Is HSP70 upregulation crucial for cellular proliferative response in simulated microgravity?

Astronauts are susceptible to a variety of conditions such as motion sickness, muscular atrophy, bone demineralization and cardiovascular deconditioning. These findings suggest that the adaptation to the absence of gravity is due, at least in part, to the effects exerted by microgravity at the cellular level. Indeed, a number of studies have indicated that gravity affects mammalian cell growth and differentiation through the modulation of gene expression. We have characterized the behaviour of endothelial cells and of the human monocytic cell line U937 cultured in the NASA-developed bioreactor

Effects of clinostat-microgravity on bone and calcium metabolism in rats.

Decreases in bone minerals and tissue volume after space flight have been observed in humans and animals, with a variety of results. Such data obtained from space flight experiments have given unsatisfactory results due to short periods of space flight and differences in age, body weights, and strain of animals used. Therefore, ground-based animal models have been developed in order to elucidate changes in bone affected by space flight. For example, a tail-suspended rat model has been established to study the effects of microgravity on bones by producing hind limb unloading. However, problems

Effect of head-down-tilt bed rest and hypovolemia on dynamic regulation of heart rate and blood pressure.

Adaptation to head-down-tilt bed rest leads to an apparent abnormality of baroreflex regulation of cardiac period. We hypothesized that this "deconditioning response" could primarily be a result of hypovolemia, rather than a unique adaptation of the autonomic nervous system to bed rest. To test this hypothesis, nine healthy subjects underwent 2 wk of -6 degrees head-down bed rest. One year later, five of these same subjects underwent acute hypovolemia with furosemide to produce the same reductions in plasma volume observed after bed rest. We took advantage of power spectral and transfer functi

Modeled microgravity increases filamentation, biofilm formation, phenotypic switching, and antimicrobial resistance in Candida albicans.

Candida albicans is an opportunistic fungal pathogen responsible for a variety of cutaneous and systemic human infections. Virulence of C. albicans increases upon exposure to some environmental stresses; therefore, we explored phenotypic responses of C. albicans following exposure to the environmental stress of low-shear modeled microgravity. Upon long-term (12-day) exposure to low-shear modeled microgravity, C. albicans transitioned from yeast to filamentous forms at a higher rate than observed under control conditions. Consistently, genes associated with cellular morphology were differential

Cardiorespiratory responses to physical work during and following 17 days of bed rest and spaceflight.

To determine the influence of a 17-day exposure to real and simulated spaceflight (SF) on cardiorespiratory function during exercise, four male crewmembers of the STS-78 space shuttle flight and eight male volunteers were studied before, during, and after the 17-day mission and 17 days of -6 degrees head-down-tilt bed rest (BR), respectively. Measurements of oxygen uptake, pulmonary ventilation, and heart rate were made during submaximal cycling 60, 30, and 15 days before the SF liftoff and 12 and 7 days before BR; on SF days 2, 8, and 13 and on BR days 2, 8, and 13; and on days 1, 4, 5, and 8

Effects of artificial gravity on the vascular system during 60 days of strict head-down tilt bedrest.

During spaceflight, the lack of gravity-induced hydrostatic pressure gradients in the body results in a headward fluid shift hypothesized to play a role in pathophysiological changes in the cardiovascular, ocular, and central nervous systems. Here, 24 subjects (n = 8 control) underwent 60 days of strict 6° head-down tilt (HDT) bedrest, an analog of spaceflight where we tested the ability of daily 30-min artificial gravity (AG) exposure via short-arm human centrifugation (intermittent AG, n = 8, and continuous AG, n = 8) to reverse the HDT-induced headward fluid shift. Effect of lower body negative pressure against orthostatic intolerance induced by 21 days head-down tilt bed rest.

BACKGROUND: Exposure to actual or simulated weightlessness is known to induce orthostatic intolerance in humans. Many different methods have been suggested to counteract orthostatic hypotension. The repetitive or prolonged application of lower body negative pressure (LBNP) has shown beneficial effects to counter orthostatic intolerance, but devoting so much time to countermeasures is not compatible with space mission objectives or costs. The purpose of the present study was to assess the effects of brief LBNP sessions against orthostatic intolerance during a 21-d head-down tilt (HDT) bed rest.

A high protein diet does not improve protein synthesis in the nonweight-bearing rat tibialis anterior muscle.

We recently demonstrated that a high protein intake partially prevented the decrease in protein synthesis in the atrophied dark soleus muscle of rats that were hindlimb suspended (HS) for 21 d. To study the possible role of protein intake in a muscle more representative of the whole musculature, we measured the effect of a high protein (HP) (30%) and a medium protein (MP) (15%) diet on protein synthesis in the pale fast-twitch tibialis anterior muscle of HS rats. The HS animals were suspended by the tail for 21 d so that only their front legs were able to rest on the floor. The fractional rate

Characteristics of metabolism during prolonged water immersion.

The effect on the organism of a 12-day stay in a water immersion medium (head on the water surface) was studied on 10 subjects. The condition of the metabolic processes in the subjects was judged from the investigation of the parameters of protein and carbohydrate metabolism, acid-base equilibrium, the activity of a number of enzymes and steroid hormones. The venous blood and diurnal urine served as the material for conducting the corresponding biochemical determinations. The stay in an immersion medium is accompanied by a significant increase in the content of residual nitrogen in the blood,

Orthostatic hypotension in patients, bed rest subjects, and astronauts.

Orthostatic hypotension after even short space flights has affected a significant number of astronauts. Given the need for astronauts to function at a high level of efficiency during and after their return from space, the application of pharmacologic and other treatments is strongly indicated. This report addresses the clinical problem of orthostatic hypotension and its treatments to ascertain whether pharmacologic or physiologic treatment may be useful in the prevention of orthostatic hypotension associated with space flight. Treatment of orthostatic hypotension in patients now includes incre

Sex Difference in Orthostatic Tolerance During Simulated Partial Gravity Head-Up Tilts and Efficacy of a Lower Body Compression Garment Countermeasure.

After adaptation to weightlessness, astronauts will experience head-to-foot accelerations (G) during descent to and ascent from the lunar surface that may provoke symptoms of orthostatic intolerance (OI). There were no reports consistent with OI among Apollo astronauts who experienced up to 10 minutes of 0.4-0.6 G, while standing without OI countermeasures during dynamic flight phases, though heart rate (HR) was elevated. The likelihood of developing OI in future Artemis crews, which will include females, may be greater given observations of increased OI in females and potentially greater acce

Ankle Somatosensation and Lower-Limb Neuromuscular Function on a Lunar Gravity Analogue.

Background/Objectives: The adverse effects of low gravity on human physiology are well documented; however, much of the literature is directed at changes which occur in microgravity (µg: weightlessness) with relatively less documented on changes in hypogravity (<1 g; >µg: gravity less than Earth's but more than microgravity). Somatosensation and neuromuscular control may be of particular importance for astronauts as they prepare for future missions to walk on the Moon. This study aimed to explore the effect of reduced weight bearing (to simulate conditions of hypogravity) on ankle somatosensat

From Collapse to Active Self-Repair: Integrative Multi-Omics and Machine Learning Analysis Map the Hepatic Metabolic Adaption in Response to Simulated Spaceflight Stress.

Long-term spaceflight poses substantial challenges to human physiology, with the liver being highly susceptible due to its central metabolic role. To determine whether hepatic alterations represent transient stress or sustained remodeling, we performed an integrated multi-omics analysis in a rat model simulating chronic space radiation and microgravity. Herein, we applied an integrated multi-omics and AI-driven analytical framework combining histopathology, cytokine and miRNA profiling, proteomics, metabolomics and Western blot validation. After 21 days of simulated space conditions, rats exhi

The change in phase shift between respiratory oscillations of mean arterial pressure and heart rate in head-up tilt test: The effect of simulated microgravity.

PURPOSE: We studied the synchronization of mean arterial pressure (MAP) and heart rate (HR) high frequency (HF) respiratory oscillations in young men in the supine position and during head-up tilt (HUT), and then analyzed the effect of blood redistribution on these parameters in the lower body negative pressure (LBNP) test, sit-to-stand test and HUT test performed after a 7-day dry immersion (DI, on-ground model of microgravity). METHODS: 75 young male volunteers participated in the study. Blood pressure and ECG were continuously recorded, to obtain mean arterial pressure (MAP), heart rate (H

Moxifloxacin plus Cordyceps polysaccharide ameliorate intestinal barrier damage due to abdominal infection via anti-inflammation and immune regulation under simulated microgravity.

BACKGROUND: Currently, there is limited research on the impact of abdominal infection on intestinal damage under microgravity conditions. Cordyceps polysaccharide (CPS), the main active ingredient of Cordyceps, has demonstrated various pharmacological effects, including anti-inflammatory, antioxidant, and immunomodulatory properties. Moxifloxacin (MXF) is a fourth-generation quinolone antibiotic that is believed to have a dual regulatory effect on immune system activation and suppression. Our objective was to investigate the effects of MXF plus CPS on the intestinal barrier damage due to abdom

Synergistic role of hydroxyapatite nanoparticles and pulsed electromagnetic field therapy to prevent bone loss in rats following exposure to simulated microgravity.

The purpose of the present study was to use capacitive coupling of pulsed electromagnetic field (CC-PEMF) and hydroxyapatite nanoparticles (HAp) as a countermeasure to prevent osteoporosis induced by simulated microgravity. We used the hind-limb suspension (HLS) rat model to simulate microgravity-induced bone losses for 45 days. In order to compare the resulting changes, mineralogical (bone mineral density [BMD], calcium [Ca], and phosphorus [P]), biochemical (osteocalcin, alkaline phosphatase [ALP], and type I collagen), and histological (scanning electron microscopy) parameters were adopted.

Effects of simulated microgravity on the morphology and function of neonatal porcine cell clusters cultured with and without Sertoli cells.

Human islet allografts are well known to induce full and sustained remission of hyperglycemia, with complete normalization of key metabolic parameters. Nevertheless, acquiring human islets, even from cadaveric human donor pancreases, remains a significant impediment to successful transplantation therapy for diabetes. To overcome this difficulty, neonatal porcine cell clusters (NPCCs) have been considered for human islet substitutes because they are easily obtained by collagenase digestion of the neonatal piglet pancreas. Currently, the major hurdle in using NPCCs for xenograft is the delay (ti

Microgravity tissue engineering.

Tissue engineering studies were done using isolated cells, three-dimensional polymer scaffolds, and rotating bioreactors operated under conditions of simulated microgravity. In particular, vessel rotation speed was adjusted such that 10 mm diameter x 2 mm thick cell-polymer constructs were cultivated in a state of continuous free-fall. Feasibility was demonstrated for two different cell types: cartilage and heart. Conditions of simulated microgravity promoted the formation of cartilaginous constructs consisting of round cells, collagen and glycosaminoglycan (GAG), and cardiac tissue constructs
