Archaeoparasitological and biomolecular analysis of the wells from the Black Sea city of Phanagoria during the Khazar period (8th-9th centuries A.D.).

Ancient water supply systems provide important insights into historical sanitation practices and disease transmission patterns. This study examined soils from 2 wells of the medieval city of Phanagoria (8th-9th centuries AD) to reconstruct parasite contamination patterns during the Khazar period. Soil samples from wells 33 and 46 were analysed using light microscopy for parasite egg identification and gas chromatography-mass spectrometry (GC-MS) for faecal biomarker detection. Microscopic analysis identified 6 types of parasite eggs: Trichuris trichiura/suis, Trichuris muris, Ascaris lumbricoi

Leishmania vaccine development: A comprehensive review.

Infectious diseases like leishmaniasis, malaria, HIV, tuberculosis, leprosy and filariasis are responsible for an immense burden on public health systems. Among these, leishmaniasis is under the category I diseases as it is selected by WHO (World Health Organization) on the ground of diversity and complexity. High cost, resistance and toxic effects of Leishmania traditional drugs entail identification and development of therapeutic alternative. Since the natural infection elicits robust immunity, consistence efforts are going on to develop a successful vaccine. Clinical trials have been conduc

Kocuria rosea Sepsis in an Immunocompromised Patient: A Case Report.

Kocuria species, classified within the phylum Actinomycetota, class Actinomycetes, order Micrococcales, family Micrococcaceae, are Gram-positive coccoid bacteria. They bear morphological resemblance to Staphylococci spp and Micrococci spp, which often leads to misidentification and oversight as contaminants, given their presence as normal flora on human and animal skin and mucous membranes. Accurate identification of these organisms typically relies on automated systems such as MALDI-TOF-MS, Vitek-2 System, and 16S rRNA studies. Once considered rare, there is increasing recognition of Kocuria

Transcriptome study reveals tick immune genes restrict Babesia microti infection.

A systems biology approach was employed to gain insight into tick biology and interactions between vectors and pathogens. Haemaphysalis longicornis serves as one of the primary vectors of Babesia microti, significantly impacting human and animal health. Obtaining more information about their relationship is crucial for a comprehensive understanding of tick and pathogen biology, pathogen transmission dynamics, and potential control strategies. RNA sequencing of uninfected and B. microti-infected ticks resulted in the identification of 15 056 unigenes. Among these, 1 051 were found to be differe

Inflammatory Mediators of Axon Regeneration in the Central and Peripheral Nervous Systems.

Although most pathways in the mature central nervous system cannot regenerate when injured, research beginning in the late 20th century has led to discoveries that may help reverse this situation. Here, we highlight research in recent years from our laboratory identifying oncomodulin (Ocm), stromal cell-derived factor (SDF)-1, and chemokine CCL5 as growth factors expressed by cells of the innate immune system that promote axon regeneration in the injured optic nerve and elsewhere in the central and peripheral nervous systems. We also review the role of ArmC10, a newly discovered Ocm receptor,

Perinatal asphyxia: a review from a metabolomics perspective.

Perinatal asphyxia is defined as an oxygen deprivation that occurs around the time of birth, and may be caused by several perinatal events. This medical condition affects some four million neonates worldwide per year, causing the death of one million subjects. In most cases, infants successfully recover from hypoxia episodes; however, some patients may develop HIE, leading to permanent neurological conditions or impairment of different organs and systems. Given its multifactor dependency, the timing, severity and outcome of this disease, mainly assessed through Sarnat staging, are of difficult

Pathogenic, diagnostic and vaccine potential of leptospiral outer membrane proteins (OMPs).

Pathogenic Leptospira species are important human and animal pathogen that causes leptospirosis, with more than half a million cases reported annually but little is known regarding the true incidence of leptospirosis due to the limitations in diagnosis. Proteins embedded in the outer membrane are found to be prime drug targets due to its key role as receptors for cellular communication and gatekeepers for iron and substrate transport across cell membranes. The major key issues to be addressed to overcome the disease burden of leptospirosis are: need to identify the genes that turn on in vivo;

Genetics of panic disorder: focus on association studies and therapeutic perspectives.

There is evidence for either genetic heterogeneity or complex inheritance with an interaction of environmental factors and multiple single genes in the etiology of panic disorder. Although linkage analyses of panic disorder have implicated several chromosomal regions including 1q, 2q, 4q, 7p, 9q, 12q, 13q, 15q and 22q, they so far have not been able to identify a major gene responsible for panic disorder. Several genes of classical candidate neurotransmitter systems have been reported to be associated with panic disorder. Genetic variation in genes of monoamine oxidase A, catechol-O-methyltran

Multidrug-resistant Staphylococcus pettenkoferi isolated from cat in India.

BACKGROUND AND AIM: Coagulase-negative staphylococci (CoNS) are considered to be one of the emerging pathogens in human and animals in recent times. Staphylococcus pettenkoferi, a novel pathogen under CoNS, is discovered in 2002 in humans with multiple clinical manifestations in various patients. To date, the pathogens have not yet been reported from any animals. The present study reported the first ever isolation, identification, and characterization of multidrug-resistant S. pettenkoferi from a cat with peritonitis in India. MATERIALS AND METHODS: Peritoneal fluid was collected aseptically

Asking the Right Questions With Animal Models: Methionine- and Choline-Deficient Model in Predicting Adverse Drug Reactions in Human NASH.

In the past few decades, great conceptual and technological advances have been made in the field of toxicology, but animal model-based research still remains one of the most widely used and readily available tools for furthering our current knowledge. However, animal models are not perfect in predicting all systemic toxicity in humans. Extrapolating animal data to accurately predict human toxicities remains a challenge, and researchers are obligated to question the appropriateness of their chosen animal model. This paper provides an assessment of the utility of the methionine- and choline-defi

ECVAM and new technologies for toxicity testing.

The development of alternative empirical (testing) and non-empirical (non-testing) methods to traditional toxicological tests for complex human health effects is a tremendous task. Toxicants may potentially interfere with a vast number of physiological mechanisms thereby causing disturbances on various levels of complexity of human physiology. Only a limited number of mechanisms relevant for toxicity ('pathways' of toxicity) have been identified with certainty so far and, presumably, many more mechanisms by which toxicants cause adverse effects remain to be identified. Recapitulating in empiri

Evolving concepts in molecular pathology.

During the last decade, an understanding of the causes of many human diseases has progressed rapidly, in large measure because of the development of technologies that allow us to identify the genes that are involved. Identification of a gene that is suspected to play an important role in a particular disease opens up a whole new dimension of research to understand the molecular events that underlie the cause of that disorder. A crucial step in this process is often the development of an animal model of the disease. Again, the last decade has seen rapid advances in our ability to create such mo

Understanding error culture in veterinary medicine: a survey among veterinarians across German-speaking countries.

INTRODUCTION: Errors are assumed to occur frequently in veterinary practice and may affect animal health, client trust, and the well-being of veterinary staff. Their handling is shaped by the prevailing error culture within practices and institutions. While error culture has been investigated in human medicine and extensively in other high-risk fields such as aviation, little is known about how it is perceived within veterinary medicine. METHODS: This study therefore examined practising veterinarians' perceptions of errors, contributing factors, reporting practices, and institutional response

Taphonomic Analysis of the Faunal Assemblage Associated with the Hominins (Australopithecus sediba) from the Early Pleistocene Cave Deposits of Malapa, South Africa.

Here we present the results of a taphonomic study of the faunal assemblage associated with the hominin fossils (Australopithecus sediba) from the Malapa site. Results include estimation of body part representation, mortality profiles, type of fragmentation, identification of breakage patterns, and microscopic analysis of bone surfaces. The diversity of the faunal spectrum, presence of animals with climbing proclivities, abundance of complete and/or articulated specimens, occurrence of antimeric sets of elements, and lack of carnivore-modified bones, indicate that animals accumulated via a natu

Heat and chemical stressors: Independent and interactive impacts on the pig ovary.

Seasonal infertility as a phenotypic consequence of heat stress has been recognized in production animals for decades and more recently in women. Systemic impacts of heat stress include compromised intestinal integrity, and elevated prolactin and insulin. These effects alone can compromise female fertility. There are also ovarian endocrine changes and molecular sequelae that have been identified. This review summarizes the current literature with a focus on the pig as a model. In addition to direct thermal effects on fecundity, the influence of heat on the ovarian capacity to respond to chemic

Commentary - In silico acute oral toxicity models are fit for inclusion in GHS and DG Model regulations classification criteria.

This commentary advocates for the inclusion of in silico acute oral toxicity (AOT) models within the classification criteria of the Globally Harmonized System (GHS) and Dangerous Goods (DG) Model Regulations. AOT data, traditionally derived from animal studies, are central to hazard classification and regulatory compliance. However, these studies involve severe outcomes including significant toxicity and death as an endpoint and are conducted primarily to support categorical classification rather than for mechanistic understanding. Recent cross-sector evaluations demonstrate that current in si

Eradication of feline dermatophytosis in a shelter: a field study.

Enzootic dermatophytosis in a shelter with approximately 140 cats was treated according to a protocol combining identification, isolation and treatment of subclinical carrier and affected animals in accordance with a three-area system: healthy animals (no lesions and negative cultures), subclinical carrier animals (no lesions but with positive cultures) and clinically affected animals (lesions and positive cultures). The cats were examined and inspected under a Wood's lamp and had samples taken for fungal culture every 2 weeks. Thirty-three per cent of the cats had a positive fungal culture at

Teaching a new mouse old tricks: Humanized mice as an infection model for Variola virus.

Smallpox, caused by the solely human pathogen Variola virus (VARV), was declared eradicated in 1980. While known VARV stocks are secure, smallpox remains a bioterrorist threat agent. Recent U.S. Food and Drug Administration approval of the first smallpox anti-viral (tecovirimat) therapeutic was a successful step forward in smallpox preparedness; however, orthopoxviruses can become resistant to treatment, suggesting a multi-therapeutic approach is necessary. Animal models are required for testing medical countermeasures (MCMs) and ideally MCMs are tested directly against the pathogen of interes

Molecular detection and genetic analysis of Candida species isolated from bovine clinical mastitis in India.

Bovine clinical mastitis is an economically important disease in dairy industry worldwide resulting in reduction of milk yield and quality. Among mycotic mastitis, Candida spp. are commonly occurring opportunistic mycosis in immunocompromised animals. The micro-organism's causing mastitis has high zoonotic potential and has been linked with rapid growth and introduction of antimicrobial resistance between animals and humans. The present study was conducted to isolate and identify the common pathogenic Candida spp. from bovine mastitis cases in India. The isolates were phenotypically characteri
