Methane production from a field-scale biofilter designed for desulfurization of biogas stream.

The development of a simple and low maintenance field-scale biotrickling filter (BTF) for desulfurization of swine wastewater-derived biogas stream that was also capable of increasing biomethane concentrations was investigated. BTF was continuously fed with wastewater effluent from an air sparged nitrification-denitrification bioreactor installed downgradient from an UASB-type digester. BTF maximum removal efficiency (RE) of 99.8% was achieved with a maximum elimination capacity (EC) of 1,509 g H2S m(-3) h(-1). Average EC obtained with inlet biogas flow rates of 0.024, 0.036 and 0.048 m(3) h(-

Cryo-EM study and in vivo chemical mapping of the Methanosarcina acetivorans ribosome and its dimerization via a repurposed enzyme and translation factor.

Despite the overall conservation of ribosomes across all domains of life, differences in their 3D architecture, rRNA sequences, ribosomal protein composition, and translation factor requirements reflect lineage-specific adaptations to environmental niches. In the domain Archaea, structural studies have primarily focused on nonmethanogenic thermophiles and halophiles, leaving it unclear whether these represent the broader Archaea domain. Here, we report the cryo-electron microscopy (cryo-EM) structure of the ribosome from Methanosarcina acetivorans, a previously unreported high-resolution struc

Algal Organic Matter Drives Methanogen-Mediated Methylmercury Production in Water from Eutrophic Shallow Lakes.

Algal blooms bring massive amounts of algal organic matter (AOM) into eutrophic lakes, which influences microbial methylmercury (MeHg) production. However, because of the complexity of AOM and its dynamic changes during algal decomposition, the relationship between AOM and microbial Hg methylators remains poorly understood, which hinders predicting MeHg production and its bioaccumulation in eutrophic shallow lakes. To address that, we explored the impacts of AOM on microbial Hg methylators and MeHg production by characterizing dissolved organic matter with Fourier transform ion cyclotron reson

Effects of Per- and Polyfluoroalkyl Compounds (PFASs) on Anaerobic Granular Sludge: Methane Production and Microbial Community Composition.

PFASs are a group of pollutants ubiquitous in the environment, for which their effects on the microbial community composition and activity of anaerobic granular sludge are still poorly understood. With our study, we aimed to provide insight into the impact of four PFASs on the methane yield of anaerobic granular sludge (AGS) using acetate as the substrate. Anaerobic granular sludge was exposed to different concentrations of 1H,1H,2H,2H-Tridecafluorooctan-1-ol (6:2FTOH), 1H,1H,2H,2H-Perfluoro-1-decanol (8:2FTOH), Tridecafluorohexane-1-sulfonic acid (PFHxS) and Perfluorooctanoic acid (PFOA). Comprehensive analyses of archaeal viral genomes reveal genomic characteristics, divergence, and host interactions.

BACKGROUND: The ecological significance of bacteriophages has been extensively investigated, while the role of archaeal viruses across different environments remains poorly understood. RESULTS: Here, we present the Archaeal Viral Genome Database (AVGD), a comprehensive survey of archaeal viruses across eight distinct habitat types, including 3708 archaeal viral genomes, with genome sizes ranging from 3 to 188 kb, identified from 64,521,709 putative viral genomes using 40 public metagenomic datasets, an integrated public viral genome database (IGN), and pig gut viral databases. Predicted shifts in microbial functional potential across the late glacial to holocene transition: a 16S rRNA-based inference from southeastern Arabian Sea sediments.

Marine sediments preserve valuable records of ancient microbial communities. In this exploratory study, direct environmental amplicon sequencing reconstruction is often constrained by DNA degradation. In this exploratory study, we applied predictive functional profiling (PICRUSt2) to 16S rRNA gene amplicon data from five discrete sediment horizons spanning ~14.400 years (Bølling-Allerød, Younger Dryas, Early Holocene, Mid-Holocene, and Late Holocene) in the southeastern Arabian Sea. Inferred functional profiles based on Clusters of Orthologous Groups (COGs) pointed to variations in the predict

Nitrification Couples Microbial CO2 Fixation to Warming and Drought Responses in Alpine Grassland Soils.

Microbial CO2 fixation in alpine grassland soils is highly sensitive to warming and drought. Nitrogen inputs from grazing may stimulate autotrophic nitrifiers, including ammonia-oxidizing archaea (AOA), ammonia-oxidizing bacteria (AOB), and nitrite-oxidizing bacteria (NOB). These nitrifiers assimilate carbon through CO2 fixation, but how their activity is associated with microbial CO2 fixation under warming and drought remains unclear. Here, we investigated microbial CO2 fixation and nitrification under altered temperature and moisture using urea-amended soil microcosms with 13CO2 labeling, DN

Resistance of the Archaeal Community to a Severe Disturbance in an Extreme Alkaline Saline Soil.

An extreme disturbance, i.e. a 1-day chloroform fumigation, had a strong impact on the bacterial community structure in an extreme saline alkaline soil, but how Archaea respond to such an event is largely unknown. Three alkaline saline soils with electrolytic conductivity (EC) between 139 and 157 dS m- 1 and pH 10.0-10.3 were chloroform fumigated for one day and the recovery of the archaeal community determined after 1, 5 and 10 days, while an unfumigated soil served as control. Six archaeal phyla dominated by Candidatus Halobacterota (relative abundance 96.9%) were detected, while Natronorubr

Microbial Communities Drive Methane Fluxes From Floodplain Lakes-A Hydrological Gradient Perspective.

This study examined the impact of methanotrophic bacteria and methanogenic archaea on CH4 fluxes from floodplain lakes at various successional stages, analysing their interactions with physicochemical properties of water. Seasonal microbiological and hydrochemical studies of 10 floodplain lakes in the Łyna River floodplain, characterised by varying hydrological connectivity, revealed that methanotrophic bacteria (MOB) and mGen significantly influenced CH4 and CO2 emissions. The microbial structure, expressed as the MOB/mGen ratio, was associated with a gradient of CH4 flux rates specific to ea

A translation proofreader of archaeal origin imparts multi-aldehyde stress tolerance to land plants.

Aldehydes, being an integral part of carbon metabolism, energy generation, and signalling pathways, are ingrained in plant physiology. Land plants have developed intricate metabolic pathways which involve production of reactive aldehydes and its detoxification to survive harsh terrestrial environments. Here, we show that physiologically produced aldehydes, i.e., formaldehyde and methylglyoxal in addition to acetaldehyde, generate adducts with aminoacyl-tRNAs, a substrate for protein synthesis. Plants are unique in possessing two distinct chiral proofreading systems, D-aminoacyl-tRNA deacylase1

Metabolically diverse microorganisms mediate methylmercury formation under nitrate-reducing conditions in a dynamic hydroelectric reservoir.

Brownlee Reservoir is a mercury (Hg)-impaired hydroelectric reservoir that exhibits dynamic hydrological and geochemical conditions and is located within the Hells Canyon Complex in Idaho, USA. Methylmercury (MeHg) contamination in fish is a concern in the reservoir. While MeHg production has historically been attributed to sulfate-reducing bacteria and methanogenic archaea, microorganisms carrying the hgcA gene are taxonomically and metabolically diverse and the major biogeochemical cycles driving mercury (Hg) methylation are not well understood. In this study, Hg speciation and redox-active

Aspartate-phobia of thermophiles as a reaction to deleterious chemical transformations.

Prokaryotes growing at high temperatures have a high proportion of charged residues in their proteins to stabilize their 3D structure. By mining 175 disparate bacterial and archaeal proteomes we found that, against the general trend for charged residues, the frequency of aspartic acid residues decreases strongly as natural growth temperature increases. In search of the explanation, we hypothesized that the reason for such unusual correlation is the deleterious consequences of spontaneous chemical transformations of aspartate at high temperatures. Our subsequent statistical analysis supported t

Water skating: How polymerase sliding clamps move on DNA.

Polymerase sliding clamps are ring-shaped proteins that encircle duplex DNA and hold polymerases to DNA for high processivity during synthesis. The crystal structure of clamp-DNA complex reveals that the DNA is highly tilted through the clamp with extensive interaction with the clamp inner surface. In contrast to the tilted clamp-DNA interaction without DNA polymerases, recent structures of replicative polymerases of bacteria, eukaryotes, and archaea that are bound to the clamp and DNA show that the polymerase positions DNA straight through the clamp without direct protein-DNA contacts. Microbial metabolite fluxes in a model marine anoxic ecosystem.

Permanently anoxic regions in the ocean are widespread and exhibit unique microbial metabolic activity exerting substantial influence on global elemental cycles and climate. Reconstructing microbial metabolic activity rates in these regions has been challenging, due to the technical difficulty of direct rate measurements. In Cariaco Basin, which is the largest permanently anoxic marine basin and an important model system for geobiology, long-term monitoring has yielded time series for the concentrations of biologically important compounds; however, the underlying metabolite fluxes remain poorl

Diversity of phosphorus reserves in microorganisms.

Phosphorus compounds are indispensable components of the Earth's biomass metabolized by all living organisms. Under excess of phosphorus compounds in the environment, microorganisms accumulate reserve phosphorus compounds that are used under phosphorus limitation. These compounds vary in their structure and also perform structural and regulatory functions in microbial cells. The most common phosphorus reserve in microorganism is inorganic polyphosphates, but in some archae and bacteria insoluble magnesium phosphate plays this role. Some yeasts produce phosphomannan as a phosphorus reserve. Thi

Structure-specific DNA nucleases: structural basis for 3D-scissors.

Structure-specific DNA nucleases play important roles in various DNA transactions such as DNA replication, repair and recombination. These enzymes recognize loops and branched DNA structures. Recent structural studies have provided detailed insights into the functions of these enzymes. Structures of Holliday junction resolvase revealed that nucleases are broadly diverged in the way in which they fold, however, are required to form homodimers with large basic patches of protein surfaces, which are complementary to DNA tertiary structures. Many nucleases maintain structure-specific recognition m

Microbes as Key Players During Evolution: I From the Beginnings to the Appearance of Eukaryotes.

Life on Earth originated in the Precambrian, around 4 billion years ago. This review focuses on microbes (viruses, bacteria, archaea) which developed in these prehistoric times and their roles during the later evolution of protists (eukaryotes). Among the different theories of life evolution, the author favors the theory of the RNA world as having been first. Over the past few decades, new molecular biological technologies and findings have changed the paradigm of mutation and selection as being the main driving force of biological evolution. Instead, microbes and microbe-derived genetic eleme

Diversity and ecology of the prokaryotic microbiome associated with marine sponges across Antarctica.

Antarctic sponges host diverse and functionally relevant microbial communities that play central roles in the structure and resilience of polar benthic ecosystems. This review provides a focused analysis of the prokaryotic microbiomes associated with Antarctic sponges, with an emphasis on three ecologically significant species: Mycale (Oxymycale) acerata, Dendrilla antarctica, and Hymeniacidon torquata. Drawing from recent molecular studies, we examine the composition, predicted functional potential, and environmental responsiveness of these bacterial and archaeal communities. Comparative anal
