Poloxamer 407 (P407)-based nanoparticles were produced by the high pressure homogenization method for the encapsulation and delivery of methotrexate (MTX), aiming intravenous therapeutic applications. The surface of these nanoparticles was functionalized by conjugation of P407 with folic acid (FA) or with MTX, which served as targeting ligand agents. MTX-P407 conjugate was also developed to increase the final drug cargo. Two hydrophobic derivatives of MTX, MTX di-ethylated ester (MTX-OEt) and the ionic complex MTX-dimethyldioctadecylammonium bromide (MTX-DODAB) were produced and entrapped onto

In this paper, we report the successful growth of pure, Cu(2+) ions and Cd(2+) ions doped on ninhydrin single crystals by slow solvent evaporation technique. The presence of Cu(2+) and Cd(2+) ions in the specimen of ninhydrin single crystal has been determined by atomic absorption spectroscopy. The powder X-ray diffraction analysis was done to calculate the lattice parameters of the pure and doped crystals. The percentage of transmittance of the crystal was recorded using the UV-Vis Spectrophotometer. Thermal behaviors of the grown crystals have been examined by the thermal gravimetric/differe

OBJECTIVE: Differences in dietary habits may be one of the reasons that the incidence of inflammatory bowel disease has remained lower in Japan than in Western countries. We investigated whether a traditional Japanese medicine (Strong Wakamto), based on Aspergillus oryzae koji, would exert any effect on experimental colitis in rats.
MATERIAL AND METHODS: Colitis was induced using an enema of trinitrobenzene sulfonic acid (TNBS) and ethanol. Strong Wakamto was administered for 28 days before induction of colitis and for 7 days thereafter, and the effect of this medicine was evaluated.
RESULTS

A proteinase that hydrolyses clupeine and salmine at acidic pH, called aorsin, was found in the fungus Aspergillus oryzae. Purified aorsin also hydrolysed benzyloxycarbonyl-Arg-Arg-4-methylcoumaryl-7-amide optimally at pH 4.0. The specificity of aorsin appeared to require a basic residue at the P(1) position and to prefer paired basic residues. Aorsin activated plasminogen and converted trypsinogen to trypsin. The trypsin-like activity was inhibited strongly by antipain or leupeptin, but was not inhibited by any other standard inhibitors of peptidases. To identify the catalytic residues of aor

Both 1,3-beta-D-glucan (BDG) and galactomannan (GM) are polysaccharide components of the fungal cell wall. Although elevated levels of serum BDG and Aspergillus GM suggest invasive fungal infection or Pneumocystis pneumonia and aspergillosis, respectively, it is also necessary to consider the possibility of false-positives. We herein report a 68-year-old man with marked elevation in serum BDG and GM levels accompanied by Mendelson's syndrome after rice aspiration. With the improvement of Mendelson's syndrome, his serum BDG and GM levels decreased. The false-positive serum BDG and GM findings m

Mycotoxins are produced by some filamentous fungi that can contaminate food products and are toxic to humans and animals. The aim of this study was to evaluate the efficiency of extracts rich in bioactive compounds from one coffee industrial processing by-product (coffee husk) against two mycotoxigenic fungi and their mycotoxins. In vitro experiments showed that extracts from coffee husk had anti-fungal effect against Fusarium verticillioides and Aspergillus flavus, and the inhibition was directly proportional to the concentration of the extract. In addition, the extract had a significant anti

Laying Japanese quails were fed on diet of 1:1 mixture of basal diet and rice cultures of 2 strains of Aspergillus candidus. During 20 days of feeding, egg production was significantly reduced in these groups compared with the birds fed unmolded rice. The liver of the quails sacrificed at 20 days showed fatty degeneration of hepatic cells. Following withdrawal of the moldy rice, rate of egg production recovered in 5 days. The suppressive effect of egg production seems to be secondary to hepatic injury. In the other experiment, xanthoascin, a purified mycotoxin of A. candidus, was given subcuta

In this study, eight nonconserved residues with exposed surfaces and flexible conformations of the homotetrameric PGUS (β-glucuronidase from Aspergillus oryzae Li-3) were identified. Single-point mutation into cysteine enabled the thiol-maleimide reaction and site-specific protein assembly using a two-arm polyethylene glycol (PEG)-maleimide crosslinker (Mal2 ). The Mal2 (1k) (with 1 kDa PEG spacer)-crosslinked PGUS assemblies showed low crosslinking efficiency and unimproved thermostability except for G194C-Mal2 (1k). To improve the crosslinking efficiency, a lengthened crosslinker Mal2 (2k) (

Mucormycosis (a.k.a. zygomycosis) is an often-life-threatening disease caused by fungi from the ancient fungal division Mucoromycota. Globally, there are nearly a million people with the disease. Rhizopus spp., and R. delemar (R. oryzae, R. arrhizus) in particular, are responsible for most of the diagnosed cases. Pulmonary, rhino-orbito-cerebral, and invasive mucormycosis are most effectively treated with amphotericin B (AmB) and particularly with liposomal formulations (e.g., AmBisome®). However, even after antifungal therapy, there is still a 50% mortality rate. Hence, there is a critical ne

Both sulfonamides (SAs) and copper (Cu(II)) were frequently detected together in swine wastewater. In this study, the regulation of Cu(II) on SAs adsorption and release of dissolved organic matters (DOMs) by fungi-microalgae pellets (FM-pellets) were investigated. Aspergillus oryzae pellets were prepared for combination with Chlorella vulgaris and the optimal conditions were at agitation speed of 130 rpm, fungi to microalgae ratio of 10:1 and the combined time of 3 h with the highest combination efficiency of 98.65%. The results showed that adsorption was the main mechanism for SAs removal. FM

BACKGROUND: Acute invasive fungal rhinosinusitis (AIFRS) is aggressive morbidity affecting immunocompromised patients. Coronavirus disease 2019 (COVID-19) may allow secondary fungal disease through a propensity to cause respiratory infection by affecting the immune system leading to dysregulation and reduced numbers of T lymphocytes, CD4+T, and CD8+T cells, altering the innate immunity. The aim of this study is to evaluate the incidence of acute invasive fungal rhinosinusitis (AIFRS) in COVID-19 patients.
METHODOLOGY: Data for acute invasive rhinosinusitis was obtained from the Otorhinolaryng

Zearalenone (ZEA) and aflatoxin B₁ (AFB₁) are two main kinds of mycotoxins widely existing in grain and animal feed that cause a lot of economic loss and health problems for animals and humans. In order to alleviate the cytotoxic effects of AFB₁ and ZEA on swine jejunal epithelial cells (IPEC-J2), the combination of a cell-free supernatant of compound probiotics (CFSCP) with mycotoxin degradation enzymes (MDEs) from Aspergillus oryzae was tested. The results demonstrated that coexistence of AFB₁ and ZEA had synergetic toxic effects on cell viability. The cell viability was decreased with mycot

INTRODUCTION: The low incidence of oesophageal adenocarcinoma (EAC) in Barrett's oesophagus (BE) patients reinforces the need for risk stratification tools to make BE surveillance more effective. Therefore, we have undertaken a systematic review and meta-analysis of published studies on immunohistochemical (IHC) biomarkers in BE to determine the value of IHC biomarkers as neoplastic predictors in BE surveillance.
MATERIALS AND METHODS: We searched MEDLINE, EMBASE, Web of Science, CENTRAL, Pubmed publisher, and Google scholar. All studies on IHC biomarkers in BE surveillance were included. ORs

alpha-Amylase is a starch cleaving enzyme often used in the baking industry as a flour additive. It is usually of fungal origin, produced by Aspergillus oryzae. One previous report has shown IgE antibodies and positive skin prick test against alpha-amylase in asthmatic bakers. This paper describes four alpha-amylase sensitised index cases with occupational asthma or rhinitis and the results of a cross sectional study of 20 workers from the same factory who were also exposed to alpha-amylase powder. Air sampling detected airborne alpha-amylase at a concentration of 0.03 mg/m3. Significantly mor

BACKGROUND: Lower respiratory tract infections represent prevalent ailments. Nonetheless, current comprehension of the microbial ecosystems within the lower respiratory tract remains incomplete and necessitates further comprehensive assessment. Leveraging the advancements in metagenomic next-generation sequencing (mNGS) technology alongside the emergence of machine learning, it is now viable to compare the attributes of lower respiratory tract microbial communities among patients across diverse age groups, diseases, and infection types.
METHOD: We collected bronchoalveolar lavage fluid sample

This study investigated the effects of microwave puffing-induced soy protein structural modification on the dynamic changes in physicochemical properties and umami formation during soybean paste fermentation. The results indicated that optimal microwave puffing (400 W, 90 s) disrupts protein non-covalent bonds, depolymerizes protein aggregates, exposes the internal hydrophobic groups to enhance surface hydrophobicity, forms a loose porous structure, and creates favorable conditions for enzymatic hydrolysis during soybean paste fermentation. Conversely, excessive treatment (400 W, >90 s) induce

Droplet-based microfluidics has been frequently used for high-throughput single-cell microbial screening, where individual cells are encapsulated into nanoliter or picoliter-sized droplets and sorted based on measurable phenotypes such as fluorescent proteins or enzyme activity using fluorescence or absorbance-based substrates. However, using droplet microfluidics with filamentous fungi poses a unique challenge: fungal strains tend to escape from droplets as their hyphae grow, causing individual droplets to merge and eventually burst which prevents measuring individual phenotypes. In our work,
