Role of group B streptococcal capsular polysaccharides in the induction of septic arthritis.

The ability of different serotypes of group B streptococci (GBS) to induce septic arthritis in mice was compared. Types II, III, IV, V, VI and VII GBS were investigated. A highly capsulate strain of type III GBS, COH1, and its mutants, COH1-11 (lacking capsular sialic acid) and COH1-13 (non-capsulate), obtained by transposon insertional mutagenesis, were used to assess the role of type-specific polysaccharide on the induction of arthritis. At an intravenous dose of 10(7) cfu/mouse, reference strains of types II, III, IV, VI and VII and type III strain COH1 induced arthritis with an incidence r

A study of pathogenic factors of Streptococcus pneumoniae strains causing meningitis.

Pneumococcal meningitis in St. Petersburg in the period 1985-1991 occurred in 1.7-2.3 children per 100,000 annually. The most common serotypes among pneumococcal strains isolated from patients with meningitis were 19, 1, 6, 15, and 2, whereas, among the capsulated strains isolated from carriers, type 3 predominated. Only one third of strains from cases of meningitis were highly virulent for mice (types 1, 2, 3). Hyaluronidase was produced by all the 39 studied strains, 22 (84.6 +/- 7.1%) out of 26 strains from patients with otitis media, and only by 15 (11.5 +/- 2.8%) out of 130 strains isolat

[The efficiency of probiotics in the prevention of recurrent lower urinary tract infections and bacterial vaginosis].

INTRODUCTION: The literature data indicate that bacterial vaginosis (BV) increases the risk of development of lower urinary tract infections (LUTI). Thus, it is reasonable to assume that the use of probiotics, in particular Lactoginal, can be effective in preventing relapses of LUTI. AIM: To study the efficiency of probiotics, particularly Lactoginal, for the prevention of recurrent LUTI and BV. MATERIALS AND METHODS: A total of 120 women with chronic recurrent bacterial cystitis in the acute stage and concomitant bacterial vaginosis were randomized into two groups of 60 patients. In group 1

Deficiency of hasB accelerated the clearance of Streptococcus equi subsp. Zooepidemicus through gasdermin d-dependent neutrophil extracellular traps.

Streptococcus equi subsp. zooepidemicus (S. zooepidemicus, SEZ) is an essential zoonotic bacterial pathogen that can cause various inflammation, such as meningitis, endocarditis, and pneumonia. UDP-glucose dehydrogenase (hasB) is indispensable in synthesizing SEZ virulence factor hyaluronan capsules. Our study investigated the infection of hasB on mice response to SEZ by employing a constructed capsule-deficient mutant strain designated as the ΔhasB strain. This deficiency was associated with a reduced SEZ bacterial load in the mice's blood and peritoneal lavage fluid (PLF) post-infection. Bes

Optical Coherence Tomography Examination of the Anterior Segment in a Case of Corneal Perforation and Lens Trauma by Chestnut Burr.

Chestnut burrs, the thorny encapsulation of chestnut fruit, can sometimes cause corneal injuries and ulceration, with poor prognoses. We report a case of corneal perforation and damaged anterior lens capsule due to a chestnut burr, using anterior segment optical coherence tomography (AS-OCT). A 67-year-old woman with a chestnut burr injury in her right eye was referred to our hospital. Her right best-corrected visual acuity (BCVA) was 0.8. Slit-lamp examination and AS-OCT showed perforation involving the endothelial layer at the center of the cornea. The iris and anterior lens capsule were dam

Association between hospital bed supply and health outcomes: an ecological analysis and narrative review with insights from Japan.

Lactococcus cremoris subsp. cremoris FC (L. cremoris FC) improves bowel movement; however, its associations with the intestinal microbiota remain unclear. To address this gap, this post hoc analysis of a four-week, placebo-controlled, parallel-group trial evaluates the associations between L. cremoris FC intake and gut microbiota profiles. We analyzed fecal samples from 83 healthy Japanese adults with mild constipation who received either placebo capsules containing corn starch (n=20) or L. cremoris FC capsules (50, 75, or 100 mg; n=21 each). Using 16S rRNA gene amplicon sequencing, we evaluat

SP-A enhances phagocytosis of Klebsiella by interaction with capsular polysaccharides and alveolar macrophages.

We found that surfactant protein A (SP-A) enhances phagocytosis of Klebsiella pneumoniae K21a but not of K2 serotypes by alveolar macrophages. SP-A interacted with the capsule of K21a (containing Man alpha1 Man sequences) as shown by SP-A-induced agglutination of the bacteria, by binding of SP-A-coated particles onto the bacterial surface, and by binding of SP-A to immobilized parent K21a strain and recombinant strains that switched their capsule from K2 to K21a. In contrast, only marginal binding of SP-A to K2 parent strain (lacking this sequence) could be detected. Furthermore, binding of ca

Macrolides Inhibit Capsule Formation of Highly Virulent Cryptococcus gattii and Promote Innate Immune Susceptibility.

Cryptococcus gattii is a fungal pathogen, endemic in tropical and subtropical regions, the west coast of Canada, and the United States, that causes a potentially fatal infection in otherwise healthy individuals. Because the cryptococcal polysaccharide capsule is a leading virulence factor due to its resistance against innate immunity, the inhibition of capsule formation may be a promising new therapeutic strategy for C. gattii Macrolides have numerous nonantibiotic effects, including immunomodulation of mammalian cells and suppression of bacterial (but not fungal) pathogenicity. Thus, we hypot

Biochemical and biophysical characterization of the sialyl-/hexosyltransferase synthesizing the meningococcal serogroup W135 heteropolysaccharide capsule.

Neisseria meningitidis (Nm) is a leading cause of bacterial meningitis and sepsis. Crucial virulence determinants of pathogenic Nm strains are the polysaccharide capsules that support invasion by hindering complement attack. In NmW-135 and NmY the capsules are built from the repeating units (→ 6)-α-D-Gal-(1 → 4)-α-Neu5Ac-(2 →)n and (→ 6)-α-D-Glc-(1 → 4)-α-Neu5Ac-(2 →)n, respectively. These unusual heteropolymers represent unique examples of a conjugation between sialic acid and hexosyl-sugars in a polymer chain. Moreover, despite the various catalytic strategies needed for sialic acid and hexo

Global mapping of Streptococcus pneumoniae sequence types associated with multidrug resistance and capsular switching: A systematic review.

BACKGROUND: Streptococcus pneumoniae is a significant bacterial pathogen responsible for invasive and non-invasive infections. One of its most concerning traits is capsular switching, which may contribute to multidrug resistance (MDR) and emergence of difficult-to-treat infections. This systematic review examined the sequence types of S. pneumoniae associated with MDR and/or capsular switching, focusing on their genetic relatedness. METHODOLOGY: A systematic review was conducted following PRISMA guidelines and registered in PROSPERO. A comprehensive literature search was performed across PubM

Interaction of Neisseria meningitidis Group X N-acetylglucosamine-1-phosphotransferase with its donor substrate.

Neisseria meningitidis Group X is an emerging cause of bacterial meningitis in Sub-Saharan Africa. The capsular polysaccharide of Group X is a homopolymer of N-acetylglucosamine α(1-4) phosphate and is a vaccine target for prevention of disease associated with this meningococcal serogroup. We have demonstrated previously that the formation of the polymer is catalyzed by a phosphotransferase which transfers N-acetylglucosamine-1-phosphate from UDP-N-acetylglucosamine to the 4-hydroxyl of the N-acetylglucosamine on the nonreducing end of the growing chain. In this study, we use substrate analogs

Comparison of DNA dot blot hybridization and lancefield capillary precipitin methods for group B streptococcal capsular typing.

Group B streptococci (GBS) (Streptococcus agalactiae) are a major cause of sepsis and meningitis in neonates and infants and of invasive disease in pregnant women, nonpregnant, presumably immunocompromised adults, and the elderly. Nine GBS serotypes based on capsular polysaccharide antigens have been described. The serotype distributions among invasive and colonizing isolates differ between pediatric and adult populations and have changed over time. Thus, periodic monitoring of GBS serotype distributions is necessary to ensure the proper formulation and application of an appropriate GBS vaccin

Divergent regulation of virulence factors by FimK in classical and hypervirulent Klebsiella pneumoniae pathotypes.

Klebsiella pneumoniae is an opportunistic bacterial pathogen that is increasingly recognized as a major global threat. Type 1 pili, encoded by the fim operon, are virulence factors imperative in K. pneumoniae urinary tract infection. FimK, a regulatory protein, has been shown to repress type 1 fimbrial expression in specific K. pneumoniae strains, but its role across diverse K. pneumoniae pathotypes has not been fully elucidated. In this study, we investigate the influence of fimK on key virulence factors and phenotypes, including pili, capsule, and hypermucoviscosity (HMV), across five classi

Staphylococcus aureus serotype 5 capsular polysaccharide is antiphagocytic and enhances bacterial virulence in a murine bacteremia model.

Controversy persists over the role that the capsular polysaccharide plays in the pathogenesis of Staphylococcus aureus infections. To address this issue, we compared the mouse virulence of S. aureus Reynolds and capsule-defective mutant strains cultivated under conditions of high or low capsule expression. Strain Reynolds cells cultivated on Columbia salt agar plates expressed approximately 100-fold more type 5 capsular polysaccharide than did cells cultivated in Columbia salt broth. The relative virulence of strain Reynolds and its capsule-defective mutants after growth on either solid or liq

Meningococcal vaccines.

BACKGROUND: The 5 major pathogenic serogroups of the Gram-negative encapsulated bacterium Neisseria meningitidis are A, B, C, Y, and W135. In the 1960s, vaccines consisting of purified capsular polysaccharide antigens were developed against serogroups A, C, Y, and W135. These vaccines were highly effective among adults but were not efficacious among infants and young children. Capsular polysaccharide-protein conjugate vaccines were subsequently developed. METHODS: The development of second-generation capsular polysaccharide glycoconjugate vaccines that are effective among infants and children

Insight on Bacterial Newborn Meningitis Using a Neurovascular-Unit-on-a-Chip.

Understanding the pathogenesis of bacterial infections is critical for combatting them. For some infections, animal models are inadequate and functional genomic studies are not possible. One example is bacterial meningitis, a life-threatening infection with high mortality and morbidity. Here, we used the newly developed, physiologically relevant, organ-on-a-chip platform integrating the endothelium with neurons, closely mimicking in vivo conditions. Using high-magnification microscopy, permeability measurements, electrophysiological recordings, and immunofluorescence staining, we studied the d
