Cellular zinc metabolism and zinc signaling: from biological functions to diseases and therapeutic targets.

Zinc metabolism at the cellular level is critical for many biological processes in the body. A key observation is the disruption of cellular homeostasis, often coinciding with disease progression. As an essential factor in maintaining cellular equilibrium, cellular zinc has been increasingly spotlighted in the context of disease development. Extensive research suggests zinc's involvement in promoting malignancy and invasion in cancer cells, despite its low tissue concentration. This has led to a growing body of literature investigating zinc's cellular metabolism, particularly the functions of

Pertussis Infection and Bilateral Optic Neuropathy Leading to Blindness in a 68-Year-old Male Patient: Possible Link or a Coincidence?

Whooping cough is a deadly infection that mainly targets infants, but its incidence is also on the rise in adults. Bilateral optic neuropathy is rare and can be due to genetic factors, infections or autoimmune disorders. We present a case of a 68-year-old man who presented to a UK hospital with worsening vision in both eyes for three days. His vital signs including blood pressure were within normal limits. He was reviewed by acute internal medicine, ophthalmology and neurology teams. His investigations were unremarkable except serological evidence of recent pertussis infection. Despite steroid

Optimising DTwP-containing vaccine infant immunisation schedules in Uganda and Nepal (OptImms): two open-label, non-inferiority, randomised controlled trials.

BACKGROUND: Few data are available comparing the WHO diphtheria-tetanus-pertussis (DTP)-containing vaccine schedule with reduced-dose or delayed three-dose schedules in infants. We aimed to identify an alternative schedule that is non-inferior or superior to the WHO schedule in maintaining early pertussis antibody levels before booster, while creating space for integration of new vaccines. METHODS: In two parallel, open-label, randomised, non-inferiority trials in Uganda and Nepal, healthy infants aged 42-50 days were randomised in a 4:4:4:3:3 ratio via an online system to five diphtheria-tet

Variability in the Diagnostic Criteria for Severe Pertussis: A Systematic Review.

IMPORTANCE: Pertussis, or whooping cough, can be life threatening in infants, yet the concept of severe pertussis remains poorly defined in the literature. Lack of standardization may impair research and public health interventions. OBJECTIVE: To assess the variability in terminology and criteria used to define severe pertussis in the published literature. EVIDENCE REVIEW: A systematic review of the literature drawn from PubMed and Web of Science databases was performed. Studies involving infants with pertussis that provided a definition for severe pertussis and published from database incep

Clinical characteristics of cases during the 2024 pertussis epidemic in France, January 2024 to December 2024.

The French national surveillance of pertussis consists of several units, including general practitioners (GPs). Here, we report on the clinical characteristics of cases from January to December 2024 from primary care surveillance in France. During this period, a total of 689 pertussis cases were reported by GPs participating in the surveillance. The national incidence rate in general practice was estimated to be 244 cases per 100,000 inhabitants (95 % CI: 224-264). Sixty-one per cent of cases were female and 86 % had received at least one dose of pertussis vaccine. Among cases vaccinated with

Is It Time to Consider Hydroxyurea as a Potential Therapeutic Option in the Management of Malignant Pertussis - A Case Report and Literature Review.

Malignant pertussis is a life-threatening condition in infants, for which no effective evidence-based treatment strategies have been established, owing to the rarity of the condition. We present the case of a four-month-old infant with paroxysmal coughing spells followed by a whooping sound and shortness of breath with a leukocyte count of 41.82 × 109/L and lymphocytosis (32.25 × 109/L). A diagnosis of malignant pertussis, based on the severe paroxysmal cough, significant lymphocytosis, and progressive respiratory symptoms, was established, and treatment was initiated with azithromycin and hyd

Success and limitations of the French public health strategy after 2 years of large implementation of respiratory syncytial virus prophylaxis: experience of a tertiary hospital (2018-2025).

OBJECTIVES: France has gradually implemented prophylaxis against RSV infections in infants since 2023 and, more recently, in the elderly. This study described the recent evolution of the RSV-hospital burden in a French tertiary hospital. METHODS: Characteristics of patients admitted during the 2023/2024 and 2024/2025 RSV seasons with a RSV-lower respiratory tract infection (LRTI) were compared to those of patients admitted during three preprophylaxis RSV seasons. RESULTS: Overall, 1114 pediatric inpatients and 41 adults with RSV-LRTI were enrolled, of whom 36% had ≥1 chronic complex conditio

Whooping Cough Cases Increase in Central Italy after COVID-19 Pandemic.

Pertussis continues to be a highly contagious respiratory infection, especially in children, with cyclical peaks of disease spread every three to five years. Here, we report relevant cases of B. pertussis infection between August 2023 and January 2024, and compare them with B. pertussis prevalence in pediatric patients admitted to the Reference Italian Pediatric Hospital, located in Rome, from January 2015 to July 2023. A total of 5464 tests for B. pertussis were performed during the study period, and 6.9% were positive. At the time of the COVID-19 pandemic, there was a sharp decrease in the p

Pertussis vaccination in adults in France: Overview and suggestions for improvement.

In France, the goal of the pertussis vaccination program is to protect newborns. All infants are vaccinated under the program and then given booster shots up to the age of 25 years. Pregnant women are likewise vaccinated, with the cocooning strategy reserved for infants born to unvaccinated mothers. Real-world data shows (i) inadequate coverage among adolescents and adults under 25; (ii) improper use of the tetanus, diphtheria, and polio (Td/IPV) vaccine in children under six years, adolescents, and young adults; and (iii) underdiagnosis of pertussis in adults. Older patients or those with spe

Pertussis vaccination in adults: a behavioral study of physicians from the US, France, and Germany.

BACKGROUND: Pertussis is a highly contagious respiratory tract infection that affects all ages, though it is most severe in young infants. Adults, especially those with respiratory conditions or other chronic illnesses can also suffer serious consequences of pertussis. Pertussis vaccination is the best method of disease prevention in a lifetime. This behavioral study aimed to assess physicians' attitudes towards pertussis vaccination in adults and the importance of pertussis vaccination for disease prevention, especially in those with chronic illnesses, and to determine the impact of the COVID

Maternal Tetanus Toxoid, Reduced Diphtheria Toxoid, and Acellular Pertussis Vaccination During Pregnancy: Impact on Infant Anti-Pertussis Antibody Concentrations by Maternal Pertussis Priming Series.

BACKGROUND: Acellular pertussis (aP) vaccines replaced whole-cell pertussis (wP) vaccines for the US childhood primary series in 1997. As women primed with aP vaccines enter childbearing age, protection of infants through tetanus toxoid, reduced diphtheria toxoid, and acellular pertussis (Tdap) vaccination during pregnancy may be impacted. METHODS: Term infants born to women vaccinated with Tdap during pregnancy were included. Geometric mean concentrations (GMCs) of pertussis-specific immunoglobulin G antibodies (international units per milliliter) in cord blood of infants born to women born

Pertussis surveillance results from a French general practitioner network, France, 2017 to 2020.

IntroductionIn France, three complementary surveillance networks involving hospitals and paediatrician practices currently allow pertussis surveillance among infants (<1 year old) and children (1-12 years old). Data on incidences among adolescents (13-17 years old) and adults (≥ 18 years) are scarce. In 2017, a sentinel surveillance system called Sentinelles network, was implemented among general practitioners (GPs).AimThe purpose of Sentinelles network is to assess pertussis incidence, monitor the cases' age distribution and evaluate the impact of the country's vaccination policy. We present
