Notch1 regulates the effects of matrix metalloproteinase-9 on colitis-associated cancer in mice.

BACKGROUND & AIMS: Inflammatory bowel disease increases the risks of colon cancer and colitis-associated cancer (CAC). Epithelial cell-derived matrix metalloproteinase (MMP)-9 mediates inflammation during acute colitis and the cleavage and activation of the transcription factor Notch1, which prevents differentiation of progenitor cells into goblet cells. However, MMP-9 also protects against the development of CAC and acts as a tumor suppressor. We investigated the mechanisms by which MMP-9 protects against CAC in mice. METHODS: C57/B6 wild-type mice were given a single dose of azoxymethane an

Effect of dietary resistant starch and protein on colonic fermentation and intestinal tumourigenesis in rats.

Protein as well as starch is fermented in the colon, but the interaction between protein and starch fermentation and the impact on colonic oncogenesis is unknown. High-protein diets increase delivery of protein to the colon and might promote oncogenesis through generation of toxic products. We investigated the interaction of resistant starch (RS) with digestion-resistant potato protein (PP) on colonic fermentation events and their relationship to intestinal tumourigenesis. Male Sprague-Dawley rats were fed an AIN-76A-based diet for 4 weeks and intestinal neoplasms were induced by azoxymethane.

Increased Production of Angiopoietin-like Protein 2 in a Ligature- and LPS-Induced Periodontitis Mouse Model May Promote Colorectal Tumor Progression.

Background/Objectives: Recent studies suggest that angiopoietin-like protein 2 (ANGPTL2) is one of the factors contributing to disease progression in distant organs associated with periodontitis. We previously reported that periodontitis promotes hepatocellular carcinoma and that ANGPTL2 may be involved in tumor progression. Based on these findings, we herein investigated the role of periodontitis-induced ANGPTL2 in the progression of colorectal cancer (CRC) in mice. Methods: Male C57BL/6 mice were divided into control and periodontitis groups. Colorectal tumors were induced using azoxymethane

Putrescine Supplementation Limits the Expansion of pks+ Escherichia coli and Tumor Development in the Colon.

UNLABELLED: Escherichia coli that harbor the polyketide synthase (pks) genomic island produce colibactin and are associated with sporadic colorectal cancer development. Given the considerable prevalence of pks+ bacteria in healthy individuals, we sought to identify strategies to limit the growth and expansion of pks+ E. coli. We found that culture supernatants of the probiotic strain E. coli Nissle 1917 were able to inhibit the growth of the murine pathogenic strain pks+ E. coli NC101 (EcNC101). We performed a nontargeted analysis of the metabolome in supernatants from several E. coli strains

CD73 promotes colitis-associated tumorigenesis in mice.

Patients with inflammatory bowel disease (IBD) are at a higher risk of developing colitis-associated colorectal cancer. The aim of the present study was to investigate the role of CD73 in IBD-associated tumorigenesis. A mouse model of colitis-associated tumorigenesis (CAT) induced by azoxymethane and dextran sulfate sodium was successfully constructed. Model mice were injected with CD73 inhibitor or adenosine receptor agonist. Colon length, body weight loss and tumor formation were assessed macroscopically. Inflammatory cytokine measurement and RNA sequencing on colon tissues were performed. I

HDAC up-regulation in early colon field carcinogenesis is involved in cell tumorigenicity through regulation of chromatin structure.

Normal cell function is dependent on the proper maintenance of chromatin structure. Regulation of chromatin structure is controlled by histone modifications that directly influence chromatin architecture and genome function. Specifically, the histone deacetylase (HDAC) family of proteins modulate chromatin compaction and are commonly dysregulated in many tumors, including colorectal cancer (CRC). However, the role of HDAC proteins in early colorectal carcinogenesis has not been previously reported. We found HDAC1, HDAC2, HDAC3, HDAC5, and HDAC7 all to be up-regulated in the field of human CRC.

Dietary selenium deficiency exacerbates DSS-induced epithelial injury and AOM/DSS-induced tumorigenesis.

Selenium (Se) is an essential micronutrient that exerts its functions via selenoproteins. Little is known about the role of Se in inflammatory bowel disease (IBD). Epidemiological studies have inversely correlated nutritional Se status with IBD severity and colon cancer risk. Moreover, molecular studies have revealed that Se deficiency activates WNT signaling, a pathway essential to intestinal stem cell programs and pivotal to injury recovery processes in IBD that is also activated in inflammatory neoplastic transformation. In order to better understand the role of Se in epithelial injury and

Targeting the Lcn2/Lcn2R-Grb2 cascade ameliorates mitochondrial bioenergetic failure and neuronal death in hepatic encephalopathy.

Hepatic encephalopathy (HE) is a severe neuropsychiatric complication of liver failure characterized by a spectrum of cognitive and motor impairments. Lipocalin-2 (Lcn2) is a multifunctional secretory protein and inflammatory mediator predominantly released by glial cells in response to brain insults, whereas its receptor, Lcn2R, is expressed in cells. Nevertheless, the precise role of this Lcn2/Lcn2R axis in HE pathogenesis remains poorly understood. This study investigated the cellular mechanisms by which the Lcn2/Lcn2R axis triggers neurotoxicity in an azoxymethane-induced mouse model of HE

Secondary bile acid production by gut bacteria promotes Western diet-associated colorectal cancer.

BACKGROUND: Western diet and associated production of secondary bile acids (BAs) have been linked to the development of sporadic colorectal cancer (CRC). Despite observational studies showing that secondary BAs produced by 7α-dehydroxylating (7αDH+) gut bacteria are increased in CRC, a causal proof of their tumour-promoting effects is lacking. OBJECTIVE: Investigate the causal role of BAs produced by 7αDH+ gut bacteria in CRC. DESIGN: We performed feeding studies in a porcine model of CRC combined with multi-omics analyses and gnotobiotic mouse models colonised with 7αDH+ bacteria or a genet

Pharmacological Targeting of PARP-1-NLRP3 Inflammasome Pathway in Colitis-Associated Colorectal Neoplasia: Effects of 3-aminobenzamide and Olaparib.

Colitis-associated colorectal neoplasia is the third most life-threatening consequence of prolonged ulcerative colitis. In the present investigation, we evaluated the efficacy of PARP-1 inhibitors, 3-aminobenzamide and Olaparib, in a rodent model of colitis-associated colorectal cancer (CACC) induced by azoxymethane (AOM) and Dextran sulphate sodium (DSS). For model induction, male BALB/c mice were provided DSS (3% w/v) in three cycles within drinking water following an injection of AOM (10 mg/kg, intraperitoneally). A week after the DSS treatment, 3-aminobenzamide (5, 10 and 20 mg/kg; i.p.) a

FTO-mediated m6A demethylation of CSF3 suppresses NETosis via downregulation of RLN2 expression in colorectal cancer.

CSF3 exerts a significant function in the progression of colorectal cancer (CRC). N6-methyladenosine (m6A) modification is now considered the main driving factor of RNA influence for maintaining homeostasis in cancer cells. Nevertheless, how m6A mediates the role of CSF3 and its influence in pathogenesis of CRC is still elusive. After neutrophil isolation from bone marrow, the purity and survival rate of neutrophils were assessed. Azoxymethane (AOM)/Dextran Sodium Sulfate (DSS) was employed to construct the CRC mice model. Both loss-of-function and gain-of-function experiments were conducted t

Loss of proton-sensing TDAG8 increases tumor progression in mouse models of colon cancer.

T-cell death-associated gene 8 (TDAG8) is a proton-sensing G-protein-coupled receptor that is activated by neutral to acidic extracellular pH. TDAG8 is primarily expressed in leukocytes and leukocyte-rich tissues, including the spleen, lymph nodes, and thymus. Genome-wide association studies have identified TDAG8 as a risk gene for inflammatory bowel disease (IBD). Patients with IBD have an increased risk of developing colorectal cancer (CRC), and TDAG8 has been implicated in regulating anti-tumor immunity. We used the MC38 and azoxymethane/dextran sodium sulfate (AOM/DSS) mouse models of CRC

Cantharidin analogue NCTD-01 inhibits AOM/DSS-induced orthotopic colorectal carcinogenesis in mice by suppressing inflammation-driven tumorigenesis.

Although cantharidin (CTD) exhibits significant clinical efficacy against various cancers, its clinical utility is severely hampered by profound systemic toxicity. Thus, structural modification of CTD to obtain analogues with potent anticancer activity and lower toxicity is essential for advancing CTD-based therapeutics. Building on our previous synthesis of a series of C-5 alkynyl-substituted CTD analogues with potent anti-inflammatory properties, this study investigates the anticancer potential of a new analogue NCTD-01 demonstrated a favorable safety profile in mice, with the maximum tolera

SN38 PLGA nanoparticles provide sustained antitumor efficacy and ameliorate colitis severity in a murine colitis-associated cancer model.

Irinotecan is a cornerstone therapy for metastatic colorectal cancer (CRC); however, the poor solubility and instability of its active metabolite, SN38, limit its therapeutic efficacy. This study aimed to improve SN38 delivery using poly(lactic-co-glycolic acid) (PLGA) nanoparticles and to evaluate their efficacy and safety in vitro and in a murine colitis-associated CRC model. SN38-PLGA nanoparticles exhibited a mean diameter of 170.7 ± 0.95 nm with a polydispersity index of 0.18 ± 0.04. In vitro, SN38-PLGA exhibited selective cytotoxicity-potent against HCT116 CRC cells but less toxic to HEK

Abdominal ultrasound attenuates early-stage aberrant proliferation and depressive behavior in AOM/DSS-induced colorectal cancer: regulatory mechanisms of the Wnt/β-catenin pathway and HPA axis.

Chronic inflammation is a key factor promoting colorectal cancer (CRC) development and progression. Abdominal ultrasound stimulation (AUS) has shown potential in alleviating inflammatory reactions and behavioral impairments in dextran sodium sulfate (DSS)-induced colitis models by modulating the gut microbiota composition and mitigating damage to colonic tissues. This study observed the protective effects of AUS on colitis and mice with early-stage CRC and clarified the mechanism potentially underlying these effects. CRC was induced in mice using azoxymethane (AOM)/DSS. Low-intensity pulsed ul

Deficiency in Epithelium RAD50 Aggravates UC via IL-6-Mediated JAK1/2-STAT3 Signaling and Promotes Development of Colitis-Associated Cancer in Mice.

BACKGROUND: Ulcerative colitis (UC) is one of the most important risk factors for developing colitis-associated cancer (CAC). Persistent DNA damage increases CAC risk and has been observed in patients with UC. We aimed to identify the regulatory role of RAD50, a DNA double-strand breaks (DSBs) sensor, in UC progression to CAC. METHODS: DSBs and RAD50 expression in inflammatory bowel disease (IBD) and CAC cell and mouse models were assessed. Mice with intestinal epithelial RAD50 deletion (RAD50IEC-KO) were used to examine the role of RAD50 in colitis and CAC. RESULTS: Along with the increased

Loss of WNT2B Increases Progression from Dysplasia to Colorectal Cancer.

Colorectal cancer (CRC) is the third most common cancer and the second leading cause of cancer-related deaths in the United States, and upregulation of the WNT pathway is a primary driver in most cases. However, the role of individual WNT proteins in the development of CRC remains poorly understood. Our previous studies demonstrated that WNT2B loss-of-function leads to severe intestinal enteropathy in humans and increases chemically-induced colitis in mice, suggesting a protective function in the colon. Therefore, we investigated how loss of WNT2B affects CRC development. We used azoxymethane

Heart failure-induced microbial dysbiosis contributes to colonic tumour formation in mice.

AIMS: Heart failure (HF) and cancer are the leading causes of death worldwide. Epidemiological studies revealed that HF patients are prone to develop cancer. Preclinical studies provided some insights into this connection, but the exact mechanisms remain elusive. In colorectal cancer (CRC), gut microbial dysbiosis is linked to cancer progression and recent studies have shown that HF patients display microbial dysbiosis. This current study focussed on the effects of HF-induced microbial dysbiosis on colonic tumour formation. METHODS AND RESULTS: C57BL/6J mice were subjected to myocardial infar
