Rapid field detection assays for Bacillus anthracis, Brucella spp., Francisella tularensis and Yersinia pestis.

Rapid detection is essential for timely initiation of medical post-exposure prophylactic measures in the event of intentional release of biological threat agents. We compared real-time PCR assay performance between the Applied Biosystems 7300/7500 and the RAZOR instruments for specific detection of the causative agents of anthrax, brucellosis, tularemia and plague. Furthermore, an assay detecting Bacillus thuringiensis, a Bacillus anthracis surrogate, was developed for field-training purposes. Assay sensitivities for B. anthracis, Brucella spp., Francisella tularensis and Yersinia pestis were

Real-time PCR system targeting a chromosomal marker specific for Bacillus anthracis.

Specific identification of Bacillus anthracis and differentiation from closely related Bacillus cereus and Bacillus thuringiensis strains is still a major diagnostic problem. Commercially available diagnostic kits targeting plasmid-markers cannot differentiate between B. anthracis, non-anthracis Bacillus species harbouring anthrax-specific virulence plasmids, and plasmidless B. anthracis strains. A TaqMan PCR assay was designed targeting sequences of gene locus BA_5345 of the B. anthracis strain Ames. Specificity was determined by using a panel of 328 Bacillus strains; sensitivity was determin

Growth characteristics of Bacillus anthracis compared to other Bacillus spp. on the selective nutrient media Anthrax Blood Agar and Cereus Ident Agar.

Anthrax Blood Agar (ABA) and Cereus Ident Agar (CEI) were evaluated as selective growth media for the isolation of Bacillus anthracis using 92 B. anthracis and 132 other Bacillus strains from 30 species. The positive predictive values for the identification of B. anthracis on ABA, CEI, and the combination of both were 72%, 71%, and 90%, respectively. Thus, less than 10% of all species were misidentified using both nutrient media. Species which might be misidentified as B. anthracis were B. cereus, B. mycoides, and B. thuringiensis. Particularly, 30% of B. weihenstephanensis strains were miside

Biological control of malaria vectors.

A comprehensive review is presented of the potentiality of biocontrol agents viz. entomophagus bacteria (Bacillus thuringiensis var. israelensis and Bacillus sphaericus), fungi, microsporidians, predators and parasites against malaria vectors in the field condition. Unlike insecticides, these control agents are host specific and safer to the environment. However, barring fishes which are being used in certain situations, other biocontrol agents have not yet reached the operational stage. Two spore forming bacteria B. thuringiensis var. israelensis and B. sphaericus have been extensively tested

Spore-Associated Proteins Involved in c-di-GMP Synthesis and Degradation of Bacillus anthracis.

Bis-(3'-5')-cyclic-dimeric GMP (c-di-GMP) is an important bacterial regulatory signaling molecule affecting biofilm formation, toxin production, motility, and virulence. The genome of Bacillus anthracis, the causative agent of anthrax, is predicted to encode ten putative GGDEF/EAL/HD-GYP-domain containing proteins. Heterologous expression in Bacillus subtilis hosts indicated that there are five active GGDEF domain-containing proteins and four active EAL or HD-GYP domain-containing proteins. Using an mCherry gene fusion-Western blotting approach, the expression of the c-di-GMP-associated protei

Real-Time PCR Identification of Unique Bacillus anthracis Sequences.

Bacillus anthracis is a spore-forming, Gram-positive microorganism. It is a causative agent of anthrax, a highly infectious disease. It belongs to the "Bacillus cereus group", which contains other closely related species, including Bacillus cereus, Bacillus thuringiensis, Bacillus mycoides, Bacillus weihenstephanensis, and Bacillus pseudomycoides. B. anthracis naturally occurs in soil environments. The BA5345 genetic marker was used for highly specific detection of B. anthracis with TaqMan probes. The detection limit of a real-time PCR assay was estimated at the level of 16.9 copies (CI95% - 3

Purification and Thioflavin T Staining of Bacillus spp. Endospores for Fluorescence-Based Assays.

We present a simple, low-cost, and efficient protocol for fluorescent staining of Bacillus spp. endospores using the thioflavin T (ThT) dye. The article provides step-by-step procedures for culturing Bacillus paranthracis, Bacillus thuringiensis (2 strains), and Bacillus subtilis, purifying endospores from pure cultures, and staining them with ThT. ThT-stained endospores retain fluorescence for at least 2 years when stored at 4°C in the dark and remain fluorescent following formaldehyde fixation. Importantly, ThT staining does not impair spore viability, and stained endospores germinate normal

Characterization of Bacillus cereus Group Isolates From Human Bacteremia by Whole-Genome Sequencing.

Members of the Bacillus cereus group are spore-forming organisms commonly associated with food poisoning and intestinal infections. Moreover, some strains of the group (i.e., B. cereus sensu stricto and Bacillus thuringiensis) can cause bacteremia in humans, mainly in immunocompromised individuals. Here we performed the genetic characterization of 17 human clinical strains belonging to B. cereus group isolated from blood culture. The whole-genome sequencing (WGS) revealed that the isolates were closely related to B. cereus sensu stricto and B. thuringiensis-type strain. Multilocus sequence typ

Sweet Corn Sentinel Monitoring for Lepidopteran Field-Evolved Resistance to Bt Toxins.

As part of an insect resistance management plan to preserve Bt transgenic technology, annual monitoring of target pests is mandated to detect susceptibility changes to Bt toxins. Currently Helicoverpa zea (Boddie) monitoring involves investigating unexpected injury in Bt crop fields and collecting larvae from non-Bt host plants for laboratory diet bioassays to determine mortality responses to diagnostic concentrations of Bt toxins. To date, this monitoring approach has not detected any significant change from the known range of baseline susceptibility to Bt toxins, yet practical field-evolved

The Mossie-Buster: a hose-driven insecticide delivery tool for the control of container-breeding mosquitoes.

A novel insecticide delivery tool, the Mossie-Buster, was recently developed to control larval populations from urban breeding sites in Townsville, Australia. This functional and user-friendly control device directly targets the main breeding sites of Aedes aegypti (L.) with a focused delivery of insecticides. The Mossie-Buster comprises a commercially available device and attachments that mix an insecticide solution into the flow of water emitted from a hose. Attached to the device is a trigger for controlled insecticide release. Preliminary laboratory and field trials demonstrated the tool t

Combining sterile insect releases with refuge areas to delay the evolution of resistance to Bt sugarcane: an agent-based modeling approach.

The strategic use of refuge areas is a well-known method for delaying the development of pest resistance to Bacillus thuringiensis (Bt) crop. A lesser-known method to control against resistance development is sterile insect releases. In this article, an agent-based simulation model is used to test the effectiveness of combining the use of Bt sugarcane, refuge areas, and sterile insect releases as an integrated strategy against Eldana saccharina Walker (Lepidoptera: Pyralidae) infestation and resistance development. Individual insects are modeled with their own genetic traits on a simulated sug

Laboratory and field survival of Spodoptera frugiperda (Lepidoptera: Noctuidae) on Bt and non-Bt maize and its susceptibility to insecticides.

BACKGROUND: Field-evolved resistance of fall armyworm (FAW), Spodoptera frugiperda (Smith), has been reported to Bt maize technologies in Brazil. The control failures of FAW by Bt maize increased the use of insecticides for their control. However, no information is available on the interaction between resistant FAW and their response to insecticides. Here, we evaluated the survival of FAW strains on Bt and non-Bt maize in laboratory and field conditions, and their susceptibility to insecticides. RESULTS: In the laboratory, resistant FAW larvae reared on Bt and non-Bt maize showed a similar su

Diversity, Antimicrobial Activity, and Antibiotic Susceptibility Pattern of Endophytic Bacteria Sourced From Cordia dichotoma L.

Endophytic bacteria isolated from medicinal plants are crucial for the production of antimicrobial agents since they are capable of possessing bioactive compounds with diverse structures and activities. Cordia dichotoma, a plant of medicinal importance native to the Jammu region of India, was selected for the isolation and characterization of culturable endophytic bacteria and evaluation of their antimicrobial activities. Standardized surface sterilization methods were employed to isolate thirty-three phenotypically distinguishable endophytic bacteria from the root, stem, and leaf parts of the

Bacillus subtilis strain engineered for treatment of soil-transmitted helminth diseases.

Soil-transmitted helminths (hookworms, whipworms, and large roundworms) are agents of intestinal roundworm diseases of poverty that infect upwards of 2 billion people worldwide. A great challenge in treating these diseases is the development of anthelmintic therapeutics that are inexpensive, can be produced in great quantity, and are capable of delivery under varied and adverse environmental conditions. A potential solution to this challenge is the use of live bacteria that are acceptable for human consumption, e.g., Bacillus subtilis, and that can be engineered with therapeutic properties. In

D-cycloserine or similar physiochemical compounds may be uniquely suited for use in Bacillus anthracis spore decontamination strategies.

AIMS: As observed in the aftermath of the anthrax attacks of 2001, decontamination and remediation of a site contaminated by the accidental or intentional release of Bacillus anthracis spores is difficult, costly and potentially damaging to the environment. The identification of novel strategies that neutralize the threat of spores while minimizing environmental damage remains a high priority. We investigated the efficacy of d-cycloserine (DCS), an antibiotic and inhibitor of the spore-associated enzyme (alanine racemase) responsible for converting l-alanine to d-alanine, as a spore germinatio

Evaluation of larvicides for the control of Simulium damnosum s.l. (Diptera: Simuliidae) in West Africa.

The Onchocerciasis Control Program of the World Health Organization is carrying out an extensive screening program in a search for new larvicides to be used for control of Simulium damnosum s.l. Emphasis has been given to finding a pyrethroid and a carbamate to supplement the organophosphates currently in use. These chemicals with differing modes of action, together with Bacillus thuringiensis H-14, are being used in an attempt to cope with the development and spread of resistance to the organophosphates temephos and chlorphoxim.

Polymicrobial infections: Do bacteria behave differently depending on their neighbours?

Despite the number of examples that correlate interspecies interactions in polymicrobial infections with variations in pathogenicity and antibiotic susceptibility of individual organisms, antibiotic therapies are selected to target the most relevant pathogen, with no consideration of the consequences that the presence of other bacterial species may have in the pathogenicity and response to antimicrobial agents. In this issue of Virulence, Garcia-Perez et al. [ 10 ] applied replica plating of used wound dressings to assess the topography of distinct S. aureus types in chronic wounds of patients

Sentinel monitoring for resistance to Bt toxins in European corn borer (Lepidoptera: Crambidae) in Canada.

Transgenic corn Zea mays (L.) producing Bacillus thuringiensis insecticidal toxins (Bt corn) has successfully controlled the European corn borer Ostrinia nubilalis (Lepidoptera: Crambidae), one of the most destructive corn pests in North America, for over 25 years. However, field-evolved resistance to Cry1Fa toxin was first documented in Nova Scotia, Canada, in 2018, with subsequent detections in Quebec and Manitoba. Before these discoveries, no resistance had been reported in O. nubilalis populations to Bt toxins, despite ongoing resistance monitoring mandated by regulatory agencies. Sentinel
