How infection can trigger autoimmunity.

The role of infection in the development of autoimmunity is discussed in view of recent advances in the field of immunologic tolerance. Infectious agents may directly damage immune function by impairing the mechanisms of discrimination between self and non-self antigens; alternatively they may "deceive" the immune system by presentation of self-determinants in an immunogenic form or by uncovering of normally sequestered antigenic determinants. The first of these two pathogenetic mechanisms may be responsible for generalized and the second for selective autoimmune reactions. Different clinical

Autoimmunity in a rural community.

As part of a long term prospective study, sera from 3492 subjects, over 90% of the adult population of the rural Australian town of Busselton, were tested by indirect immunofluorescence for various autoantibodies, and by latex particle agglutination for rheumatoid factor. Autoantibodies were detected in 21·6% of all subjects and the overall prevalence increased with age in both sexes. A dip in the age prevalence curve of autoantibodies amongst subjects aged 75–80 years preceded a terminal sharp rise. The greater prevalence in females, 27·5%, than in males, 13·7%, was accounted for by autoantib

Selective IgA deficiency and autoimmunity.

Laboratory and clinical data are presented in fifteen patients under the age of sixteen years with selective IgA deficiency, all of whom had normal serum IgG, IgM, IgD and IgE. Results indicate a high incidence of autoimmune disease, autoimmune phenomenon or unusual antibody formation. Three patients had thyroiditis, one cerebral vasculitis, one pulmonary haemosiderosis, one cystic fibrosis and nine had recurrent upper respiratory tract infections. Ten out of fifteen were positive for one or more `autoantibodies.' Selective IgA deficiency cannot always be considered a benign entity and individ

Vasculitis and infections: contribution to the issue of autoimmunity reviews devoted to "autoimmunity and infection".

Infections are associated with secondary forms of vasculitis. However, there is increasing evidence that microbial agents play a role also in primary systemic vasculitides. For a long time it has been noted that Hepatitis B virus (HBV) is involved in polyarteritis nodosa (PAN) although the incidence of HBV-associated PAN seems to decline. Cryoglobulinemic vasculitis has been shown to be strongly associated with Hepatitis C Virus (HCV) infection, but this is most striking in Southern Europe and less in Northern Europe. Different microbial agents have been suggested to influence disease expressi

Epigenetics in human autoimmunity. Epigenetics in autoimmunity - DNA methylation in systemic lupus erythematosus and beyond.

Epigenetic mechanisms are essential for normal development and function of the immune system. Similarly, a failure to maintain epigenetic homeostasis in the immune response due to factors including environmental influences, leads to aberrant gene expression, contributing to immune dysfunction and in some cases the development of autoimmunity in genetically predisposed individuals. This is exemplified by systemic lupus erythematosus, where environmentally induced epigenetic changes contribute to disease pathogenesis in those genetically predisposed. Similar interactions between genetically dete

Thyroid autoimmunity and the subsequent development of islet and celiac autoimmunity in the TEDDY study.

We aimed to determine if thyroid autoimmunity is associated with a child's risk for subsequent development of islet or celiac autoantibodies. Children at high genetic risk of type 1 diabetes were followed for thyroid autoimmunity (thyroid peroxidase antibodies [TPOAb] and thyroglobulin antibodies [TGAb]), islet autoimmunity (IA), and celiac disease autoimmunity (CDA) in The Environmental Determinants of Diabetes in the Young study. Out of 5482 children tested for thyroid autoimmunity, IA, and CDA, 39% developed at least one autoantibody. At age 14 years, thyroid autoimmunity co-occurred with I

Does thyroid autoimmunity affect the reproductive outcome in women with thyroid autoimmunity undergoing assisted reproductive technology?

PROBLEM: Our study aims to investigate whether the anti-thyroperoxidase antibody (TPO-Ab) and TSH level in euthyroid women have any association with reproductive outcomes after the ART cycle. METHODS OF STUDY: A total of 1107 patients who were enrolled in the study were divided into four groups based on serum TSH level and TPO-Ab status: group A, 0.3 ≤ TSH < 2.5 mIU/L and TPO-Ab- ; group B, 0.3 ≤ TSH < 2.5 mIU/L and TPO-Ab+ ; group C, 2.5 ≤ TSH < 4.2 mIU/L, and TPO-Ab- ; and group D, 2.5 ≤ TSH < 4.2 mIU/L, TPO-Ab+ . The differences in ART cycles and pregnancy outcomes were analyzed between st

Antigen-specific T cells and autoimmunity.

Autoimmune diseases (ADs) showcase the intricate balance between the immune system's protective functions and its potential for self-inflicted damage. These disorders arise from the immune system's erroneous targeting of the body's tissues, resulting in damage and disease. The ability of T cells to distinguish between self and non-self-antigens is pivotal to averting autoimmune reactions. Perturbations in this process contribute to AD development. Autoreactive T cells that elude thymic elimination are activated by mimics of self-antigens or are erroneously activated by self-antigens can trigge

Mitochondrial extracellular vesicles, autoimmunity and myocarditis.

For many decades viral infections have been suspected as 'triggers' of autoimmune disease, but mechanisms for how this could occur have been difficult to establish. Recent studies have shown that viral infections that are commonly associated with viral myocarditis and other autoimmune diseases such as coxsackievirus B3 (CVB3) and SARS-CoV-2 target mitochondria and are released from cells in mitochondrial vesicles that are able to activate the innate immune response. Studies have shown that Toll-like receptor (TLR)4 and the inflammasome pathway are activated by mitochondrial components. Autorea

Silicone prosthetics and anti-thyroid autoimmunity.

INTRODUCTION: In esthetic surgery, the use of silicone implants is a topic of hot discussion. MATERIAL AND METHODS: An analysis of 119 esthetic surgical interventions on the mammary gland was performed. A study of the immune and endocrine parameters after mammoplasty was carried out. RESULTS: The phenomenon of an increase in the levels of autoantibodies to the TSH receptor was revealed. The phenomenon of pre-operative growth of prolactin and TSH levels has been confirmed. An increase in thyroid autoimmunity after silicone mammoplasty is interpreted as a result of a silicone adjuvant action.

Autoimmunity in people with cystic fibrosis.

Cystic fibrosis (CF) clinicians may see patients who have difficult-to-manage symptoms that do not have a clear CF-related etiology, such as unusual gastrointestinal (GI) complaints, vasculitis, or arthritis. Alterations in immunity, inflammation and intraluminal dysbiosis create a milieu that may lead to autoimmunity, and the CF transmembrane regulator protein may have a direct role as well. While autoantibodies and other autoimmune markers may develop, these may or may not lead to organ involvement, therefore they are helpful but not sufficient to establish an autoimmune diagnosis. Autoimmun

Autoimmunity and Infection in Glomerular Disease.

The ongoing glomerular damage of infections is not limited to the most widely known form of post-streptococcal glomerulonephritis, which is today less common in the Western world; other forms of glomerulonephritis are associated with several bacterial, viral and parasitic pathogens. The mechanisms responsible range from the direct damage of glomerular cells to the formation and deposition of immunocomplexes to molecular mimicry to the secretion of superantigens. Similarly, in the course of glomerular disease, infections are more frequent than in the general population due to the loss of immuno

Interferon and autoantigens: intersection in autoimmunity.

Interferon (IFN) is a key component of the innate immune response. For reasons that remain incompletely understood, the IFN system is upregulated in several rheumatic diseases, particularly those that feature autoantibody production, such as SLE, Sjögren's syndrome, myositis and systemic sclerosis. Interestingly, many of the autoantigens targeted in these diseases are components of the IFN system, representing IFN-stimulated genes (ISGs), pattern recognition receptors (PRRs), and modulators of the IFN response. In this review, we describe features of these IFN-linked proteins that may underlie

Adipokines and Autoimmunity in Inflammatory Arthritis.

Adipokines are adipose tissue-derived factors not only playing an important role in metabolism but also influencing other central processes of the body, such as inflammation. In autoimmune diseases, adipokines are involved in inflammatory pathways affecting different cell types. Many rheumatic diseases belong to the group of autoimmune diseases, for example rheumatoid arthritis (RA) and psoriatic arthritis. Due to the autoimmune responses, a chronic inflammatory milieu develops, which affects the whole body, including adipose tissue. Metabolic alterations such as obesity influence inflammatory

Iodoprophylaxis and thyroid autoimmunity: an update.

Adequate iodine intake is necessary for normal thyroid function. Iodine deficiency is associated with serious complications, but also iodine excess can lead to thyroid dysfunction, and iodine supplementation aimed to prevent iodine deficiency disorders has been associated with development of thyroid autoimmunity. The epidemiology of thyroid diseases has undergone profound changes since the implementation of iodoprophylaxis, notably by means of iodine-enriched salt, specifically resulting in decreased prevalence of goiter and neonatal hypothyroidism, improved cognitive function development in i
