This work addresses the changes in the phytohormonal signature in the recognition of the necrotrophic fungal pathogen Alternaria brassicicola by susceptible Brassica juncea and resistant Sinapis alba. Although B. juncea, S. alba and Arabidopsis all belong to the same family, Brassicaceae, the phytohormonal response of susceptible B. juncea towards this pathogen is unique because the latter two species express non-host resistance. The differential expression of the PR1 gene and the increased level of salicylic acid (SA) indicated that an SA-mediated biotrophic mode of defence response was trigg

Fungal pathogens, such as Fusarium spp. and Alternaria spp., cause substantial yield losses worldwide through root and fruit rot, wilting, and leaf and fruit spots, and increasing restrictions on chemical pesticides have intensified interest in sustainable alternatives such as essential oils, plant extracts, and microbial biocontrol agents. However, these three treatment categories are rarely compared directly under identical experimental conditions, which limits conclusions about their relative efficacy. This study directly compared, for the first time under the same in vitro conditions, the

Alternaria infectoria causes opportunistic human infections and is a source of allergens leading to respiratory allergies. In this work, we prepared cell wall nanoparticles (CWNPs) as a novel approach to study macrophage immunomodulation by fungal hyphal cell walls. A. infectoria was grown in the presence of caspofungin, an inhibitor of β(1,3)-glucan synthesis; nikkomycin Z, an inhibitor of chitin synthases; and pyroquilon, an inhibitor of dihydroxynaphthalene (DHN)-melanin synthesis. Distinct CWNPs were obtained from these cultures, referred to as casCWNPs, nkCWNPs, and pyrCWNPs, respectively

Tannic acid (TA), a naturally occurring polyphenol, exhibits broad-spectrum antimicrobial and antioxidant activities. This study elucidates the mechanisms by which TA controls Alternaria alternata in Korla fragrant pears. In vitro, TA at 10 mg mL-1 directly inhibited fungal growth by inducing hyphal deformation. In vivo, TA treatment significantly attenuated blackhead disease development. The underlying protective mechanism involved two coordinated pathways: first, TA enhanced the activity and gene expression of key antioxidant enzymes [Ascorbate peroxidase (APX), Glutathione reductase (GR), C

Sphingolipids play an important role in signal transduction pathways that regulate physiological functions and stress responses in eukaryotes. In plants, recent evidence suggests that their metabolic precursors, the long-chain bases (LCBs) act as bioactive molecules in the immune response. Interestingly, the virulence of two unrelated necrotrophic fungi, Fusarium verticillioides and Alternaria alternata, which are pathogens of maize and tomato plants, respectively, depends on the production of sphinganine-analog mycotoxins (SAMs). These metabolites inhibit de novo synthesis of sphingolipids in

This study investigated the effects of repeated dielectric barrier discharge (DBD) plasma applications on the morphological and physiological characteristics of pathogenic Alternaria and Fusarium species. Fungi, including both culture collection strains and environmental isolates, were exposed to sublethal doses of DBD plasma. The results demonstrated that the plasma exposure time required to achieve 90% cell mortality varied significantly among microorganisms, ranging from 2 min and 39 s for Fusarium culmorum DSM 1094 to 5 min and 19 s for Alternaria alternata DSM 62010. INTRODUCTION: Pseudostellaria heterophylla, a valuable medicinal herb in traditional Chinese medicine, is severely threatened by leaf spot disease caused by Alternaria alternata, leading to significant yield losses and mycotoxin contamination that compromises root quality and safety. This study aimed to characterize Bacillus amyloliquefaciens strain D-1 as a potential multifunctional biocontrol agent to mitigate this pathogen.
METHODS: Strain D-1 was isolated from the rhizosphere soil of P. heterophylla. Its antifungal efficacy was assessed through in vitro dual-culture assays and in planta p

Alternaria alternata infects grapes postharvest and causes harmful fruit rot. This study evaluated Tetraclinis articulata essential oil (TA-EO) nanoemulsion as a sustainable biocontrol strategy for Vitis vinifera cv. Red Globe. After storage, grapes were inoculated with A. alternata and treated with TA-EO nanoemulsions at different concentrations. Disease severity index (DSI), total polyphenol, flavonoid contents, and antioxidant activity were evaluated after 3, 6, 9, and 12 days. The EO, rich in α-pinene, camphor, and limonene, was successfully encapsulated into a stable nanoemulsion (358-392

The pressing need for sustainable, plant-based alternatives is highlighted by the growing resistance of agricultural pests to synthetic pesticides. This study examined the pesticidal potential of phytocompounds from C. tora discovered by GC-MS analysis against important tomato insect (T. absoluta) and fungal pathogen (A. solani). The binding stability and interaction dynamics of specific metabolites with fungal virulence (polygalacturonase, MAP kinase HOG1, and effector AsCEP50) and insect neuromuscular (ryanodine receptor and sodium channel protein) targets were assessed using molecular docki

Infection by fungal pathogens is the main factor leading to postharvest rot and quality deterioration of fruit and vegetables. Rotting caused by Alternaria alternata is a concerning disease for numerous crops in both production and postharvest stages, especially tomato black spots. In this study, the double Petri dish assay showed that the VOCs of Ceratocystis fimbriata WJSK-1 and Mby inhibited the mycelial growth of fungal pathogen A. alternata, with a percentage inhibition of 52.2% and 42.9%. Then, HS-SPME-GC-MS technology was used to analyze the volatiles produced by two strains of C. fimbr

The tangerine pathotype of Alternaria alternata can withstand high-level reactive oxygen species (ROS). By analyzing loss- and gain-of-function mutants, this study demonstrated that a Cys2His2 zinc finger-containing transcription regulator, A. alternata Stress Response Regulator 1 (AaSRR1), plays a negative role in resistance to peroxides and singlet-oxygen-generating compounds. AaSRR1 plays no role in cellular susceptibility or resistance to superoxide-producing compounds. AaSRR1 also negatively regulates conidiogenesis, maintenance of cell wall and membrane integrities, and chitin biosynthes

BACKGROUND: Aspergillus fumigatus and Alternaria alternata are ubiquitous environmental fungal allergens that can exacerbate airway inflammation and contribute to the disease process in patients with chronic rhinosinusitis (CRS). These antigens have been shown to induce human sinonasal epithelial cells (HSNECs) to promote a proinflammatory response, but what is unclear is a means by which to reduce these effects. Inhaled pathogens can induce HSNECs to produce reactive oxygen species (ROS) that trigger cytokine production.
OBJECTIVE: This study aimed to determine whether the free radical scave

Fungal infections are rare in the general population but are an emerging cause of disease in immunosuppressed patients, especially solid organ transplant recipients. Here, we report the case of a female Caucasian liver transplant patient who developed pulmonary nocardiosis two months after an episode of liver rejection. At the time of lung nocardiosis, she was being treated with tacrolimus and corticosteroids and suffered from diffuse papular skin lesions. She was initially suspected of having a cutaneous nocardial infection but culture examination revealed the presence of a dematiaceous fungu

Mold spoilage is a major challenge in semi-hard cheese production, causing economic losses and quality deterioration. This study evaluated the antifungal activity and mechanisms of essential oils (EOs) from Origanum vulgare (cultivar CARVA) and Thymus vulgaris (cultivar VARICO) against A. alternata isolated from Caciocavallo cheese alongside a preliminary assessment of their olfactory impact. A cheese agar medium (CAM) was developed to mimic the cheese matrix. The MICs of O. vulgare and T. vulgaris EOs were 0.94% and 0.12% (v/v), respectively, and both concentrations prevented fungal growth on

The synthesis of rhamnosylated compounds has gained great importance since these compounds have potential therapeutic applications. The enzymatic approaches for glycosylation of bioactive molecules have been well developed; however, the enzymatic rhamnosylation has been largely hindered by lacking of the glycosyl donor for rhamnosyltransferases. Here, we employed an α-L-rhamnosidase from Alternaria sp. L1 (RhaL1) to perform one-step rhamnosylation of anticancer drugs, including 2'-deoxy-5-fluorouridine (FUDR), cytosine arabinoside (Ara C), and hydroxyurea (Hydrea). The key synthesis conditions

The antifungal activities of many sulfur-containing defense compounds suggest a connection between pathogen infection, primary sulfur metabolism and sulfate nutritional status of plants. This relationship was investigated using Arabidopsis thaliana plants that were cultivated under different sulfur regimes and challenged by Alternaria brassicicola. Plants grown with 500 μM sulfate were significantly less infected compared to plants grown on 50 μM sulfate. Upon infection, the formation of the sulfur-containing defense compound camalexin and the gene expression of the sulfur-rich defense peptide

Tomato black spot disease, caused by the fungus Alternaria alternata, leads to plant wilting and chlorosis and results in sunken, black lesions on harvested fruit, thereby causing significant economic losses in the tomato industry. To mitigate issues related to environmental contamination and the emergence of resistant pathogens, this study investigated a high-efficacy, low-toxicity biocontrol strategy. The marine-derived fungus Q8-1 inhibited the growth of A. alternata by up to 87.04 %. Based on morphological features and ITS rDNA sequencing, the strain was identified as Epicoccum nigrum. Gen
