Influence of different histamine receptor agonists and antagonists on apomorphine-induced licking behavior in rat.

The effects of different histamine receptor agonists and antagonists on apomorphine-induced licking behavior in rats were investigated. Subcutaneous (s.c.) injection of various doses of apomorphine (0. 125-1.25 mg/kg) induced licking. The licking response was counted by direct observation and recorded for a 75-min period. Intracerebroventricular (i.c.v.) or intraperitoneal (i.p.) injection of the histamine H(1) or H(2) receptor agonist, HTMT (6-[2-(4-imidazolyl)ethylamino]-N-(4-trifluoromethylphenyl) heptanecarboxamide) (50 and 100 microg per rat), or dimaprit (10 and 15 mg/kg, i.p.), respecti

Selective metabolic activation by apomorphine in striosomes of denervated striatum in MPTP-induced hemiparkinsonian monkeys.

Results of previous studies have suggested differences in the regulatory mechanisms in striatal striosomes and matrix following interruption of dopaminergic input to the striatum by MPTP in the monkey. In the present study we have investigated the possibility that stimulation of dopamine receptors by apomorphine modifies glucose metabolism differentially in the two striatal compartments of unilaterally MPTP-lesioned monkeys. Apomorphine treatment was found to result in higher rates of glucose utilization in the denervated striatum than in the intact hemisphere. Furthermore, the effect was more

Effect of apomorphine on rat motor activity induced by fencamfamine and other indirect psychostimulant drugs.

We evaluated the effects of low doses of apomorphine on the stimulant behavioral effects induced by amphetamine (2.5 mg/kg), fencamfamine (6.0 mg/kg) and cocaine (15.0 mg/kg). Rats received 0.02 mg/kg of apomorphine (sc) and 30 min later were injected with one of the stimulants. Motor activity including locomotion, rearing and sniffing was quantified in the animals home cages for 60 min at 15-min intervals. All stimulant drugs induced hyperactivity. When apomorphine was administered before cocaine, but not when administered before fencamfamine or amphetamine, distinctive changes occurred. The

Effects of apomorphine, physostigmine and vasoactive intestinal peptide on penile erection and yawning in diabetic rats.

The present report describes for the first time the effects of systemic administration of apomorphine and of physostigmine, as well as the effects of central and systemic administration of vasoactive intestinal peptide (VIP), on penile erection and yawning in rats with streptozotocin-induced diabetes. Systemic administration of apomorphine induced both penile erection and yawning in non-diabetic rats but not in diabetic rats, while that of physostigmine induced only yawning in non-diabetic rats, and neither yawning nor penile erection in diabetic rats. Intracerebroventricular administration of

Differential release of vasopressin and oxytocin in response to abdominal vagal afferent stimulation or apomorphine in the ferret.

The aim of this study was to investigate whether direct afferent stimulation of the abdominal vagus could promote release of the neurohypophyseal hormones. The nucleus of the solitary tract is the major recipient of vagal afferent information, and this region of the brainstem may also be activated by stimulation of the area postrema. For this reason apomorphine, a D2 dopaminergic agonist which acts on the area postrema, and can evoke vasopressin secretion in man, was also investigated for its effect on vasopressin and oxytocin release. Our results show that vasopressin, but not oxytocin is rel

Involvement of GABAergic systems and benzodiazepine receptors in the jumping behavior induced by harmine and apomorphine in rats.

Harmine (HA) induces a jumping behavior in rats when the central dopaminergic function has been activated. Possible involvement of the GABAergic systems and the benzodiazepine receptors (BZA-R) in the jumping behavior induced by HA and apomorphine (APO) was investigated. Pretreatment with GABAergic agonists, muscimol (0.05-0.2 mg/kg) and diazepam (0.2-1.0 mg/kg), and antagonists, picrotoxin (0.1-0.5 mg/kg) and bicuculline (0.1-0.5 mg/kg), suppressed the jumping behavior in a dose-dependent manner in rats treated with HA (10 mg/kg) and APO (2 mg/kg). After treatment with 2, 5 or 10 mg/kg of HA

Desenkephalin-gamma-endorphin restores the impaired sensitivity of hypophysectomized rats to small doses of apomorphine.

Hypophysectomized male rats, tested 7 days after removal of the pituitary, showed a reduced sensitivity to small doses of apomorphine. In these rats, subcutaneous (s.c.) treatment with apomorphine (25, 100 or 250 micrograms/kg) did not elicit any reduction in locomotor activity measured 5 min after injection in contrast to that observed in sham-operated control rats. The enhancement of locomotor activity and stereotyped behavior, 20 min after the s.c. administration of apomorphine, was similar in hypophysectomized rats and in control animals. Prolactin does not seem to be implicated in this al

The effects of apomorphine, d-amphetamine and chlorpromazine on the aggressiveness of isolated Aequidens pulcher (Teleostei, Cichlidae).

Apomorphine, d-amphetamine, chlorpromazine and dopamine, added to the aquarium water, were tested for their effects on the aggressive responses of isolated fish to mirrors and to models, compared with the responses of the same fish as non-treated controls tested at the start of, in the middle, and at the end of the experiment (Experiment 1). The effects of d-amphetamine administered by intracranial injection were also evaluated for responses to mirror presentations (Experiment 2). The three sets of controls in Experiment 1 indicate that the responses of the fish apparently changed over the 32-

Relationship of psychotic symptom clusters in schizophrenia to neuroleptic treatment and growth hormone response to apomorphine.

The authors propose an alternative model for relating clinically rated psychotic symptoms to biological measures in schizophrenic patients. They suggest that clinical presentation in schizophrenic patients comprises at least four distinct psychotic symptom clusters and that at most one or two of the symptom clusters are closely associated with central dopamine (DA) activity as measured by growth hormone (GH) response to apomorphine. Factor and cluster analytic techniques both identified the same four psychotic symptom clusters, three of which were similar to the major subtypes of schizophrenia

S(+)Apomorphines. Selective inhibition of excitatory effects of dopamine injected into the limbic system of the rat.

Dopamine (DA) injected unilaterally into the corpus striatum of the brain of the rat after pretreatment with a monoamine oxidase inhibitor, induced contralateral deviation of the head (ED50 = 23 micrograms). Dopamine, administered similarly into the nucleus accumbens septi, induced strong locomotor arousal effects (ED50 = 32 micrograms), as reported by others. Systemic administration of the S(+) isomers of apomorphine (APO) and its N-n-propyl analog (NPA) had no significant action on the effects on posture of DA given intrastriatally, even in large doses. In striking contrast, both agents sele

Interaction of drugs with apomorphine, tryptamine and norepinephrine. A new 'in vivo' approach: the ATN-test in rats.

A new experimental test procedure is described for the 'in vivo' study of drug interactions with dopamine (DA), 5 hydroxy-tryptamine (5-HT) and norepinephrine (NE). The ultimate aim of this study is to provide an empirical evaluation of the relative specificity with which drugs may affect particular neurotransmitter systems, and to allow a classification of new drugs in this respect. The data indicate that, within certain dosage limits, compounds may modulate specifically a single component of the test procedure and may thus be considered as interacting specifically to modify one neurotransmit

Intrastriatal injection of opioid receptor agonists inhibits apomorphine-induced behavior in 6-hydroxydopamine-treated mice.

The effects of intrastriatal (i.st.) injections of mu-, delta-, and kappa-selective opioid receptor agonists on the augmentation of apomorphine-induced behaviors were determined in 6-hydroxydopamine-treated mice by using multidimensional behavioral analyses. 6-Hydroxydopamine (16 mu g/mu l, i.st.) was unilaterally injected into the striatum 30 min after pretreatment with desipramine (25 mg/kg, s.c). Mice were tested 14 days after injection of 6-hydroxydopamine. Apomorphine (0.5 mg/kg, s.c.) produced a marked increase in linear locomotion, contralateral circling and/or rearing behavior in 6-hyd

Apomorphine produced more yawning in Sprague-Dawley rats than in F344 rats: a pharmacological study.

Apomorphine induced yawning in both Sprague-Dawley and F344 rats in the same dose range, but F344 rats emitted only about 1/4 as many yawns as did Sprague-Dawley rats. At higher doses, rats of both strains exhibited stereotypic behavior with a comparable intensity. Pretreatment with either SCH 23390 [R(+)-8-chloro-2,3,4,5-tetrahydro-3-methyl-5-phenyl-1H-3-benzazepine-7-o l] or pindolol increased apomorphine-induced yawning further in Sprague-Dawley rats, but had little effect on the low yawning score produced by apomorphine in F344 rats. The low yawning response to apomorphine in F344 rats is,

A single report of hemiplegic arm stretching related to yawning: further investigation using apomorphine administration.

We observed a stroke patient with an infarct of the internal capsule interrupting the pyramidal tract who stretched his hemiplegic arm during spontaneous and apomorphine-induced yawning. The putative mechanism by which yawning can induce the paradoxical motor response of the plegic arm in the patient might be the functional efficiency of a pathway projecting directly from the stimulated basal ganglia to lower motor systems in the brainstem.

Caffeine produces contralateral rotation in rats with unilateral dopamine denervation: comparisons with apomorphine-induced responses.

Like the dopamine agonist apomorphine, the methylxanthines caffeine, theophylline and theobromine produced dose-dependent contralateral rotation in rats with unilateral 6-hydroxydopamine denervation, a response considered to be dependent upon dopamine receptors rendered supersensitive. This response was also observed after the injection of the substances into the denervated striatum. Indeed, intrastriatal administration of caffeine into the dopamine denervated striatum produced, dose-dependently (1.0-50.0 micrograms/ul), contralateral rotation. However, while apomorphine produced ipsilateral r

Clonidine sensitizes mice for apomorphine-induced stereotypic gnawing: antagonism by neuroleptics and cholecystokinin-like peptides.

In mice sensitized for apomorphine by either scopolamine or teflutixol, clonidine antagonized the antistereotypic effect of ceruletide and haloperidol. The same effect of clonidine occurred in normal mice with methylphenidate-induced gnawing. In naive mice, clonidine alone had a sensitizing effect for the action of apomorphine leading to wire-gnawing. Yohimbine and rauwolscine (but not corynanthine) antagonized this effect of clonidine. The gnawing-inducing effect of methylphenidate was also enhanced by clonidine but not to the same extent as that of apomorphine. The stereotypic effect of apom

Iatrogenic hemoperitoneum and sigmoid colon injury from malpositioned apomorphine infusion device in advanced Parkinson's disease.

Apomorphine pumps are increasingly used for the management of patients with advanced Parkinson's Disease. We report a unique case of hemoperitoneum and penetrating colonic injury resulting from malpositioning of a subcutaneous apomorphine device-a complication not previously described in the literature. Seventy-six-year-old man with advanced Parkinson's Disease presented with acute abdominal pain 3-hours post the re-siting of his apomorphine infusion device. Contrast-enhanced computed tomography (CT) of the abdomen and pelvis revealed high-density fluid (47-67 Hounsfield Units), consistent wit
