Illumina-Based Metagenomic Insights into the Gut Microbiome of Amblyomma helvolum (Koch, 1844) Parasitizing Xenochrophis trianguligerus from Great Nicobar Island, India.

During a faunal survey in Great Nicobar Island, we collected four Amblyomma helvolum ticks infesting the snake Xenochrophis trianguligerus and processed for gut-metagenomic analysis using Illumina paired-end sequencing. A total of 8.7 million high-quality reads were generated, revealing that the gut microbiome was dominated by Bacteria (∼99.9%), primarily represented by Proteobacteria (∼95.7%), followed by minor fractions of Firmicutes, Actinobacteria, and Bacteroidetes. The predominant bacterial families were Alcaligenaceae, Bradyrhizobiaceae, Boseaceae, and Rickettsiaceae, with Achromobacter

Evaluation of different chemical groups used for the control of Rhipicephalus microplus against ticks from the Amblyomma cajennense complex parasitizing cattle.

The objective of this study was to evaluate the efficacy of different chemical groups, approved as acaricide to Rhipicephalus microplus, against ticks from the Amblyomma cajennense complex parasitizing cattle under laboratory and field conditions. Laboratory trials (LT) used calves artificially infested with Amblyomma tonelliae, while field trials (FT) involved cattle naturally infested with Amblyomma sculptum. Bovines were treated with commercial formulations of amitraz (only FT), doramectin, deltamethrin, fipronil, ivermectin, or a combination of fipronil and ivermectin, using doses recommen

Metataxonomic profiles of bacterial and parasitic communities in Amblyomma spp. ticks collected from wildlife in Colombia: Implications for tick-borne diseases.

As a tropical country, Colombia hosts a wide range of arthropods that can act as vectors of disease-causing pathogens, particularly those carrying hemopathogens. Ticks play a crucial role in the transmission of zoonotic pathogens, impacting both human and veterinary health. The pathogen load of ticks from wildlife is of particular concern, as it can contribute to the spillover of infectious agents to domestic animals and humans, highlighting the need for surveillance and control strategies to mitigate emerging tick-borne diseases. Therefore, this study aimed to determine the presence of microo

Babesia bigemina (smith and Kilbourne, 1893) detection in Amblyomma sculptum (Berlese, 1888) ticks in the Mato Grosso do Sul state, Brazil.

Ticks parasitize various hosts, including humans, and are known to transmit pathogens that can be harmful not only to animals but also to humans. To evaluate the possible presence of pathogens in ticks, we aimed to collect and identify tick fauna specimens in Lagoa Comprida Municipal Natural Park, an anthropogenic urban area located in Aquidauana, Mato Grosso do Sul, Brazil. A total of 1216 ticks, of which 51.2% were Amblyomma sculptum, 1.2% were Amblyomma dubitatum, and 41% were Amblyomma spp. were collected. These results show that the prevalence of A. sculptum is significantly higher than t

The Lone Star Tick, Amblyomma americanum, salivary factors exacerbate the clinical outcome of Heartland virus disease in a small animal model.

Heartland virus was first isolated in 2009 from two patients in Missouri and is transmitted by the lone star tick, Amblyomma americanum. To understand disease transmission and pathogenesis, it is necessary to develop an animal model that utilizes the natural transmission route and manifests in a manner similar to documented human cases. Herein we describe our investigations on identifying A129 mice as the most appropriate small animal model for HRTV pathogenesis that mimics human clinical outcomes. We further investigated the impact of tick saliva in enhancing pathogen transmission and clinica

Lethal Effects of a Silica Gel + Thyme Oil (EcoVia) Dust and Aqueous Suspensions on Amblyomma americanum (Ixodida: Ixodidae) Larvae and Nymphs.

Ixodids suck blood from host animals and transmit pathogens that cause important medical and veterinary diseases. As synthetic conventional acaricide resistance becomes increasingly common, alternative tactics are likely to enhance control efforts. Laboratory bioassays assessed the lethality of CimeXa, a commercial silica gel desiccant dust, and EcoVia, a commercial product containing silica gel + thyme oil, against lone star tick, Amblyomma americanum (L.), larvae and nymphs. Both life stages were completely killed by CimeXa by 24 h, and EcoVia achieved total larval mortality within 1 h when

High prevalence of Rickettsia africae variants in Amblyomma variegatum ticks from domestic mammals in rural western Kenya: implications for human health.

Tick-borne spotted fever group (SFG) rickettsioses are emerging human diseases caused by obligate intracellular Gram-negative bacteria of the genus Rickettsia. Despite being important causes of systemic febrile illnesses in travelers returning from sub-Saharan Africa, little is known about the reservoir hosts of these pathogens. We conducted surveys for rickettsiae in domestic animals and ticks in a rural setting in western Kenya. Of the 100 serum specimens tested from each species of domestic ruminant 43% of goats, 23% of sheep, and 1% of cattle had immunoglobulin G (IgG) antibodies to the SF

Detection of Rickettsia parkeri and Candidatus Rickettsia andeanae in Amblyomma maculatum Gulf Coast ticks collected from humans in the United States.

Rickettsia parkeri, a spotted fever group (SFG) rickettsia recently found to be pathogenic to humans, causes an eschar-associated febrile illness. The R. parkeri rickettsiosis, Tidewater spotted fever, has been misdiagnosed as Rocky Mountain spotted fever due to serologic cross reactivity and the lack of specific diagnostic methods. Candidatus Rickettsia andeanae, also a SFG rickettsia, is a recently described agent of unknown pathogenicity originally identified in ticks collected from domestic animals during a fever outbreak investigation in northern Peru. Among 37 Amblyomma maculatum (collec

Efficacy of pheromone-acaricide-impregnated tail-tag decoys for controlling the bont tick, Amblyomma hebraeum (Acari: Ixodidae), on cattle in Zimbabwe.

A large-scale field test using pheromone-acaricide-impregnated plastic tail-tag decoys demonstrated excellent efficacy of these devices for control of the bont tick, Amblyomma hebraeum, on cattle in Zimbabwe. The tail tags were impregnated with a mixture containing o-nitrophenol, methyl salicylate, 2,6-dichlorophenol and phenylacetaldehyde and one of three different acaricides (cyfluthrin, flumethrin or alphacypermethrin). o-Nitrophenol and methyl salicylate are components of the A. hebraeum attraction-aggregation-attachment pheromone, while 2,6-dichlorophenol and phenylacetaldehyde are proven

Repeated feeding of Amblyomma herbraeum (Acarina: Ixodidae) immatures on laboratory hosts. Host effects on tick yield, engorged weight and engorgement period.

Rabbits and sheep are unable to acquire a resistance to larvae and nymphs of Amblyomma hebraeum. After repeated infestation of hosts there is no progressive decline in either the tick yield or the engorged weight of fed ticks. Seasonal fluctuations in engorged weights do occur however, with the weight declining in early to midwinter and increasing in early to midsummer. Changes in hose physiology as a result of low temperature acclimatization appear to be responsible for the fluctuations. Tick yield is determined by the amount of grooming undertaken by hosts. The feedings periods of larvae and

Tissue-specific and sex-dependent microbiome structure in East African cattle ticks (Amblyomma gemma and Rhipicephalus pulchellus) and their bovine hosts.

Ticks and their vertebrate hosts share intimate ecological interactions that shape microbial community composition, with implications for tick fitness and pathogen transmission. However, the extent and specificity of tick-host microbial interactions remain poorly understood, particularly in East African tick species. Here, we characterised microbial communities across multiple tissue types from two economically important tick species (Amblyomma gemma and Rhipicephalus pulchellus) and their shared bovine host using 16S rRNA V1-V2 gene sequencing. We profiled haemolymph, midgut, salivary glands,

cAmbly, a peptide derived from the star tick Amblyomma sculptum salivary gland transcriptome, attenuates inflammatory pain by activating endogenous opioid pathways.

Pain affects more than 1.5 billion people worldwide and remains a leading cause of disability, but currently available analgesics often have limited efficacy and significant adverse effects. In this context, bioactive molecules derived from animal venoms and secretions have emerged as promising novel therapeutic agents. Here, we investigated the antinociceptive and anti-inflammatory properties of cAmbly, a cell-penetrating peptide derived from the C-terminal region of Amblyomin-X, a protein from the salivary gland transcriptome of the Amblyomma sculptum tick. Beyond the usual suspects: Uncovering less-recognized pathogenic bacteria in Ghanaian blood-feeding Amblyomma variegatum ticks using 16S rRNA amplicon sequencing.

Ticks are important vectors of bacterial pathogens affecting both human and animal health. In Ghana, Amblyomma variegatum is the predominant cattle-infesting tick, yet most studies have focused on a limited range of well-characterized pathogens, potentially overlooking a broader diversity of less-recognized, emerging, or opportunistic bacteria. In this study, we used 16S rRNA amplicon sequencing to characterize the bacteriome of partially blood-fed Am. variegatum ticks, with emphasis on underexplored taxa. As ticks were blood-fed at the time of collection, some detected microorganisms may repr

First report of Anaplasma spp., Ehrlichia spp., and Rickettsia spp. in Amblyomma gervaisi ticks infesting monitor lizards (Varanus begalensis) of Pakistan.

Ticks pose significant health risks to both wildlife and humans due to their role as vectors for various pathogens. In this study, we investigated tick infestation patterns, tick-associated pathogens, and genetic relationships within the tick species Amblyomma gervaisi, focusing on its prevalence in monitor lizards (Varanus bengalensis) across different districts in Pakistan. We examined 85 monitor lizards and identified an overall mean intensity of 19.59 ticks per infested lizard and an overall mean abundance of 11.98 ticks per examined lizard. All collected ticks (n = 1019) were morphologica

Rickettsia amblyommatis-infected Amblyomma coelebs parasitizing a human traveler in Rio Grande do Sul, southern Brazil, after returning from the Amazon.

With increased national and international transportation, humans as well as domestic and wild animals are able to get from one end to the other within hours, accidently carrying transboundary vectors and pathogens. We report here, the introduction of Amblyomma coelebs in Rio Grande do Sul, the southernmost state of Brazil, via a man travelling to the Carajás National Forest in the state of Pará (2661 km away). Upon return to his residence in Porto Alegre, RS, he realized that he was parasitized by one tick. The tick was identified as a nymph of Amblyomma sp., placed in a Biological Oxygen Dema

Rickettsia rickettsii (Rickettsiales: Rickettsiaceae) Infecting Amblyomma sculptum (Acari: Ixodidae) Ticks and Capybaras in a Brazilian Spotted Fever-Endemic Area of Brazil.

The bacterium Rickettsia rickettsii is the agent of Brazilian spotted fever (BSF), a highly fatal disease that is transmitted in Brazil mainly by the tick Amblyomma sculptum Berlese, which uses capybaras (Hydrochoerus hydrochaeris Linnaeus) as major hosts. In 2015, we captured nine capybaras in a BSF-endemic area of southeastern Brazil. From each capybara, we collected blood sera that were tested through the immunofluorescence assay using Rickettsia spp. antigens, and A. sculptum ticks, processed for isolation of R. rickettsii through guinea pig inoculation. All capybaras (100%) were seroreact

Genetic diversity of Rickettsia africae isolates from Amblyomma hebraeum and blood from cattle in the Eastern Cape province of South Africa.

Rickettsia africae is a re-emerging tick-borne pathogen causing African tick bite fever (ATBF) in humans. Amblyomma variegatum is the principal vector in most sub-Sahara African countries, whereas in South Africa it is A. hebraeum. Reports of high genetic heterogeneity among R. africae isolates in southern Africa have prompted the need for molecular investigations of isolates form South Africa. Therefore, this study aimed to determine the prevalence and genetic diversity of R. africae in A. hebraeum collected from cattle, grazing pasture, as well as from blood of cattle in the Eastern Cape Pro

A putative novel strain of Ehrlichia infecting Amblyomma tigrinum associated with Pampas fox (Lycalopex gymnocercus) in Esteros del Iberá ecoregion, Argentina.

The current work evaluated road-killed Pampas foxes (Lycalopex gymnocercus) and their ticks for the presence of vector-borne agents in the ecoregion Esteros del Iberá in northeastern Argentina. Spleen, lung and blood samples and Amblyomma tigrinum adult ticks collected from the foxes were tested by polymerase chain reaction (PCR) assays targeting bacteria of the genera Ehrlichia, Anaplasma, and Rickettsia. All foxes tested were negative for the three genera, but evidence of Ehrlichia and Rickettsia infection was detected in the ticks. One A. tigrinum (out of 12 tested) was infected by an ehrli

Influence of hosts on the oviposition and hatching of engorged females Amblyomma cajennense sensu stricto (Acari: Ixodidae) with notes on new host records.

The parasite-host relationships of Amblyomma cajennense sensu stricto, one of the species of the Amblyomma cajennense complex that occurs in the state of Maranhão, are poorly known and are restricted to its association with horses and humans. This study evaluated, for the first time, the effect of hosts on the oviposition of A. cajennense engorged females, naturally collected from wild animals and humans, as well as those experimentally fed on cattle and highlights the importance of the relationship between A. cajennense and different hosts in maintaining tick populations in the Amazon biome.
