinases. After three rounds of selection, the activity of variant cystatins against papain and cysteine proteinases

Cholesterol oxidase is a bacterial FAD-containing flavooxidase that catalyzes the first reaction in cholesterol catabolism. Indeed, this enzyme catalyzes two reactions: the oxidation of the C(3)-OH group of cholesterol (and other sterols) to give cholest-5-en-3-one; and its isomerization to cholest-4-en-3-one. In the past several years, the structural and functional characterization of cholesterol oxidase has been developed together with its application as a biological tool. Cholesterol oxidase has been used in biocatalysis for the production of a number of steroids, as an insecticidal protein

The ambrosia beetle Xyleborus volvulus Fabricius has been reported as a potential vector of the plant pathogen Raffaelea lauricola T.C. Harr., Fraedrich & Aghayeva that is affecting avocado orchards in South Florida. In this study, we examined its life cycle, process of gallery formation, gallery structure, and fungal associates by rearing one generation on avocado sawdust medium under control conditions. The adult foundress excavated a vertical tunnel that constituted the main gallery with a length of 2.5 cm, followed by the construction of up to six secondary galleries with a total length of

Ambrosia beetles have emerged as significant pests of avocado ((Persea americana Mill. [Laurales: Lauraceae])) due to their association with pathogenic fungal symbionts, most notably Raffaelea lauricola T.C. Harr., Fraedrich & Aghayeva (Ophiostomatales: Ophiostomataceae), the causal agent of the laurel wilt (LW) disease. We evaluated the interaction of ambrosia beetles with host avocado trees by documenting their flight height and daily flight periodicity in Florida orchards with LW. Flight height was assessed passively in three avocado orchards by using ladder-like arrays of unbaited sticky t

Studies of symbioses have traditionally focused on explaining one-to-one interactions between organisms. In reality, symbioses are often much more dynamic. They can involve many interacting members, and change depending on context. In studies of the ambrosia symbiosis-the mutualism between wood borer beetles and fungi-two variables have introduced uncertainty when explaining interactions: imprecise symbiont identification, and disregard for anatomical complexity of the insects. The black twig borer, Xylosandrus compactus Eichhoff, is a globally invasive ambrosia beetle that infests >200 plant

Flight and emergence of the redbay ambrosia beetle, Xyleborus glabratus Eichhoff, were monitored from March 2011 through August 2012 using Lindgren funnel traps baited with manuka oil and emergence traps attached over individual beetle galleries on infested redbay (Persea borbonia (L.) Sprengel) trees. Of the 432 gallery entrances covered with emergence traps, 235 (54.4%) successfully produced at least two adults. Gallery success rates and time until adult emergence were highly variable and strongly depended on time of year galleries were initiated. Successful galleries produced 23.4 ± 2.50 (x

Xyleborus affinis Eichthoff is a neotropical ambrosia beetle that, in certain regions such as the United States and Mexico, has been associated with exotic phytopathogenic fungi causing extensive tree mortality. Although studies relating its olfactory response to volatile compounds and trapping systems have been published, factors such as the insect's physiological condition, which can affect its recognition or response to odors, have not been studied. Here, we evaluated the electroantennographic (EAG) response of wild and laboratory reared X. affinis females to 70% ethanol, a compound known t

Predatory long-legged flies of the genus Medetera are important, but currently understudied, natural enemies of Scolytinae bark beetles such as Ips typographus. Medetera flies lay eggs on beetle-infested trees, where the developing larvae find their prey, but the chemical cues used by Medetera to locate infested trees are currently unknown. To identify odors attracting Medetera signaticornis, a species in Europe, headspace samples were collected at several time-points through different stages of I. typographus attacks on logs of Norway spruce (Picea abies). The headspace samples were analyzed

Temperature has strong effects on metabolic processes of individuals and demographics of populations, but effects on ecological communities are not well known. Many economically and ecologically important pest species have obligate associations with other organisms; therefore, effects of temperature on these species might be mediated by strong interactions. The southern pine beetle (Dendroctonus frontalis Zimmermann) harbors a rich community of phoretic mites and fungi that are linked by many strong direct and indirect interactions, providing multiple pathways for temperature to affect the sys

INTRODUCTION: Sweet potato weevils (Cylas formicarius) are devastating pest that seriously affect sweet potato production, and their feeding can significantly reduce sweet potato yield and quality.
METHODS: To elucidate the molecular mechanisms underlying sweet potato responses to C. formicarius herbivory, the resistant cultivar J2 and the susceptible cultivar G12 were selected for integrated transcriptomic and metabolomic analyses of leaves and storage roots during the early stage of infestation.
RESULTS: The results demonstrated that, in storage roots, the resistant J2 exhibited a greater

The development of sustainable routes for metallic nanocatalysts remains challenging due to the reliance on toxic reagents and harsh synthesis conditions. To address this, a green and efficient method was established for the preparation of palladium nanoparticles (PdNPs@Fu) using fucoidan, a naturally occurring polysaccharide with several hydroxyl and sulfate groups as a stabilizing and reducing agent. Fucoidan can rapidly reduce Pd2+ to Pd0 in aqueous medium under microwave irradiation (450 W, 10 min). The resulting PdNPs@Fu exhibited uniform crystalline nanoparticles (sizes ranging from 2-6

The performances of traps used for pest monitoring are not well understood. Variable trap sizes and shapes make cross-comparisons difficult. We tested with four Scolytinae species (Coleoptera, Curculionidae) a set of traps of the same model (fan-traps) in four sizes (310, 536, 832.5, 1,200 cm2), and a larger trap with another design (a four-vane trap, 4,500 cm2). A significantly linear relationship was observed between trap sizes and catches for Pityogenes chalcographus, but the catches of Ips typographus, Trypodendron lineatum and T. signatum tapered off. In the literature, comparable taperin

Florivores consume floral structures with negative effects on plant fitness and pollinator attraction. Several studies have evaluated these consequences in hermaphroditic plants, but little is known about the effects on monoecious and dioecious species. We characterize the florivory and its effects on floral visitors and reproductive success in a monoecious population of Sagittaria lancifolia. Five categories of florivory were established according to the petal area consumed. Visits were recorded in male and female flowers within the different damage categories. Reproductive success was evalua

Vigna unguiculata is an important source of proteins and energy for humans and animals. However, postharvest losses caused by Callosobruchus maculatus can reach from 20 to 100% of stored seeds. In this study, the insecticide potential of compounds extracted from Himatanthus drasticus latex was assessed. The latex was extracted with ethanol (70%) and then partitioned through sequential use of hexane and chloroform. These fractions were investigated by chromatography to determine their chemical composition. Plumieride, identified in a hydroalcoholic subfraction, was tested for insecticidal activ

Digestive alpha-amylases play an essential role in insect carbohydrate metabolism. These enzymes belong to an endo-type group. They catalyse starch hydrolysis, and are involved in energy production. Larvae of Zabrotes subfasciatus, the Mexican bean weevil, are able to infest stored common beans Phaseolus vulgaris, causing severe crop losses in Latin America and Africa. Their alpha-amylase (ZSA) is a well-studied but not completely understood enzyme, having specific characteristics when compared to other insect alpha-amylases. This report provides more knowledge about its chemical nature, inclu

The obligate intracellular bacterium, Wolbachia pipientis (Rickettsiales), is a widespread, vertically transmitted endosymbiont of filarial nematodes and arthropods. In insects, Wolbachia modifies reproduction, and in mosquitoes, infection interferes with replication of arboviruses, bacteria and plasmodia. Development of Wolbachia as a tool to control pest insects will be facilitated by an understanding of molecular events that underlie genetic exchange between Wolbachia strains. Here, we used nucleotide sequence, transcriptional and proteomic analyses to evaluate expression levels and establi
