Comparative proteomics reveals hydrogenase-centered steel-dependent growth and corrosion in Desulfovibrio vulgaris and Desulfovibrio ferrophilus.

Comparative proteomic analyses including ecologically distinct sulfate-reducing strains and different cellular fractions are scarce, limiting our understanding of corrosion-related mechanisms under anoxic conditions and the identification of protein signatures relevant to microbiologically influenced corrosion (MIC). To capture both freshwater and marine adaptations, we investigated the well-established SRB model strains Desulfovibrio vulgaris and Desulfovibrio ferrophilus. We compared energy metabolism, electron transport pathways, and growth patterns in both strains cultivated with lactate,

Streptococcus thermophilus DM287 and DM294 as candidate oral probiotic strains with anti-biofilm activity against cariogenic pathogens.

Streptococcus mutans and Streptococcus sobrinus are key contributors to dental caries within a broader polymicrobial biofilm community, driving pathogenicity through biofilm and dental plaque formation rather than direct tissue invasion. Probiotic-based strategies that target colonization rather than viability represent a promising non-antibiotic approach to caries prevention. Streptococcus thermophilus is a food-grade organism with established GRAS status and a long history of safe human consumption, yet it remains largely unexplored as an oral care ingredient. In this study, two S. thermophi

Nonlinear response mechanisms of microbial communities to coral reef biogeomorphy in the South China Sea.

UNLABELLED: To resolve how planktonic microbial composition, assembly mechanisms, and interaction architectures vary across coral reef biogeomorphic states, we sought to identify process-relevant indicators of reef resilience. We analyzed size-fractionated microbiomes from 57 stations in the South China Sea using 16S/18S rRNA amplicon sequencing. A distance-decay model defined a near-coral reef influence zone within 588 m, and reefs were classified into Poor, Moderate, and Good states based on coral density metrics. Results showed that microbial composition diverged significantly between near-

Mfd interacts with RNA polymerase to modulate flagellum-dependent motility, chemotaxis, and population heterogeneity in the stationary phase of Bacillus subtilis.

Bacillus subtilis is a model for cell differentiation, capable of transitioning between distinct states: a sessile, chained state (often associated with biofilm formation), and a motile, planktonic state. This transition is governed by a complex regulatory network that includes alternative sigma factor-D (σD), which drives the expression of genes for autolysins, flagellar biosynthesis, and chemotaxis, resulting in separated, flagellated, motile cells. Although the conserved transcription-coupled repair factor mutation frequency decline (Mfd) is best known for resolving stalled RNA polymerase (

Large-scale toxic bloom of Dinophysis acuminata complex following a major flood disaster in southern Brazil.

Dinophysis species are the main producers of diarrhetic shellfish toxins (DSTs) in the Southwestern Atlantic, where recurrent blooms frequently lead to shellfish harvesting bans. In 2024, an extreme rainfall event over Rio Grande do Sul State, southern Brazil, caused one of the most severe floods ever recorded in the country, and generated an anomalous freshwater discharge through the Patos Lagoon estuary. The resulting low-salinity plume spread northward along the southern Brazilian coast, reaching aquaculture areas hundreds of kilometers away from the estuary. Here, we integrate hydrological

Profound cell wall remodeling in Candida parapsilosis during systemic infection confers simultaneous tolerance to echinocandins and host immunity.

Antifungal tolerance can promote the emergence of resistance yet often incurs fitness costs for fungal pathogens. How tolerant populations compensate for these deficits and how they may be therapeutically targeted remain poorly understood. Here, we investigate four sequential Candida parapsilosis isolates recovered from a patient with persistent candidemia and failure of micafungin therapy. The infection was ultimately cleared with liposomal amphotericin B (LAMB). Whole-genome sequencing (WGS) confirmed clonal relatedness and the absence of known resistance mutations. Later isolates displayed

Assessment of Enterococcus faecalis biofilm response to amoxicillin and ciprofloxacin using a dynamic in vitro pharmacokinetic-pharmacodynamic model of catheter-associated urinary tract infection.

BACKGROUND: Catheter-associated urinary tract infections (CAUTIs) are among the most common types of healthcare-associated infections. Among uropathogens, Enterococcus faecalis is particularly notable for its strong capacity to form biofilms on urinary catheters, thereby decreasing its susceptibility to antibiotics commonly used to treat CAUTIs. However, current antibiotic dosing strategies still rely on planktonic phenotypes, limiting their applicability to biofilm-associated infections. To address this limitation, a dynamic in vitro pharmacokinetic-pharmacodynamic model was developed to asse

Novel compounds derived from AR-12 that demonstrate host-directed clearance of intracellular Salmonella enterica Serovar Typhimurium.

Salmonella infections, including typhoid and paratyphoid fevers, impose a substantial burden in low-income countries, contributing to significant morbidity and mortality associated with enteric and systemic infections. Developing effective treatments for Salmonella infections requires further consideration, especially due to widespread antibiotic resistance. Host-targeted therapies, such as AR-12, offer a promising solution to combat drug resistance. AR-12 has demonstrated broad-spectrum antimicrobial activity against various bacterial pathogens, including Salmonella enterica serovar Typhimuri

Anti-biofilm and anti-quorum sensing activities of galloylquinic acid against clinical isolates of multidrug-resistant Pseudomonas aeruginosa in open wound infection: in vitro and in vivo efficacy studies.

BACKGROUND: Pseudomonas aeruginosa can proliferate in immunocompromised individuals, forming biofilms that increase antibiotic resistance. This bacterium poses a significant global health risk due to its resistance to human defenses, antibiotics, and various environmental stresses. The objective of this study was to evaluate the antibacterial, anti-biofilm, and anti-quorum sensing activities of galloylquinic acid compounds (GQAs) extracted from Copaifera lucens leaves against clinical isolates of multidrug-resistant (MDR) P. aeruginosa. We have investigated the optimal concentration of GQAs ne

Repurposing bunamidine hydrochloride as a potent antimicrobial agent targeting vancomycin-resistant Enterococcus membranes.

Vancomycin-resistant enterococci (VRE) are a major cause of hospital-acquired infections, with limited treatment options due to rising antibiotic resistance. Targeting bacterial membranes offers a promising alternative to conventional therapies. In this study, we investigated the antimicrobial activity and mechanism of action of bunamidine hydrochloride (BUN) against VRE. BUN exhibited potent bactericidal effects against both VRE and vancomycin-susceptible enterococci (VSE), with minimum inhibitory concentrations (MICs) ranging from 2 to 4 µg/mL. BUN significantly inhibited biofilm formation a

Synergistic Antibacterial Effect of ZnO Nanoparticles and Antibiotics against Multidrug-resistant Biofilm Bacteria.

BACKGROUND: The misuse of antibiotics leads to a global increase in antibiotic resistance. Therefore, it is imperative to search for alternative compounds to conventional antibiotics. ZnO nanoparticles (Zn NP) are one of these alternatives because they are an effective option to overcome biofilm bacterial cells and a novel way to overcome multidrug resistance in bacteria. The current research study aims to characterize the efficacy of ZnO nanoparticles alone and in combination with other antibacterial drugs against bacterial biofilms. METHODS: ZnO NPs were prepared by co-precipitation method,

Novel yellow light-responsive SnO2/SnS2 piezo-photocatalysts with excellent performances for tooth whitening and biofilm eradication.

Oral problems caused by pathogens and tooth discoloration have posed great threats to public health in recent years. Timely killing of cariogenic bacteria and removing surface pigments are the key points to treat yellow teeth to restore healthy whitening. Piezo-photocatalysis has been proved to be an effective strategy for treating yellow teeth. However, few effective and safe nanomaterials have been developed to address this issue in the oral field. Herein, yellow light-responsive SnO2/SnS2 heterostructures are proposed for piezo-photocatalytic biofilm eradication and tooth whitening for the

Enzyme-Linked Lipid Nanocarriers for Coping Pseudomonal Pulmonary Infection. Would Nanocarriers Complement Biofilm Disruption or Pave Its Road?

INTRODUCTION: Cystic fibrosis (CF) is associated with pulmonary Pseudomonas aeruginosa infections persistent to antibiotics. METHODS: To eradicate pseudomonal biofilms, solid lipid nanoparticles (SLNs) loaded with quorum-sensing-inhibitor (QSI, disrupting bacterial crosstalk), coated with chitosan (CS, improving internalization) and immobilized with alginate lyase (AL, destroying alginate biofilms) were developed. RESULTS: SLNs (140-205 nm) showed prolonged release of QSI with no sign of acute toxicity to A549 and Calu-3 cells. The CS coating improved uptake, whereas immobilized-AL ensured >

Dry powder inhalation containing muco-inert ciprofloxacin and colistin co-loaded liposomes for pulmonary P. Aeruginosa biofilm eradication.

Pseudomonas aeruginosa (PA), a predominant pathogen in lung infections, poses significant challenges due to its biofilm formation, which is the primary cause of chronic and recalcitrant pulmonary infections. Bacteria within these biofilms exhibit heightened resistance to antibiotics compared to their planktonic counterparts, and their secreted toxins exacerbate lung infections. Diverging from traditional antibacterial therapy for biofilm eradication, this study introduces a novel dry powder inhalation containing muco-inert ciprofloxacin and colistin co-encapsulated liposomes (Cipro-Col-Lips) p

Equine bone marrow-derived mesenchymal stromal cells reduce established S. aureus and E. coli biofilm matrix in vitro.

Biofilms reduce antibiotic efficacy and lead to complications and mortality in human and equine patients with orthopedic infections. Equine bone marrow-derived mesenchymal stromal cells (MSC) kill planktonic bacteria and prevent biofilm formation, but their ability to disrupt established orthopedic biofilms is unknown. Our objective was to evaluate the ability of MSC to reduce established S. aureus or E. coli biofilms in vitro. We hypothesized that MSC would reduce biofilm matrix and colony-forming units (CFU) compared to no treatment and that MSC combined with the antibiotic, amikacin sulfate

Gramine sensitizes Klebsiella pneumoniae to tigecycline killing.

BACKGROUND: The presence of plasmid-mediated resistance-nodulation-division (RND) efflux pump gene cluster tmexCD1-toprJ1 and its related variants has been associated with heightened resistance to tigecycline, thus diminishing its effectiveness. In this study, we explored the potential of gramine, a naturally occurring indole alkaloid, as an innovative adjuvant to enhance the treatment of infections caused by K. pneumoniae carrying tmexCD-toprJ-like gene clusters. METHODS: The synergistic potential of gramine in combination with antibiotics against both planktonic and drug-tolerant multidrug-
