Research progress and prospect of β-carotene hydroxylase gene mediating plant carotenoid metabolism in response to abiotic stress.

Carotenoid metabolism plays a significant role in plants response to abiotic stress. The β-carotene hydroxylase gene (BCH) is an important gene located downstream of the carotenoid metabolic pathway. Its main function is to hydroxylate β-carotene to form zeaxanthin, which is the direct valve regulating the carotenoid xanthophyll cycle and also an important controller in the upstream of ABA synthesis. Most carotene components contain a molecular structure with conjugated double bonds, which determines that they have a powerful antioxidant function. When the BCH gene is overexpressed, Reactive o

Lutein Modulates Stress-Responsive Signaling Pathways in THLE-2 Human Hepatocytes Under Intestinal Failure-Associated Liver Disease Conditions.

Intestinal dysfunction and parenteral nutrition (PN) can trigger a spectrum of liver disorders collectively referred to as intestinal failure-associated liver disease (IFALD), for which therapeutic options remain limited. In the present study, we investigated the modulatory effects of the bioactive xanthophyll carotenoid lutein in an in vitro IFALD model utilizing human THLE-2 hepatocytes exposed to lipopolysaccharide and Intralipid to mimic PN-associated inflammatory and metabolic stress. Because lutein is poorly water-soluble and patients receiving PN lack enteral intake of this compound, we

Astaxanthin and Male Reproductive Health: Mechanisms, Experimental Evidence, and Clinical Perspectives.

Oxidative stress is a primary driver of male infertility, underscoring the critical importance of effective antioxidant defense systems in maintaining reproductive function. Astaxanthin, a xanthophyll carotenoid with potent free radical-scavenging capacity and membrane-stabilizing properties, has garnered interest as a therapeutic agent for male reproductive health. Experimental evidence indicates that astaxanthin modulates cellular redox homeostasis, preserves mitochondrial integrity, and attenuates lipid peroxidation in spermatozoa and testicular cells. Through these mechanisms, it potential

Mechanism of Lutein to meso-Zeaxanthin Isomerization by RPE65 Catalysis.

The macular pigments lutein (L), zeaxanthin (Z), and meso-zeaxanthin (MZ) protect the human retina from light and oxidative stress. While L and Z are abundant in the human diet, MZ is nearly absent. We previously demonstrated MZ is derived from L in chicken embryos precisely timed with expression of RPE65. Herein, we show that RPE65 from mouse, an animal that does not concentrate MZ in the eye, catalyzes L to MZ isomerization similarly as found for RPE65 from chicken and human, when expressed in cultured cells. Co-expression with xanthophyll-binding proteins had no impact on MZ yield. L and MZ

Haematococcus pluvialis-Derived Astaxanthin Is a Potential Neuroprotective Agent against Optic Nerve Ischemia.

Astaxanthin, a xanthophyll belonging to the family of carotenoids, is a potent antioxidant. However, much less is known about its protective effects on the oxidative stress of ischemic optic nerve. We hypothesized that astaxanthin treatment could protect retinal ganglion cells (RGCs) from death via anti-oxidative and anti-apoptotic responses. Adult male Wistar rats were fed astaxanthin (100 mg/kg/day) by daily gavage for seven consecutive days, either before or after inducing oxidative stress in the retina by photodynamic treatment. The visual function, RGC apoptosis, macrophage infiltration i

Metal resistant PGPR lowered Cd uptake and expression of metal transporter genes with improved growth and photosynthetic pigments in Lycopersicon esculentum under metal toxicity.

Plant growth promoting rhizobacteria (PGPRs) are very effective in immobilization of heavy metals and reducing their translocation in plants via precipitation, complex formation and adsorption. The present study was therefore designed to understand the role of Pseudomonas aeruginosa and Burkholderia gladioli in mitigation of Cd stress (0.4 mM) in 10-days old L. esculentum seedlings. The present work investigated growth characteristics, photosynthetic pigments, metal tolerance index, metal uptake and the contents of metal chelating compounds (protein bound and non-protein bound thiols, total th

Histone deacetylase 9 plays a role in the antifibrogenic effect of astaxanthin in hepatic stellate cells.

Activation of hepatic stellate cells (HSCs) is critical for liver fibrosis development. Previously, we showed that astaxanthin (ASTX), a xanthophyll carotenoid, has antifibrogenic effects in LX-2 cells, a human HSC cell line. We sought to determine the effect of ASTX on HSC activation, and to identify molecular mediators that are critically involved in the processes. ASTX prevented the activation of mouse primary HSCs, as evidenced by attenuated induction of procollagen type I α1. In human primary HSCs, ASTX also inhibited transforming growth factor β1 (TGFβ1)-induced fibrogenic gene expressio

From shrimp reddening to spoilage indication: Shrimp-shell-derived crustacyanin as a multi-trigger responsive chromoprotein.

Natural colorimetric freshness indicators are commonly based on pH-sensitive pigments, although pH changes do not always match food quality deterioration. Here, shrimp-shell-derived crustacyanin was investigated as a multi-trigger spoilage-responsive chromoprotein rather than a conventional pH indicator. The recovery process was designed to obtain sufficient protein while preserving the astaxanthin-apoprotein binding environment required for color response. Using Macrobrachium rosenbergii shells, salting-out extraction under mild conditions gave a crustacyanin yield of 1.49 ± 0.11 mg/g and a t

pH-responsive emulsion gels based on low acyl gellan gum stabilized by yeast protein for enhanced astaxanthin protection.

Astaxanthin (AXT), a powerful natural antioxidant, suffers from limited oral bioavailability owing to its chemical lability and weak water solubility. Microbial-derived biopolymers have received increasing attention for sustainable emulsification in food delivery systems. In this work, highly bioactive AXT was extracted from Haematococcus pluvialis using soybean oil as a green solvent. pH-responsive emulsion gels were fabricated using heat-treated yeast proteins (HYPs) as Pickering stabilizers and low-acyl gellan gum (LAG) as an in situ gelling agent to enhance AXT stabilization. Integrated analyses of hepatopancreatic response induced by high temperature and its alleviation by astaxanthin in Sinosolenaia oleivora.

High temperature (HT) is a growing threat to freshwater mollusks under climate change, yet its toxicological mechanisms and mitigation strategies remain poorly understood. This study investigated the hepatopancreatic response of an endangered bivalve Sinosolenaia oleivora to acute HT stress (35 °C for 6 h) using transcriptomic, metabolomic, and physiological biomarker analyses. Significant hepatopancreatic injury was demonstrated by TUNEL assay and histological analysis. Moreover, HT triggered severe oxidative stress, indicated by elevated antioxidant enzyme activities (SOD, CAT, GSH, T‑AOC) a

Astaxanthin administration attenuates capillary regression in soleus muscles of rats with streptozotocin-induced diabetes.

AIMS: Diabetes induces skeletal muscle atrophy and capillary regression, especially in oxidative muscles, such as the soleus (SOL). Capillary regression impairs oxygen and substrates delivery, and contributes to metabolic dysfunction and vascular complications. Although protective effects of antioxidants have been demonstrated in disuse models, their role in diabetes-induced capillary regression remains unclear. We examined whether dietary astaxanthin supplementation attenuates capillary regression in the SOL muscles of rats with streptozotocin-induced diabetes. METHODS: Twenty-four male Wist

The Glow Up: Optimizing Growth Conditions and Carotenoid Profiling in Euglena gracilis.

Until now, only five photoactive pigments have been identified in Euglena gracilis and there has been contradictory reporting of the major carotenoid. The impact of various environmental factors on the carotenoid content of E. gracilis has not been well studied. Using an improved HPLC UV-VIS method, combined with mass spectrometry, we have now identified 12 photoactive pigments from E. gracilis extracts and confirmed diadinoxanthin as the major carotenoid produced in E. gracilis. After testing cultivation media and major carbon sources (addition of glucose-2-20 g/L, ethanol-0.2%-2.0% w/v), we

Multilamellar nanocochleates as novel co-encapsulation systems for enhancement of the physicochemical properties and controlled release of astaxanthin and phycocyanin.

Nanoliposomes that co-encapsulate astaxanthin and phycocyanin were treated with calcium ions (Ca2+) to generate nanocochleates. Zeta potential results revealed an electrostatic attraction between Ca2+ and the negatively charged nanoliposomes. Dynamic light scattering, SEM, TEM, FTIR, and DSC results demonstrated the effective deposition of Ca2+ onto the surface of nanoliposomes. Laurdan fluorescence spectroscopy and X-ray diffraction patterns showed that Ca2+ can reduce membrane fluidity and improve lateral lipid packing. Additionally, the antioxidant activity of the control nanoliposomes (22.

[Search for a new geroprotector: study of the effect of fucoxanthin on the main markers of aging in mice.].

Today, one of the key tasks of gerontology is the search for geroprotectors that are highly effective and safe. This paper presents data on the effect of fucoxanthin, a natural carotenoid, on aging markers in mice. Results are presented demonstrating the potential geroprotective activity of fucoxanthin. In particular, a decrease in the concentration of malondialdehyde in the liver by 17%, an increase in the total antioxidant activity of blood plasma by 24%, and a decrease in the levels of p16, p21 and p53 in the myocardium and spleen by 13-20% were observed. In addition, a decrease in proinfla

Unlocking the anti-aging potential: In silico analysis of astaxanthin, curcumin, quercetin, and resveratrol in modulating skin aging pathways.

Multiple studies have linked aging to a result of the inflammatory response. Thus, there is a recognized need for cosmeceuticals that modulate inflammation pathways to prevent and treat aging. In this sense, four bioactive compounds were selected for their documented anti-inflammatory/antioxidant properties. Therefore, this study aimed to evaluate whether the bioactive compounds astaxanthin, curcumin, quercetin, and resveratrol are effective in treating the effects of skin aging, using in silico analyses. Protein-protein interaction networks (PPINs) related to skin aging and the bioactive comp

From Ecological Threat to Bioactive Resource: The Nutraceutical Components of Blue Crab (Callinectes sapidus).

Native to the western Atlantic, the Atlantic blue crab Callinectes sapidus (CS) has spread to the Mediterranean, affecting local ecosystems and mussel aquaculture and leading to severe ecological and financial losses in Italy and other areas. Given its rapid spread and socio-economic impacts, several countries have begun to exploit CS commercially, but the consumers' interest is still limited. In this study, we analysed both nutrient and nutraceutical profiles of CS meats, evaluating potential differences related to sex and meat typology. We found that CS meats are rich in high quality protein

Engineered Microalgal Extracellular Vesicles for Enhancing Mitochondrial Homeostasis in Radiodermatitis Prevention.

Radiodermatitis, one of the most prevalent side effects of cancer radiotherapy, is characterized by cellular oxidative stress, mitochondrial damage, and inflammatory responses. In this study, we isolated extracellular vesicles (EVs) from the natural microalgae Spirulina platensis (SP) and engineered them by loading astaxanthin (AST) into SP-EVs, resulting in the formation of SP-EVs@AST. This engineered system significantly enhanced the solubility and stability of AST while preserving the structural integrity and biological activity of SP-EVs, thereby enabling the complementary and synergistic

Astaxanthin ameliorates oxidative stress in lens epithelial cells by regulating GPX4 and ferroptosis.

Ferroptosis is a form of regulated cell death closely associated with oxidative stress and mitochondrial dysfunction and is characterised by the accumulation of reactive oxygen species (ROS) and lipid species and iron overload. Damage to human lens epithelial cells (LECs) is associated with age-related cataract progression. Astaxanthin (ATX), a carotenoid with natural antioxidant properties, counteracts ferroptosis in the treatment of various degenerative diseases. However, this mechanism has not been reported with respect to cataract treatment. In this study, the differential expression level

Effects of astaxanthin on metastasis suppressors in ductal carcinoma. A preliminary study.

BACKGROUND: Breast cancer (BC) is a major public health problem diagnosed in more than 2 million women worldwide in 2018, causing more than 600,000 deaths. 90% of deaths due to breast cancer are caused by metastasis. Metastasis is a complex process that is divided into several steps, including separation of tumor cells from the primary tumor, invasion, cell migration, intravasation, vasculature survival, extravasation, and colonization of the secondary site. Astaxanthin (AXT) is a marine-based ketocarotenoid that has many different potential functions such as anti-oxidant, anti-inflammatory an

Ameliorative effects of astaxanthin on brain tissues of alzheimer's disease-like model: cross talk between neuronal-specific microRNA-124 and related pathways.

Alzheimer's disease (AD) is a chronic, progressive, multifactorial, and the most common neurodegenerative disease which causes dementia and mental deterioration in the elderly. The available treatments for AD are not disease-modifying drugs and only provide symptomatic relief. Astaxanthin (ATX), a second-generation antioxidant, is a dark red carotenoid and exhibits the highest antioxidant capacity, anti-inflammatory, neuroprotective, and antiapoptotic effects. In this study, we investigated the therapeutic effect of different doses of ATX on the cerebral cortex and hippocampus of AD-like rats.
