Identification and characterization of DdPDE3, a cGMP-selective phosphodiesterase from Dictyostelium.

In Dictyostelium cAMP and cGMP have important functions as first and second messengers in chemotaxis and development. Two cyclic-nucleotide phosphodiesterases (DdPDE 1 and 2) have been identified previously, an extracellular dual-specificity enzyme and an intracellular cAMP-specific enzyme (encoded by the psdA and regA genes respectively). Biochemical data suggest the presence of at least one cGMP-specific phosphodiesterase (PDE) that is activated by cGMP. Using bioinformatics we identified a partial sequence in the Dictyostelium expressed sequence tag database that shows a high degree of amin

The microbial communities of a tomato crop grown in Veggie under different lighting regimes on the International Space Station.

Customized lighting treatments are being investigated to help optimize space crop production. The VEG-05 experiment on the International Space Station (ISS) investigated the effect of red-rich and blue-rich lighting in the Veggie plant-growth chamber on the microbial communities of a dwarf tomato variety Solanum lycopersicum cv. Red Robin. The microbial communities were investigated using bacterial 16S rRNA and fungal internal transcribed spacer (ITS) sequencing methods to identify bacterial and fungal communities from tomato fruit, leaves, roots, rooting substrate, and Veggie chamber surfaces

Cloning of two novel P-III class metalloproteinases from Trimeresurus stejnegeri venom gland.

Hemorrhagic toxins are widely distributed in viperid and crotalid snake venoms. Envenomation of Trimeresurus stejnegeri, a member of Crotalidae family, caused potent systemic and local hemorrhage. Up to now, there is no report on hemorrhage toxins from this venom. In this work, we cloned two cDNAs of P-III metalloproteinase precursors, designated as stejnihagin-A and stejnihagin-B, respectively, from T. stejnegeri venom gland. Both cDNAs encode an opening reading frame of 600 amino acid residues, containing a signal sequence, a proprotein domain, a metalloproteinase domain, a disintegrin-like

Seasonal variation in the bacterial microbiome of questing nymphal ticks in Missouri, United States.

BACKGROUND: Seasonal environmental variation may influence the composition of tick-associated bacterial communities. This study assessed seasonal differences in the microbiome of questing nymphal ticks collected from Missouri, United States. METHODS: Questing ticks were collected during early and late seasonal periods at a livestock-associated site in central Missouri. To minimize confounding by developmental stage, microbiome analyses were restricted to nymphal ticks. Bacterial communities were characterized using 16S rRNA gene sequencing. Alpha diversity (richness, Shannon, and Simpson indi

Longitudinal study of the early-life fecal and nasal microbiotas of the domestic pig.

BACKGROUND: The mammalian microbiota plays a key role in host health and disease susceptibility. However, knowledge of the early-age microbiota of pigs is lacking. The purpose of this study was to use high-throughput next-generation sequencing to characterize the fecal and nasal microbiotas of pigs during early life. RESULTS: Ten commercially-raised pigs were randomly enrolled at birth and sampled throughout the first 7 weeks of life. DNA was extracted from fecal and nasal samples and the hypervariable region V4 of the 16S rRNA gene was amplified. The product was sequenced using the Illumina

A new proline-rich protein precursor expressed in the salivary glands of the rat is encoded by a gene homologous to the gene coding for the prohormone-like protein SMR1.

A gene encoding a prohormone-like protein (SMR1) has previously been characterized and shown to be expressed in the rat submandibular glands under androgenic control (Rosinski-Chupin, I., Tronik, D., and Rougeon, F. (1988) Proc. Natl. Acad. Sci. U.S.A. 85, 8553-8557). This gene, now named VCS-alpha 1, belongs to a multigene family (Rosinski-Chupin, I., and Rougeon, F. (1990) DNA Cell Biol. 9, 553-559). We now describe the structure and the expression of a second member (VCS-beta 1) of this family. The two genes differ principally in the protein-coding region, therefore we have named these rela

WH2 and proline-rich domains of WASP-family proteins collaborate to accelerate actin filament elongation.

WASP-family proteins are known to promote assembly of branched actin networks by stimulating the filament-nucleating activity of the Arp2/3 complex. Here, we show that WASP-family proteins also function as polymerases that accelerate elongation of uncapped actin filaments. When clustered on a surface, WASP-family proteins can drive branched actin networks to grow much faster than they could by direct incorporation of soluble monomers. This polymerase activity arises from the coordinated action of two regulatory sequences: (i) a WASP homology 2 (WH2) domain that binds actin, and (ii) a proline-

Cyathostomin species richness and alpha diversity at egg reappearance following fecal egg count-based selective ivermectin treatment.

Cyathostomin nematodes are ubiquitous in grazing horses. The overuse of anthelmintics has led to increasing problems with drug resistance to benzimidazoles and pyrantel. Although macrocyclic lactones (ML), such as ivermectin, generally retain the highest efficacy, reduced ML efficacy has increasingly been reported. A shortened egg reappearance period (ERP) is frequently used as an indicator of reduced susceptibility. However, the species-specific response dynamics after ivermectin treatment are inadequately characterized in cyathostomins. The objective of this study was to compare the cyathost

Importance of anthropogenic sources at shaping the antimicrobial resistance profile of a peri-urban mesocarnivore.

Anthropogenically derived antimicrobial-resistant bacteria (ARB) and antimicrobial resistance genes (ARG) have been detected in wildlife. The likelihood of detecting ARB and ARG in wildlife increases with wildlife exposure to anthropogenic sources of antimicrobial resistance (AMR). Whether anthropogenic sources also increase the risk for AMR to spread in bacteria of wildlife is not well understood. The spread of AMR in bacteria of wildlife can be estimated by examining the richness of ARB and ARG, and the prevalence of ARB that have mobilizable ARG (i.e., ARG that can be transferred across bac

Ml proteins from Mesorhizobium loti and MucR from Brucella abortus: an AT-rich core DNA-target site and oligomerization ability.

Mesorhizobium loti contains ten genes coding for proteins sharing high amino acid sequence identity with members of the Ros/MucR transcription factor family. Five of these Ros/MucR family members from Mesorhizobium loti (Ml proteins) have been recently structurally and functionally characterized demonstrating that Ml proteins are DNA-binding proteins. However, the DNA-binding studies were performed using the Ros DNA-binding site with the Ml proteins. Currently, there is no evidence as to when the Ml proteins are expressed during the Mesorhizobium lo ti life cycle as well as no information conc

Normal Facial Nerve Enhancement on Volumetric Interpolated Breath-Hold Examination MRI Sequence.

BACKGROUND AND PURPOSE: Enhancement of the facial nerve can be seen on MRI due to its rich arteriovenous plexus. Classically, enhancement of the facial nerve beyond the geniculate ganglion has been described as a normal finding, while enhancement of the canalicular and labyrinthine segments is considered abnormal. We hypothesize facial nerve enhancement of the canalicular and labyrinthine segments is a normal finding on the postcontrast T1-weighted, fat-saturated volumetric interpolated breath-hold examination (VIBE) sequence on both 1.5T and 3T MRI scanners. MATERIALS AND METHODS: Fifty pati

DNA Binding and Cleavage by the Human Parvovirus B19 NS1 Nuclease Domain.

Infection with human parvovirus B19 (B19V) has been associated with a myriad of illnesses, including erythema infectiosum (Fifth disease), hydrops fetalis, arthropathy, hepatitis, and cardiomyopathy, and also possibly the triggering of any number of different autoimmune diseases. B19V NS1 is a multidomain protein that plays a critical role in viral replication, with predicted nuclease, helicase, and gene transactivation activities. Herein, we investigate the biochemical activities of the nuclease domain (residues 2-176) of B19V NS1 (NS1-nuc) in sequence-specific DNA binding of the viral origin

Self-Assembling Gemini-Type Amphiphiles From Helix-Loop-Helix Peptides.

Amphiphilic peptides can be designed to self-assemble into various nanostructures, such as nanorods, nanotubes, and vesicles, depending on their sequence and secondary structure. We have designed a self-assembling amphiphilic peptide that folds into a hairpin-like helix-loop-helix (HLH) motif. The α-helical segments are rich in hydrophobic leucine residues, while the loop segment is positively charged. The morphology of the peptide self-assemblies can be controlled by the preparation method. When prepared from an ethanolic solution at room temperature, these amphiphilic HLHPs self-assemble int

Comparative analysis of mitochondrial genome data for Necator americanus from two endemic regions reveals substantial genetic variation.

Necator americanus is a blood-sucking, intestinal nematode of major human health importance in many tropical and subtropical regions of the world. The aim of the present study was to compare the complete mitochondrial genome sequence from one N. americanus individual from Togo with another from China, in order to estimate the magnitude of genetic variability for different mitochondrial genes and non-coding regions. For the 12 protein genes, this comparison revealed sequence differences at both the nucleotide (3-7%) and amino acid (1-7%) levels. The most conserved of these was the nad4L gene, w

Plasma extracellular vesicles promote follicular T helper cell expansion in primary Sjögren's syndrome.

Primary Sjögren's syndrome (pSS) is a prevalent autoimmune disease characterized by exocrine gland dysfunction, with hallmarks of B cell and T cell overactivation, whose underlying mechanism remains largely unknown. Herein, we show that pSS plasma contained more extracellular vesicles (EVs) than HC plasma, which promoted CD4+ T cell activation, Th1, and follicular T helper cell (Tfh) differentiation, aggravating pSS immunopathology. Notably, pSS plasma EVs were enriched with miR-501-3p, mediating CD4+ T cell activation and Tfh cell differentiation. Furthermore, miR-501-3p downregulated special

HiNet3D: Integrating patient hierarchical aggregation network and molecular 3D-aware for medication recommendation.

Accurate medication recommendation from electronic health records (EHR) is crucial for clinical decision support. However, prior methods often treat EHR as a flat visit sequence and encode drugs with 2D molecular graphs, which limits capturing inadequate patient representation and drug molecular semantics. Therefore, we propose a framework named HiNet3D for the recommendation task. Specifically, HiNet3D builds a patient history graph by representing each visit as a node and performs cross-visit pairwise aggregation to form a layer of virtual nodes, enabling rich cross-visit context modeling to

Role of signal sequence in vaccine-induced protection against experimental coccidioidomycosis.

The vaccine efficacy of the gene sequence encoding the signal peptide of the antigen known as antigen 2 or proline-rich antigen (Ag2/PRA), an immunodominant antigen present in the cell wall of the fungal pathogen Coccidioides immitis, was investigated in a murine model of coccidioidomycosis. Expression plasmids for Ag2/PRA(1-18) DNA (signal sequence), Ag2/PRA(19-194) DNA (lacking the signal sequence), and Ag2/PRA(1-194) DNA (full length) were inserted in the pVR1012 vector, and the constructs were used to vaccinate the highly susceptible BALB/c mouse strain. Characterization and expression analysis of Toll-like receptor 3 cDNA from Atlantic salmon (Salmo salar).

Innate pathway activation is fundamental for early anti-viral defense in fish, but currently there is insufficient understanding of how salmonid fish identify viral molecules and activate these pathways. The Toll-like receptor (TLR) is believed to play a crucial role in host defense of pathogenic microbes in the innate immune system. In the present study, the full-length cDNA of Salmo salar TLR3 (ssTLR3) was cloned. The ssTLR3 cDNA sequence was 6071 bp long, containing an open reading frame of 2754 bp and encoding 971 amino acids. The TLR group motifs, such as leucine-rich repeat (LRR) domains

Structural and biochemical characterisation of VaF1, a P-IIIa fibrinogenolytic metalloproteinase from Vipera ammodytes ammodytes venom.

A high molecular mass metalloproteinase with α-fibrinogenolytic activity, termed VaF1, was purified from nose-horned viper (Vipera ammodytes ammodytes) venom. Subcutaneous injection of 9 μg of VaF1 did not induce bleeding in rats. Nevertheless, in vitro it degraded collagen IV, nidogen and fibronectin, components of the extracellular matrix, although with low efficacy and narrow specificity. VaF1 would be expected to exert anti-coagulant action, due to its hydrolysis of fibrinogen, factor X, prothrombin and plasminogen, plasma proteins involved in blood coagulation. The enzyme is a single-chai

cDNA cloning, sequencing and in situ localization of a transcript specific to both sublingual demilune cells and parotid intercalated duct cells in mouse salivary glands.

A cDNA clone derived from mouse sublingual gland was isolated from lambda-phage cDNA library. Northern blot hybridization indicated that the transcript from which it was derived was approx. 700 nucleotides in length. This mRNA encoded a protein of about 20 kDa, as determined by hybrid selection and cell-free translation. Conceptual translation of the cDNA clones showed that p20 is 170 amino acids in length. The putative protein is hydrophobic in nature, is neither a mucin-like protein nor does its amino acid sequence or composition resemble the other known mouse proteins. However, the amino ac
