Integrin ligation initiates intracellular signaling events, among which are the activation of protein tyrosine kinases. The related adhesion focal tyrosine kinase (RAFTK), also known as PYK2 and CAKbeta, is a tyrosine kinase that is homologous to the focal adhesion kinase (FAK) p125FAK. The structure of RAFTK is similar to p125FAK in that it lacks a transmembrane region, does not contain Src homology 2 or 3 domains, and has a proline-rich region in its C terminus. Here we report that RAFTK is a target for beta1-integrin-mediated tyrosine phosphorylation in both transformed and normal human B c

Neural Wiskott-Aldrich syndrome protein (N-WASP) has been implicated in endocytosis; however, little is known about how it interacts functionally with the endocytic machinery. Sucrose gradient fractionation experiments and immunofluorescence studies with anti-N-WASP antibody revealed that N-WASP is recruited together with clathrin and dynamin, which play essential roles in clathrin-mediated endocytosis, to lipid rafts in an epidermal growth factor (EGF)-dependent manner. Endophilin A (EA) binds to dynamin and plays an essential role in the fission step of clathrin-mediated endocytosis. In the

Mutations in PKD1, the gene encoding for the receptor Polycystin-1 (PC-1), cause autosomal dominant polycystic kidney disease (ADPKD). The cytoplasmic C-terminus of PC-1 contains a coiled-coil domain that mediates an interaction with the PKD2 gene product, Polycystin-2 (PC-2). Here we identify a novel domain in the PC-1 C-terminal tail, a polyproline motif mediating an interaction with Src homology domain 3 (SH3). A screen for interactions using the PC-1 C-terminal tail identified the SH3 domain of nephrocystin-1 (NPHP1) as a potential binding partner of PC-1. NPHP1 is the product of a gene th

Casitas B-lineage lymphoma-b (Cbl-b), a pivotal negative regulator of TCR signaling, is highly expressed in immune cells. Its inhibition potentiates immune-mediated antitumor effects. Herein, we constructed a potent fluorescent tracer, Tracer 2, based on the Cbl-b peptide inhibitor Pep 1. Utilizing this tracer, we established a fluorescence polarization (FP) assay with enhanced sensitivity, discriminative capacity, and precision for the activity evaluation and high-throughput screening of orthosteric Cbl-b Src homology 2 (SH2) domain inhibitors. Leveraging this FP assay, we derived a potent ph

Liver fibrosis is characterised by the activation of hepatic stellate cells (HSCs) and matrix deposition. Accumulating evidence has revealed that the oncogenic protein tyrosine phosphatase Src homology 2 domain-containing phosphatase 2 (SHP2) acts as a therapeutic target of fibrosis. Although several SHP2 inhibitors have reached early clinical trials, there are currently no FDA-approved drugs that target SHP2. In this study, we aimed to identify novel SHP2 inhibitors from an in-house natural product library to treat liver fibrosis. Out of the screened 800 compounds, a furanogermacrane sesquite

SOCS7 is a member of the suppressor of cytokine signaling (SOCS) family of proteins (SOCS1-SOCS7 and CIS). SOCS proteins are composed of an N-terminal domain of variable length, a central Src homology 2 domain, and a C-terminal SOCS box. Biochemical and genetic studies have revealed that SOCS1, SOCS2, SOCS3, and CIS play an important role in the termination of cytokine and growth factor signaling. However, the biological actions of other SOCS proteins are less well defined. To investigate the physiological role of SOCS7, we have used gene targeting to generate mice that lack expression of the

The Wiskott-Aldrich syndrome is an inherited X-linked immunodeficiency characterized by thrombocytopenia, eczema, and a tendency toward lymphoid malignancy. Lymphocytes from affected individuals have cytoskeletal abnormalities, and monocytes show impaired motility. The Wiskott-Aldrich syndrome protein (WASP) is a multi-domain protein involved in cytoskeletal organization. In a two-hybrid screen, we identified the protein Cdc42-interacting protein 4 (CIP4) as a WASP interactor. CIP4, like WASP, is a Cdc42 effector protein involved in cytoskeletal organization. We found that the WASP-CIP4 intera

c-Abl is a key regulator of cell signaling and is under strict control via intramolecular interactions. In this study, we address changes in the intramolecular dynamics coupling within the c-Abl regulatory unit by presenting its N-terminal segment (N-Cap) with an alternative function in the cell as c-Abl becomes activated. Using small angle x-ray scattering, nuclear magnetic resonance, and confocal microscopy, we demonstrate that the N-Cap and the Src homology (SH) 3 domain acquire μs-ms motions upon N-Cap association with the SH2-L domain, revealing a stabilizing synergy between these segment

The rotational and translational diffusion behaviors of proteins are closely related to the processes of contact and complex formation between proteins. Previous studies have indicated that allosteric inhibitors of SH2 domain-containing phosphatase 2 (SHP2) could suppress its activation induced by the PD-1 protein. Nevertheless, the underlying mechanism governing this allosteric regulation remained poorly understood. In this study, we utilized nonequilibrium molecular dynamics (NEMD) simulations to investigate the potential relationship between the diffusion behavior of SHP2 and its allosteric

Airway epithelial mitochondrial injury plays a critical role in the pathogenesis of chronic obstructive pulmonary disease (COPD). The p66Shc adaptor protein is a newly recognized mediator of mitochondrial dysfunction. However, little is known about the effect of p66Shc on airway epithelial damage in the development of COPD. The aim of the present study is to investigate the roles of p66Shc and its upstream regulators in the mitochondrial injury of airway epithelial cells (Beas-2b) induced by cigarette smoke extract (CSE). Our present study revealed that CSE increased p66Shc expression and its

Chikungunya virus (CHIKV), a mosquito-borne alphavirus, causes febrile disease, muscle and joint pain, which can become chronic in some individuals. The non-structural protein 3 (nsP3) plays essential roles during infection, but a complete understanding of its function is lacking. Here we used a microscopy-based approach to image CHIKV nsP3 inside human cells. The SNAP system consists of a self-labelling enzyme tag, which catalyses the covalent linking of exogenously supplemented synthetic ligands. Genetic insertion of this tag resulted in viable replicons and specific labelling while preservi

X-linked lymphoproliferative syndrome (XLP) is an inherited immunodeficiency characterized by increased susceptibility to Epstein-Barr virus (EBV). In affected males, primary EBV infection leads to the uncontrolled proliferation of virus-containing B cells and reactive cytotoxic T cells, often culminating in the development of high-grade lymphoma. The XLP gene has been mapped to chromosome band Xq25 through linkage analysis and the discovery of patients harboring large constitutional genomic deletions. We describe here the presence of small deletions and intragenic mutations that specifically

Human parvovirus B19 infects erythroid progenitor cells and inhibits erythropoiesis. The small 11-kDa (s11-kDa) protein of the virus interacts with the host adaptor Grb2 (growth factor receptor-bound protein 2) and suppresses downstream kinase phosphorylation to promote viral genome replication; however, the underlying mechanism remains unclear. Here, we show that s11-kDa competitively disrupts the Grb2-SOS (Son of Sevenless) interaction by binding to the N-terminal SH3 domain of Grb2. Structural modeling indicated that the proline-rich region of the s11-kDa protein engage Grb2 molecules, pote

Growth factor receptor-bound protein 2 (Grb2) plays a critical role in proliferative diseases by binding phosphotyrosine residues on target proteins leading to activation of key pathways. The phosphotyrosyl motif of Focal Adhesion Kinase (FAK) has been utilized as a pharmacophore to identify small molecules that disrupt its interaction with the Grb2 Src Homology 2 (SH2) domain. In this study, we report the hit-to-lead optimization of 11,12,479 synthesizable analogs of previously identified Grb2-SH2 antagonists via virtual screening using AutoDock Vina v1.2.0. In budding yeast, partitioning of the cytoplasm during cytokinesis can proceed via a pathway dependent on the contractile actomyosin ring, as in other eukaryotes, or alternatively via a septum deposition pathway dependent on an SH3 domain protein, Hof1/Cyk2 (the yeast PSTPIP1 ortholog). In dividing yeast cells, Hof1 forms a ring at the bud neck distinct from the actomyosin ring, and this zone is active in septum deposition. We previously showed the yeast Wiskott-Aldrich syndrome protein (WASP)-interacting protein (WIP) ortholog, verprolin/Vrp1/End5, interacts with Hof1 and facilitates Hof1 rec

BACKGROUND: The Src homology 2 (SH2) domains of cytoplasmic signaling proteins generally bind phosphotyrosine (pTyr) sites in the context of carboxy-terminal residues. SAP (also known as SH2D1A or DSHP), the product of the gene that is mutated in human X-linked lymphoproliferative (XLP) disease, comprises almost exclusively a single SH2 domain, which may modulate T-cell signaling by engaging T-cell co-activators such as SLAM, thereby blocking binding of other signaling proteins that contain SH2 domains. The SAP-SLAM interaction can occur in a phosphorylation-independent manner.

The protein tyrosine phosphatase family comprises a large number of members. Among them, Src homology 2 (SH2) domain-containing protein tyrosine phosphatases (commonly referred to as the SHP family). Members of the SHP family regulate multiple signaling pathways by catalyzing the dephosphorylation of substrate proteins, thereby participating in fundamental biological processes such as immune regulation and cell proliferation. Their normal function is essential for maintaining organismal homeostasis. Extensive evidence indicates that dysregulation of SHP family members is closely associated wit

Clathrin-mediated endocytosis is a major cellular pathway for internalization of proteins and lipids and for recycling of synaptic vesicles. The GTPase dynamin plays a key role in this process, and the proline-rich domain of dynamin participates in various protein-protein interactions to ensure a proper coordination of endocytic processes. Although dynamin is not directly associated with actin, several dynamin-binding proteins can interact with actin or with proteins that regulate actin assembly, thereby coordinately regulating actin assembly and trafficking events. This article summarizes dyn
