Protective effect of apigenin on d-galactosamine/LPS-induced hepatocellular injury by increment of Nrf-2 nucleus translocation.

Apigenin has a protective effect on D-galactosamine (D-GalN)/lipopolysaccharide (LPS)-induced mouse liver injury through the increments of hepatic nuclear factor erythroid 2-related factor 2 (Nrf-2) and peroxisome proliferator-activated receptor γ (PPARγ) expressions, but its exact mechanisms are still uncertain. This study aimed to further verify its protective effect on hepatocytes and to determine its target of action. The results showed that after treatment of D-GalN/LPS-stimulated hepatocytes with 2.5-20 μM apigenin, the supernatant alanine aminotransferase, aspartate aminotransferasein,

Apigenin: Selective CK2 inhibitor increases Ikaros expression and improves T cell homeostasis and function in murine pancreatic cancer.

Pancreatic cancer (PC) evades immune destruction by favoring the development of regulatory T cells (Tregs) that inhibit effector T cells. The transcription factor Ikaros is critical for lymphocyte development, especially T cells. We have previously shown that downregulation of Ikaros occurs as a result of its protein degradation by the ubiquitin-proteasome system in our Panc02 tumor-bearing (TB) mouse model. Mechanistically, we observed a deregulation in the balance between Casein Kinase II (CK2) and protein phosphatase 1 (PP1), which suggested that increased CK2 activity is responsible for re

5-Fluorouracil combined with apigenin enhances anticancer activity through mitochondrial membrane potential (ΔΨm)-mediated apoptosis in hepatocellular carcinoma.

The development of chemoresistance may reduce the efficacy of chemotherapeutic drugs for treating hepatocellular carcinoma (HCC). In the present study, the effects of apigenin on intensifying the chemosensitivity of HCC cells and an HCC xenograft model in response to 5-fluorouracil (5-FU) were investigated. Sub-toxic concentrations of apigenin (4 μmol/L) significantly enhanced the cytotoxicity of 5-FU (100 μg/mL) in HCC cells. In vivo, combined treatment with apigenin (20 mg/kg, five times/week for 3 weeks) and 5-FU (20 mg/kg for 5 consecutive days) significantly inhibited the growth of HCC xe

Apigenin blocks induction of vascular endothelial growth factor mRNA and protein in progestin-treated human breast cancer cells.

OBJECTIVE: The results of recent clinical trials indicate that hormone therapy with estrogen and progestin is associated with higher risk of breast cancer in postmenopausal women than treatment with estrogen alone or placebo. This observation is consistent with studies showing that progestins stimulate the expression of vascular endothelial growth factor (VEGF), which in turn stimulates angiogenesis. The objective of this study was to examine whether apigenin, a natural flavone, inhibits the progestin-dependent induction of VEGF in human breast cancer cells. METHODS: T47-D human breast cancer

Repurposed metformin-apigenin combination therapy for metabolic dysfunction-associated steatohepatitis: from integrative multi-omics discovery to in vivo validation.

BACKGROUND: Metabolic dysfunction-associated steatohepatitis (MASH) is a chronic liver disease. Although recent drugs have been approved, the complex pathophysiology of MASH limits monotherapy efficacy and generates considerable inter-patient heterogeneity. The purpose of this research was to identify new molecular targets and develop a rational combination therapeutic approach for MASH. METHODS: To determine the molecular signatures of MASH mouse models and healthy controls, we conducted bulk RNA sequencing on liver tissues and then analyzed the data with a variety of Gene Ontology, Kyoto En

Apigenin Upregulates Regulatory B Cells, Suppresses Class Switch Recombination, and Restores Immune Homeostasis to Treat Autoimmune Arthritis and Sepsis.

BACKGROUND: Restoring immune homeostasis is crucial for inflammation resolution, in which B cells play dual regulatory roles. A promising therapeutic strategy involves simultaneously suppressing proinflammatory B-cell activity while enhancing regulatory B cells (Bregs). Natural anti-inflammatory compounds, such as the flavonoid apigenin (API), may achieve this dual immunomodulation, yet the precise mechanisms of API on B cells remain incompletely understood. PURPOSE: This study aimed to elucidate how API modulates B cells to exert immunoregulatory and protective effects in inflammatory condit

Effect of dietary apigenin on colonic ornithine decarboxylase activity, aberrant crypt foci formation, and tumorigenesis in different experimental models.

The efficacy of dietary apigenin, a dietary flavonoid, in colon cancer prevention was investigated by evaluating the inhibition of the ornithine decarboxylase (ODC) activity and the formation of aberrant crypt foci (ACF) and by studying the ability of apigenin to block colon carcinogenesis in two mouse models. First, the activity of ODC was measured in colon cancer cells (Caco-2) and in the colon epithelium of CF-1 mice. Apigenin at 10 and 30 muM significantly inhibited the ODC activity of Caco-2 cells by 26% and 57%, respectively. Colonic ODC activity in CF-1 mice was reduced with 0.1% dietar

The Effects of Resveratrol and Apigenin on Jejunal Oxidative Injury in Ducks and on Immortalized Duck Intestinal Epithelial Cells Exposed to H2O2.

Oxidative stress increases the apoptosis of intestinal epithelial cells and impairs intestinal epithelial cell renewal, which further promotes intestinal barrier dysfunction and even death. Extensive evidence supports that resveratrol and apigenin have antioxidant, anti-inflammatory, and antiproliferative properties. Here, we investigated the ability of these two compounds to alleviate diquat-induced jejunal oxidative stress and morphological injury, using the duck as a model, as well as the effects of apigenin on oxidative stress induced by H2O2 in immortalized duck intestinal epithelial cell
