Indigenous soil bacteria with the combined potential for hydrocarbon consumption and heavy metal resistance.

INTRODUCTION: Transconjugant bacteria with combined potential for hydrocarbon utilization and heavy metal resistance were suggested by earlier investigators for bioremediation of soils co-contaminated with hydrocarbons and heavy metals. The purpose of this study was to offer evidence that such microorganisms are already part of the indigenous soil microflora. METHODS: Microorganisms in pristine and oily soils were counted on nutrient agar and a mineral medium with oil as a sole carbon source, in the absence and presence of either sodium arsenate (As V), sodium arsenite (As III) or cadmium sul

Synechocystis sp. PCC6803 possesses a two-component polyhydroxyalkanoic acid synthase similar to that of anoxygenic purple sulfur bacteria.

During cultivation under storage conditions with BG11 medium containing acetate as a carbon source, Synechocystis sp. PCC6803 accumulated poly(3-hydroxybutyrate) up to 10% (w/w) of the cell dry weight. Our analysis of the complete Synechocystis sp. PCC6803 genome sequence, which had recently become available, revealed that not only the open reading frame slr1830 (which was designated as phaC) but also the open reading frame slr1829, which is located colinear and upstream of phaC, most probably represent a polyhydroxyalkanoic acid (PHA) synthase gene. The open reading frame slr1829 was therefor

Analysis of a repetitive DNA sequence from Bordetella pertussis and its application to the diagnosis of pertussis using the polymerase chain reaction.

A tandemly repeated 1,046-base-pair (bp) ClaI DNA fragment from Bordetella pertussis was cloned into Escherichia coli by using the vector pUC19. This fragment, when isolated, hybridized strongly to DNA from all 100 clinical isolates of B. pertussis tested. It was shown to have homology to single-copy sequences in Bordetella bronchiseptica but not Bordetella parapertussis and did not hybridize to lysate blots of a wide range of other bacteria, including members of the closely related genera Pasteurella, Alcaligenes, and Haemophilus. The 1,046-bp fragment was sequenced, and complementary synthet

Investigating the Prevalence of Bacterial Infections in Patients with Coronavirus Disease 2019 Hospitalized in Intensive Care Unit and Determining their Antibiotic Resistance Patterns.

BACKGROUND: COVID-19 patient hospitalization, particularly in intensive care units, exposes them to bacterial and fungi co-infections, which can have very serious consequences, including increased mortality. In addition, antibiotic resistance among pathogens is a hidden threat behind COVID-19. METHODS: In the period from 2020 September to 2021 August, bacterial isolates from COVID- 19 patients admitted to the ICU of Sina Hospital in Hamadan, Iran, were collected and identified based on standard biochemical tests. COVID-19 cases were confirmed based on clinical symptoms, computed tomography, a

Quantification of multi-antibiotic resistant opportunistic pathogenic bacteria in bioaerosols in and around a pharmaceutical wastewater treatment plant.

Pharmaceutical wastewater treatment plants (WWTPs) are thought to be a "seedbed" and reservoirs for multi-antibiotic resistant pathogenic bacteria which can be transmitted to the air environment through aeration. We quantified airborne multi-antibiotic resistance in a full-scale plant to treat antibiotics-producing wastewater by collecting bioaerosol samples from December 2014 to July 2015. Gram-negative opportunistic pathogenic bacteria (GNOPB) were isolated, and antibiotic susceptibility tests against 18 commonly used antibiotics, including 11 β-lactam antibiotics, 3 aminoglycosides, 2 fluor

Antibacterial activity of lantibiotics, alone and in combination with colistin, against Gram-negative bacteria.

In this study, the potential of the use of class I antimicrobial cationic peptides, also known as lantibiotics, nisin, warnerin and hominin, combined with subinhibitory concentrations of colistin against Gram-negative bacteria (GNB) was investigated. The minimum inhibitory concentration (MIC) and fractional inhibitory concentration index were determined via the microbroth dilution and checkerboard techniques, respectively. We showed synergistic effects of the combination of all the lantibiotics + colistin against Escherichia coli, Enterobacter aerogenes, Pseudomonas fluorescens, clinical multi

Molecular Characterization and Diversity of Bacteria Isolated from Fish and Fish Products Retailed in Kenyan Markets.

Fish products are highly vulnerable to microbial contamination due to their soft tissues, making them perishable and harmful to consumers. The clinical and subclinical infections reported by fish consumers are mainly associated with pathogenic microorganisms in fish products. Therefore, this study aimed at establishing the molecular profiles and diversity of the bacterial isolates from fish and fish products obtained from Kirinyaga County markets in Kenya. A total of 660 samples were randomly sampled in six Kirinyaga County markets and transported to Kenyatta University for bacterial isolation

Screening methane-oxidizing bacteria from municipal solid waste landfills and simulating their effects on methane and ammonia reduction.

Municipal solid waste landfills are not only a crucial source of global greenhouse gas emissions; they also produce large amounts of ammonia (NH3), hydrogen sulfide, and other odorous gases that negatively affect the regional environment. Several types of methane-oxidizing bacteria (MOB) were proved to be effective in mitigating methane emission from landfills. Nevertheless, more MOB species and their technical parameters for best mitigating methane still need to be explored. In landfills, methane is simultaneously generated with ammonia, which may impede the CH4 bio-oxidizing process of MOB.

Effectiveness of biodegradation of petroleum products by mixed bacterial populations in liquid medium at different pH values.

The possibility of using mineral oils as a carbon source by bacteria adapted to high oil concentrations was tested in liquid media with different pH values (pH = 5, 7 and 9). Two types of inocula were tested: inoculum I consisted of selected strains used in the bioremediation of oil-contaminated soils and inoculum II contained bacteria isolated from soil samples previously bioremediated at pH = 5, 7 and 9. Biodegradation was observed in all the investigated media independently of initial pH value and type of inoculum used. After 21 days of cultivation the reduction of oil content reached 60-70

Endoscopically guided aerobic cultures in postsurgical patients with chronic rhinosinusitis.

BACKGROUND: There is considerable amount of debate in the literature regarding the microbial flora of normal, acutely infected, and chronically infected paranasal sinuses. Few studies have specifically looked at the microbial flora of healthy and infected sinus cavities after functional endoscopic sinus surgery. METHODS: One hundred thirty-four cultures were studied. All cultures were obtained using a standard technique. The nasal cavities were decongested and anesthetized, and nasal endoscopy was performed. When purulent secretions were identified, specimens of purulent secretions were obtai

Enzymatically derived chitooligosaccharides exhibit enhanced therapeutic efficacy against estrogen receptor-positive and triple-negative breast cancer cells.

This study investigates the enzymatic production of chitooligosaccharides (E-NACOS) from Alcaligenes faecalis chitinase and evaluates their potential therapeutic relevance in breast cancer models. Electrophoretic protein profiling combined with activity-based zymography of the crude supernatant identified four chitinase isoforms (∼45-150 kDa) exhibiting complementary endo- and exo-hydrolytic activities. HPLC analysis of colloidal chitin hydrolysates confirmed the generation of N-acetylglucosamine (GlcNAc) and short-chain oligomers (dimers to tetramers), reflecting the broad substrate specifici

Regulated intestinal microbiota and gut immunity to ameliorate type 1 diabetes mellitus: A novel mechanism for stem cell-based therapy.

Although stem cell transplantation is an effective strategy in the treatment of type 1 diabetes mellitus (T1DM), the mechanisms underlying its therapeutic effects remain unclear. We hypothesized that stem cells target gut microbiota and intestinal mucosal immunity to promote therapeutic effects against T1DM. We investigated the effects of human amniotic mesenchymal stem cells (hAMSCs) on intestinal microbiota and mucosal immunity in streptozotocin-induced T1DM mice. hAMSCs promoted significant reductions in blood glucose levels and increased the number of insulin-secreting cells in the T1DM mo

Trafficking of antigens from gut to sacroiliac joints and spine in reactive arthritis and spondyloarthropathies: Mainly through lymphatics?

Bacterial trafficking from gut to mesenteric lymph nodes is physiologic only for a few commensal species, like Alcaligenes which produces antimicrobial-substances inhibiting growth of pathogenic bacteria. In reactive arthritis, some living bacteria transiently manage to travel from gut to joints/enthesis within dendritic cells and/or macrophages. Migration of dead or dormant bacteria outside the gut in spondyloarthropathies, including those associated with Crohn's disease, can occur either through blood or lymphatics. Migration through lymphatics instead of blood depends on the host, but also
