[USE OF THE REAL-TIME PCR FOR STUDY OF THE PERIODONTAL MICROBIOME IN PATIENTS WITH COMBINED PATHOLOGY OF GASTRODUODENAL ZONE AND CHRONIC PERIODONTITIS].

The total of 54 patients with chronic periodontitis of different severity was tested using real-time PCR (Dentoflor kit). The group included 38 patients with chronic gastritis. For the first time, a higher prevalence of Treponema denticola in periodontium of males in comparison with females was demonstrated. The patients with chronic gastritis had more human genome DNA at their periodontium than healthy individuals. Non-parametric statistical analysis demonstrated high association of periodontium colonization with. T. forsythensis and T. denticola (but not Aggregatibacter actinomycetemcomitans

Bacterial colonization patterns of periodontal pockets in different ages.

The aim of this study was to investigate subgingival bacterial composition of untreated Italian subjects with aggressive and chronic periodontitis. The total bacterial load, pathogenic bacteria belonging to "red" and "orange" complexes and Aggregatibacter actinomycetemcomitans were determined by Real-Time PCR in 1216 patients. Data were analysed by looking for relationships between bacteriological parameters, age and periodontal probing depth. The obtained results showed a significant higher number of red complex bacteria in older rather than in younger patients. The total number of bacteria a

Concentration-dependent antimicrobial effects of two mouthwashes on key periodontal pathogens: An in vitro study.

Periodontal diseases, caused by pathogenic dental plaque biofilm, are often treated with chlorhexidine, but its long-term use can lead to undesirable side effects. Therefore, it is of interest to compare the antimicrobial efficacy of two herbal mouthwashes, Oralife and Ora-T, against periodontal pathogens Porphyromonas gingivalis and Aggregatibacter actinomycetemcomitans using the disc diffusion method. Results showed that Oralife exhibited significantly larger zones of inhibition than Ora-T at all tested concentrations. No inhibitory effect was observed at the lowest concentration. Thus, we s

Probiotics and periodontal health.

Periodontitis is one of the most common chronic inflammatory diseases. The etiology is clearly bacterial and a number of putative bacterial pathogens have been associated with the disease, including Aggregatibacter actinomycetemcomitans, Tannerella forsythus and Porphyromonas gingivalis. Comparatively, little attention has been paid to the identification of health-associated and potentially beneficial bacterial species that may reside in the gingival sulcus. Probiotic technology represents a breakthrough approach to maintaining oral health by using natural beneficial bacteria, commonly found i

[A.a.: the bacterium with the long name].

The introduction of Kodak's chairside test kit, Evalusite, for Actinobacillus actinomycetemcomitans (A.a.) and two other oral bacteria has stimulated this study of the relation between A.a. and periodontal destruction. A.a. has been associated with oral abscesses, brain abscesses, endocarditis and localized juvenile periodontitis. However, in all periodontal studies A.a. also has been observed in 'ordinary' periodontal pockets and subjects without active periodontal destruction. Based on longitudinal studies it is concluded, that there is no proof for a causal relationship between A.a. and per

Pediatric acute dacryocystitis due to Eikenella corrodens: A case report.

Eikenella corrodens is a facultatively anaerobic gram-negative rod bacterium in the oropharynx and respiratory tract. It is a member of HACEK (Haemophilus spp., Aggregatibacter spp., Cardiobacterium hominis, E. corrodens, and Kingella kingae) group commonly associated with endocarditis and craniofacial infections. It is usually susceptible to penicillin, second and third-generation cephalosporins, and carbapenem, but has variable susceptibility to first-generation cephalosporin. We herein provide a description of the first case of pediatric acute dacryocystitis caused by E. corrodens. The pati

Orthodontic treatment in the presence of aggressive periodontitis.

INTRODUCTION: Aggressive periodontitis causes periodontal destruction, with loss of supporting alveolar bone. The common symptom is rapid attachment loss in the first molar and incisor area, in young adults. OBJECTIVE: The aim of this study was to discuss the challenges, implications and the impact of orthodontic treatment in patients affected by severe periodontal problems, specifically aggressive periodontitis. DISCUSSION: In addition to other bacteria, the main pathogen involved in aggressive periodontitis is the Aggregatibacter actinomycetemcomitans. However, the susceptibility to the di

Periodontitis and rheumatoid arthritis: What have we learned about their connection and their treatment?

Rheumatoid arthritis and periodontitis are chronic inflammatory diseases defined respectively by the destruction of the articular cartilage and tooth-supporting periodontal tissues. Although the epidemiologic evidence for an association between these two diseases is still scarce, there is emerging scientific information linking specific bacterial periodontal pathogens, such as Porphyromonas gingivalis and Aggregatibacter actinomycetemcomitans, in the citrullination process, leading to autoantibody formation and compromised immunotolerance of the susceptible patient to rheumatoid arthritis. Nanofibers with Incorporated Autochthonous Bacteria as Potential Probiotics for Local Treatment of Periodontal Disease.

The conventional treatment of periodontal disease does not solve the high incidence of recolonization of periodontal pockets by pathogens. Here, we introduce an innovative concept of incorporating autochthonous bacteria as potential probiotics into nanofibers for local treatment. We selected and isolated the strain 25.2.M from the oral microbiota of healthy volunteers. It was identified as Bacillus sp. based on 16S rRNA sequence analyses. The strain is nonpathogenic, produces antimicrobial substances, and can grow over the periodontal pathogen Aggregatibacter actinomycetemcomitans in vitro, ma

Bacterial composition of subgingival plaque in crowded and noncrowded teeth.

INTRODUCTION: Patients with dental crowding, even with good oral hygiene, may show increased bacterial presence in the crowded area. This study aimed to determine the microbial composition of subgingival plaque in crowded and noncrowded dental regions. METHODS: Forty dental students with anterior dental crowding, who were aware of correct brushing techniques and oral hygiene, were enrolled in this study. After orthodontic records were taken and the periodontal examination was performed, subgingival plaque samples were collected from the crowded (experimental) and noncrowded regions (control)

Current concepts in juvenile periodontitis.

Juvenile periodontitis is one of the more debilitating periodontal diseases. The disease entity includes two manifestations of aggressive bone destruction, a localized and a generalized form, which affect adolescents at puberty. Recent studies have defined the prevalence of the disease with regard to locale, gender, and ethnicity. In addition, significant advances have been made in evaluating host response effects in juvenile periodontitis. Studies have identified changes in cell surface receptors on neutrophils, which appear to be related to both receptor density and structure. Various molecu

Ocular Surface Microbiota Alterations Following FS-LASIK and Their Association With Postoperative Dry Eye.

PURPOSE: Given the limited evidence on ocular surface microbiota (OSM) changes after femtosecond laser-assisted in situ keratomileusis (FS-LASIK) and their link to dry eye (DE), this study aimed to compare microbial profiles in patients with and without postoperative DE, offering a basis for early detection and targeted treatment. METHODS: Patients undergoing FS-LASIK were evaluated 3 months postoperatively and stratified into DE (N = 30) and non-DE groups (N = 30) based on ocular surface disease index (OSDI), TBUT, and the Schirmer I test. Corneal nerve alterations were assessed using IVCM.

Association between oral microbiota and inflammatory bowel disease: A 2-sample Mendelian randomization study in East Asian populations.

Inflammatory bowel disease (IBD), including ulcerative colitis (UC) and Crohn's disease (CD), is increasingly prevalent in East Asia. Its pathogenesis is linked to microbiota dysbiosis via the oral-gut axis, but population-specific causal evidence remains scarce. This study aimed to clarify the causal associations between oral microbiota and UC/CD in East Asian populations using Mendelian randomization (MR), providing evidence for IBD etiology and precise prevention/treatment. A 2-sample MR approach was adopted, using genome-wide association study data of East Asian populations. Single nucleot

Lactobacillus lysates protect oral epithelial cells from pathogen-associated damage, increase secretion of pro-inflammatory cytokines and enhance barrier integrity.

Periodontitis is a chronic gum disease characterised by inflammation and the loss of bone. We have explored the potential prophylactic effects of lysates from four Lactobacillus strains against the toxic effects of three periodontal pathogens (Porphyromonas gingivalis, Fusobacterium nucleatum, and Aggregatibacter actinomycetemcomitans). TR146 oral epithelial cells were pre-treated with Lactobacillus lysates (L. rhamnosus - GG, L. rhamnosus - SD11, L. reuteri and L. plantarum) and then challenged with pathogenic material (live cells, lysates, or supernatants). Cytokine analysis was performed on

Impact of smokeless and smoking tobacco on subgingival microbial composition: A comparative study.

BACKGROUND: Environmental perturbations such as tobacco use causes increased bacterial diversity in the subgingival microbiome. Despite the recognized impact of tobacco on oral health, there is a notable gap in the literature regarding the specific characteristics of the subgingival microbiome among Indian tobacco users. OBJECTIVE: This study seeks to provide a comparative analysis of subgingival microbial profile of smokeless tobacco users and smokers with an otherwise healthy periodontal environment. MATERIAL AND METHODS: This cross-sectional study at a Tertiary Dental Hospital in India re

Identification of a glutathione transporter in A. actinomycetemcomitans.

Microbes produce a large array of extracellular molecules, which serve as signals and cues to promote polymicrobial interactions and alter the function of microbial communities. This has been particularly well studied in the human oral microbiome, where key metabolites have been shown to impact both health and disease. Here, we used an untargeted mass spectrometry approach to comprehensively assess the extracellular metabolome of the pathogen Aggregatibacter actinomycetemcomitans and the commensal Streptococcus gordonii during mono- and co-culture. We generated and made publicly available a me
